Alteration of Runt-related Transcription Factor 3 Gene Expression and Biologic Behavior of Esophageal Carcinoma TE-1 Cells after 5-Azacytidine Intervention

  • 5-Azacytidine (5-azaC) was originally identified as an anticancer drug (NSC102876) which can cause hypomethylation of tumor suppressor genes. To assess its effects on runt-related transcription factor 3 (RUNX3), expression levels and the promoter methylation status of the RUNX3 gene were assessed. We also investigated alteration of biologic behavior of esophageal carcinoma TE-1 cells. MTT assays showed 5-azaC inhibited the proliferation of TE-1 cells in a time and dose-dependent way. Although other genes could be demethylated after 5-azaC intervention, we focused on RUNX3 gene in this study. The expression level of RUNX3 mRNA increased significantly in TE-1 cells after treatment with 5-azaC at hypotoxic levels. RT-PCR showed 5-azaC at $50{\mu}M$ had the highest RUNX3-induction activity. Methylation-specific PCR indicated that 5-azaC induced RUNX3 expression through demethylation. Migration and invasion of TE-1 cells were inhibited by 5-azaC, along with growth of Eca109 xenografts in nude mice. In conclusion, we demonstrate that the RUNX3 gene can be reactivated by the demethylation reagent 5-azaC, which inhibits the proliferation, migration and invasion of esophageal carcinoma TE-1 cells.

A Study Related to Adolescent Students' School Uniform Behavior and Attitude toward Appearance (청소년의 교복행동과 외모에 대한 태도와의 관련 연구)

  • This study examined the relativity between school uniform behavior(especially attitude toward school uniform, school uniform satisfaction, school uniform alteration) and attitude toward appearance in the adolescence. From November to December in 2007, 898 middle school and high school students from four schools in Jeollabuk-do Province were analyzed and the influence of relevant variables was understood to gather basic information about the fashion life in the adolescence. This study used SPSS 11.5 for Windows Program to conduct frequency analysis, factor analysis, reliability analysis, ${\chi}^2$-test, t-test, One-way ANOVA, Duncan's multiple comparison test, and Pearson's correlation. The following summarizes the findings of this study: The results of analysing the factors to the response attitude toward uniforms and attitude toward appearance and clothing attitude emerged four dimensions(fashion, symbolism, cleanliness, alternation), three dimensions(needs value conformity toward appearance). In attitude toward uniform, female students were more actively altering their uniforms than male students and middle school students were more sensitive to trends than high school students. However, high school students were more aware of the cleanliness and alteration of uniforms. Overall, most students were not very satisfied with their uniforms (design, color, texture). When students' attitude toward appearance and demographical characteristics were examined, it was found that female students were more aware of desire, value, and conformity in appearances than male students. By age, it was found that high school students were more aware than middle school students. Also, students receiving KRW 30,000 or more for monthly allowance were more aware than others who receive a lower amount between KRW 10,000 and KRW 20,000. Therefore, most students(62.1%) have experiences in altering uniforms. Especially, more female students and more high school students had such experiences. Uniform alteration is more related to attitude to appearance. In other words, students who choose to alter their uniforms are highly aware of desire, value, and conformity toward appearance. Students who are satisfied with all factors regarding attitude toward uniform, except for alteration, did not alter their uniforms that much. In addition, when the relativity between students' attitude toward appearance with uniform was considered, students that are not very satisfied with their uniforms were more aware of desire, value, and conformity toward appearance. In conclusion, it was found that students' attitude toward appearance and school uniform behavior are closely related and their interests in appearance lead to alteration of uniforms, the clothing in which they spend most time of the day. From now on, students' opinions shall be considered when selecting or designing school uniforms.

Fashion consumer's environmental awareness, pursuing values in disposal, sustainable fashion consumption attitude and fashion disposal behavior (패션 소비자의 환경의식, 처분 추구가치, 지속가능한 패션소비태도와 패션처분행동에 관한 연구)

  • This study identified fashion consumers disposal behavior and analyzed the effects of consumers pursuing values in disposal, environmental awareness, and sustainable fashion consumption attitudes in regards to fashion disposal behavior. A survey questionnaire was developed and data were obtained from 460 consumers in their 20's to 50's in Korea who had experienced fashion disposal behaviors during last 12 months. As a result, there were four different fashion disposal behaviors such as economical, practical, and social disposal as well as hoarding behaviors. Consumers pursuing values in disposal affected fashion disposal behavior. Practical and economical values had positively impacted economical disposal and hoarding behaviors. While hedonic value had a negative impact on economical disposal behavior, it had a positive impact on social disposal behavior. Also, environmental-social values had positively impacted practical and social disposal behaviors. Fashion-related environmental knowledge had positively impacted economical and practical disposal behaviors and PCE affected social disposal behavior, while environmental concerns had a negative impact on economical disposal behavior. Consumers attitude toward used-fashion items, fashion recycling, and fashion innovativeness affected all of fashion disposal behaviors. Although hoarding behavior has been an under researched area, the finding implied that hoarding behavior was affected by consumer's pursuing value in disposal and sustainable consumption attitude. Also, environmental-social values and attitudes toward used-fashion items would induce practical disposal behavior such as reuse by alteration or reform. Consumers economical and hedonic values can promote donations or exchange/resale of unwanted fashion items, which can lead to sustainable consumption.

A comparison of two differential methods for nutrition education in elementary school: lecture- and experience-based learning program

  • BACKGROUND/OBJECTIVES: This research was conducted to compare lecture- and experience-based methods of nutritional education as well as provide fundamental data for developing an effective nutritional education program in elementary schools. SUBJECTS/METHODS: A total of 110 students in three elementary schools in Jeollanam-do were recruited and randomly distributed in lecture- and experience-based groups. The effects of education on students' dietary knowledge, dietary behaviors, and dietary habits were analyzed using a pre/post-test. RESULTS: Lecture- and experience-based methods did not significantly alter total scores for dietary knowledge in any group, although lecture-based method led to improvement for some detailed questions. In the experience-based group, subjects showed significant alteration of dietary behaviors, whereas lecture-based method showed alteration of dietary habits. CONCLUSIONS: These outcomes suggest that lecture- and experience-based methods led to differential improvement of students' dietary habits, behaviors, and knowledge. To obtain better nutritional education results, both lectures and experiential activities need to be considered.

Dynamic behavior of clayey sand over a wide range using dynamic triaxial and resonant column tests

  • Deformations in soils induced by dynamic loads cause damage to the structures above the soil layers. It is important for geotechnical engineering practice that how the soil behaves due to repeated loads and the necessary precautions to be taken accordingly. Turkey is one of the most important seismic regions in Europe and earthquake studies to be conducted in this area are intended to reduce the damage as a result of taking the necessary measures. To determine the properties of soils under dynamic loads, stress-controlled dynamic triaxial and resonant column tests can be performed. In this study, these experiments were implemented in the laboratory on the clayey sand soil samples obtained from Bilecik Söğüt. To evaluate the effects of the confining pressure and rate of loading on the dynamic behavior of soils, samples were dynamically loaded by different rates at varying confining pressures. As a result, the changes in stress-strain properties of soils under dynamic loads were investigated. The alteration in behavior in terms of modulus reduction and damping ratios was obtained to vary a lot with the change of the lateral pressure on soil along with the frequency of the load.

Treatment and Management of Conversion Disorder (전환장애의 치료와 관리)

  • Conversion disorder is a psychologically produced alteration or loss of physical functioning suggestive of a physical disorder. Conversion symptoms are often superimposed on organic disease and can be overlooked. Psychological techniques are central to the management include the following: avoiding confrontation with the patients; avoiding reinforcement or trivializing the symptoms; reviewing results of tests and exams and creating an expectation of recovery; educating the patient before a treatment is begun; evaluating the patient's emotional adjustment and considering it at a treatment; using caution in labeling the condition; considering referral for psychotherapy; establishing particularly a treatment plan and making a definite treatment program; adjusting patient' s environment; letting participate a family at appropriate time; developing a reinforcement program for a treatment of chronic symptoms; developing a home program for outpatients. Use behavior therapy reinforcement may be helpful with more chronic or resistant symptoms, especially when there is a history of vague or excessive somatic complaints or significant secondary gain.

An Empirical Study on the Housing Affordability of Multi-Family Dwellers

  • The purpose of this study is to investigate the housing affordability of multi-family dwelling in Seoul and to systemize the housing expenditure patterns and housing affordability according to household and housing characteristics. Housing expenditures are basically composed of the payments of four major items. including utilities. home alteration and interior furnishing. home mortgage debt. and monthly rent. Housing affordability index as operationally defined in this study is Housing Expenditure-to-Income Ratio (HEIR). The ratios were computed for each of the four housing expenditure items. Data for 465 multi-family dwellers were gathered from the structured questionnaire. The results showed the differences between household and housing characteristics on housing expenditure levels and HEIR. In addition. this differences had influence on future housing choice behavior. The analysis on the variables of housing choice behavior showed the result that user cost considering capital gains were less important. comparing those of early 1990's.

Effects of Enriched Enviroment on Expression of BDNF and Motor Performence after Alcohol-induced Brain Injury in Neonatal Rats (신생흰쥐의 알콜성 뇌손상 후 환경강화가 BDNF 발현 및 운동수행력에 미치는 영향)

  • Alcohol exposure during development leads to significant long-term neurobehavior dysfunction and central nervous system alteration. The purpose of this study was to determine the effects of enriched enviroment in developmental period through motor behavior test and expression of BDNF. Neonatal rat exposed to alcohol on postnatal days 4 through 10 were studied. Female Sprague-Dawley pups were assigned to two groups. Experimental group(EG) via 4.5 g kg-1day-1 of ethanol was housed in enriched enviornment for 9 weeks. The main result of this study were as follows: 1. There was significant difference in the mean of weight change between control and experimental group. 2. In motor behavior test, there was significant difference in the mean of weight change between control and experimental group. 3. Regarding the immunoreactivity of BDNF were higher appeared experimental group than control group. In conclusion, the present results reveals that enriched enviroment in developmental period is to be extremely useful in neuronal reprganization and motor behavior improvement after alcohol exposure.

Synthesis and Characterization of pH-sensitive and Self-oscillating IPN Hydrogel in a pH Oscillator (pH 진동계 안에서 pH 감응성 자기진동 IPN 하이드로젤의 합성과 분석)

  • A self-oscillating interpenetrating polymer network (IPN) poly(acrylic acid)/poly(ethylene glycol) (PAA/PEG) hydrogel was prepared by using radical polymerization with a two-step method. The IPN hydrogel was characterized by FTIR spectroscopy and morphological analysis. The results indicated that the chains of PEG and PAA twined to form porous structure which is beneficial to water molecules entering inside of the hydrogel. In addition, the pH-responsive behavior, salt sensitivity, swelling/de-swelling oscillatory behaviors and self-oscillation in a closed pH oscillator were also studied. The results showed that the prepared hydrogel exhibited pH-sensitivity, good swelling/de-swelling reversibility and excellent salt sensitivity. The self-oscillating behavior of swelling/de-swelling for the prepared hydrogel was caused by pH alteration coupled with the external media. This study may create a new possibility as biomaterial including new self-walking actuators and other related devices.

Enhanced Efficacy of the Commissural Transmission between Lateral Giants in the Sensitization of Crayfish Escape Behavior

  • Lateral giant (LG)-mediated escape response of crayfish is sensitized by natural traumatic events. Such sensitization has previously been shown to be associated with increased transmission between primary afferents and sensory interneurons at the cholinergic synapse of LG escape reflex circuit. In the present study, it was firstly investigated as to whether transmission is also altered at other synapses of the LG-escape reflex circuit by traumatic shock-induced sensitization. Evidence that traumatic shock also directly affects the excitability of lateral giants is now provided by the finding that traumatic shock produces a significant reduction of the time needed for LG to recruit its contralateral homologue, which is defined as commissural delay. Octopamine, a naturally occurring neuromodulator in the crayfish nerve cord, has also been shown to enhance transmission at the cholinergic synapse between primary afferents and sensory interneurons, and has been conjectured to mediate sensitization. Like traumatic shock, $octopamine\;(10^{-5}-5{\times}10^{-4}\;M)$ also enhanced the efficacy of commissural transmission between lateral giants, as indicated by a significant reduction of commissural delay. This effect was blocked by an octopamine antagonist phentolamine, suggesting a specific action of octopamine on the octopamine receptor present on LGs. These observations suggest that both traumatic shocks and octopamine may cause a rather broad alteration in the excitability of the crayfish nervous system that contributes to the sensitization of the LG escape response.
