Recently, one of the most interesting topic in the field of wastewater treatment is the disposal of excess sludge. The new concept of excess sludge reduction with recirculation of solubilized sludge via effective microorganisms for cell disruption within the wastewater treatment process has been developed in this study. The alkalophiles for degradation of sludge cell wall were isolated as Exiguobacterium sp., which could be more effectively solubilized sludge in the anaerobic condition. The SCOD of solubilized excess sludge by Exiguobacterium sp. was up to about 2,000mg/L and average TN and TP concentration of solubilized component were 117mg/L and 58mg/L, respectively and C/N ratio was more than 17. To investigate the effects of solubilized sludge by alkalophiles on excess sludge reduction and nutrient removal efficiency, the pilot plant of $DF^{(S)}-MBR$ process, combined with membrane bioreactor and sludge solubilization tank, was operated. In the control run(without sludge solubilization), the daily sludge production was about 4.54 kgMLVSS/day. However, in the $DF^{(S)}-MBR$(with sludge solubilization), the daily sludge production was decreased to 1.39kgMLVSS/day. The effluent quality satisfied the effluent regulation in both cases. Furthermore, the $DF^{(S)}-MBR$ showed relatively better TN removal efficiency in spite of high influent loading. So we concluded that the solubilized excess sludge by alkalophiles was effectively degraded in the MBR process as the carbon source and 70% of sludge reduction efficiency can be achieved.
Myocardial protection against ischemic and reperfusion injuries is still in troublesome eventhough couples of the way of myocardial protection have been applied since 1970's. One of the possibility in myocardial protection is adding Fructose-l,6-diphosphate(FDP) in cardioplegic solution. It is assumed that FDP can promote ATP production under anaerobic condition as well as inhibiting the supressing effect of lactate on phosphofructokinase. We compared the myocardial protecting effects of FDP in crystalloid cardioplegic solution (St. Thomas formula, 10$^{\circ}C$, pH = 7.4) and reperfusate using isolated rat hearts in modified Langendorf apparatus by the parameters of preischemic and post reperfusing heart rate, time to first beat, occurance of arrhythmia, time to stabilization, and the rate of left ventricular pressure developing. Group A (n = 10), containing no FDP in cardioplegic and reperfusing solutions was control. Group B (n = 5), containing FDP in cardioplegic solution, showed statistically significant superiority of postischemic left ventricular pressure development than the control group. Group C (n = 5), containing FDP in reperfusate, showed statistically significant myocardial depressing effect than the controls. Other parameters were unremarkable. The cause is uncertain, but it is assumed that the negative feedback inhibition of FDP in energy metabolism or unknown blocking effect of FDP on certain transmembrane ionic currents is present. In conclusion, 1) FDP in cardioplegic solution has beneficial effect on postischemic left ventricular preservation. 2) FDP is strong acid when is hydrolyzed, so precise acid titration is neccessary. 3) FDP in reperfusate has negative left ventricular preservation, otherwise the mechanism is still uncertain.

In order to provide recommendations on the most useful forage species to smallholder farmers, eleven grass and eleven legume forages grown in Abomey-Calavi in Republic of Benin were investigated for nutritive value (i.e. chemical composition and energy content) and fermentation characteristics (i.e. gas and volatile fatty acid production, organic matter degradability). The in vitro gas production technique was used, incubating the forages for 120 h under anaerobic condition with buffalo rumen fluid. Compared to legume, tropical grass forages showed lower energy (8.07 vs 10.57 MJ/kg dry matter [DM]) and crude protein level (16.10% vs 19.91% DM) and higher cell wall content (neutral detergent fiber: 63.8% vs 40.45% DM), respectively. In grass forages, the chemical composition showed a quite high crude protein content; the in vitro degradability was slightly lower than the range of tropical pasture. The woody legumes were richer in protein and energy and lower in structural carbohydrates than herbaceous plants, however, their in vitro results are influenced by the presence of complex compounds (i.e. tannins). Significant correlations were found between chemical composition and in vitro fermentation characteristics. The in vitro gas production method appears to be a suitable technique for the evaluation of the nutritive value of forages in developing countries.
Under sulfur deprived conditions, PS II and photosynthetic $O_2$ evolution by Chlamydomonas reinhardtii UTEX 90 are inactivated, resulting in shift from aerobic to anaerobic condition. This is followed by hydrogen production catalyzed by hydrogenase. We hypothesized that the photosynthetic capacity and the accumulation of endogenous substrates such as starch for hydrogen production might be different according to cell age. Accordingly, we investigated (a) the relationships between hydrogen production, induction time of sulfur deprivation, increase of chlorophyll after sulfur deprivation, and residual PS II activity, and (b) the effect of initial cell density upon sulfur deprivation. The maximum production volume of hydrogen was 151 ml $H_2$/l with 0.91 g/l of cell density in the late-exponential phase. We suggest that the effects of induction time and initial cell density at sulfur deprivation on hydrogen production, up to an optimal concentration, are due to an increase of chlorophyll under sulfur deprivation.
The inulinase gene (INU1) from Kluyveromyces marxianus NCYC2887 was overexpressed by using the GAL10 promotor in a ${\Delta}ga180$ strain of Saccharomyces cerevisiae. The inulinase gene lacking the original signal sequence was fused in-frame to a mating factor ${\alpha}$ signal sequence for secretory expression. Use of the ${\Delta}ga180$ strain allowed for the galactose-free induction of inulinase expression using a glucose-only medium. Shake-flask cultivation in YPD medium produced 34.6 U/ml of the recombinant inulinase, which was approximately 13-fold higher than that produced by K. marxianus NCYC2887. It was found that the use of the ${\Delta}ga180$ strain improved the expression of inulinase in the recombinant S. cerevisiae in both aerobic and anaerobic conditions by about 2.9- and 1.7-fold, respectively. A 5-l fed-batch fermentation using YPD medium was performed under aerobic condition with glucose feeding, which resulted in the inulinase production of 31.7 U/ml at the $OD_{600}$ of 67. Ethanol fermentation of dried powder of Jerusalem artichoke, an inulin-rich biomass, was also performed using the recombinant S. cerevisiae expressing INU1 and K. marxianus NCYC2887. Fermentation in a 5-l scale fermentor was carried out at an aeration rate of 0.2 vvm, an agitation rate of 300 rpm, and with the pH controlled at 5.0. The temperature was maintained at $30^{\circ}C$ and $37^{\circ}C$, respectively, for the recombinant S. cerevisiae and K. marxianus. The maximum productivities of ethanol were 59.0 and 53.5 g/l, respectively.
The activated paper mill sludge was treated with WF and some additives (sodium sulfide, nickel nitrate, ethyl acetate) for methane fermentation at $35^{\circ}C$. Optmum C/N ratio was 60 out of three conditions (20, 30 and 60). The Period of 40% of methane content possibly ignition, was 2 days shorter than with non-treatment during 10 days. Nevertheless, the total amount of methane production showed the 1/8 level of control far the same period. The yield and content of methane were increased by the addition of sodium Sulfur and ethyl acetate. Sulfur was an essential factor in methane fermentation of paper mill sludge.

This study examined the feasibility of phosphorus removal from dewatering centrate/filtrate in wastewater treatment by struvite formation. Since dewatering centrate/filtrate obtained after anaerobic digestion contains a high content of ammonia, magesium addition and pH adjustment could remove phosphorous. The optimum magnesium dose, reaction time, settling time, and pH value was found at 2 mol Mg/mol $PO_4^{3-}$, 10 min, 120 min, and 8.5, respectively. A bench-scale continuous operation at the optimum condition showed 80% of total phosphorus removal and 82% of phosphate removal in dewatering centrate. The elemental composition and crystal structure of the chemical precipitate was similar to the reported values of struvite.

The objective of this study was to examine the hydrogeochemical and microbiological characteristics for stable maintenance of coastal LPG storage cavern. Cavern seepage water and groundwater in the vicinity of the LPG storage cavern were sampled and analyzed (February, May, August and October 2007). Groundwater samples of propane cavern showed high pHs of 8.1~12.4 due to dissolution of cement grouting materials that had been applied during the well and cavern construction. The EC values showed distinctive seasonal variation. The negative ORP indicated reduced condition. The seepage and surrounding groundwater are classified as Na-Cl type, which represents seawater effects. All of aerobic bacteria, anaerobic bacteria, slime forming bacteria and sulfate reducing bacteria were less than 500 CFU/mL, which indicated that there was no abrupt increase of bacteria in the cavern. Therefore there exist no hydrochemical symptoms to indicate unstable conditions of the cavern operation. However, regular and continuous monitoring is essentially required.

In order to improve applicability of a microbial fuel cell the laboratory-scaled study has been performed by adopting an air-cathode MFC system with high concentrated anaerobic slugies in this study. The concentrations of microbes are grouped into three types, Type A (TS 1.7%), Type B (TS 1.1%) and Type C (TS 0.51%). The open circuit voltage $(V_{oc})$ characteristics showed that the medium microbes concentration of 1.10% (Type B) kept a constant voltage of 1.0 V for 150 hours, which showed the longest time among three types (Type A and Type C). The discharge charge curves for a closed circuit with $500 \Omega$ also showed that Type B generated a stable discharge voltage of 0.8 V for a longer time as in the open circuit voltage case. This could be explained by the relatively large amount of the attached microbes. Under the $V_{oc}$condition the COD removal efficiency of Type B was found to be low for a long time, but those of Type A and C were found to be high for a short period of time. Therefore, the suspended microbes could decrease the coulombic efficiency. It was concluded that the high $V_{oc}$ was caused by low COD and the $V_{oc}$ became low after the COD removal. The COD reduction resulted in an unstable and low working voltage. From the polarization characteristics Type A was found to show the highest power density of $193\;mW/m^2$ with a fill factor of 0.127 due to the relatively high remaining COD even after the MFC reaction.

The objective of this study was to evaluate several types of uterine bacteria in Hanwoo. uterine bacteria from randomly selected 5 uterus was collected by flushing methods into a sterilized 1.5 ml centrifuge tube and was inoculated onto MacConkey agar and blood agar, respectively. After being incubated for 5% $CO_2$, aerobic or anaerobic condition at $37^{\circ}C$ during 48h, bacterial colonies were selected and re-inoculated onto blood agar plates. Re-cultured colonies were identified by Gram staining and finally identified using Vitek system. The identified bacteria were Staphylococcus lentus, Staphylococcus sciuri, Staphylococcus vitulinus, Staphylococcus warneri of Gram (+) and Rhizobium radiobacter, Sphingomonas paucimobilis of Gram (-) bacteria. Although, pathogenicity of identified bacteria was unclear, the bacteria can have an effect on the uterine microenvironment. Therefore, repetitive research will be required to determine the effects of bacteria in cattle exposed to a various environment.
