유당분해효소에 의한 Glycoside의 Aglycone으로 전환을 위한 최적 조건 확립 (Optimum Conditions for Glycoside Conversion to Aglycone by ${\gamma}-Galactosidase$)

  • This study was designed to find the optimum conditions for converting isoflavone glycoside to aglycone by ${\beta}-galactosidase$. Three different forms of the enzyme were tested and the optimum enzyme concentration, incubation temperature, pH, and incubation time were determined. Before treatment with enzyme, isoflavone contained 89.4% glycoside including daidzin, glycitin and genistin, and only 10.6% aglycone including daidzein, glycitein and genistein. Among the enzymes tested, the highest rate of isoflavone hydrolysis to aglycone, 35%, was observed when 3 unit/g Fungal Lactase (Amano Enzyme) was used. Higher incubation temperatures resulted in a higher rate of hydrolysis along with a greater loss of isoflavone mass. Therefore, body temperature $(37^{\circ}C)$ may be adequate for isoflavone conversion, with 44.9% hydrolysis and less than 10% loss of mass. As expected, a higher amount of aglycone was produced at pH 7 compared with other pH values. During 5hr of incubation, the conversion of glycoside to aglycone increased dramatically from 0 to 1hr, and plateaued thereafter. In addition, commercial soy-based milk was hydrolyzed more effectively with ${\beta}-galactosidase$ when incubated for 5hr. Based on the above results, the optimum conditions for isoflavone hydrolysis by ${\beta}-galactosidase$ were for 3 hr at $37^{\circ}C$, pH 7 with 3 unit/g Fungal Lactase (Amano Lactase), yielding an average total amount of aglycone ranging from 40 to 47%.

Effects of Uphill Exercise on Digestible Energy Intake and Energy Expenditure during Exercise in Yearling Horses

  • The objectives of this study were to evaluate the digestible energy intake and energy expenditure in yearling horses on different training protocols (uphill- and level-track exercise training protocols). Twenty-four thoroughbred yearlings (12 males and 12 females, aged 27.0$\pm$0.9 months) were divided into two groups based on their training on two different tracks: the uphill (with a gradient of about 3%) training group (uphill training) and the level training group (level training). The digestible energy (DE) intake and energy expenditure (EE) during exercise were measured in both the groups. It was found that the DE intake in the uphill training and the level training groups was 5.1$\pm$3.1 and 36.9$\pm$4.8 Mcal/day, respectively. The EE during exercise in the two groups was 3.05$\pm$0.51 and 2.07 $\pm$0.56 Mcal, respectively. Thus, there was a significant difference in the EE (p<0.05), but not in the DE intake between the animals of the two training groups. The EE for a given intensity of exercise was greater in the uphill training group than in the level training group, but the DE intake was not affected by the exercise intensity. The DE intake was not generally affected by the intensity of exercise in this study, but a daily negative gain of body weight was observed in the uphill training group, particularly in the females. Thus, the energy requirement may be higher in yearlings undergoing uphill training than in those undergoing level training.

Influences of Long-Term Feeding of Japanese Green Tea Powder on Laying Performance and Egg Quality in Hens

  • Influences of Japanese green tea powder (GTP) supplementation to commercial diet on laying performance and egg quality were studied by using 60 laying hens. The experimental diet with or without 0.6% GTP was given ad libitum to the birds during the period from 6 to 71 weeks of age. The birds started egg production from 21 wk of age regardless GTP feeding. Body weight, feed intake, egg weight tended to decrease with GTP supplementation, while egg production rate tended to increase. Haugh unit score was significantly increased with GTP, which accompanied with the increased albumen height. These were observed almost throughout the laying period over 50 wk. Gel proportion in thick albumen was decreased as storage time is prolonged, then higher values of the gel proportion were recorded in the eggs from GTP group. Thick albumen of the eggs from GTP-fed layers had more carbohydrate than that from control layers. All these indicate physical stability of thick albumen in the eggs from GTP group. Thiobarbituric acid content of egg yolk tended to remain lower in the eggs from GTP group during 5-10 days of storage at room temperature. Levels of egg yolk cholesterol and yolk lipid were significantly reduced by GTP feeding. There were no significant differences in eggshell weight, shell thickness and shell strength between the two groups. Thyroid gland and liver from hens slaughtered at 71 wk of age did not differ in weight irrespective of GTP feeding. The present results suggest that GTP could modify components of edible part of egg, leading to the characteristics favourable to consumers such as high durability of thick albumen and less cholesterol in yolk, without altering general performance of the layers throughout this year round experiment.

Effect of Season, Parity and Lactation on Reproductive Performance of Sows in a Tropical Humid Climate

  • The aim of this study was to analyze post-weaning reproductive performance of Large White sows in relation to season, parity and their lactation performance under tropical conditions in Guadeloupe (French West Indies, $16^{\circ}$ Lat. N, $61^{\circ}$ Long. W.). This work was based on data recorded in the experimental unit of INRA from January 1993 to December 2003. Two seasons were determined a posteriori from climatic parameters recorded continuously in a station close to the experimental unit. Mean ambient temperature was higher during the hot season than the warm season ($26^{\circ}C$ vs. $24^{\circ}C$) but relative humidity was comparable for both seasons (i.e. 87% on average). Season had a significant effect on all reproductive parameters analyzed. Primiparous sows weaned in the hot season had a higher probability of a prolonged weaning to estrus interval, WEI (odds ratio was 4.1; p<0.01) but multiparous sows were not affected. A higher probability of a prolonged weaning to conception interval, WCI (odds ratio >2.5, p<0.01) and a lower subsequent farrowing rate (-10%, p<0.01) were found for sows weaned in the hot season. A higher daily feed intake during lactation reduced the probability of a prolonged WEI (p<0.05). Body weight and average back-fat thickness at farrowing affected WEI and WCI (p<0.05), whereas body weight and average backfat thickness change in lactation did not. This study confirms the negative effects of the hot season on primiparous reproductive performance. It also indicates that lactation performance influences sow non-productive period.

Effect of Fermented Soybean "Natto" Supplement on Egg Production and Qualities

  • Natto is a Japanese traditional soybean product fermented by Bacillus natto. The effects of dried fermented soybean (natto) supplement on egg production and egg qualities of layer chickens was studied with regard to the effective use of various waste foods in Japan. Dried natto, prepared by heating at $60^{\circ}C$, was added to a basic diet at a level of up to 3%. Forty 166-wk-old layer chickens (Rhode Island Red) were randomly divided into 4 groups and five layer chickens were used in each group with two replicates. Layer chickens in group 1 were fed a basic diet as the control. The remaining 3 groups were fed the basic diet supplemented with dried natto at levels of 1, 2, and 3% (w/w), respectively. The result did not show improvements in egg production or feed conversion ratio of layer chickens even when 3% dried natto was added to the control diet. The egg qualities including egg weight, eggshell strength and thickness, yolk color, yolk weight, albumen height, and Haugh unit were also not improved. However, the feeding of dried natto changed the cholesterol content in the egg yolk. The supplementation of dried natto showed the tendency to decrease the yolk cholesterol after 12-wk of feeding compared to the control diet though it did not change plasma cholesterol levels in the blood. On the other hand, yolk cholesterol decreased significantly after 12-wk of feeding 3% dried natto (p<0.05).

환경성 발암물질의 용량-반응모델의 이론적 근거와 응용에 관한 연구 - 음용수 중 chloroform을 중심으로 (Scientific rationale and applicability of dose-response models for environmental carcinogens)

  • This study described methods to predict human health risk associated with exposure to environmental carcinogens using animal bioassay data. Also, biological assumption for various dose-response models were reviewed. To illustrate the process of risk estimate using relevant dose-response models such as Log-normal, Mantel-Bryan, Weibull and Multistage model, we used four animal carcinogenesis bioassy data of chloroform and chloroform concentrations of tap water measured in large cities of Korea from 1987 to 1995. As a result, in the case of using average concentration in exposure data and 95% upper boud unit risk of Multistge model, excess cancer risk(RISK I) was about $1.9\times10^{-6}$, in the case of using probability distribution of cumulative exposure data and unit risks, those risks(RISK II) which were simulated by Monte-Carlo analysis were about $2.4\times10^{-6}\;and\;7.9\times10^{-5}$ at 50 and 95 percentile, respectively. Therefore risk estimated by Monte-Carlo analysis using probability distribution of input variables may be more conservative.

로봇에 의한 유두컵 착탈 시스템 개발을 위한 기초 연구 (Basic Study for the Development of Teat Cup Handling System Operated by a Robot)

  • 본 연구는 로봇착유기 개발을 위한 기초연구로서 유두위치 측정장치와 로봇제어 시스템을 구성하고 그 성능을 분석하고자 수행되었으며 그 결과는 다음과 같다. 1. 유두인식은 스테레오 측량법에 기초한 이미지 장치로 처리했으며 유두인식오차는 (x, y, z) = (0.83, 1.95, 0.81)mm이다. RCS로 좌표값을 변환했을 때 시스템 전체의 오차는 x = 0.9mm, y = 2.0mm, z = 0.9mm였다. 2. 로봇 유두컵 착탈 시스템의 로봇 착탈 성공률은 평균 91.5%로 나타났다. 작업시간은 27.8sec였으며, 이미지 처리를 포함한 전체 작업시간은 86.1sec로 나타났다.

돼지 체외수정란의 발달에 미치는 Mouse Leukemia Inhibitory Factor의 영향 (Effect of Mouse Leukemia Inhibitory Factor on the Development of In Vitro Produced Pig Embryos)

  • 본 연구는 돼지 난포란으로부터 생산된 수정란의 체외발달에 미치는 아미노산, 우태아혈청 (FBS)과 Leukemia Inhibitry Factor의 영향을 조사하였다. 돼지난포란은 도살된 돼지의 난소로부터 회수하여 39$^{\circ}C$, 5% CO2 배양조건하에서 1$\mu\textrm{g}$/ml FSH-p, 1$\mu\textrm{g}$/ml Estradiol-17$\beta$와 10%FBS가 첨가된 TCM-199 배양액내에서 42시간동안 성숙시켰다. 사출된 정자의 수정능 획득은 45와 90% Percoll density gradient법을 통한 원심분리에 의해 얻었으며, 이들 수정능획득된 정자를 성숙된 난포란이 함유된 배양액에 3$\times$105/ml의 농도로 주입하여 10$\pm$1시간동안 배양함으로서 체외수정을 유도하였다. 수정된 난포란은 ; 1) 10% FBS가 함유된 TCM-199, DMEM, mKRB 또는 CR1aa 배양액, 2) 아미노산 또는 10% FBS가 첨가된 CR1 배양액, 3) STO 세포 또는 mLIF (1,000 unit/ml) 첨가된 CR1aa(10%FBS) 배양액, 4) mLIF (1,000 unit/ml)를 수정 직후 또는 8-세포기 이후에 첨가된 CR1aa(10%FBS)의 네가지 배양조건에서 각각 분리 배양하였다. 그결과 체외수정란의 배반포까지 발달율은 아미노산과 10%FBS가 포함된 CR1 배양액에서 다른 배양액에서보다 양호하였고, 특히 8-세포기 이후에 mouse LIF를 첨가한 CR1aa(10% FBS) 배양액에서는 다른 배양조건보다 현저히 높은 결과를 보였으며, 부화 배반포까지도 배발달을 유도할 수 있었다. 따라서 돼지수정란의 발달에 있어서 배양액내에 아미노산과 FBS 및 mouse LIF의 첨가는 효과가 있으며, 특히 8-세포기 이후에 있어서 mouse LIF의 첨가는 돼지의 수정란을 배반포 이후의 단계에까지 발달시킬 수 있었다.

비닐하우스용 돈분뇨 액비살포장치 개발 (Development of Swine Liquid Manure Spread System for Greenhouse)

  • 비닐하우스용 소형 액비살포기를 설계, 제작하였으며, 납작호스용 일체형과 견고한 둥근호스 분리형으로 되어 있다. 살포장치의 구성은 권취기, 유압모터, 유압실린더, 배출장치, 호스, 3점링크히치, 호스고정장치, 프레임 등으로 되어있다. 살포 시스템은 일체형과 분리형으로 나누어진다. 일반적으로 납작호스는 일체형에 사용되고 둥근호스는 분리형에 사용된다. 분리형은 좁은 공간에서 선회 등을 할 때 작업의 활동성을 개선할 수 있다. 액비살포기를 사용함으로써 작업효율을 5배 정도 높일 수 있었으며, 인력소요는 1/3로 감소되었다. 원하는 살포량에 따른 작업속도를 맞추어주면 면적당 목표 살포량을 얻을 수 있다. 관리자에 대한 작업환경이 개선되었다.

Conjugated Linoleic Acid(CLA)의 급여가 계란의 품질에 미치는 영향 (Effects of Dietary Conjugated Linoleic Acid (CLA) on the Egg Quality)

  • 본 실험은 CLA급여수준과 기간이 계란의 품질에 미치는 영향에 대하여 연구하였다. 계란의 무게는 CLA 급여에 따른 유의적인 차이를 나타내지 않았으며, 난백의 높이는 급여 7주에서 CLA 급여구가 유의적(P<0.05)으로 낮은 값을 나타내었다. Haugh unit는 CLA 급여에 따른 유의적인 차이는 나타나지 않았다. 난황내 CLA축적량은 급여수준이 높을수록 유의적(P4O.05)으로 높은 축적량을 나타내었다. 그러나 저장 2주 이후에는 증가폭이 크지 않았다. CLA급여에 의한 난황내 지방산 조성을 보면 포화지방산이 증가되고 불포화 지방산이 감소하였는데 포화지방산인 palmitic acid의 증가와 불포화지방산인 oleic acid의 증가가 주요 원인인 것으로 사료된다. 난황색 (Roche)은 CLA 급여에 의해 유의적(P<0.05)으로 높게 나타났으며, 난황의 경도 또한 CLA급여에 의해 유의적(P<0.05)으로 높게 나타났다. 이와 같은 결과는 CLA 급여가 계란의 이화학적인 성질에 영향을 미친다는 것을 알 수있다. 그러나 CLA급여에 의한 계란 품질의 변화가 소비자의 관능적 특성에 미치는 영향에 관해서는 많은 연구가 필요할 것으로 사료된다.
