알파-아밀라제 저해제 생성 Streptomyces DMCJ-49의 동정과 저해제의 분리 (Identification of Streptomyces DMCJ-49 Producing the alpha-Amylase Inhibitors and the Isolation of the Inhibitor)

  • To find ${\alpha}-amylase$ inhibitors produced by microorganisms from soil, a strain which had a strong inhibitory activity against bacterial ${\alpha}-amylase$ was isolated from the soil sample collected in Korea. The morphological and physiological characteristics of this strain on several media and its utilization of carbon sources showed that it was one of Streptomyces species according to the International Streptomyces Project method. The amylase inhibitor of this strain was purified by active carbon adsorption, silicagel column chromatography, SP-Sephadex C-25 column chromatography, adsorption on Amberlite XAD-2. The inhibitor was oligosaccharide which was composed of glucose. The inhibitor had inhibitory activity against other amylase such as salivary ${\alpha}-amylase$, pancreatic ${\alpha}-amylase$, fungal ${\alpha}-amylase$ and gluco-amylase.

환삼덩굴로부터 분리한 Luteolin-7-O-$\beta$-D-Glucoside의 활성산소 소거능 (Active Oxygen Scavenging Activity of Luteolin-7-O-$\beta$-D-Glucoside Isolated from Humulus japonicus)

  • We investigated the active oxygen scavenging activity and active compound from Humulus japonicus. The ethyl acetate and butanol fraction exhibited strong scavenging effects on hydroxyl radical. Furthermore active compound was isolated and purified using Amberlite XAD-2 column chromatography and preparative HPLC from ethyl acetate fracton, and major compound was identified as a luteolin-7-O-$\beta$-D-glucoside by the MS, UV, 1H-NMR and 13C-NMR analyses. Luteolin-7-O-$\beta$-D-glucoside exhibited strong scavenging effect on active oxygens such as superoxide radical, hydroxyl radical and hydrogen peroxide.

  • As one of the serial studies on the specific and indigenous plants of Mt. Chiri the constituents of aerial part from filipendula formosa (Rosaceae) were investigated. From of the MeOH extract, five flavonol glycosides, kaempferol-3-O-$\beta$-D-galactopyranoside, querecetin-3-O-$\beta$-D-galactopyranoside, quercetin-3-O-$\alpha$-Lrhamopyranosyl (1 6)-$\beta$-D-galactopyranoside, kaempferol-3-O-$\alpha$-L-rhamnopyranosyl (1 6)-$\beta$-D-galactopyranoside and quercetin-7-O-$\beta$-D-glucopyranosy-3-O-$\beta$-D-galactopyranoside were isolated by column chromatographic separation using Amberlite XAD-2 and Sephadex LH-20, and identified physicochemical evidences (IR, FAB-Mass, $^1H,{\;}^{13}C-NMR$).

수종생약의 Brine Shrimp 독성에 관한 연구(I) (Studies on the Brine Shrimp Toxicity of Crude Drugs (I))

  • Brine shrimp toxicity of water extracts of twenty three crude drugs was tested by the brine shrimp bioassay. Eight crude drugs were shown to have 100% mortality in $1000{\;}{\mu}g/ml$. They were fractionated by Amberlite XAD-2 column chromatography and were tested by the brine shrimp bioassay. Methanol fraction of Rhei Rhizoma $(LC_{50}=49.60{\;}{\mu}g/ml)$ and Cantharis $(LC_{50}=54.08{\;}{\mu}g/ml)$ were shown to have potent brine shrimp toxicity in this test.

  • 맥문동 괴근을 80% MeOH 수용액으로 추출하고, 얻어진 추출물을 EtOAc, n-BuOH 및 $H_2O$로 용매 분획하였다. 암세포 A549, SK-OV-3, SK-Mel-2, XF-498 및 HCT-15에 대한 각 분획물의 생장억제활성을 SRB 법으로 측정한 결과 n-BuOH 분획에서 암세포 생장억제활성이 높게 나타났다. n-BuOH 분획으로부터 Amberlite XAD-II 및 silica gel column chromatography를 반복하여 2종의 사포닌을 분리하였고, NMR, IR 데이터의 해석과 가수분해반응을 이용하여 spicatoside A와 B로 동정하였다. Spicatoside A가 암세포에 대한 생장억제활성을 보여 A549, SK-OV-3, SK-Mel-2, XF-498 및 HCT-15에 대한 $IC_{50}$ 값이 각각 17.3, 21.7, 14.9, 18.8 및 $15.6\;{\mu}g/ml$를 나타내었다.

  • 식물성장촉진 호르몬인 auxin, 식물병원성 진균의 생육을 억제하는 siderophore, 그리고 세포벽이 cellulose로 구성된 식물병원성 진균의 세포벽을 분해하는 cellulase를 동시에 생산하는 PGPR균주이자 생물방제균주로 선발된 Bacillus licheniformis K11이 생산하는 항진균성 siderophor의 특성을 확인한 결과 catechol type의 siderophore로 확인하였다. 또한 B. licheniformis K11의 siderophore 생산조건을 조사한 결과 nutrient broth(pH 9.0)를 $20^{\circ}C$에서 96 h 배양시 가장 많이 생산하였다. 그리고 탄소원과 질소원으로는 각각 trehalose와 $NH_4Cl$ 첨가시 siderophore를 가장 많이 생산하였다. 그리고 $siderophore_{K11}$의 정제는 Amberlite XAD-2과 sephadex LH-20 column chromatography를 통해 수행하였으며, HPLC를 통해 단일 물질임을 확인하였다. 정제된 $siderophore_{K11}$은 2, 3-dihydroxybenzoate의 유도체로 추정하며, P. capsici의 zoospores와 F. oxysporum의 conidia의 발아를 강하게 억제하였다.

장독성 대장균 eKT-53 균주의 내열성 장독소 정제 (Purification of Heat-Stable Enterotoxin of Enterotoxigenic Escherichia coli eKT-53)

  • 장독성대장균은 영유아설사 및 여행자설사의 주요한 원인균의 하나로서 이들이 생산하는 내열성장독소는 설사 유발 원인물질로 이들 생산균주는 비병원성 대장균과 비교하여 검정할 수 있다. 본 실험에서는 이러한 내열성장독소를 순수정제하였다. 저자들이 보고한 바 있는 형질전환균주 eKT-53을 사용하여 10L의 succinate salts 배지에 배양한 배양액을 30,000g로 원심분리하여 조 ST 용액을 사용하여 몇 단계의 정제과정을 통하여 정제하였다. 즉 Amberlite XAD-2 chromatography, DEAE-Sephacel anion exchange chromatography, Bio-Gel P-6 get filtration, Polyacrylamide slab gel 전기영동을 통하여 정제하고 장독소의 정제도 및 장독소의 생물학적 최소단위량을 조사하였다. 최종적으로 정제되어진 ST는 조 ST 용액에 비해 113배 더 정제되었고 회수율은 11%였다. 또한 생물학적 최소단위량이 2.8ng이였고 SDS-polyacrylamide disc gel상에서 단일 band를 나타내어 거의 순수정제된 것으로 보이며 면역을 위한 항원으로 이용할 수 있을 것으로 생각된다.

해양으로부터 분리한 Pseudomonas sp. CHCS-2가 생산하는 Biosurfactant의 정제 및 특성에 관한 연구 (Purification and Characterizationn of Biosurfactant from Marine Pseudomonas sp. CHCS-2)

  • 해양의 유류유출이 잦은 지역으로부터 crude oil 분해능이 뛰어난 미생물을 분리하여 통정한 결과 Pseudomoans 속으로 판명되였으며, 이를 Pseud$\sigma$ moans sp. CHCS-2로 명 명하였다. 이 균주가 배양 중에 생산하는 biosurf actant의 생생 최적 pH 및 NaCI 농도는 각각 8.0 및 3.0% 였으며, 질소원인 peptone에 영향을 받았다. 2% Kuwait crude oil이 첨가된 배양액을 48, 96, 132시간별로 gas chroma­t tography를 이용, 잔류 oil을 분석한 결과 Kuwait crude oil의 C10-CI4부위에 biosurf actant가 작용하여 분해하였으며, 배양 상층액으로부터 Amberlite XAD-7을 이용한 흡착 chromatography와 Sepha­d dex G-100을 이용한 gel chromatography, 그리고 HPLC를 이용하여 biosurf actant를 분리. 정제한 결과 유화력이 뛰어난 단일 peak를 얻었다. Bio-sur­f f actant 유화력은 $40^{\circ}C$에서 가장 좋았으며, 안정성 은 $30^{\circ}C$에서 $60^{\circ}C$까지의 넓은 온도 범위에서 유지 되었다. 또한, 정제된 biosurf actant를 이용하여 계 연장력에 미치는 영향을 검토한 결과, 상엽적으로 널리 판매되고 있는 Tween 80, Tween 60 그리고 SDS보다 표연장력 저하능력이 뛰어난 것으로 밝혀졌다.

의이인이 3T3-Ll Adipocytes에서 인슐린성 작용과 인슐린 민감성에 미치는 영향 (Effect of Insulin-like Action and Insulin Sensitizing on 3T3-Ll Adipocytes from Coicis Semen)

  • Objectives : In this study, water extracts from Coix lachryma-jobi Linne var. mayuen Stapf. were investigated for their effects on insulin-like action and glucose uptake in 3T3-Ll cells. Methods : We examined the effects of insulin-like action on the differentiation of 3T3-Ll fibroblasts. The Coicis Semen was treated with hot water and the extract was freeze-dried. The hot water extract was chromatographed on nonionic polymer resin (Amberlite XAD-4, Sigma) with distilled water (Fr. 1), 20% (Fr. 2), 40% (Fr. 3), 60% (Fr. 4), 80% (Fr. 5), and 100% EtOH (Fr. 6), successively. Results : Total extract of Coicis Semen was fractionated into 0 to 100% MeOH with Amberlite XDA-4 column. Treatment of cells with $10{\;}\mu\textrm{g}/ml$ of total extracts of Coicis Semen significantly increased the differentiation (p<0.05). At $1{\;}\mu\textrm{g}/ml$ level of insulin, the differentiation was accelerated (p<0.01). The effect of extracts plus insulin on the differentiation was greater than that of insulin alone. In 3T3-Ll adipocytes, glucose uptake was higher by 2.7 times with 5 uM of total extract in low dosage of insulin (3 ng/ml) than without total extract. 5 and 50 uM of water and 40% MeOH fractions increased glucose uptake by 3.5 times in 3T3-Ll adipocytes (p<0.00l). Conclusions : Coicis Semen contains compounds which playa role of insulin-like action and insulin sensitizer.

잔디밭과 나지에 산포된 주요 잔디밭용 제초제의 휘산 (Volatility of Herbicides Sprayed in Zoysia japonica Turf and Bare Soil)

  • This experiment was conducted to investigate the loss of various herbicides by means of vola-tility from the turfgrass field and the hare ground with the different soil moisture contents and temperatures. Different herbicides were applied at the rates of 375 g a.i. /l0a of pendimethalin,250 g a.i. /l0a of napropamide, and 96.4 g a.i. /l0a of dicamba with 200 \ulcorner/10a of spray volume in the turfgrass(Zoysia japonica cut off 5cm) grown in pots(265.8 $cm^2$) and bared soil. The pots were placed in the growth chamber with 10,000 lux of light intensity(12h per day) at 25 and 35˚C for 7days. Amberlite XAD polymeric resin(20/50 mesh) was used as sampling media for herbicide airborne residues. Air flow was maintained at 10 \ulcorner /min by vacuum pump regulated with a factory calibrated flow meter. Herbicide airborne residues were extracted from the XAD resin with 300 ml of 1:1 acetone and hexane. The extracts were concentrated by rotary evaporation at 35˚C and dissolved in 1 ml MeCN for HPLC analysis. The airborne losses of the herbicide applied in the turfgrass and bare soil increased as the temperature and soil moisture contents were increased, regardless of the kinds of herbicide. Higher airborne residues was observed in the turfgrass pots than the bare soil pots. Pendimethalin and dicamba with higher vapor pressure gave rise to the increased loss of airborne herbicides, showing that 6.26 and 6.4% of average airborne loss in pendimethalin and dicamba, respectively, compared to 0.56% in napropamide. The amount of airborne losses in turfgrass was highest at one day after application and then a declined trend was observed as the time was prolonged. Key words.
