  • 마이크로칩에서의 생체물질 분석에 있어서 재현성은 매우 중요한 사항이다. 이전부터 많은 연구자들에 의해서 분석시 재현성을 향상시키고자 많은 연구가 선행되었다. 재현성을 향상시킬 수 있는 한 방안으로 겔 전기영동이 이용되고 있다. 본 연구에서도 겔 전기영동을 마이크로칩에 접목시켜 재현성을 향상시키는 실험을 진행하였다. DNA 시료로 100bp부터 1500bp 길이의 DNA 단편들을 사용한 결과 인산완충식 염수 (PBS)만을 사용하였을 경우보다 인산완충식염수(PBS)와 5% 폴리아크릴아미드 겔 (5% polyacrylamide)과 같이 사용하였을 경우 더 향상된 재현성을 확보하였다.

Electrochemical Biosensors for Biomedical and Clinical Applications: A Review

  • There are strong demands for accurate, fast, and inexpensive devices in the medical diagnostic laboratories, such as biosensors and chemical sensors. Biosensors can provide the reliable and accurate informations on the desired biochemical parameters, which is an essential prerequisite for a patient before going for a treatment. They can be used for continuous measurements of metabolites, blood cations, gases, etc. Of these, electrochemical biosensors play an important role in the improvement of public health, because rapid detection, high sensitivity, small size, and specificity are achievable for clinical diagnostics. In this paper, the clinical applications with electrochemical biosensors are reviewed. An attempt is also made to highlight some of the trends that govern the research and developments of the important biosensors that are associated to clinical diagnosis.

A Dipstick-Type Electrochemical Immunosensor for The Detection of The Organophosphorus Insecticide Fenthion

  • A dipstick-type immunochemical biosensor for the detection of the organophosphorus insecticide fenthion was developed using a screen-printed electrode system as an amperometric transducer with polyclonal antibodies against fenthion as a bioreceptor. The assay of the biosensor involved competition between the pesticide in the sample and pesticide-glucose oxidase conjugate for binding to the antibody immobilized on the membrane. This was followed by measurement of the activity of the bound enzyme by the supply of the enzyme substrate (glucose) and amperometric determination of the enzyme reaction product ($H_2O_2$). The activity of the bound enzyme was inversely proportional to the concentration of pesticide. The optimized sensor system showed a linear response against the logarithm of the pesticide concentration ranging from $10^{-2}$ to $10^3\;{\mu}g/L$.

Voltammetric Analysis on a Disposable Microfluidic Electrochemical Cell

  • A microfabricated electrochemical cell comprising PDMS-based microchannel and in-channel gold microelectrodes was fabricated as a sensitive and a miniature alternative to the conventional electroanalytical systems. A reproducible fabrication procedure enabled patterning of multiple microelectrodes integrated within a PDMS-based fluidic network. The active area of each electrode was $200{\mu}m{\times}200{\mu}m$ with a gap of $200{\mu}m$ between the electrodes which resulted in a higher signal to noise ratio. Also, the PDMS layer served the purpose of shielding the electrical interferences to the measurements. Analytes such as potassium ferrocyanide; amino acid: cysteine and nucleoside: guanosine were characterized using the fabricated cell. The microchip was comparable to bulk electrochemical systems and its applicability was also demonstrated with flow injection based rapid amperometric detection of DNA samples. The device so developed shall find use as a disposable electrochemical cell for rapid and sensitive analysis of electroactive species in various industrial and research applications.

Development of Single-layer-structured Glucose Biosensor

  • In this paper, we fabricated a low-cost glucose sensor with a simpler structure and fabrication process than the existing glucose sensor. The currently used glucose sensor has a three-layer structure with upper, middle, and bottom plates; here, we fabricated a single-layer glucose sensor using only a printing and dispensing process. We successfully fabricated the glucose sensor using a simple method involving the formation of an electrode and insulator layer through a 2- or 3-step printing process on plastic or paper film, followed by the dispensing of glucose oxidase solution on the electrode. Cyclic voltammetry (CV) and cyclic amperometry (CA) measurements were used to evaluate the characteristics of the fabricated single-layer glucose sensor. Also, its sensitivity was analyzed through glucose-controlled blood measurements. Hence, a low-cost single-layer glucose sensor was fabricated with evaluation of its characteristics demonstrating that it has useful application in medicine.

Flow Injective Determination of Thiourea by Amperometry

  • The amperometric responses of thiourea were studied in 0.1 M NaOH by flow injection analysis. D. C. amperometric and pulsed amperometric detection methods were applied for the determination of thiourea at novel metal electrodes such as Pt and Au. Triple-step potential waveforms were adopted in the pulsed amperometric detection. With an optimized pulsed waveform, the current for the oxidation of thiourea was examined with the variation of flow rate of carrier solution and with the change in the amount of sample injected. Gold working electrode turned out to be better in sensitivity and signal to noise ratio than Pt electrode in the pulsed amperometric detection of thiourea. Detection limit is estimated to be 5.33 ${\times}$ 10$^{-5}$ M with this detection method.

백금 도금된 탄소반죽 전극을 이용한 일회용 글루코오스 센서 (Disposable Glucose Sensor Based on Platinised Carbon Paste Electrode)

  • Carbon paste전극을 백금도금 한 후 이 전극을 사용한 일회용 전류법 glucose센서를 제작하였다. 전극에 시료용액을 가한 후, voltammetry을 이용하여 본 센서의 감응성을 연구하였다. screen printing된 carbon paste전극 표면 위에 전기도금 방법으로 도입된 백금은 센서의 분석성능과 mediator의 전기 화학적 가역성을 크게 향상시켜 준다. 제작된 센서가 $[Fe(CN)_6]^{4-/3-}$에 대한 heterogeneous rate constant는 $1.45\times10^{-2}cm{\cdot}s^{-1}$이며, 이 센서는 적용전위 0.3V vs. Ag/AgCl에서 glucose에 대한 좋은 감응성을 보여주었다. 이 때 glucose에 대한 Michaelis-Menten상수는 24.5mM이다. 일회용 glucose 스트립 센서의 분석성능을 평가하기 위하여 NOVA S.P. Ultra M analyzer와 비교실험을 수행하였다. 이 때 시료용액으로서 $80\~297 mg/dL$ 범위의 glucose를 함유하고 있는 30여 종류의 혈청을 사용하였고, 상관계수는 0.983으로서 정밀도와 정확도가 비교적 우수함을 보여주었다.

A Facile Electrochemical Fabrication of Reduced Graphene Oxide-Modified Glassy Carbon Electrode for Simultaneous Detection of Dopamine, Ascorbic Acid, and Uric Acid

  • This paper describes the simple fabrication of an electrode modified with electrochemically reduced graphene oxide (ERGO) for the simultaneous electrocatalytic detection of dopamine (DA), ascorbic acid (AA), and uric acid (UA). ERGO was formed on a glassy carbon (GC) electrode by the reduction of graphene oxide (GO) using linear sweep voltammetry. The ERGO/GC electrode was formed by subjecting a GO solution ($1mg\;mL^{-1}$ in 0.25 M NaCl) to a linear scan from 0 V to -1.4 V at a scan rate of $20mVs^{-1}$. The ERGO/GC electrode was characterized by Raman spectroscopy, Fourier transform infrared spectroscopy, contact angle measurements, electrochemical impedance spectroscopy, and cyclic voltammetry. The electrochemical performance of the ERGO/GC electrode with respect to the detection of DA, AA, and UA in 0.1 M PBS (pH 7.4) was investigated by differential pulse voltammetry (DPV) and amperometry. The ERGO/GC electrode exhibited three well-separated voltammetric peaks and increased oxidation currents during the DPV measurements, thus allowing for the simultaneous and individual detection of DA, AA, and UA. The detection limits for DA, AA, and UA were found to be 0.46, 77, and $0.31{\mu}M$ respectively, using the amperometric i-t curve technique, with the S/N ratio being 3.

Metal Fiber의 전기적 특성 평가와 이온제거 Filter로의 응용 (Evaluate the Metal Fiber's Electrical Character and Applicance removal of Ion Derivatives)

  • 일반적으로 중금속이나 질산성 질소와 같은 이온성 물질들은 사람의 건강이나 생태계에 좋지 않은 영향을 미칠 수 있다. 이들 오염물질들은 그 종류에 따라 건강에 피해를 주는 정도에 차이가 있지만 안전한 생태환경을 유지하기 위해서는 이들 물질들을 효과적으로 처리할 수 있는 필터가 필요하다. 전극 필터에 최대한 많은 이온들을 흡착시키기 위해서는 전극의 비표면적을 높이는 것이 매우 중요하다. Metal Fiber Filter의 제작과 제작된 Filter의 전기적 특성과 비표면적 향상, 고정성 및 통전성, 축전용량과 전기흡착 및 탈착 경향을 관찰하기 위해 3전극 시스템을 이용하여 Cyclic Voltammetry(CV)와 Chrono-amperometry(CA)를 측정하고 표면의 기공 구조를 관찰하기 위해 주사전자현미경 (SEM;Scanning Electron Microscope)을 이용하여 전극에 생성된 미세 기공의 크기 및 크기 분포를 측정한다. 또한 제작된 Filter 전극을 이용하여 전기장 하에서 수질 속에 있는 질산성이온의 선택적 제거 효율을 측정하여 보고, 측정된 데이터 값을 통해 Metal Fiber Filter의 전기적 특성과 중금속이나 질산성 이온 물질들을 선택적으로 제거할 수 있는 Filter 전극 설계 근거를 제시하고자 한다.

잔류염소 측정용 전기화학센서의 유효성 (Effectiveness of the Electrochemical Sensor for the Free Chlorine Measurement)

  • Sodium hypochlorite is used worldwide as a water disinfectant and in bleaching agent. Sodium hypochlorite applied to water initially undergoes hydrolysis to form free chlorine consisting of hypochlorous acid(HOCl) and hypochlorite ion($OCl^-$). For free chlorine determination, an electrochemical method is simple due to the electroactivity of free chlorine; it measures current and is free of most reagents. Amperometric free chlorine sensor has been developed with gold (Au)-based electrode. The 3-electrode free chlorine sensor whose working and counter electrodes were Pt exhibited excellent response to HClO at +400mV vs. Ag/AgCl/sat. KCl. In addition, the use of a pH error correction algorithm provided a reliable measurement of residual free chlorine in water sample without any pretreatment in the normal pH range(pH 6~8) of municipal water supply. The free chlorine sensor installed in on-line monitoring system could be used to continually monitor the level of residual free chlorine in real samples.
