인지증을 유발하는 원인은 알츠하이머병(Alzheimer's Disease: AD)과 혈관성 인지증(vascular Dementia: VD)이 가장 높은 비율을 차지한다. 본 논문에서는 측정된 라만 스펙트럼에서 AD, VD, 정상(NOR: normal)을 분류하기 위해 변별력 있는 영역을 조사하고, 특징 변환을 이용한 분류 실험 결과를 제시하였다. 혈소판으로부터 측정한 라만 스펙트럼은 먼저 smoothing을 적용한 다음 배경 잡음을 제거하고 스펙트럼의 기준 피크를 중심으로 그 위치를 정렬하였고 minmax 방법을 사용하여 정규화 하였다. 전처리를 거친 스펙트럼은 AD와 VD, NOR를 변별하기 가장 용이한 영역을 결정하기 위해 조사되었으며, 그 결과 725-777, 1504-1592, 1632-1700 $cm^{-1}$ 영역에서 스펙트럼이 많은 차이를 보임을 확인하였다. 분류 실험은 선택한 각 영역에 대하여 PCA(principal component analysis)와 NMF(nonnegative matrix factorization) 방법을 적용하여 얻은 특징을 이용하여 행하였다. 총 327개의 라만 스펙트럼에 대한 MAP(maximum a posteriori probability) 분류 실험 결과에 따르면, 본 연구에서 제안된 국부 영역 변환 특징을 사용했을 때 평균 92.8 %의 분류율을 보임을 알 수 있었다.
본 논문은 혈소판으로부터 측정한 라만 스펙트럼을 알츠하이머병(AD: Alzheimer's Disease) 진단에 사용하는 연구에 관한 것이다. 실험에 사용한 라만 스펙트럼은 다음과 같은 몇 가지 전처리 과정을 거친 다음 분류기를 이용하여 질병의 유무를 판별하였다. 먼저 스펙트럼은 간단한 smoothing을 거친 다음 스펙트럼의 각 피크 크기를 쉽게 측정할 수 있도록 기준선 (baseline)의 왜곡을 차감하였고, 그 다음 기준 피크를 중심으로 그 위치를 정렬한다. 이와 같은 전처리 과정을 수행한 라만 스펙트럼으로부터 AD과 정상상태(NOR: normal)를 구분할 수 있는 특징을 조사하였는데 그 결과 여러 피크들 중 743과 757 $cm^{-1}$ 영역의 피크 비와 1658 $cm^{-1}$ 영역의 피크 크기가 AD와 NOR 스펙트럼에서 가장 변별력 있는 특징을 나타내는 것을 확인하였다. 이 특징들을 MAP(maximum a posteriori)와 MLP(multi-layer perceptron) 분류기에 사용하여 총 278개의 스펙트럼에 적용한 결과 MLP에서 평균 약 95.5%의 분류율을 보였다. 이 결과는 혈소판 라만 스펙트럼이 알츠하이머병 진단에 매우 효과적으로 사용될 수 있음을 보여준다.

Alzheimer's disease (AD) is a neurological condition that is recognized as one of the primary causes of memory loss. AD currently has no cure. Therefore, the need to develop an efficient model with high precision for timely detection of the disease is very essential. When AD is detected early, treatment would be most likely successful. The most often utilized indicators for AD identification are the Mini-mental state examination (MMSE), and the clinical dementia. However, the use of these indicators as ground truth marking could be imprecise for AD detection. Researchers have proposed several computer-aided frameworks and lately, the supervised model is mostly used. In this study, we propose a novel 3D Convolutional Neural Network Multilayer Perceptron (3D CNN-MLP) based model for AD classification. The model uses Attention Mechanism to automatically extract relevant features from Magnetic Resonance Images (MRI) to generate probability maps which serves as input for the MLP classifier. Three MRI scan categories were considered, thus AD dementia patients, Mild Cognitive Impairment patients (MCI), and Normal Control (NC) or healthy patients. The performance of the model is assessed by comparing basic CNN, VGG16, DenseNet models, and other state of the art works. The models were adjusted to fit the 3D images before the comparison was done. Our model exhibited excellent classification performance, with an accuracy of 91.27% for AD and NC, 80.85% for MCI and NC, and 87.34% for AD and MCI.
In machine learning, the performance of the system depends upon the nature of input data. The efficiency of the system improves when the behavior of the input data changes from un-normalized to normalized form. This paper experimentally demonstrated the performance of KNN, SVM, LDA and NB on Alzheimer's dataset. The dataset undertaken for the study consisted of 3 classes, i.e. Demented, Converted and Non-Demented. Analysis shows that LDA and NB gave an accuracy of 89.83% and 88.19% respectively in both the cases whereas the accuracy of KNN and SVM improved from 46.87% to 82.80% and 53.40% to 88.75% respectively when input data changed from un-normalized to normalized state. From the above results it was observed that KNN and SVM show significant improvement in classification accuracy on normalized data as compared to un-normalized data, whereas LDA and NB reflect no such change in their performance.
Alzheimer's Disease (AD) is a cognitive disorder characterized by memory impairment that can be assessed at early stages based on administering clinical tests. However, the AD pathophysiological mechanism is still poorly understood due to the difficulty of distinguishing different levels of AD severity, even using a variety of brain modalities. Therefore, in this study, we present a hybrid EEG-fNIRS modalities to compensate for each other's weaknesses with the help of Machine Learning (ML) techniques for classifying four subject groups, including healthy controls (HC) and three distinguishable groups of AD levels. A concurrent EEF-fNIRS setup was used to record the data from 41 subjects during Oddball and 1-back tasks. We employed both a traditional neural network (NN) and a CNN-LSTM hybrid model for fNIRS and EEG, respectively. The final prediction was then obtained by using majority voting of those models. Classification results indicated that the hybrid EEG-fNIRS feature set achieved a higher accuracy (71.4%) by combining their complementary properties, compared to using EEG (67.9%) or fNIRS alone (68.9%). These findings demonstrate the potential of an EEG-fNIRS hybridization technique coupled with ML-based approaches for further AD studies.
본 연구는 주기적인 알츠하이머 병의 중증도 모니터링을 위해 스마트 인솔을 통한 보행 특징 추출과 머신러닝 기반 중증도 분류의 성능에 대해 살펴보았다. 최근 고령화가 가속화되는 추세에 있어 치매 환자가 급증하고 있으며, 중증도가 심해질수록 필요한 치료 비용 및 노력이 급증하기 때문에 조기 진단이 최선의 치료 전략으로 보여진다. 환자 친화적이고 저비용의 관성 측정 장치가 내장된 스마트 인솔만을 사용하여 다양한 보행 실험 패러다임에서 환자의 보행 특징을 추출하고, 이를 알츠하이머 병의 중증도 진단을 위한 머신러닝 기반 분류기를 훈련시켜 성능을 평가한 결과, 숫자세기와 같이 뇌에 부하를 주는 하위 작업이 포함된 복합 보행을 측정한 데이터셋을 사용하여 훈련된 분류 모델이 일반 걷기 데이터셋을 사용한 모델보다 성능이 높게 나타나는 것이 관찰되었다. 본 연구는 안전하고 환경적 제약이 적은 방법을 사용하여 시기 적절한 진단뿐만 아니라 주기적인 중증도 모니터링 시스템의 일환으로 활용될 수 있을 것이다.
Background: Dementia is one of important social and economic healthcare issues in the aging age. Therefore, it signifies to analyze the relationship between chronic disease or cardiovascular drug use and the incidence of dementia to establish a basis for increasing or preventing the risk of dementia. The purpose of this study was to investigate the correlation between the prevalence of chronic diseases and the use of cardiovascular drugs in patients diagnosed with dementia. Methods: In this study, we used data from sample of elderly patients from the Health Insurance Review and Assessment Service. We analyzed by logistic regression analysis with age, gender, and medication as covariates. KCD-7 was used to diagnosis of the disease, and drugs were analyzed using ATC codes and Korean standardized drug classification codes. Results: A total of 1,276,331 patients were analyzed in the sample of the elderly population, of which 532,075 (41.7%) were male and 744,256 (58.3%) were female. The patients have the higher risk of dementia in the older, women, and lower socioeconomically status. Cerebral infarction and ischemic heart disease increases risk of dementia. Patients taking statins, angiotensin converting enzyme inhibitor (ACEI) or angiotensin II receptor antagonists (ARB) showed low incidence of dementia. Conclusion: This study has been shown that ACEI, ARB, and statin drugs may associate with lower incidence of Alzheimer's and other dementia except vascular dementia.

Background and Purpose: Analyzing brain amyloid positron emission tomography (PET) images to access the occurrence of β-amyloid (Aβ) deposition in Alzheimer's patients requires much time and effort from physicians, while the variation of each interpreter may differ. For these reasons, a machine learning model was developed using a convolutional neural network (CNN) as an objective decision to classify the Aβ positive and Aβ negative status from brain amyloid PET images. Methods: A total of 7,344 PET images of 144 subjects were used in this study. The 18F-florbetaben PET was administered to all participants, and the criteria for differentiating Aβ positive and Aβ negative state was based on brain amyloid plaque load score (BAPL) that depended on the visual assessment of PET images by the physicians. We applied the CNN algorithm trained in batches of 51 PET images per subject directory from 2 classes: Aβ positive and Aβ negative states, based on the BAPL scores. Results: The binary classification of the model average performance matrices was evaluated after 40 epochs of three trials based on test datasets. The model accuracy for classifying Aβ positivity and Aβ negativity was (95.00±0.02) in the test dataset. The sensitivity and specificity were (96.00±0.02) and (94.00±0.02), respectively, with an area under the curve of (87.00±0.03). Conclusions: Based on this study, the designed CNN model has the potential to be used clinically to screen amyloid PET images.
본 논문에서는 특이값 분해(SVD: singular value decomposition)에 의한 기저 스펙트럼의 합성을 통해 혈소판 라만 스펙트럼에서 알츠하이머병(AD: Alzheimer's disease)을 검출하는 방법을 제안하였다. AD가 유도된 형질 전환 실험용 쥐의 혈소판에서 측정한 라만 스펙트럼은 가산 잡음과 배경 잡음의 제거와 정규화로 구성된 전처리 과정을 수행한다. 각 데이터 행렬의 열벡터는 AD와 정상(NR: normal)의 라만 스펙트럼으로 구성한다. 이 데이터 행렬을 SVD로 분해한 다음 각 행렬의 열벡터 12개를 AD와 NR의 기저 스펙트럼으로 결정한다. 분류 과정은 각 클래스의 기저 스펙트럼을 선형 합성한 스펙트럼과 분류 스펙트럼의 평균제곱근오차(root mean square error)가 최소인 클래스를 선택하는 것으로 완료된다. 278개의 혈소판 라만 스펙트럼을 사용한 실험에 따르면 제안한 방법의 평균 분류율은 약 97.6%로 주성분 분석(principle components analysis)으로 추출한 특징에 MLP(multi-layer perceptron)를 이용한 경우보다 약 6.1% 정도의 우수한 성능을 보였다. 이 결과에서 SVD에 의한 기저 스펙트럼이 혈소판 라만 스펙트럼에서 AD의 검출에 적합하게 사용될 수 있음을 확인하였다.

There is an emerging interest in brain functional connectivity (FC) based on functional Magnetic Resonance Imaging in Alzheimer's disease (AD) studies. The complex and high-dimensional structure of FC makes it challenging to explore the association between altered connectivity and AD susceptibility. We develop a pipeline to refine FC as proper covariates in a penalized logistic regression model and classify normal and AD susceptible groups. Three different quantification methods are proposed for FC refinement. One of the methods is dimension reduction based on common component analysis (CCA), which is employed to address the limitations of the other methods. We applied the proposed pipeline to the Alzheimer's Disease Neuroimaging Initiative (ADNI) data and deduced pathogenic FC biomarkers associated with AD susceptibility. The refined FC biomarkers were related to brain regions for cognition, stimuli processing, and sensorimotor skills. We also demonstrated that a model using CCA performed better than others in terms of classification performance and goodness-of-fit.
