Evaluation of Antimicrobial Activities of Sequential Spray Applications of Decontamination Treatments on Chicken Carcasses

  • The objective of this study was to evaluate the effects of sequential applications of ${\varepsilon}$-polylysine (EPL) or lauramide arginine ethyl ester (LAE) sprays followed by an acidic calcium sulfate (ACS) spray on inoculated chicken carcasses to reduce Salmonella (Salmonella enterica serovars including Salmonella typhimurium and Salmonella enteritidis) contamination during 6 days of storage ($4.4^{\circ}C$). Secondly, reductions of the resident microflora were studied on uninoculated chicken carcasses following the sequential application of the treatments, chilling and 10 days of storage at $4.4^{\circ}C$. The treatment of Salmonella inoculated carcasses with 300 mg/L EPL followed by 30% ACS (EPL300-ACS30) sprays reduced Salmonella counts initially by 1.5 log cfu/mL and then by 1.2 log cfu/mL (p<0.05) following 6 days of storage at $4.4^{\circ}C$. Likewise, 200 mg/L LAE followed by 30% ACS (LAE200-ACS30) treatment reduced initial Salmonella counts on poultry carcasses by 1.8, 1.4 and 1.8 log cfu/mL (p<0.05), respectively, after 0, 3, and 6 days storage. Immediately after the treatments, EPL300-ACS30 and LAE200-ACS30 both reduced Escherichia coli counts significantly by 2.6 and 2.9 log cfu/mL, respectively. EPL300-ACS30 and LAE200-ASC30 were effective in lowering psychrotroph counts by 1 log cfu/mL on day 10 when compared to the control and distilled water treatments. This study demonstrated that EPL300-ACS30 and LAE200-ACS30 were effective in reducing Salmonella on inoculated chicken carcasses both after treatment and during the storage at $4.4^{\circ}C$ for up to 6 days. In addition, reductions in psychrotroph counts indicated that these treatments might have the potential to increase the shelf-life of poultry carcasses.

Comparison of Water Retention and Loss of Chicken Carcasses by Different Water Chilling Condition (침지냉각 조건에 따른 닭 도체의 수분 흡수 및 감량 비교)

  • This study was conducted to investigate whether water chilling had effect on water retention, freshness or internal temperature of chicken carcasses during chilling or storage in two different chicken processing plants (designated as A and B). A total of 240 carcasses from chicken processing plants (n = 120 per chicken processing plant) was randomly sampled and evaluated the effect of water chilling on carcass characteristics (i.e., water retention, water loss or freshness during chilling or storage). Torrymeter value was used as an indicator of freshness in chicken carcasses. Water chilling did not affect carcass water retention between the processing plants. However, chicken carcasses processing in B plant exhibited significantly higher freshness (p<0.05) compared with those in A plant. This difference in freshness was mainly due to the longer transit time through the water chiller in A versus B plants. Water loss of carcasses during storage was not different between plants. It was found that carcass freshness can be affected by water chilling time as manifested in this study. Further study is warranted to see whether freshness or microbiological status of chicken carcasses may be affected depending on the chilling methods, i.e., air or water chilling.

Comparison of crude fat content and intramuscular fat score at different position of loin from Hanwoo (한우 등심의 위치별 조지방 함량 및 근내지방도 비교)

  • This study was conducted to make certain that the intramuscular fat score at 13th rib could represent the fat content of whole loin from Hanwoo in Korea beef grading system. A total 42 loins from 23 carcasses of grade $1^{{+}{+}}$, 16 carcasses of grade $1^+$, and 3 carcasses of grade 1 of Hanwoo were used for this study. The crude fat content and intramuscular fat score at the $1^{st}$, $6^{th}$, and $13^{th}$ rib, and the last lumbar position of loin were measured. The crude fat content was the highest at the $6^{th}$ rib of loin from the carcasses of grade $1^{{+}{+}}$ and $1^+$ (p<0.05). There were no significant difference of the intramuscular fat scores measured by image analysis and beef marbling score (BMS) between the 6th and $13^{th}$ rib of loin from the carcasses of $1^{{+}{+}}$ and $1^+$ and that were significantly higher than intramuscular fat scores at the $1^{st}$ rib and the last lumbar position of the loin. From the results, we conclude that the intramuscular fat score at the $13^{th}$ rib can represent the fat content of loin when meat graders decide the grade of Hanwoo carcass.

A survey of the relation between fatty lesions after slaughter inspection results and cattle carcasses quality grades in Korea (도축검사 후 지방성 병변과 축산물 품질 연관성 조사)

  • The survey was conducted to investigate the relation between slaughter inspection results and the quality of cattle carcasses by inquiring animal products grading service results of Korea Institute for Animal Products Quality Evaluation (KAPE) about partial-disposed cattle carcasses after dismantling inspection. Of the 13,586 slaughtered cattle, 1,343 (9.9%) cattle heads were partially disposed from January to August, 2012 at slaughterhouses in Gwangju area, Korea. Of the 1,343 partial disposed cattle, 613 (45.6%) cattle carcasses were graded into above "Grade 1". Among partially disposed 1,171 Hanwoo heads, 613 (52.3%) cattle carcasses were graded into above "Grade 1". However, none of dairy and beef cattle carcasses were graded into above "Grade 1" out of 163 and 9 partial disposals, respectively. Of the 221 partial disposals by enteric fat necrosis, 139 carcasses were graded into above "Grade 1", and the ratio; 62.9% (P=0.001), was higher than that (50.3%) of total cattle carcass grade which is above "Grade 1". From these results, it is suggested that quality of cattle carcasses which is mainly determined by marbling might be related to the fatty lesions such as fatty liver and enteric fat necrosis.

INFLUENCE OF DIETARY ENERGY AND POSTMORTEM ELECTRICAL STIMULATION ON MEAT QUALITY AND COLLAGEN CHARACTERISTICS OF LAMB CARCASSES

  • Sixty ram lambs, weighting 23.5 kg, were randomly assigned in a $2{\times}3$ factorial arrangement of two dietary energy (high; 11.7 and low; 9.0 MJ ME/kg DM) and three levels of poultry offal meal supplementation (0, 5 and 10%). Lambs were fed ad libitum for 120-day before slaughter. At slaughter, half the lambs in each dietary treatment group were randomly selected for electrical stimulation of their undressed carcasses. The M. Biceps femoris pH and temperatures were monitored at 1, 3, 5, 8 and 24 h postmortem. At 24 h postmortem, the M. biceps femoris was removed from the fight side of each carcass and steaks were obtained for determination of Warner-Bratzler shear force, collagen content and collagen solubility. The results showed that temperature and pH values during the 24-h postmortem were consistently higher (p < .01) and lover (p < .01), respectively, for M. biceps femoris from lambs fed high energy diets than for those fed on low energy diets. Muscles from high energy fed lambs had lower (p < .01) shear force values and higher (p < .01) percent soluble collagen than for low energy fed lambs; total collagen content was not significantly influenced by dietary energy level. Increased the level of poultry offal meal supplementation in the diet to 10% was associated with concomitant increases (p < .01) in muscle tenderness and percent soluble collagen. Electrical stimulation (ES) of carcasses resulted in a lower shear force values for the M. biceps femoris than in non-stimulated carcasses (Non-ES); total collagen content and percent soluble collagen were not significantly affected by ES treatment.

Prevalence and characterization of pathogenic Escherichia coli from carcasses and environmental samples of cattle and pig slaughterhouses (소와 돼지 도축장 도체 및 환경에서 분리된 병원성대장균 분포 및 특성조사)

  • We investigated the virulence genes, O-serotypes, antimicrobial resistance of pathogenic E. coli isolated from carcasses (n=455) and environmental (n=372) samples of 11 cattle and 12 pig slaughterhouses from December 2020 to December 2021. E. coli were isolated from nine carcasses (2.0%), three slaughter facilities (1.4%), two utensils (2.7%) and three abattoir workers (3.5%) from four cattle and four pig slaughterhouses. Among all isolates, 13 STEC (76.5%) were identified, followed by four EPEC (23.5%). As a result of the antibiotic susceptibility test, all isolates were resistant to at least one antimicrobial, of which 70.6% isolates showed multidrug resistance patterns. The serotypes were diverse in pigs compared to cattle, with serotypes O18, O66, O109 in cattle and O9, O76, O85, O100, O153, and O159 in pigs. In a single cattle slaughterhouse, eight STEC O66 were isolated from various types of sample (4 slaughter animal surfaces, 3 gloves, and 1 knife) with two antimicrobial resistance patterns (CHL-FIS-STR and CHL-FIS). Those two types of strain were suspected cross-contamination from utensils to slaughter animal surfaces. These results showed that pathogenic E. coli were detected in carcasses and various environmental samples in cattle and pig slaughterhouses. Nationwide monitoring and hygiene management are required to prevent cross-contamination of STEC isolate slaughterhouses.

Chemical Components and Meat Quality Traits Related to Palatability of Ten Primal Cuts from Hanwoo Carcasses

  • To determine chemical components and meat quality traits related to palatability of 10 primal cuts, 25 Hanwoo carcasses were selected from 5 carcasses × 5 quality grades and used to obtain proximate data and meat quality characteristics. Significant differences (p<0.05) in chemical component and meat quality were found among the 10 primal cuts. The highest fat content was found in the kalbi, followed by dungsim, yangjee, chaekeut, ansim, abdari, suldo, moksim, udun, and satae. Protein and moisture contents in the 10 primal cuts were in reverse order of fat content. Moksim had the highest drip loss % and cooking loss % than all other primal cuts while kalbi showed the lowest (p<0.05) percentage of drip and cooking loss. Ansim had the longest sarcomere length but the lowest shear force values than all other cuts (p<0.05). The highest (p<0.05) score for overall acceptability was observed in ansim. Moksim, udun, abdari, and satae were rated the lowest (p<0.05) in overall acceptability among the 10 primal cuts from Hanwoo carcasses. In conclusion, ansim, dungsim, chaekeut, and kalbi had the highest overall acceptability due to their higher fat contents and lower shear force values.

Isolation of Listeria monocytogenes from animal carcasses and environmental specimens in slaughter house level (도축처리 단계별 도체 및 환경재료에서 Listeria monocytogenes의 분리)

  • To invastigate the epidemiological trait of listeriosis, Listeria monocytogenes were isolated from the carcasses of pigs and cattle, and environmental specimens in slaughter house. Also serotype of isolates were classified by rapid slide agglutination test. In the carcasses of pigs, Listeria sp were isolated from the carcasses after bleeding(62%), after dismemberment(60.0%) and before shipping(76.0%), and L monocytogenes were present in 8% of the carcasses after dismemberment and in 14% of the carcasses before shipping. However, few Listeria sp were isolated from the living body skin and the carcasses after scalding. In the carcasses of cattle, Listeria sp were isolated from the carcasses after bleeding(10%), after dismemberment(36.7%) and before shipping(63.3%), L monocytogenes were present in 3.3% of the carcasses after dismemberment and in 10% of the carcasses before shipping. Overall, L monocytogenes, L innocua, L welshimen, L grayi, and L murrayi were present In 4.8, 40, 2.3, 2.6 and 0.3% of all the carcasses, respectively. Prevalence of Listeria sp in environmental specimens were found to be 80% in slaughter house floors and 100% in sewage, and L monocytogenes were present in 15% of sewage. However, few Listeria sp were isolated from chilled water and from scalding water. Overall, L monocytogenes, L innocua, and L welshimeri were present in 3.8, 45 and 6.3% of all the environmental specimens, respectively. A total 27 strains of L monocytogenes were isolated from samples tested and all of the strains were classified into serotype 1.

Characteristics and Cutability of Farmed Rusa Deer (Cervus timorensis) Carcasses for Marketing of Venison

  • Rusa deer are the only viable commercial tropical deer species for farming in the tropics because of their hardiness, adaptability and prolific characteristics. Twelve entire rusa stags were slaughtered according to halal procedure and used for carcass evaluation and cutability studies. Three carcass categories; large (46 to 55 kg), medium (36 to 45 kg) and small (25 to 35 kg) were developed for rusa stags. This study indicated that entire Moluccan rusa deer stags with mean live weight ranges from 50 to 80 kg and age groups of 15 to 29 months showed dressing percentage of 58 to 62%. Carcass conformation of rusa stags showed significantly (p<0.05) higher forequarters portion than hindquarters for medium (mean weight, 40.8 kg) and large (mean weight, 50.0 kg) carcass categories. The large carcass category was significantly (p<0.05) bigger in hindquarters portion (47.3% vs. 45.4%) than the medium carcass category. This study showed that medium and large carcass categories are more suitable for boneless cuts since the muscles were larger than small carcasses and easy for deboning. Stag carcasses showed higher proportion of the musculature in the high-priced areas of the carcass such as in round cuts. Bone-in cuts are more suitable for small carcasses since the muscles were smaller and difficult to debone (mean weight, 30.5 kg). About 90% of total deer carcasses in the small carcass category were developed into retail bone-in cuts (excluding 9.9% of breast, shank and trimming). The carcass characteristics and cutability information derived from this study can be used as a basis for a venison marketing strategy for deer farming and production in the tropics.

Effects of Different Levels of Concentrate in the Diet on Physicochemical Traits of Korean Native Black Goat Meats

  • The effects of feeding of diets based on roughage supplemented with concentrate on the carcass and meat quality attributes of Korean black goats were investigated. The 40 male goats at 5 mon age were divided into four treatment groups; T1 (1.5%), T2 (2.0%), T3 (2.5%) and T4 (ad libitum) feeding of concentrate with ad libitum rice straw. Forty bucks at the end of the feeding were fasted for 12 h before slaughter and were then slaughtered. For carcass measurement, cold buck carcasses were examined after 24 h of chilling at $5^{\circ}C$. The carcasses were chilled in refrigerator at $2{\pm}2^{\circ}C$ for 24 h, then the longissimus dorsi muscle was removed from the carcasses. The samples were separately vacuum packaged, frozen and stored at $-20^{\circ}C$ for up to 1 mon, prior to physicochemical evaluations. Carcass yields were greatly affected by rate of concentrate feeding. T4 showed the heaviest live and carcass weights and the highest dressing and fat percentages (p<0.05). The moisture contents were lower in T4 compared with other treatments, while the crude protein and fat contents were higher (p<0.05). mono-unsaturated fatty acid in T4 was significantly higher than others, which showed the highest percentages of oleic acid (C18:1). For the sensory test, T4 was tenderer and produced better flavor scores than others. Increasing the level of concentrate in the diet resulted in an improvement in growth performance, and carcass and physicochemical characteristics of goat meats.
