송류화분(松類花粉)의 아미노산(酸) 조성(組成) 및 화분발아(花粉發芽)에 따른 그 조성(組成)의 변화(變化) (On the Amino Acid Composition in the Pollen of Pines and Changes of the Composition during Germination of Pollen)

  • In the present experiment the amino acid composition in the pollen of Pinus taeda and Pinus rigida and methbolic changes of the composition occurring in germinating pollen were examined by the method of paper chromatography. The results obtained in this study are as follows. 1. The mature pollen of Pinus taeda and Pinus rigida contained 12.00%, 13.19% crude protein respectively and sixteen protein-constituting amino acids and three unknowns were detected on the chromatogram. 2. Main components of the protein-constituting amino acids were leucines, valine, alanine, glutamic acid and aspartic acid. 3. The free amino acids such as leucines, valine, tyrosine, alanine and glutamic acid increased evidently during germination of the pine pollen.

  • The amino acid composition of human and cow's milk represents a standard of reference in infantnutrition. The amino acid compositions determined by automatic amino acid analyzer, Yanagimote Model LC-5. Protein in the human and cow's milk were found to be hydrolyzed to yield free amino acids. Qualitative data for free amino acids in the milk are as follows: 1) Amounts of acidic amino acids such as glutamic and aspartic acid in cow's milk were obserbed to be about 2 times compared with human milk and it is considered that the abundance in these amino acids may contribute significantly to the specific flavor of cow's milk. 2) It is much interesting that in the human milk the contents of sulfur-containing amino acids were high comparatively better than cow's milk; cystine was found to be 3 times ana methionine, 2 times. 3) In the human milk a high content of some essential amino acids such as threonine, isoleucine and leucine was demonstrated and a specific flavor sweet amino acids. 4) Large amounts of basic amino acid such as histidine was found to occur in human milk and arginine in cow's milk.

Amino Acid Profiles of Tropical Legumes, Cooper (Glycine wightii), Tinaroo (Neonotonia wightii) and Siratro (Macroptilium atropurpureum), at Pre-blooming and Blooming Stages

  • An experiment was conducted to investigate amino acid composition of three tropical legumes (Cooper (Glycine wightii), Tinaroo (Neonotonia wightii) and Siratro (Macroptilium atropurpureum)) at two different stages (pre-blooming and blooming stages). Chemical composition and totally 16 amino acids of these plants were analysed for comparison of their composition among species at different growing stages and characterizing the amino acid pattern of these legumes. Crude protein content of the plants ranged from 16% to 27% on a dry matter basis. The total amount of 16 amino acids analyzed in this experiment was highest at 89.7 mg/16 g N in Cooper at pre-blooming and lowest at 80.9 mg/16 g N in Glycine at blooming stage. Total amount of amino acids in each legume species tended to slightly decrease with their maturity but no statistical difference was found. The percentage of aspartic acid, glutamic acid and proline in the total amount of amino acids was dominant at 9% to 13%, and that of methionine was less than 1.6%. In this experiment it was concluded that three tropical legumes were rich in crude protein content and characterized by 16 different amino acids with lower sulfur-containing amino acid as methionine.

Thin Layer Chromatogram by an Extracellular ${\beta}$-Amylase of Bacillus sp. KYJ 963 and its Amino Acid Composition

  • Bacillus sp. KYJ 963, which was isolated from Korean salt-fermented anchovy (anchovy-jeot), produces an extracellular ${\beta}$-amylase. The analysis of the digestion products of substrates by thin layer chromatography from the purified protein revealed that the enzyme could not hydrolyze maltose or ${\alpha}$-cyclodextrin. In the amino acid composition analysis, the major characteristic of the ${\beta}$-amylase was the high proportion of amino acids that possess short side chain such as glycine and alanine.

성게 알의 아미노산과 지방산 조성 (The Composition of fatty acid and amino acid for sea urchin)

  • The composition of fatty acid and amino acid for Seong-ge-al(sea urchin) were analysed by the automatic analyser. There are two kind of samples such as raw and saited, fermented one. The results obtained are as follows. 1. In the general composition, crude protein, crude lipid and ash as 15.8%, 3.5% and 2.2%, respectively. The raw sample showed pH 5.6 value. 2. In case of amino acid, it contained 17 different kind of amino acids. Among them, a lot of glutamic acid, aspartic acid, arginine, glycine and valine were contained. 3. In case of fermentation period, threonine, histidine, lysine and arginine were increased a little bit. 4. In case of fatty acid, it contained 13 different kind of fatty acids. Among them, a lot of palmitic acid, behnic acid, linoleic acid and arachidonic acid were contained.

Prediction of Protein Secondary Structure Content Using Amino Acid Composition and Evolutionary Information

  • There have been many attempts to predict the secondary structure content of a protein from its primary sequence, which serves as the first step in a series of bioinformatics processes to gain knowledge of the structure and function of a protein. Most of them assumed that prediction relying on the information of the amino acid composition of a protein can be successful. Several approaches expanded the amount of information by including the pair amino acid composition of two adjacent residues. Recent methods achieved a remarkable improvement in prediction accuracy by using this expanded composition information. The overall average errors of two successful methods were 6.1% and 3.4%. This work was motivated by the observation that evolutionarily related proteins share the similar structure. After manipulating the values of the frequency matrix obtained by running PSI-BLAST, inputs of an artificial neural network were constructed by taking the ratio of the amino acid composition of the evolutionarily related proteins with a query protein to the background probability. Although we did not utilize the expanded composition information of amino acid pairs, we obtained the comparable accuracy, with the overall average error being 3.6%.

Amino Acid Composition Analysis of the 32 kDa Sperminogen

  • Boar sperminogen was purified from the acid extracts of the washed epididymal spermatozoa by gel filtration through a Sephadex G-100 column, followed by preparative SDS-PAGE. The 32 kDa sperminogen band was sliced out from the preparative SDS-PAGE and 32 kDa sperminogen was eluted from the gel matrix. The purified 32 kDa sperminogen was subjected to amino acid composition analysis. The amino acid composition of the 32 kDa boar sperminogen showed significant differences from that of either boar proacrosin or ${\beta}-acrosin$, which signifies that 32 kDa sperminogen might not be a breakdown product of proacrosin-acrosin system and that the 32 kDa sperminogen is a different protein from proacrosin-acrosin system.

  • 나무딸기의 화학적 성분의 조성(組成)을 알기 위하여 유리 아미노산은 아미노산 자동분석계, 유기산은 GC 유리당은 ppC를 통하여 분석 조하여 다음과 같은 결론(結論)을 얻었다. 1. 유리아미노산은 18종(種) 발견되었으며 특히 lysine의 함량(含量)이 많았다. 필수(必須)아미노산을 고르게 함유하여 타 과실에 비하여 좋은 아미노산 조성(組成)을 이루고 있다. 2. 유기산은 citric acid, tartaric acid, maleic acid, fumaric acid가 발견 되어졌으며 일반적으로 그 함량은 적었다. 3. 당은 glucose, fructose, mannose, maltose lactose등을 함유하고 있다.

산삼 배양액 급여 돈육의 지방산, 아미노산 조성 및 관능적 특성 (Fatty Acid, Amino Acid Composition and Sensory Traits of Pork from Pigs Fed Artificial Culture Medium of Wild Ginseng)

  • 본 시험은 산삼 배양액 급여가 성별(암퇘지와 거세돼지)에 따른 돈육 품질에 미치는 영향을 조사하기 위해 $60{\pm}3kg$의 버크셔 120두를 공시하여 산삼 배양액을 급여하지 않은 대조구와 산삼 배양액을 1일 두당 1 L를 출하 시(약 110 kg)까지 약 70일 동안 음수로 급여한 처리구로 하여 실시하였다. 지방산 조성은 대조구보다 처리구가, 암퇘지보다 거세돼지가 단가 불포화 및 포화지방산 비율은 증가하고 상대적으로 불포화지방산 비율은 감소하였다. 아미노산 조성은 대조구보다 처리구가 필수 아미노산 함량은 적은 반면 황함유 및 방향족 아미노산 함량은 많았으며, 암퇘지보다 거세돼지가 필수 및 맛 관여 아미노산 함량은 적은 반면, 감미계, 황함유 및 방향족 아미노산 함량은 많은 결과였다. 신선육의 관능평가 결과 처리 간에는 암퇘지의 육색만 대조구보다 처리구가 높게 나타났으며, 성별 간에는 대조구의 모든 항목과 처리구의 drip loss와 marbling은 암퇘지보다 거세돼지가 높게 나타났다. 가열육의 관능 평가 결과 성별 간에는 대조구의 향과 전체적 기호도만이 암퇘지보다 거세돼지가 높게 나타났다.

Organic Solvent-Tolerant Esterase from Sphingomonas glacialis Based on Amino Acid Composition Analysis: Cloning and Characterization of EstSP2

  • Organic solvent-tolerant (OST) enzymes are widely applied in various industries for their activity and stability in organic solvents, for their higher substrate solubility, and for their greater stero-selectivity. However, the criteria for identifying OST enzymes largely remain undefined. In this study, we compared the amino acid composition of 19 OST esterases with that of 19 non OST esterases. OST esterases have increased the ratio of Ala and Arg residues and decreased the ratio of Asn, Ile, Tyr, Lys, and Phe residues. Based on our amino acid composition analysis, we cloned a carboxylesterase (EstSP2) from a psychrophilic bacterium, Sphingomonas glacialis PAMC 26605, and characterized its recombinant protein. EstSP2 is a substrate specific to p-nitrophenyl acetate and hydrolyzed aspirin, with optimal activity at $40^{\circ}C$; at $4^{\circ}C$, the activity is approximately 50% of its maximum. As expected, EstSP2 showed tolerance in up to 40% concentration of polar organic solvents, including dimethyl sulfoxide, methanol, and ethanol. The results of this study suggest that selecting OST esterases based on their amino acid composition could be a novel approach to identifying OST esterases produced from bacterial genomes.
