Protective Effects of Angelica koreana on Experimentally Induced Liver Injury (실험적 간장해에 대한 강활의 보호효과)

  • The present study was carried out to find the possible protective effects of Angelica koreana water extract on biochemical parameters in benzo(a)pyrene (B(a)P)-induced liver injury in rats. B(a)P treatment (0.1 mg/kg, 1.p.) caused a liver damages, which led to biochemical alterations in serum and liver enzyme activities and serum lipid levels. The activities of liver marker enzymes, especially, aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP) and lactate dehydrogenase (LDH) were markedly changed in B(a)P treatment. Oral administration of Angelica koreana (50 mg/kg) recovered these biochemical Parameters to near normal levels. Therefore, the present results have revealed that Angelica koreana water extract might have the antihepatotoxic effect and consequently ameliorate liver damage associated with B(a)P in rats.

  • Silica gel column chromatography on the ether extract from the fruits of Angelica koreana $M_{AXIMOWICZ}$ gave two kinds of crystalline products of coumarin. The following furocoumarins-imperatorin and bergapten has been identified by elemental analysis, IR, NMR spectra and physico-chemical tests.

  • The root of Angelica koreana $M_{AXIMOWICZ}$ (Umbelliferae) known as "Kang-whal" in Korea used for the treatment of common cold, headache and gout etc. The comparative anatomy of Angelica Koreana $M_{AXIMOWICZ}$ and A. purpuraefolia $C_{HUNG}$ was presented. A great part of the report was dealed with the differences in the comparative histology of root, stem, main vein in leaf and fruits.

Falcarindiol from Angelica koreana Down-regulated IL-8 and Up-regulated IL-10 in Colon Epithelial Cells

  • Angelica koreana is an important medicinal plant for some locals in East Asia including Korea. A few reports have shown the efficacy of its phytochemical constituents. We have isolated and purified one compound falcarindiol (FAL) from the methanolic extract of A. koreana roots. At concentrations from to $1{\mu}M$ to $25{\mu}M$, the FAL isolated from the roots of A. koreana exerted no significant cytotoxicity and down-regulated LPS-stimulated pro-inflammatory cytokine IL-8 in colon epithelial cells, while up-regulating anti-inflammatory cytokine IL-10. In addition, the FAL decreased the expression of LPS-induced inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 protein by Western blot analysis. Colon epithelial cells play pivotal roles in regulating the colon immune system and thus FAL is expected to be candidate agent as therapeutic potential for the treatment of inflammatory bowel disease (IBD) by modulating LPS-induced inflammation in colon epithelial cells.

  • Silica gel column chromatography of the ether extract of the root of Angelica Koreana $M_{AXIMOWICZ}$ (Umbelliferae) gave five kinds of crystalline products of coumarin. The following kinds of furocoumarins were identified by UV, IR, NMR spectra and physico-chemical tests. iso-imperatorin m.p. 108-$109^{\circ}$ $C_{16}H_{14}O_{4}$ oxypeucedanin m.p. 142-$143^{\circ}$ $C_{16}H_{14}O_{5}$ prangolarine m.p. 104-$105^{\circ}$ $C_{16}H_{14}O_{5}$ imperatorin m.p. 100-$101^{\circ}$ $C_{16}H_{14}O_{4}$ These analyses also showed that white needles m.p. 129-$130^{\circ}$ were likely to be a furocoumarin. Besides, the methanol extract of the root was found to contain sucrose.

  • A new species of Apiaceae, Angelica reflexa from Daeamsan, Gangwon-do province, Korea is described and illustrated. This new species is closely related to A. genuflexa Nutt. ex Torr. & A. Gray, having such characters as leaf rachis and petiolules refracted strongly, but distinguished from the latter by its regularly serrated leaf margins, glabrous on abaxial leaf veins, and two vittae on the commissural face of the mericarp.

  • Prostaglandins biosynthesis and nitric oxide production have been implicated in the process of inflammation. In this study we investigated on the effects of ethyl acetate extract of Angelica Koreana Radix (EAKR) on the activities of prostaglandin $E_2\;(PGE_2)$, proteoglycan (PG) degradation and nitric oxide synthase (NO) in inflammation cytokines-activated rabbit articular chondrocytes. EAKR exhibited inhibitory activities on NO production and $PGE_2$ production as $73.08\%\;and\;89.49\%$, respectively at $20{\mu}g/ml$ and inhibited the degradation of PG in a concentration-dependent manner Zelatin zymography analysis demonstrated that EAKR significantly inhibited MMP-2, 9 expression in chondrocytes. In vivo, EAKR was shown to have inhibitory effects on acetic acid-induced pain. This study suggests that modulation of $PGE_2$, NO, PG degradation and MMP-2, 9 by EARK may be important in the prevention of inflammation and osteroarthritis.

Effects of Methanolic Extract of Angelicae koreana Radix against Benzo$(\alpha)pyrene$ Induced Liver Injury in Rats (Benzo$(\alpha)pyrene$에 의해 유도된 흰쥐의 간 장해에 미치는 강활 Methanol 추출물의 효과)

  • To evaluate the preventive effects of methanolic extract of Angelica koreana(MEAK), this extract was given to rats orally at various doses of 10, 50 and 100 mg/kg before hepatotoxicant, benzo$(\alpha)pyrene$ treatment The increased serum enzyme levels of aspartate aminotransferase (AST), alanine aminotransferase(ALT) and alkaline phosphatase(ALP) by benzo$(\alpha)pyrene$ induction were significantly lowered in a dose dependent manner after pretreatment with MEAK. Furthermore, MEAK also decreased the elevated lipid levels after benzo$(\alpha)pyrene$ administration. These results revealed that MEAK could afford a significant protective action in the alleviation of benzoBenzo$(\alpha)pyrene$ induced hepatocellular injury.
