Development of an Immobilized Adsorbent for In Situ Removal of Ammonium Ion from Animal Cell Culture Media and Its Applications to Animal Cell Culture System : II. Application to Cell Culture System (동물세포 배양액으로부터 암모늄 이온의 동시제거를 위한 고정화 흡착제의 개발과 동물세포 배양 시스템에의 응용 : II. 세포배양 시스템에의 응용)

  • The possibility of application of membrane type immobilized adsorbent to the fed-batch or perfusion culture system with anchorage-independent cells as well as batch system was investigated. The improvement in cell density and cell viability due to the combination of immobilized adsorbent with each culture system was evaluated for the investigation, and the optimum culture system employing immobilized adsorbent system was suggested based on the results. It was observed that the system with immobilized adsorbent showed better cell growth and cell viability than that without immobilized adsorbent in every operation system of batch, fed-batch, and perfusion. In case of batch system, 200% improvement of maximum cell density was observed in the system where ammonium chloride was added on purpose. And 50% improvement of maximum cell density was observed in the fed-batch system where ammonium ion accumulates significantly, while small increase in maximum cell density was observed in the perfusion system where dilution of waste byproducts exists. Especially, the fed-batch system showed the most significant improvement on cell growth because both compensation of nutrient and removal of ammonium ion occurred simultaneously in the system. Therefore a combined system of immobilized adsorbent and fed-batch operation could be suggested as an optimum system with in situ removal of ammonium ion.

Applicability of Partial Post-Tension Method for Deflection Control of Reinforced Concrete Slabs (RC슬래브의 처짐제어를 위한 상향긴장식 부분PT공법의 적용)

  • Recently, it is getting into a good situation for the flat-plate slab system to be applied. The flat-plate slab without beam, however, is often too weak to control deflection properly compared to other typical slab-beam structures, for which the post-tension method is generally regarded as one of best solutions. The post-tension (PT) method can effectively control deflection without increase of slab thickness. Despite this good advantage, however, the application of PT method has been very limited due to cost increase, technical problems, and lack of experiences. Therefore, in order to reduce difficulties on applying full PT method under the current domestic circumstances and to enhance constructability of PT system, this research proposed the partial PT method with top jacking anchorage applied in a part of span as need. For the top jacking anchorage system, the efficiency of deflection control shall be considered in detail because it can vary widely depending on the location of anchorage that can be placed anywhere as need, and tensile stresses induced at back of the anchorage zone also shall be examined. Therefore, in this study, analysis were performed on the efficiency of deflection control depending on the location of anchorage and on tensile stresses or forces using finite element method and strut and tie model in the proposed top jacking anchorage system. The proposed jacking system were also applied to the floor slabs at a construction site to investigate its applicability and the analysis results of slab behavior were compared to the measured values obtained from the PT slab constructed by the partial PT method. The result of this study indicates that the partial PT method can be very efficiently applied with little cost increase to control deflection and tensile stresses in the region as a need basis where problem exists.

  • A ground anchor system is used as a load carrying element for soil structure stability The conventional systems with ground anchors bring about the anchorage loss of wedges when anchors are installed for the support of soil structures. Hence we developed the new type of anchor system using both the spacing apparatus and spring (length 60mm, diameter 6mm). In this system, we can directly check the condition of wedges and PS strands and modify the problems with the slip and anchorage of wedges under construction. For demonstrating the superiority of this system, we carried out a series of both laboratory and field test. Consequently, we can obtain satisfactory result (18.99% reduction to the loss of conventional systems). Moreover, the replacement of wedges is easy and simple when retensioning of strands.

Evaluation of Mechanical Joint Structural Performance through Actual Performance Testing of PC Connections (PC 접합부의 실물 성능실험을 통한 기계식이음 구조성능 평가)

  • In this study, the SBC system, a new mechanical joint method, was developed to improve the constructability of precast concrete (PC) beam-column connections. The reliability of the finite element analysis model was verified through the comparison of experimental results and FEM analysis results. Recently, the intermediate moment frame, a seismic force resistance system, has served as a ramen structure that resists seismic force through beams and columns and has few load-bearing walls, so it is increasingly being applied to PC warehouses and PC factories with high loads and long spans. However, looking at the existing PC beam-column anchorage details, the wire, strand, and lower main bar are overlapped with the anchorage rebar at the end, so they do not satisfy the joint and anchorage requirements for reinforcing bars (KDS 41 17 00 9.3). Therefore, a mechanical joint method (SBC) was developed to meet the relevant standards and improve constructability. Tensile and bending experiments were conducted to examine structural performance, and a finite element analysis model was created. The load-displacement curve and failure pattern confirmed that both the experimental and analysis results were similar, and it was verified that a reliable finite element analysis model was built. In addition, bending tests showed that the larger the thickness of the bolt joint surface of the SBC, the better its structural performance. It was also determined that the system could improve energy dissipation ability and ductility through buckling and yielding occurring in the SBC.

Ductility of carbon fiber-reinforced polymer (CFRP) strengthened reinforced concrete beams: Experimental investigation

  • Strength of reinforced concrete beams can easily be increased by the use of externally bonded CFRP composites. However, the mode of failure of CFRP strengthened beam is usually brittle due to tension-shear failure in the concrete substrate or bond failure near the CFRP-Concrete interface. In order to improve the ductility of CFRP strengthened concrete beams, critical variables need to be investigated. This experimental and analytical research focused on a series of reinforced concrete beams strengthened with CFRP composites to enhance the flexural capacity and ductility. The main variables were the amount of CFRP composites, the amount of longitudinal and shear reinforcement, and the effect of CFRP end diagonal anchorage system. Sixteen full-scale beams were investigated. A new design guideline was proposed according to the effects of the above-mentioned variables. The experimental and analytical results were found to be in good agreement.

Orthodontic treatment of gummy smile by maxillary total intrusion with a midpalatal absolute anchorage system

  • This article describes the orthodontic treatment of a 31-year-old Korean female patient with gummy smile and crowding. The patient showed excessive gingival display in both the anterior and posterior areas and a large difference in gingival heights between the anterior and posterior teeth in the maxilla. To correct the gummy smile, we elected to intrude the entire maxillary dentition instead of focusing only on the maxillary anterior teeth. Alignment and leveling were performed, and a midpalatal absolute anchorage system as well as a modified lingual arch was designed to achieve posterosuperior movement of the entire upper dentition. The active treatment period was 18 months. The gummy smile and crowding were corrected, and the results were stable at 21 months post-treatment.

Seismic Performance Improvement of Concrete Gravity Dam by Post-tensioned Anchors (앵커공법을 적용한 기존 콘크리트 중력식 댐의 내진성능 보강방안)

  • This paper describes the assessment of seismic performance of the concrete gravity dam seismically reinforced by post-tensioned anchors. In order to evaluate the seismic performance, the response spectrum analyses have been carried out for 7 different configurations of the post-tensioned anchors, and then their performance improvement in the maximum tensile and compressive stresses is compared to each other. The comparative results demonstrate that the layout of the post-tensioned anchors strongly influences the seismic performance of the concrete gravity dam. In this study, the slightly-inclined vertical anchorage system shows the largest improvement on the overall performance of the seismically-excited concrete gravity dam.

  • Prestressed concrete I girder bridge has been one of the most widely used bridges in the world because of its excellent construction feasibility, economic efficiency, serviceability, and safety. But in Korea, the PSC bridge has not been utilized for long span because of high girder height in its standard design. Thus, the results confirm that it is possible to applicate the continuous PSC girder with end cross beam anchorage system using multi-stage prestressing technique.

Flexural Behavior and Analysis of RC Beams Strengthened with Prestressed CFRP Plates (프리스트레스트 탄소섬유판으로 보강된 철근콘크리트 보의 휨 거동 및 해석)

  • In this paper, a total of 13 beams with bonding, anchorage system, amount of prestressing and span length as variables of experiment were tested in flexural test and analyzed in finite element analysis; one control beam, two simplified FRP-boned beams, four prestressed FRP-unbonded beams and four prestressed FRP-bonded beams. Also, a nonlinear finite element analysis of beams in the flexural test is performed by DIANA program considered material nonlinear of concrete, reinforcement and the interfacial bond-slip model between concrete and CFRP plates. The failure mode of prestressed CFRP plated-beams is not debonding but FRP rupture. RC members strengthened with external bonded prestressed CFRP plates occurred 1st and 2nd debonding of the composite material. After the debonding of CFRP plates occurs in bonded system, behavior of bonded CFRP-plated beams change into that of unbonded CFRP-plated beams due to fix of the anchorage system. Also, It was compared flexural test results and analytical results of RC members strengthened with CFRF plates. The ductility of beams strengthened by CFRP plates with the anchorage system is considered high with the ductility index of above 3. Analysis results showed a good agreement with experiment results in the debonding load, yield load and ultimate load.
