Production of Porcine Embryos in Different Culture Medium

  • Early development of porcine oocytes fertilized in vitro was examined in different culture conditions. Porcine ovaries were collected from local slaughter-house. Cumulus-oocytes complexes were aspirated from 2 to 6 mm follicles. The collected oocytes were cultured for in vitro maturation in NCSU-23 medium with 5 mM hypotaurine, 0.57 mM cystein, 10% porcine follicle fluid, 10 IU/ml PMSG and 10 IU/ml hCG for 42~44 hrs. The frozen-thawed spermatozoa were washed by centrifigation 2 times at 1, 500 rpm in D-PBS with 5.56 mM glucose, 0.33 mM Na-pyruvate, 100 IU/ml penicillin, 1$\mu\textrm{g}$/ml streptomycin and 1ng/ml BSA. The fertilization medium used mTBM with 2 mM caffeine and 2 mg/ml BSA and adjusted to a pH of 7.2 to 7.4. The final concentration of spermatozoa was adjusted to 2.5$\times$10$^{6}$ cells/ml motile sperm during fertilization in vitro. At 8hrs h after insemination, the oocytes were transferred into NCSU-23 medium with 5.0 mM hypotaurine and 4 mg/ml BSA and cultured for 7 days. In first experiment, the mean numbers of oocytes collected from 20 ovaries were 674.4 oocytes, and 4.1(27.6), 12.5(84.0), 25.4(171.6) and 57.9%(390.8) for A, B, C and D grade in morphological classification. In the second experiment, when culture medium was supplemented with various concentrations of EGF, the proportions of oocytes cleaved were 56.9, 55.7, 61.9 and 54.7% for 0, 5, 10 and 20ng/ml EGF. The higher proportions(15.1%) of oocytes developed to morular stage were obtained at concentration of 10ng/ml than 0 and 5ng/ml EGF (P<0.05). However, the proportions of embryos developed to blastocyst stage were not significantly different among concentrations of EGF. In another experiment, when the medium supplemented with catalase was used, the proportions of oocytes cleaved were higher in the concentration of 0 unit (56.5%, 61/108) than 100 and 1, 000 unit/ml of catalase (P<0.05). Although the developmental capacity of embryos was improved by medium with 0 unit/ml compared with 100, 500 and 1, 000 units/ml of catalase in oocytes developed to morula and blastocyst stages, were not significantly different among concentrations of catalase.

FRESH CASSAVA AS A FEED FOR FATTENING PIGS

  • An experiment was conducted to investigate the effects of feeding fresh cassava roots on the performance and carcass quality of pigs raised from $15{\pm}1kg$ to 85 kg live weight. Fresh unpeeled cassava roots were chopped into chips and offered separately along with a protein concentrate made up of copra cake and meat and bone meal, and a local mineral-vitamin premix. Commercial pig grower and finisher diets served as control. A total of 24 pigs were used in an incomplete randomized split-plot design experiment. Pigs fed fresh cassava-based diet grew as fast, 0.78 vs 0.77 kg/day, and were as efficient, 3.74 vs 3.77 in converting feed into body weight gain as those fed the commercial control diet. Similarly, there were no differences in carcass quality measured in terms of dressing percentage, 77 vs 77; backfat thickness, 2.76 vs 2.78 cm; loin eye muscle area, 29.2 vs $29.1cm^2$ and in the relative proportions of the different carcass cuts between the two dietary treatments. The use of fresh cassava along with the protein and the local mineral-vitamin premix however, resulted in lowered total feed cost and cost per unit of live weight gain. It is concluded that fresh cassava roots can be fed along with copra cake, meat and bone meal protein concentrate and a local mineral-vitamin premix to fattening pigs with no adverse affects on performance and carcass quality.

Estrus Behavior and Superovulatory Response in Black Bengal Goats (Capra hircus) Following Administration of Prostaglandin and Gonadotropins

  • The present study was conducted to explore the possibilities of estrus induction and superovulation in a native Indian breed of goats called 'Black Bengal'. Forty-two adult non-pregnant females were divided in two groups, of which 18 goats were subjected to a superovulatory treatment comprising of equine chorionic gonadotropin (eCG), Prostaglandin (PGF2$\alpha$) and human chorionic gonadotropin (hCG) to induce superovulation. The remaining 24 goats received no treatment and served as controls for the parameter under study as well as recipients for embryo transfer studies. The average duration of estrus was found to be significantly increased in treated goats (34.2${\pm}$3.4 h) compared to controls 3.0${\pm}$2.4 h). The average duration between PGF administration and occurrence of estrus was 2.0${\pm}$5.2 h. After mid ventral laparotomy, superovulatory responses indicated a significant increase in the number of follicles, which was 8.27${\pm}$0.37 in the treatment group compared to 4.16${\pm}$0.17 in the control group. The number of corpora lutea was also significantly increased in treated animals compared to control (2.90${\pm}$0.86 vs. 0.74${\pm}$0.04) respectively per ovary per goat.

Effects of Octacosanol Extracted from Rice Bran on the Laying Performance, Egg Quality and Blood Metabolites of Laying Hens

  • A 42-d study with 384 Hy-line brown laying hens was conducted to assess the effects of dietary octacosanol supplementation on laying performance, egg quality and blood metabolites of laying hens. Hens were randomly allocated into 4 dietary groups of 8 cages each, which were fed basal diet supplemented with 0 (Control), 9 (OCT9), 18 (OCT18), and 27 (OCT27) mg/kg diet of octacosanol isolated from rice bran, respectively. The experiment was conducted in an environmental controlled house and hens were fed twice daily for ad libitum intake. Laying performance was determined over the 42-d period, and egg quality as well as blood metabolites were estimated on d 21 and d 42. Diets in OCT18 and OCT27 increased (p<0.05) laying rate, egg weight, egg mass, egg albumen height, Haugh unit and eggshell strength on d 42, but decreased (p<0.05) feed conversion rate and levels of total cholesterol, triglyceride and low density lipoprotein cholesterol in the serum as compared to those of Control. Feed intake, yolk color, yolk diameter, eggshell thickness and high density lipoprotein cholesterol were similar (p>0.05) among treatments. Results demonstrate that supplementing 18 to 27 mg/kg diet of rice bran octacosanol can improve laying rate and egg quality and reduce blood lipid of laying hens.

Xylanase Production by Bacillus sp. A-6 Isolated from Rice Bran

  • A Bacillus sp. A-6 strain that produced xylanase was isolated from rice bran. The optimal temperature and pH for xylanase activity of the culture supernatant of Bacillus sp. A-6 were 40$^{\circ}C$ and pH 7, respectively. The optimal temperature and pH for xylanase production in the xylan medium were 30$^{\circ}C$ and pH 9, respectively. The optimal concentrations of oat spelt xylan and peptone for xylanase production were 0.5% and 1.5%, respectively. The best nitrogen sources for xylanase production was beef extract, but xylanase production was also supported comparably by tryptone and peptone. The bacterial growth in the optimal xylan medium reached stationary growth phase after 12 h of incubation. The xylanase production in the culture supernatant increased dramatically during the initial 12 h exponential growth phase and then remained constant at 23.8-24.5 unit/ml during the stationary growth phase. The pH of the culture medium decreased from 8.8 to 6.7 during the exponential growth phase and subsequently increased to 8.1 during the stationary growth phase. Rice bran, sorghum bran, and wheat bran as well as oat spelt xylan induced xylanase production. The xylanase production was repressed when glucose was added to the xylan-containing medium.

  • Each year, vast amount of agricultural crop residues are produced (about 60 percent of the total crop production), which have not been effectively utilized because they are bulky and lignocellulosic, thus having little fuel energy per unit volume. Using treated plant residues as animal feeds could result in an ultimate saving of fossil fuel energy and a more effective utilizat ion of products created by the photosynthetic process. Feeding the residues to animals would decrease the pollution potential, but these residues are difficult for even a ruminant animal to digest. If cellulosic wastes produced from cereal grain straw and wood could be digested, land now used for producing forage add grain cnuld be shifted to food crops for humans. During the past decade, considerable efforts were made to utilize crop residues. These utilization methods can be broadly grouped into for categories: (1) direct uses, (2) mechanical conversions, (3) chemical conversions and (4) biological conversions. Agricultural crop residues consist mainly of cellulose, hemicellulose, lignin, pectin, andother plant carbohydrates. The nature of the constituents of these residues can be best utilized as one of the five FS: Fuel, Fiber, Fertilizer, Feed and Food. Many processes have teen proposed and some are in industrial production stage. However, economics of the process depend on the location where availability of other competitive products are different.

EFFECT OF AGE ON THE LEVEL OF SERUM ALKALINE PHOSPHATASE ACTIVITY OF JAPANESE BLACK STEERS

  • The level of serum alkaline phosphatase activity was determined in 7 Japanese Black steers at different ages. The isoenzyme activity of non-bone origin was estimated using a heat-inactivation technique. The activity of serum alkaline phosphatase (SALP, K-A unit) decreased as age (AGE, mo.) increased: SALP = 14.15 - 0.17 (${\pm}\;0.03$) AGE, r = -0.81, P<0.01, $S.E.\;{\pm}\;0.28$. The variation of the activity was greater in younger age than the older. The temperature of $58^{\circ}C$ for the treatment of heat inactivation of bovine serum appeared to be suitable. The percentage of heat inactivated enzyme activity negatively correlated with age and positively with the level of serum alkaline phosphatase activity. The activity of SALP of non-bone origin was inferred to stay at about constant level irrespective of age and that of bone origin decreased with age.

MILK FAT CONTENT AND PRODUCTION PERFORMANCE OF HOLSTEIN DAIRY COWS FED FISH MEAL

  • Performance and production of twenty lactating Holstein cows fed diets containing either soybean meal or fish meal as the primary protein source were compared in a continuous or split feeding scheme. At 1 wk prepartum four groups of five animals were placed on each experimental diet. Animals assigned to the continuous feeding scheme were continued on these diets for 10-wk postpartum. At 4 wk postpartum, the diets for the groups assigned to switching protein sources were changed. These treatments were continued for another 6 wk. Milk production and dietary intakes were recorded daily. Milk constituents were measured every 2 d. Cows weights, rumen fluid samples and jugular blood samples were collected weekly. Data showed no effect of early lactation diet on cow performance or milk characteristics. Overall, compared to the soybean meal diet, the fish meal diet lowered the milk fat percent and increased production of milk per unit of dry matter ingested. No differences were observed for volatile fatty acid content of rumen fluid, blood mineral content, milk protein, somatic cell count, 4%-fat corrected milk, dry matter intake, or body weight.

Food Preservation Technology at Subzero Temperatures: A Review

  • Purpose: Cold storage is the most popular method used to preserve highly perishable foods such as beef and fish. However, at refrigeration temperatures, the shelf life of these foods is limited, and spoilage leads to massive food waste. Moreover, freezing significantly affects the food's properties. Ice crystallization and growth during freezing can cause irreversible textural damage to foods through volumetric expansion, moisture migration induced by osmotic pressure gradients, and concentration of solutes,which can lead to protein denaturation. Methods: Although freezing can preserve perishable foods for months, these disruptive changes decrease the consumer's perception of the food's quality. Therefore, the development and testing of new and improved cold storage technologies is a worthwhile pursuit. Results: The process of maintaining a food product in an unfrozen state below its equilibrium freezing temperature is known as supercooling. As supercooling has been shown to offer a considerable improvement over refrigeration for extending a perishable product's shelf life, implementation of supercooling in households and commercial refrigeration units would help diminish food waste. Conclusions: A commercially viable supercooling unit for all perishable food items is currently being developed and fabricated. Buildup of this technology will provide a meaningful improvement in the cold storage of perishable foods, and will have a significant impact on the refrigeration market as a whole.

Design and Fabrication of a Dual Cylindrical Microwave and Ohmic Combination Heater for Processing of Particulate Foods

  • Purpose: Dual cylindrical microwave chambers equipped with an ohmic heating tube were designed and fabricated to maximize the electric field strength for expeditious heat treatment of particulate foods. Methods: The efficacy of the combination heater was investigated by simulating the electric field distribution by using COMSOL Multiphysics software. Results: All components of the designed microwave heating unit were suitable for transmitting maximal microwave power to the load. The simulated electric field distribution implied that single-mode microwave heating would be sufficient for the steady generation of a highly localized heating zone in the cavity. During impedance matching, the calculated reflection coefficient ($S_{11}$) was small, possibly implying minimal power loss and wave reflection in the designed microwave heating chamber. Conclusions: This study demonstrates the possibility of concentrating the microwave power at the centerline for a single-frequency microwave, for thermal treatment of multiphase foods without attenuating the microwave power.
