An Anti-Virus Vaccine Selection Model Based on Stackelberg Game (슈타켈버그 게임 기반 Anti-virus 백신 선택 모형)

  • This paper deals with an information security problem that involves the strategies of both an attacker and an administrator of a web-based system. A game-theoretic model for the problem, based on an Stackelberg game environment, is presented. In the model, the administrator selects a set of anti-virus vaccines to cope with potential system attackers and the intruder chooses attacking modes that are most effective against the administrator's chosen set of vaccines. Moreover, the model considers a number of practical constraints, such as a budget limit on the vaccine purchase and a limit on the system performance. In addition, two different scenario analyses are provided, based on the results of the proposed model applied to a simulated pseudo-real-world data.

Studies on the Potato Virus X and Potato Leaf Roll Virus for Disease-free Seed Potato Production (무병종서 생산을 위한 감자X바이러스 및 엽권바이러스에 관한 연구)

  • A series of experiment was carried out to study on the production of disease-free seed potatoes at the Alpine Experiment Station from 1960 to 1968, which initiated a study of comparison on degeneration of plain warm region and high altitude products and the effect of latent potato virus X (PVX) and potato leaf roll virus(PLRV) on degeneration. Particular observations were made on some aspect of the nature of potato virus disease and its control such as concentrations of PVX, range of host plants, physical properties such as concentrations of PYX, range of host plants, physical properties and carrying effect of insects, by investigating 9 different areas of the main potato producing regions (Kimhae, Taegu, Choongju, Taejoen, Suwon, Kwangju, Chonju, Cheju and Chinju). Highly purified anti-serum was separated and tested for control of the virus disease and also various method of prevention and control of PLRV were observed, using cultivation of sprouted seed tubers, early harvesting method, and systemic chemicals. The results obtained are summarized as follows; 1. Potato yield in the plain region decreased by 32.8~66.3% in the first year cultivation of seed potatoes from colder region, and the rate of virus infection was 92.9 to 95.4%. 2. Plants of three families including, 20 species were susceptible to the PVX, and among the plants Salvia officinalis of a habits only was the carrier while the symptom of Digitalis purpurea of Screphulariaceae was masked. Necrosis and ring spot was occurred in most pJants of the Solanaceae and ring spot symptom also was observed in Nicotiana tabacum L. var. White Burley and in N. glutinosa. 3. The 8$C_2$ strain of virus had the following physical properties; thermal inactivation point, 68-$72^{\circ}C$ : dilution inactivation point, above 1, 000, 000 dilution: ageing in vitro, 240-360 days: and ageing in dry plant tissue, 30 days. 4. Myzus persicae and Oxya spp. did not transmit the 8$C_2$ strain of potato virus. 5. Virus was purified through the ammonium sulphate isolating method, and higher titer value, 1/2048 was obtained through anti-serum test. 6. Inhibition Chenopodiacae on the virus infection of potato was remarkable, and inhibition of local lesion host also was observed. 7, By earlier planting of sprouted seed tubers, growth period could be prolonged by 10 to 12 days. 8. Earlier harvest decreased much the rate of virus infection of seed potatoes. 9. According to the results of aphid control trial using systemic soil insecticides at Kangnung and Taekwanlyung, PSP 204, Disyston and Thimet was effective to aphid control. In particular, control effect of twice treatments of PSP 204 was great. 10. Treatmental effect of those chemicals lasted about 60-70 days. However, single foliar application of emulsified chemicals was not effective to potato virus control. 11. The effect of PSP 204, Disyston, and Thimet on the control of potato leaf roll virus was great, particularly in the case of two treatments of PSP 204, at Kangnung as well as at Taekwanlyung. Higher negative correlationship between the control effect of potato leaf roll virus and potato yield was observed showing the value r=-0.85 at Kangnung, and r=-0.87 at Taekwanlyung. 12. Differences in the control effects among PSP 204, Disyston, and Thimet was not noticed.

Construction and Characterization of an Anti-Hepatitis B Virus preS1 Humanized Antibody that Binds to the Essential Receptor Binding Site

  • Hepatitis B virus (HBV) is a major cause of liver cirrhosis and hepatocellular carcinoma. With recent identification of HBV receptor, inhibition of virus entry has become a promising concept in the development of new antiviral drugs. To date, 10 HBV genotypes (A-J) have been defined. We previously generated two murine anti-preS1 monoclonal antibodies (mAbs), KR359 and KR127, that recognize amino acids (aa) 19-26 and 37-45, respectively, in the receptor binding site (aa 13-58, genotype C). Each mAb exhibited virus neutralizing activity in vitro, and a humanized version of KR127 effectively neutralized HBV infection in chimpanzees. In the present study, we constructed a humanized version (HzKR359-1) of KR359 whose antigen binding activity is 4.4-fold higher than that of KR359, as assessed by competitive ELISA, and produced recombinant preS1 antigens (aa 1-60) of different genotypes to investigate the binding capacities of HzKR359-1 and a humanized version (HzKR127-3.2) of KR127 to the 10 HBV genotypes. The results indicate that HzKR359-1 can bind to five genotypes (A, B, C, H, and J), and HzKR127-3.2 can also bind to five genotypes (A, C, D, G, and I). The combination of these two antibodies can bind to eight genotypes (A-D, G-J), and to genotype C additively. Considering that genotypes A-D are common, whereas genotypes E and F are occasionally represented in small patient population, the combination of these two antibodies might block the entry of most virus genotypes and thus broadly neutralize HBV infection.

  • A novel method is developed to yield virus titers in 10 h, is easy to .perform using 96-well plates, and applicable to both any Autographa californica nucleopolyhyderovirus (AcNPV) and Bombyx mori nucleopolyhedrovirus (BmNPV)-based recombinant baculovirus. This assay uses an antibody to a DNA-binding protein to detect the infected cells via immune-staining. The titer is determined by counting foci produced due to infection of virus under a fluorescent microscopy. The required incubation period was shortened considerably because infected cells expressed viral antigens at the post infection time of 4 h. Therefore, 10 hours were enough to estimate the virus titer including virus infection time, insect cell culture, and estimation of virus titer.

  • Schwann cells and neuronal cells from dorsal root ganglion (DRG) in embryos of rat were cultured in vitro respectively. The purified neuronal cells with anti-mitotic agents and purified Schwann cells were co-cultured and then accomplished myelination processing. Infection of Semliki forest virus into this myelinated co-culture system was performed and then accomplished demyelination. We identified myelination and demyelination processing using antibody of neuropeptide Y.

  • Total saponins extracted from Panox ginseng have no effect on the cytotoxic T-cell activity, and natural killer cell activity in mice infected with A/WSN influenza virus. The saponins, however suppressed delayed-type hypersensitivity responses to the virus and to sheep erythrocytes when administered to the animal before sensitization. Thus a prophylactic anti-inflammatory action of the total saponins of ginseng is observed, which may be related to their steroid-like structure.

  • Hepatitis A virus (HAV) is a causative agent of triggering acute hepatitis which is transmitted by person-to-person contact and or fecal-oral route. In previous studies, most hepatitis A virus (HAV) isolates had been genotype IA in Korea. Recently, a small number of different genotypes were reported with an upsurge of acute hepatitis by HAV. Therefore, the distribution of HAV genotypes was investigated. RNA was extracted from anti-HAV IgM positive sera which were collected from February to August 2009, at a tertiary care hospital in eastern Jeonnam, Korea. Nested reverse transcription PCR and direct sequencing for VP1/P2A region of the HAV were performed. A total of 365 cases with suspected acute hepatitis were tested for anti-HAV IgM and positive results were obtained in 24 sera (9.0%), which were collected 2 to 15 days (median, 7 days) after the onset of symptoms. Of the 24 seropositive samples, 14 (58.3%) samples were positive for HAV RNA, among which 4 isolates (28.6%) were genotype IA and the other 10 (71.4%) were genotype IIIA. Both IA and IIIA genotypes were isolated from 5~6 neighboring administrative districts throughout the year without geographic or seasonal restrictions. HAV genotypes (IA and IIIA) were observed from the eastern Jeonnam for the studied.

Production of Polyclonal Antibody against Grapevine fanleaf virus Movement Protein Expressed in Escherichia coli

  • The genomic region of Grapevine fanleaf virus (GFLV) encoding the movement protein (MP) was cloned into pET21a and transformed into Escherichia coli strain BL21 (DE3) to express the protein. Induction was made with a wide range of isopropyl-${\beta}$-D-thiogalactopyranoside (IPTG) concentrations (1, 1.5, and 2 mM) each for duration of 4, 6, or 16 h. However, the highest expression level was achieved with 1 mM IPTG for 4 h. Identity of the expressed protein was confirmed by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) followed by Western blotting. The expressed 41 kDa protein was purified under denaturing condition by affinity chromatography, reconfirmed by Western blotting and plate-trapped antigen enzyme-linked immunosorbent assay (PTA-ELISA) before being used as a recombinant antigen to raise polyclonal antibodies in rabbits. Purified anti-GFLV MP immunoglobulines (IgGs) and conjugated IgGs detected the expressed MP and GFLV virions in infected grapevines when used in PTA-ELISA, double antibody sandwich-ELISA, and Western blotting. This is the first report on the production of anti-GFLV MP polyclonal antibodies and application for the virus detection.

Android Malware Detection using Machine Learning Techniques KNN-SVM, DBN and GRU

  • Android malware is now on the rise, because of the rising interest in the Android operating system. Machine learning models may be used to classify unknown Android malware utilizing characteristics gathered from the dynamic and static analysis of an Android applications. Anti-virus software simply searches for the signs of the virus instance in a specific programme to detect it while scanning. Anti-virus software that competes with it keeps these in large databases and examines each file for all existing virus and malware signatures. The proposed model aims to provide a machine learning method that depend on the malware detection method for Android inability to detect malware apps and improve phone users' security and privacy. This system tracks numerous permission-based characteristics and events collected from Android apps and analyses them using a classifier model to determine whether the program is good ware or malware. This method used the machine learning techniques KNN-SVM, DBN, and GRU in which help to find the accuracy which gives the different values like KNN gives 87.20 percents accuracy, SVM gives 91.40 accuracy, Naive Bayes gives 85.10 and DBN-GRU Gives 97.90. Furthermore, in this paper, we simply employ standard machine learning techniques; but, in future work, we will attempt to improve those machine learning algorithms in order to develop a better detection algorithm.

Inhibition of HIV-1 Replication by Extract of Coptis chinensis (황련추출물에 의한 HIV-1 복제 저해)

  • Natural products, total number of 175, were screened to test for their effect on the replication of human immunodeficiency virus type 1 (HIV-1). Five of them, such as Eriobotrya japonica, Eugenia caryphyllata, Cuscuta chinensis, Glycyrrhiza uralensis, and Coptis chinensis were shown to be effective in inhibiting the replication of HIV-1 in tissue culture and their selectivity indexes were 42, 40, 14, 18 and 65, respectively. To further fractionate Coptis chinensis, which is shown to be highest anti-HIV-1 activity, methanol extracts of Coptis chinensis were fractionated into methylene chloride at pH3, pH10 and water residue. The selectivity Indexes of CH$_2$C1$_2$(pH 3), CH$_2$C1$_2$(pH 10) and water residue were 50, 22 and 98 respectively. Our results show that the water residue of Coptis chinensis was the most effective for anti-HIV-1 activity.
