In this study, we investigated whether there is another amaranth GBSS isoform in an attempt to characterize the synthesis of amylose in the pericarp. We used I2/KI staining to analyze the temporal and spatial starch accumulation patterns during seed development. The spatiotemporal starch accumulation patterns in developing seeds were observed by staining with I2/KI. Starch granules were observed in the pericarp in the initial developmental stage (3 DAP). A few starch granules were detected in the perisperm in the early-late developmental stage (8 DAP), during which the pericarp starch contents rapidly decreased. Starch granules were distributed throughout the perisperm in the mid-late developmental stage (15 DAP). Similar results were reported for other cereal crops, including barley, rice, and sorghum. Starch granules in the pericarp are synthesized during the early seed developmental stages but are absent in mature seeds. We recently reported that starch deposits in the perisperm of developing amaranth seeds are detectable only after the initial developmental stage. Prior to this stage, the pericarp is the major site of starch deposition. A recent study suggested that GBSSII isoforms are responsible for amylose synthesis in pericarps.

In the present study, we compared and investigated the nutritional compositions of Amaranthus cruentus and Amaranthus hypochondriacus baby-leaves cultivated in Korea. Baby-leaves of two amaranthes consisted of more than 92% moisture, and A. cruentus contained a higher amount of moisture than A. hypochondriacus. Meanwhile, A. hypochondriacus contained higher levels of crude ash, crude protein, crude lipid, carbohydrates, and dietary fiber than A. cruentus. The major free sugars of the two amaranth baby-leaves were fructose and glucose. Fructose content of A. hypochondriacus was higher than that of A. cruentus, and glucose content of A. cruentus was higher than that of A. hypochondriacus. Acetic acid, malic acid, and fumaric acid were detected in two amaranth leaves, but succinic acid was not detected. Two amaranth leaves contained 17 amino acids except for methionine, proline, and tyrosine, and leaves contained the highest glutamic acid contents. In addition, A. cruentus and A. hypochondriacus leaves contained high contents of taurine and ${\gamma}$-aminobutyric acid and showed various biological activities. The major mineral and fatty acid of the two amaranth leaves were potassium and linolenic acid (C18:3), respectively. The ${\beta}$-carotene contents of A. cruentus and A. hypochondriacus leaves were $478.72{\mu}g/100g$ and $474.12{\mu}g/100g$, respectively. In vitamin B complex, $B_2$, $B_3$, and $B_5$ were detected in the two amaranth leaves whereas vitamins $B_1$, $B_6$, and $B_{12}$ were not detected. A. hypochondriacus contained higher amounts of vitamin C and E than those of A. cruentus. Overall, amaranth leaves contained high amounts of nutritional components. Therefore, amaranth leaves are expected to be useful for the development of a functional food. Moreover, these results will provide fundamental data for advancing sitological value, breeding new cultivars, and promoting leafy vegetable usage.

A feeding experiment was conducted on rats to investigate the inhibitive effects of baik-kimchi against the toxicity of Amaranth (Am, Food Red No.2) on growth, fecal weights and bulk, and the digestibility of protein and lipid. The body weight of the Am diet group (5% Amaranth) remained at the lowest level of all diet groups, because of diarrhea symptoms throughout the feeding period, 229.8 g of total body weight or 79.4% of the basal diet group (289.4 g). However, in the baik-kimchi diet group (10% baik-kimchi powder+5% Amaranth) no diarrhea symptoms were evident and total body weight remained at a similar level (280.3 g) to the basal diet group. It therefore appears that baik-kimchi offers an inhibitive effect against the toxicity of Am. In the baik-kimchi diet group, the total fecal weight (173.99 g/3 weeks) was 15 times that of the basal diet group (11.13 g/3 weeks) suggesting that baik-kimchi has the effect of increasing fecal bulk. The large intestine and the cecum of the baik-kimchi diet group were significantly (p<0.05) heavier than those of the Am diet group. The digestibility of protein (50.26%) and lipid (58.37%) in the Am diet group was very low in comparison with these in the baik-kimchi diet group (87.30% and 92.12%, respectively). This result indicates that the low digestibility of these nutrients in the Am diet group was directly responsible for the decrease of weight. Consequently, it is demonstrated that baik-kimchi can inhibit the effects of the toxicity of Am in rats.
The adsorption characteristics of amatanth dye by granular activated carbon were experimently investigated in the batch adsorption. Kinetic studies of adsorption of amaranth dye were carried out at 298, 308 and 318 K, using aqueous solutions with 100, 200 and 300 mg/L initial concentration of amatanth. It was established that the adsorption equilibrium of amaranth dye on granular activated carbon was successfully fitted by Langmuir isotherm equation at 298 K. The pseudo first order and pseudo second order models were used to evaluate the kinetic data and the pseudo second order kinetic model was the best with good correlation. Values of the rate constant ($k_2$) have been calculated as 0.1076, 0.0531, and 0.0309 g/mg h at 100, 200 and 300 mg/L initial concentration of amatanth, respectively. Thermodynamic parameter such as activation energy, standard enthalpy, standard entropy and standard free energy were evaluated. The estimated values for standard free energy were -5.08 - -8.10 kJ/mol over activated carbon at 200 mg/L, indicated toward a spontaneous process. The positive value for enthalpy, 38.89 kJ/mol indicates that adsorption interaction of amatanth dye on activated carbon is an endothermic process.

In this research, root extracts of Nelumbo nucifera was tested to see the possibility for functional cosmetic agent. 70-100% ethanol was used as solvent and nuciferin was confirmed as active component. To test cosmetic effect of root extracts of Nelumbo nucifera, safety effect(MTT assay), anti-wrinkle effect(elastase inhibition assay) and antioxidation effect(DPPH free radical scavenging assay) were measured. When 100% ethanol was used as extracting solvent, cell viability was over 80% at $100{\mu}g/ml$, which indicated that root extract of Nelumbo nucifera was suitable for cosmetic agent. Root extract of Nelumbo nucifera showed 40~50% elastase inhibition at $100{\mu}g/ml$ so that it had good anti-wrinkle characteristics. 50% antioxidation capacity($FSC_{50}$) was $5.0{\sim}38{\mu}g/ml$ and root extract of Nelumbo nucifera showed excellent antioxidation effect. From the research, root extracts of Nelumbo nucifera showed strong possibility for anti-wrinkle functional cosmetic agent.
DNA addicts induced by putative chemical related to carcinogenesis were detected and determined by $^{32}$P-Postlabelling assay after exposure of 4 compounds comprising two auto dyes (amaranth, new coccine) and two flavonoid compounds (rutin, quercetin) to ICR mouse. DNA was isolated from mouse liver and digested enzymatically to deoxyribonucleoside 3'-monophosphate. The postincubation of DNA digests with nuclease Pl before $^{32}$P-labelling enhanced the technique's sensitivity. Nuclease Pl cleaves deoxyribonucleoside 3'-mono-phosphates of normal nucleotides to deoxyrihonucleosides which do not serve as substrates for polynucleotide kinase, while most of addicts were found to be totally or partially resistant to the 3'-dephosphorylating action of nuclease Pl. The adducted deoxyribonucleoside 3'-monophosphate was converted to 5'-$^{32}$P-labelled deoxynucleoside 3',5'-bisphosphate by T4 polynucleotide kinase. The nucleotides were separated by anion-exchange thin layer chromatography(TLC) on polyethyleneimine cellulose by 4-dimensional or 2-dimensional TLC then detected by autoradiography. The results show that DNA addicts were detected in liver DNA of ICR mouse after administration of amaranth and quercetin by 2-dimensional and/or 4-dimensional TLC.

This study was performed to investigate pre-harvest residue limit (PHRL) in amaranth, to estimate biological half-life of emamectin benzoate and identify the characteristics of the residue. Pesticides of standard and double appplication rate, were sprayed once on amaranth at 0, 1, 2, 3, 5, 7, 10, 14 days before harvest. Amaranth sample was extracted with acetonitrile and partitioned with dichloromethane, and pesticide residues were determined with LC/MS/MS. The limit of detection of emamectin benzoate was 0.01 mg/kg. Recoveries of emamectin benzoate ($B_{1a}$, $B_{1b}$) at two fortification levels of 0.1 and 0.5 mg/kg, $B_{1a}$ were $93.3{\pm}0.7%$ and $93.2{\pm}7.7%$, $B_{1b}$ were $106.6{\pm}1.9%$ and $80.5{\pm}6.6%$, respectively. The biological half-lives of emamectin benzoate were about 2.0 days at standard application rate and 1.7 days at double application rate, respectively. The PHRL of emamectin benzoate were recommended as 0.84 mg/kg for 10 days before harvest.
