동계(冬季) 퇴비부숙열(堆肥腐熟熱) 이용(利用)에 관(關)한 연구(硏究) (Studies on the Utilization of Exothermic Heat Composting during Winter Season)

  • 동계(冬季)에 볏짚, 왕겨와 인축분뇨(人畜糞尿)를 원료(原料)로하여 재료별(材料別)로 퇴적(堆積)하고 부숙과정(腐熟過程)에서 발생(發生)되는 퇴비온도(堆肥溫度) 및 퇴비성상(堆肥性狀)의 변화(變化)를 조사(調査)한 결과(結果)는 다음과 같다. 1. 볏짚, 왕겨 1:1 혼합구(混合區)가 볏짚 단용구(單用區)에 비(比)하여 퇴비부숙발열기간(堆肥腐熟發熱期間)이 연장(延長)되었다. 2. 인산첨가구(燐酸添加區)의 퇴비온도(堆肥溫度)는 대조구(對照區)에 비(比)하여 발효초기(醱酵初期)에는 급격(急激)히 상승(上昇)하였으나 발효(醱酵) 30일(日) 이후(以後)에는 낮아지는 경향(傾向)이었다. 3. pH는 발효초기(醱酵初期)에 5.5이었으나 퇴비온도(堆肥溫度)의 급상승(急上昇)과 더불어 10일경(日傾)에 8.8까지 올라갔으며 30일 이후(日以後)부터는 거의 변화(變化)가 없었다. 4. 퇴비온도(堆肥溫度)가 $30{\sim}65^{\circ}C$로 유지(維持)되는 기간(其間)은 볏짚+계분구(鷄糞區)가 39일(日), 볏짚+왕겨+계분구(鷄糞區)가 69일(日), 볏짚+왕겨+돈분구(豚糞區)가 56일(日), 볏짚+왕겨+우분구(牛糞區)가 66일(日), 볏짚+왕겨+인분구(人糞區)가 21일(日)로서 볏짚+왕겨+계분구(鷄糞區)가 가장 길었다.

식품소재로서 자숙 다랑어류뼈의 성분 특성 (Component characteristics of cooking tuna bone as a food resource)

  • 참치통조림 가공 부산물인 다랑어류뼈를 식품소재와 같이 효율적으로 이용하기 위한 일련의 기초자료로서 가다랑어뼈 및 황다랑어뼈의 식품성분 특성에 대하여 검토하였다. 다락어류뼈의 건물당 조단백질 함량은 $29{\%}$로 두 어류뼈 간에 차이가 없었다. 이미 노산 조성비는 황다랑어뼈 콜라겐 (193잔기/1,000잔기)이 가다랑어뼈 콜라겐 (173잔기/1,000잔기)보다 높았으나, 가축뼈 콜라겐보다는 낮았다. 조지방함량, 폴리엔산/포화산 및 DHA 조성비는 황다랑어뼈가 각각 $12.5{\%}, 1.04 및 24.0{\%}$로 가다랑어뼈의 $6.6{\%}$,0.90 및 $23.9{\%}$보다 높았으나, 오징어 내장유보다는 낮았다. 다랑어류뼈의 주요 무기질은 칼슘 및 인이었고, 이들의 함량은 가다랑어뼈의 경우가 각각 36.5 g/100g ash 및 17.2g/100g ash이었고, 황다랑어뼈의 경우가 각각 38.0g/100g ash 및 18.7g/100g ash이었다. 이상의 결과로 미루어 보아 다랑어류뼈는 두 종류 모두가 콜라겐 및 지질 추출소재로 보다는 칼슘 추출소재로 적절하리라 판단되었다.

컴퓨터 정액자동분석에 의한 동결융해 한우 정액의 운동특성 연구 (Motional kinematics of Frozen-thawed Korean native cattle semen use of computer aided semen analysis(CASA) system)

  • The aim of this experiments were to assess the time-interval change of motional characteristics in frozen-thawed semen of Korean native cattle (KNC) by using computer aided semen analysis (CASA) technology. Twenty-six KNC frozen semen straws were obtained from Korean KNC improvement department, livestock improvement main division, national livestock cooperatives federation in Korea. Specimens were allowed to thaw at $37^{\circ}C$ for 30 sec in water bath. Semen analysis was performed on semen image analysis system (SIAS, Medical supply, Korea) adjusted to the gate settings and used the semen droplet ($5{\mu}l$) placed on Makler counting chamber (Sefi medical instrument, Israel) prewarmed at $37^{\circ}C$. The same person used the same micropipette to fill the Makler counting chamber. A total of 150 or more of sperms were analysed in each specimen by a single trained person by scanning at least 5 to 10 fields. The measurement parameters in SIAS were as follows ; frame rate = 30 frames per sec, image capture = 1 sec, minimum motile speed = $10{\mu}m/s$, maximum countable sperm number = 400. Statistical analysis was done by Student t-test with use of the Sigma plot program on a IBM personal computer. The dancemean(DNM) and hyperactivated sperm(HYP) of frozen-thawed KNC semen kinematics were significantly decreased(p < 0.05) after 10 min of incubation at $37^{\circ}C$ water bath. But, wobble(WOB) of same sample semen was significantly increased(p < 0.05) after 10 min of incubation and significantly decrease(p < 0.05) after 60 min of same incubation. And, after 30 mim of incubation, significantly differences were found most of motion kinematics, motifity(MOT), curvilinear velocity(VCL), straight line velocity(VSL), average path velocity(VAP), amplitude of lateral head displacement(ALH), beat cross frequency(BCF), mean angular displacement(MAD), dance(DNC), on same sample semen. The DNM of KNC semen sample was variable kinematics after 30 min of incubation. Also, the linearity(LIN) and straightness(STR) was significantly decreased(p < 0.05) from 60 min of incubation. In conclusion, the AI within 30 min after thawing of frozen semen can be an effective method for obtaining high fertility rate in KNC reproductive program.

포장 방법에 따른 즉석섭취형 함박스테이크의 냉장저장 중 품질 변화 (Quality changes in ready-to-eat hamburg steak depending on the packaging methods during chilled storage)

  • 본 연구에서는 포장방법에 따른 함박스테이크의 냉장저장 중 품질변화를 조사하였다. 대조구는 polylactide(PLA) tray를 이용한 함기포장(AC)이었으며, 처리구로는 진공포장(VAP)과 탈산소제 봉입포장(OS)이 적용되었다. 총균수는 대조구와 처리구 모두 저장기간의 경과에 따라 유의적으로 증가하였으며, 특히 AC구에서의 총균수 증가가 다른 처리구들에 비해 두드러진 것으로 나타났다. 또한 저장기간 중 TBA와 VBN값은 AC구에서 가장 빠른 증가를 보였으며 그 다음으로는 OS, 그리고 VAP포장구의 순이었다. 관능평가 결과 VAP구와 OS구 시료들은 모든 관능평가 항목에서 12일째까지 5.0점 이상의 평가를 받은 반면, AC구는 모든 항목에서 5.0 미만으로 이미 상품성을 상실한 것으로 확인되었다. 이러한 결과들은 VAP나 OS처리를 통하여 포장 내 산소를 제거함으로써 호기적 미생물의 생장과 산화작용의 억제로 인한 지방산패나 단백질부패속도가 지연된 것에 기인한 것으로 판단된다.

High Performance Flexible Inorganic Electronic Systems

  • The demand for flexible electronic systems such as wearable computers, E-paper, and flexible displays has increased due to their advantages of excellent portability, conformal contact with curved surfaces, light weight, and human friendly interfaces over present rigid electronic systems. This seminar introduces three recent progresses that can extend the application of high performance flexible inorganic electronics. The first part of this seminar will introduce a RRAM with a one transistor-one memristor (1T-1M) arrays on flexible substrates. Flexible memory is an essential part of electronics for data processing, storage, and radio frequency (RF) communication and thus a key element to realize such flexible electronic systems. Although several emerging memory technologies, including resistive switching memory, have been proposed, the cell-to-cell interference issue has to be overcome for flexible and high performance nonvolatile memory applications. The cell-to-cell interference between neighbouring memory cells occurs due to leakage current paths through adjacent low resistance state cells and induces not only unnecessary power consumption but also a misreading problem, a fatal obstacle in memory operation. To fabricate a fully functional flexible memory and prevent these unwanted effects, we integrated high performance flexible single crystal silicon transistors with an amorphous titanium oxide (a-TiO2) based memristor to control the logic state of memory. The $8{\times}8$ NOR type 1T-1M RRAM demonstrated the first random access memory operation on flexible substrates by controlling each memory unit cell independently. The second part of the seminar will discuss the flexible GaN LED on LCP substrates for implantable biosensor. Inorganic III-V light emitting diodes (LEDs) have superior characteristics, such as long-term stability, high efficiency, and strong brightness compared to conventional incandescent lamps and OLED. However, due to the brittle property of bulk inorganic semiconductor materials, III-V LED limits its applications in the field of high performance flexible electronics. This seminar introduces the first flexible and implantable GaN LED on plastic substrates that is transferred from bulk GaN on Si substrates. The superb properties of the flexible GaN thin film in terms of its wide band gap and high efficiency enable the dramatic extension of not only consumer electronic applications but also the biosensing scale. The flexible white LEDs are demonstrated for the feasibility of using a white light source for future flexible BLU devices. Finally a water-resist and a biocompatible PTFE-coated flexible LED biosensor can detect PSA at a detection limit of 1 ng/mL. These results show that the nitride-based flexible LED can be used as the future flexible display technology and a type of implantable LED biosensor for a therapy tool. The final part of this seminar will introduce a highly efficient and printable BaTiO3 thin film nanogenerator on plastic substrates. Energy harvesting technologies converting external biomechanical energy sources (such as heart beat, blood flow, muscle stretching and animal movements) into electrical energy is recently a highly demanding issue in the materials science community. Herein, we describe procedure suitable for generating and printing a lead-free microstructured BaTiO3 thin film nanogenerator on plastic substrates to overcome limitations appeared in conventional flexible ferroelectric devices. Flexible BaTiO3 thin film nanogenerator was fabricated and the piezoelectric properties and mechanically stability of ferroelectric devices were characterized. From the results, we demonstrate the highly efficient and stable performance of BaTiO3 thin film nanogenerator.

미니돼지에서 자가 피부유래 전구세포와 탈회골 및 피브린 스케폴드를 이용한 하악골 골결손부의 골재생에 대한 연구 (MANDIBULAR BONE REGENERATION USING AUTOGENOUS SKIN-DERIVED PRECURSOR CELLS WITH A MIXED DEMINERALIZED BONE AND FIBRIN GLUE SCAFFOLD IN MINIATURE PIGS)

  • Purpose: The aims of this study were to assess the in vitro co-culturing pattern of isolated skin-derived precursor cells (SKPs) with a mixed demineralized bone (DMB) and fibrin glue scaffold and to evaluate in vivo osteogenesis after transplantation of autogenous SKPs with a these mixed scaffold in the animal's mandibular defects. Materials and Methods: We isolated SKPs from the ears of adult 4 miniature pigs. The isolated SKPs were co-cultured with a mixed DMB and fibrin glue scaffold in a non-osteogenic medium for 1, 2, and 4 weeks. Histological characteristics of in vitro co-cultured cells and scaffold were evaluated. $1{\times}10^7\;cells/100\;{\mu}l$ of autogenous porcine SKPs were grafted into the mandibular defects with a DMB and fibrin glue scaffold. In the control sites, only a scaffold was grafted, without SKPs. After two animals each were euthanized at 2 and 4 weeks after grafting, the in vivo osteogenesis was evaluated with histolomorphometric and osteocalcin immunohistochemical studies. Results: Homogeneously shaped skin-derived cells were isolated from porcine ear skin after 3 or 4 weeks of primary culture. In vitro osteogenic differentiation of SKPs was observed after co-culturing with a DMB and fibrin glue scaffold in a non-osteogenic medium. Von Kossa-positive bone minerals were also noted in the co-cultured medium at 4 weeks. As the culture time progressed, the number of observable cells increased. Trabecular new bone formation and osteocalcin expression were more pronounced in the SKP-grafted group compared to the control group. Conclusion: These findings suggest that autogenous SKP grafting with a DMB and fibrin glue scaffold can serve as a useful alternative to bone grafting technique.

흰 코뿔소 배설물로부터 분리한 Bacillus pumilus H10-1의 Xylanase 활성 (Xylanase Activity of Bacillus pumilus H10-1 Isolated from Ceratotherium simum Feces)

  • Xylanase have been used to convert the polymetric xylan into fermentable sugars from the production of ethanol and xylitol from plant biomass. The aim of this study was to isolate and identify xylanolytic bacterium from herbivore feces and was to used the xylanase for enzymatic hydrolysis of biomass. Xylanolytic strains were isolated from 59 different feces of herbivores from Seoul Grand Park located in Gwacheon Gyeonggi-do. The xylanolytic strains were selected by congo red staining and DNS method. Total 67 strains isolated from the herbivores feces were tested for xylanase activity. Among the strains, H10-1, which has the highest xylanase activity, was isolated from feces of Ceratotherium simum. The H10-1 strain was identified as Bacillus pumilus based on its morphological/biochemical characteristics and partial 16S rDNA gene sequences. Culture conditions of B. pumilus H10-1 such as initial medium pH, incubation temperature and incubation time were optimized for maximum xylanase production. And also xylanase produced by B. pumilus H10-1 was applied for the saccharification of Miscanthus sacchariflorus cv. 'Geodae 1', which was pretreated with 1.5M NaOH. The optimized culture conditions of B. pumilus H10-1 were pH 9, $30^{\circ}C$ incubation temperature, and 7 day incubation time, respectively. This xylanase activity under the optimized conditions was $20.4{\pm}3.3IU$. The crude xylanase produced by B. pumilus H10-1 was used for the saccharification of xylan derived from pretreated 'Geodae 1'. The saccharification conditions were $50^{\circ}C$, 200 rpm, and 5 days. Saccharification efficiency of pretreated 'Geodae 1' by B. pumilus H10-1 was 8.2%.

가축분뇨 병합처리 바이오가스화를 위한 설계 및 운전 기술지침 마련 연구(III) 설계 및 운전 지침(안) 중심으로 (A Study on Establishment of Technical Guideline of the Installation and Operation for the Efficient Bio-gasification Facility of Pig Manure and Food Waste(III): Design and Operation Guideline)

  • 본 연구는 유기성폐자원 (가축분뇨, 음식물류폐기물, 음식물류폐수 등)을 병합 소화하는 시설을 대상으로 적정 설계 기준치를 충족하기 위한 설계 및 운전 기술지침서 마련하고자 현장조사와 정밀모니터링을 실시하였다. 현장조사시 도출된 결과를 바탕으로 설계 및 운전 사항, 모니터링 항목 및 주기, 시운전 기간 등 바이오가스화 시설의 전반적인 가이드라인을 제시하였다. 초기 혐기소화 처리용량 설계시 유입 원료별 배출 특성을 고려하여 처리용량의 10~30 %의 여유율을 적용하였다. 공정별 설계 지침의 경우, 반입 및 전처리 설비의 유기물 반입 농도를 TS 10 % 이하로 제한하고 혐기소화조 운전시 저해요인 제한 농도을 제시하였다. 또한 병합기준 유기물부하율 $1.5{\sim}4.0kgVS_{in}/(m^3{\cdot}day)$, 소화가스 이용설비는 탈황 및 제습 방법, 호기액비화조의 적정 설계 운영인자 등을 제안하였다. 바이오가스화 시설의 운전인자는 pH (산발효조 4.5~6.5, 메탄발효조 6.0~8.0), 온도변화폭 $2^{\circ}C$ 이내, 휘발성지방산과 암모니아를 각각 3,000 mg/L 이하로 유지할 것을 권장하였다.

Genetic Diversity and Phylogenetic Analysis of the Iranian Leishmania Parasites Based on HSP70 Gene PCR-RFLP and Sequence Analysis

  • Despite the broad distribution of leishmaniasis among Iranians and animals across the country, little is known about the genetic characteristics of the causative agents. Applying both HSP70 PCR-RFLP and sequence analyses, this study aimed to evaluate the genetic diversity and phylogenetic relationships among Leishmania spp. isolated from Iranian endemic foci and available reference strains. A total of 36 Leishmania isolates from almost all districts across the country were genetically analyzed for the HSP70 gene using both PCR-RFLP and sequence analysis. The original HSP70 gene sequences were aligned along with homologous Leishmania sequences retrieved from NCBI, and subjected to the phylogenetic analysis. Basic parameters of genetic diversity were also estimated. The HSP70 PCR-RFLP presented 3 different electrophoretic patterns, with no further intraspecific variation, corresponding to 3 Leishmania species available in the country, L. tropica, L. major, and L. infantum. Phylogenetic analyses presented 5 major clades, corresponding to 5 species complexes. Iranian lineages, including L. major, L. tropica, and L. infantum, were distributed among 3 complexes L. major, L. tropica, and L. donovani. However, within the L. major and L. donovani species complexes, the HSP70 phylogeny was not able to distinguish clearly between the L. major and L. turanica isolates, and between the L. infantum, L. donovani, and L. chagasi isolates, respectively. Our results indicated that both HSP70 PCR-RFLP and sequence analyses are medically applicable tools for identification of Leishmania species in Iranian patients. However, the reduced genetic diversity of the target gene makes it inevitable that its phylogeny only resolves the major groups, namely, the species complexes.

Dunaliella tertiolecta LB999 유래 바이오디젤의 산화특성 연구 (A Study on the oxidation characteristics of micro-algal bio diesel derived from Dunaliella tertiolecta LB999)

  • Bio diesel has advantages to reduce GHG(Greenhouse Gas) compare with the fossil fuel by using oil comes from plant/animal sources and even waste such as used cook oil. The diversity of energy feeds brings the positive effects to secure the national energy mix. In this circumstance, micro-algae is one of the prospective source, though some technical barriers. We analyzed the bio diesel which was derived from Dunaliella tertiolecta LB999 through the BD100 quality specifications designated by the law. From that result, it is revealed that the oxidation stability is one of the properties to be improved. In order to find the reason for low oxidation stability, we analyzed the oxidation tendency of each FAME components through some methods(EN 14111, EN14112, EN16091). In this study, we could find the higher double bond FAME portion, the more oxidative property(C18:1${\ll}C18:3$) in bio diesel and main unsaturated FAME group is acted as the key component deciding the bio diesel's oxidation stability. It is proved experimentally that C18:3 FAME are oxidized easily under the modified accelerated oxidation test. We also figure out low molecular weight hydrocarbon and FAME were founded as a result of thermal degradation. Some alcohol and aldehydes were also made by FAME oxidation. In conclusion, it is necessary to find the way to improve the micro-algal bio diesel's oxidation stability.
