Effects of Pinitol Supplementation and Strength Training on Anaerobic Performance and Status of Energy Substrates in Healthy Young Men

  • To assess the effect of pinitol supplementation and strength training for two weeks on the anaerobic capacity during and after exercise, and improvement of glucose metabolism during the recovery period of muscular fatigue with repeated acute bouts of cycling exercise, a total of 24 healthy young men were recruited and randomly and equally divided into three groups; pinitol supplementation group (PSG), placebo group (PLG), and control group (CON). Using a randomized double-blinded design, subjects in PSG were provided pinitol supplement, consumed orally 1.2 g/day, and participated in the resistance exercise program and cycling exercise for two weeks. Subjects in PLG underwent the same protocol as those in PSG but consumed the same amount of placebo. No supplementation and exercise program was given to CON. Before and after the intervention, all subjects were tested for their anaerobic capacities evaluated by Wingate test twice separated by 30 min. During the test, peak anaerobic power (PP), mean anaerobic power, total work, and fatigue index were evaluated During resting and recovery, blood samples were drawn and plasma pinitol, myo-inositol, chiro-inositol, insulin, free fatty acid, glucose, and lactate levels were analyzed After two weeks, PP and relative PP of the second biking were improved from the first biking in PSG only (p<0.05). No changes were found in all other variables of Wingate test in all groups. No statistical differences between groups and pre- and post-intervention were observed in concentrations of pinitol, myo-inositol, and chiro-inositol, but pinitol concentration was higher during recovery compared to the baseline in all groups and testings (p<0.05). Lactate level during recovery was higher than the resting level, but no other blood parameters were significantly changed. In conclusion, two weeks of pinitol supplementation in conjunction with short duration of anaerobic training in healthy young men did not induce any obvious benefits in terms of anaerobic capacity and energy metabolism Individual and/or population susceptibility may be one factor responsible for adopting pinitol supplementation.

Computational identification of significantly regulated metabolic reactions by integration of data on enzyme activity and gene expression

  • The concentrations and catalytic activities of enzymes control metabolic rates. Previous studies have focused on enzyme concentrations because there are no genome-wide techniques used for the measurement of enzyme activity. We propose a method for evaluating the significance of enzyme activity by integrating metabolic network topologies and genome-wide microarray gene expression profiles. We quantified the enzymatic activity of reactions and report the 388 significant reactions in five perturbation datasets. For the 388 enzymatic reactions, we identified 70 that were significantly regulated (P-value < 0.001). Thirty-one of these reactions were part of anaerobic metabolism, 23 were part of low-pH aerobic metabolism, 8 were part of high-pH anaerobic metabolism, 3 were part of low-pH aerobic reactions, and 5 were part of high-pH anaerobic metabolism.

  • A clinical study was designed to evaluate myocardial metabolism during continuous cold blood cardioplegia [Group A, n=10] in comparison with continuous warm blood cardioplegia [Group B, n=10], in a prospective randomized manner. Myocardial metabolism was assessed in two ways: either by collecting blood from coronary sinus before and after cardiopulmonary bypass or by collecting blood from cardioplegic affluent and effluent simultaneously at the beginning and at the end of cardioplegia. The former samples were assayed for gas analysis, lactic acid and cardiac enzyme [CK, LDH, SGOT] and the latter for gas analysis and lactic acid as a maker of anaerobic metabolism. The results were as follows. 1] Myocardial metabolism was shown to be continued in the state of cardioplegia at lower temperature as evidenced by high oxygen extraction of cardioplegic solution in Group A. 2] Anaerobic metabolism occurring at lower temperature in spite of continuous cold blood cardioplegia can be significantly reduced by continuous perfusion of normothermic blood cardioplegics as evidenced by significant reduction of lactate production in Group B [p〈0.05]. 3] Better myocardial protection can be achieved by employing continuous warm blood cardioplegia as evidenced by less cardiac enzyme release in Group B after cardiopulmonary bypass.

Co-cultured methanogen improved the metabolism in the hydrogenosome of anaerobic fungus as revealed by gas chromatography-mass spectrometry analysis

  • Objective: The purpose of this study was to reveal the metabolic shift in the fungus cocultured with the methanogen (Methanobrevibacter thaueri). Methods: Gas chromatography-mass spectrometry was used to investigate the metabolites in anaerobic fungal (Pecoramyces sp. F1) cells and the supernatant. Results: A total of 104 and 102 metabolites were detected in the fungal cells and the supernatant, respectively. The partial least squares-discriminant analysis showed that the metabolite profiles in both the fungal cell and the supernatant were distinctly shifted when co-cultured with methanogen. Statistically, 16 and 30 metabolites were significantly (p<0.05) affected in the fungal cell and the supernatant, respectively by the co-cultured methanogen. Metabolic pathway analysis showed that co-culturing with methanogen reduced the production of lactate from pyruvate in the cytosol and increased metabolism in the hydrogenosomes of the anaerobic fungus. Citrate was accumulated in the cytosol of the fungus co-cultured with the methanogen. Conclusion: The co-culture of the anaerobic fungus and the methanogen is a good model for studying the microbial interaction between H2-producing and H2-utilizing microorganisms. However, metabolism in hydrogenosome needs to be further studied to gain better insight in the hydrogen transfer among microorganisms.

Cholera Toxin Production Induced upon Anaerobic Respiration is Suppressed by Glucose Fermentation in Vibrio cholerae

  • The causative agent of pandemic cholera, Vibrio cholerae, infects the anaerobic environment of the human intestine. Production of cholera toxin (CT), a major virulence factor of V. cholerae, is highly induced during anaerobic respiration with trimethylamine N-oxide (TMAO) as an alternative electron acceptor. However, the molecular mechanism of TMAO-stimulated CT production is not fully understood. Herein, we reveal that CT production during anaerobic TMAO respiration is affected by glucose fermentation. When the seventh pandemic V. cholerae O1 strain N16961 was grown with TMAO and additional glucose, CT production was markedly reduced. Furthermore, an N16961 Δcrp mutant, devoid of cyclic AMP receptor protein (CRP), was defective in CT production during growth by anaerobic TMAO respiration, further suggesting a role of glucose metabolism in regulating TMAO-mediated CT production. TMAO reductase activity was noticeably decreased when grown together with glucose or by mutation of the crp gene. A CRP binding region was identified in the promoter region of the torD gene, which encodes a structural subunit of the TMAO reductase. Gel shift assays further confirmed the binding of purified CRP to the torD promoter sequence. Together, our results suggest that the bacterial ability to respire using TMAO is controlled by CRP, whose activity is dependent on glucose availability. Our results reveal a novel mechanism for the regulation of major virulence factor production by V. cholerae under anaerobic growth conditions.

  • Iron is an important micronutrient for plants. Iron metabolism is a complex mechanism under a delicate balance. Iron metabolism represents two major problems for plants: deficiency as a consequence of solubility problems and toxicity due to excess solubility in anaerobic conditions. In the last few years, new genes have been discovered that influence iron uptake, transport and storage. Irrigated rice is exposed to high levels of $Fe^{II}$, normally rare in aerobic soil conditions. The implications of altering iron uptake rates and the effects of newly discovered genes are discussed.

Analysis of Functional Genes in Carbohydrate Metabolic Pathway of Anaerobic Rumen Fungus Neocallimastix frontalis PMA02

  • Anaerobic rumen fungi have been regarded as good genetic resources for enzyme production which might be useful for feed supplements, bio-energy production, bio-remediation and other industrial purposes. In this study, an expressed sequence tag (EST) library of the rumen anaerobic fungus Neocallimastix frontalis was constructed and functional genes from the EST library were analyzed to elucidate carbohydrate metabolism of anaerobic fungi. From 10,080 acquired clones, 9,569 clones with average size of 628 bp were selected for analysis. After the assembling process, 1,410 contigs were assembled and 1,369 sequences remained as singletons. 1,192 sequences were matched with proteins in the public data base with known function and 693 of them were matched with proteins isolated from fungi. One hundred and fifty four sequences were classified as genes related with biological process and 328 sequences were classified as genes related with cellular components. Most of the enzymes in the pathway of glucose metabolism were successfully isolated via construction of 10,080 ESTs. Four kinds of hemi-cellulase were isolated such as mannanase, xylose isomerase, xylan esterase, and xylanase. Five $\beta$-glucosidases with at least three different conserved domain structures were isolated. Ten cellulases with at least five different conserved domain structures were isolated. This is the first solid data supporting the expression of a multiple enzyme system in the fungus N. frontalis for polysaccharide hydrolysis.

횡격막에 적용된 테이핑 처치가 30초간의 무산소 운동수행력에 미치는 영향: pilot study (The effect of taping intervention applied to the diaphragm on the performance of anaerobic exercise in 30 seconds: pilot study)

  • 본 연구는 횡격막에 적용된 테이핑 처치가 30초간의 무산소 운동수행력에 미치는 영향을 알아보고자 실시하였다. 건강한 대학생 18명을 대상으로 실험군(KT)과 대조군(Sham)에 각각 9명씩 무작위 할당되었다. 두 그룹 모두 윙게이트 검사 기반의 사전 측정이 이루어지고 1주일 휴식 후 처치와 함께 사후 측정이 실시되었다. 두 집단간 변인들의 통계는 사전, 사후값의 변화율을 Mann-Whitney U검정으로 실시하였고, 그 결과 대조군의 평균 파워(AP)에서만 통계적으로 유의한 차이가 나타났다. 이는 횡격막 테이핑이 30초 동안의 무산소 운동 수행에 영향을 미치지 않았음을 의미한다. 하지만 30초보다 긴 장기간 무산소 운동에서 유산소 대사가 증가함을 고려할 때 이후 다양한 시간 범위 내 무산소 운동 수행에 대한 추가 연구가 필요할 것으로 생각된다.

  • Studies on the carbohydrate metabolism of yeast as influenced by gamma-irradiation from cobalt-60 have been carried, then the mechanisms of radiation effect on respiration and fermentation were discussed under considerations of permeable changes of irradiated cell membrane. The cells of baker's yeast (Saccharomyces cerevisiae) which had been gamma-irradiated of 240 k.r. doses for an hour, then were put into aerobic oxidation and anaerobic fermentation without substrate. Total and fractionated carbohydrates of irradiated yeast cells were determined by calorimetric method with anthrone and orcinol reagents, the amounts of total carbohydrate, trehalose, RNA-ribose, PCA-soluble glycogen, alkali-soluble glycogen, acetic acid-soluble glycogen, mannan and glucan were determined according to the course of aerobic oxidation and anaerobic fermentation. It is found that the carbohydrates of irradiated cells leak out and amount of the losses teaches eleven times more than that of control, the volume of losses are seems to be replaced by water, it can be suggested the damage of gamma-irradiation occurs in the site of passive transport of cell membrane. The endogeneous aerobic respiration of irradiated cells are increased much more than control, the synthesis of reserve glycogen, glucan and RNA-ribose promoted much more than control. The anaerobic fermentation of irradiated cells are also increased than that of control, but the breakdown of carbohydrate is less than endogeneous respiration of irradiated cells. The synthetic rate is also less than that of aerobic oxidation. In irradiated yeast cells, trehalose is revealed to be primary substrate for endogeneous carbohydrate metabolism, so it is proved that the enzymic patterns are not changed but the activities of enzymes relating endogeneous respiration and autofermentation is activated. It is to be considerable to distiguish endogeneous respiration and autofermentation from exogeneous respiration and fermentation on irradiation, for membrane permeability changes and loses out carbohydrate by ionizing radiation.

혐기성 생물막 반응기의 초기 운전특성 (Characteristics of continuous operation of the anaerobic biofilm reactor)

  • The lab-scale anaerobic continuous reactor which was filled with the sludge of anaerobic digestion from Suyoung wastewater treatment plant was operated by feeding of various concentrations and flow rates. This experiment indicated that more than 6,870 mgCOD/L of substrate concentration was required to promote good metabolism and growth of anaerobic biomass. And increasing loading rate slowly was also required in order to treat substrate of higher concentration and higher loading rate. The substrate concentration of about 10,000 mgCOD/L was adequate to generate biogas efficiently. The pH was sharply decreased at the onset of higher leading rate, but the pH was restabilized soon at 8. During the experiment, the amount of the attached biomass was kept constant.
