마이크로파 용해장치를 활용한 토양 중 우라늄의 알파분광분석법 (Efficient Sample Digestion Method for Uranium Determination in Soil using Microwave Digestion for Alpha Spectrometry)

  • 토양 중 우라늄 동위원소의 분석을 위해서 알파분광분석법이 보편적으로 사용되고 있다. 다수의 토양시료를 분석할 경우 시료분해 시간을 단축함으로써 전체 분석시간을 효과적으로 줄일 수 있으며, 이를 위해 본 연구에서는 단시간에 시료를 완전 분해할 수 있는 마이크로파 용해장치를 활용하였다. IAEA-375를 분석 대상 토양으로 하여 용해되지 않은 시료의 양과 분석한 $^{234}U$$^{238}U$의 방사능 농도 값을 통해 시료분해 방법을 최적화 하였으며, 0.5 g의 토양시료에 최소 3 ml의 불산을 가했을 때 80분 내에 효과적으로 분해되는 것을 확인하였다. 이는 2~3일의 분해시간이 필요한 통상적인 방법(open vessel 및 closed vessel을 이용하는 방법)에 비해 시간을 단축 할 수 있어 효율적이며, 불산 사용을 최소화 하여 시료용해 시 발생할 수 있는 유해 물질의 발생 및 접촉을 줄일 수 있다는 장점이 있다.

DETERMINATION OF THE TRANSURANIC ELEMENTS INVENTORY IN HIGH BURNUP PWR SPENT FUEL SAMPLES BY ALPHA SPECTROMETRY

  • The contents of transuranic elements in high-burnup spent fuel samples were determined. The activity amounts of $^{238}Pu,\;^{239}Pu,\;^{240}Pu,\;^{241}Am,\;^{244}Cm\;and\;^{242}Cm$ were measured by alpha spectrometry using $^{242}Pu\;and\;^{243}Am$ as tracers, respectively. A spike addition method for $^{237}Np$ was established by an alpha and gamma spectrometry using $^{239}Np$ as a spike after the optimum conditions for the measurements of $^{237}Np\;and\;^{239}Np$, respectively, were obtained. A separation system using anion exchange chromatography and diethylhexylphosphoric acid extraction chromatography was applied for the separation of these elements. This method was applied to high-burnup spent nuclear fuel samples $(40{\sim}60GWD/MTU)$. The contents of the transuranic elements were compared with those by ORIGEN-2 code. Measurements and the calculations of the contents of the plutonium isotopes $^{238}Pu,\;^{239}Pu\;and\;^{240}Pu$ agreed to within 10% on average. The contents of $^{237}Np$ agreed to within approximately 5% except for one instance of a calculation, while those of $^{241}Am,\;^{244}Cm\;and\;^{242}Cm$ showed higher values by approximately 19%, 35% and 14% on average, respectively, compared to the calculations according to the burnup.

Comparative N-Linked Glycan Analysis of Wild-Type and α1,3-Galactosyltransferase Gene Knock-Out Pig Fibroblasts Using Mass Spectrometry Approaches

  • Carbohydrate antigens expressed on pig cells are considered to be major barriers in pig-to-human xenotransplantation. Even after ${\alpha}1,3$-galactosyltransferase gene knock-out (GalT-KO) pigs are generated, potential non-Gal antigens are still existed. However, to the best of our knowledge there is no extensive study analyzing N-glycans expressed on the GalT-KO pig tissues or cells. Here, we identified and quantified totally 47 N-glycans from wild-type (WT) and GalT-KO pig fibroblasts using mass spectrometry. First, our results confirmed the absence of galactose-alpha-1,3-galactose (${\alpha}$-Gal) residue in the GalT-KO pig cells. Interestingly, we showed that the level of overall fucosylated N-glycans from GalT-KO pig fibroblasts is much higher than from WT pig fibroblasts. Moreover, the relative quantity of the N-glycolylneuraminic acid (NeuGc) antigen is slightly higher in the GalT-KO pigs. Thus, this study will contribute to a better understanding of cellular glycan alterations on GalT-KO pigs for successful xenotransplantation.

Tertiary Matrices for the Analysis of Polyethylene Glycols Using MALDI-TOF MS

  • The effectiveness of tertiary matrices composed of the combination of three common matrices (dihydrobenzoic acid (DHB), ${\alpha}$-cyano-4-hydroxycinnamic acid (CHCA), and sinapinic acid (SA)) was compared with that of single or binary matrices in the analysis of polyethylene glycol (PEG) polymers ranging from 1400 to 10000 Da using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). A tertiary matrix of 2,5-DHB+CHCA+SA was the most effective in terms of S/N ratios. CHCA and CHCA+SA produced the highest S/N ratios among the single matrices and the binary matrices, respectively. The improvement observed when using a tertiary matrix in analyses of PEG polymers by MALDI-TOF MS is believed to be due to the uniform morphology of the MALDI sample spots and synergistic effects arising from the mixture of the three matrix materials.

A comparative study of different radiometric methodologies for the determination of 226Ra in water

  • An evaluation of various radiometric methods to analyze $^{226}Ra$ in water has been employed on a set of 10 standard solutions of different concentrations in the range of $1-10Bq/L^{-1}$. The analysis was carried out using well-established procedures by means of gamma-ray, alpha-particle and liquid scintillation spectrometry. The feasibility of the various methods has been quantified in terms of relative standard error and percentage error. Correlations between the various methods have been presented and discussed. In general, good agreement was found in the results of various methodologies, which assures the accuracy of the methods and allows for the validation of instrumentation and procedures. Of the different methods adopted here, a combined procedure for the determination of $^{226}Ra$ along with $^{228}Ra$ using Quantulus 1220 ultra-low level background liquid scintillation counting gave the most accurate results.

  • 소나무과의 리기다송(Pinus rigida Mill)과 적송(Pinus densiflora Sieb & Zucc)의 needle oil을 gas chromatography/mass spectrometry(GC/MS)로 분석하였다. 리기다송의 주요 성분은 ${\alpha}-pinene$, 1-hexen-3-ol formate, sabinene, ${\beta}-pinene,\;{\alpha}-terpieol$, ${\beta}-caryophyllene$ 등 이었으며, 적송은 ${\alpha}-pinene$, bornyl acetate, ${\beta}-pinene$, ${\beta}-hpellandrene$ 등이 주성분이었다. 적송은 리기다송에 비해 fresh, soft sweet하며 삼림을 연상시키는 효과가 컸는데, 리기다송에 bornyl acetate를 첨가하였더니 적송보다 더 강한 삼림향을 나타내었다.

  • The contents of transuranic elements ($^{237}Np$, $^{238}Pu$, $^{239}Pu$, $^{240}Pu$, $^{241}Am$, $^{244}Cm$, and $^{242}Cm$) in high-burnup spent fuel samples ($35.6{\sim}53.9\;GWd/MtU$) were determined by alpha spectrometry. Anion exchange chromatography and diethylhexyl phosphoric acid extraction chromatography were applied for the separation of these elements from the uranium matrix. The measured values of the nuclides were compared with ORIGEN-2 calculations. For plutonium, the measurements were higher than the calculations by about $2.6{\sim}32.7%$ on average according to each isotope, and those for americium and curium were also higher by about $35.9{\sim}63.1%$. However, for $^{237}Np$, the measurements were lower by about 52% on average for the samples.

Isolation and Structural Characterization of an Oligosaccharide Produced by Bacillus subtilis in a Maltose-Containing Medium

  • Among 116 bacterial strains isolated from Korean fermented foods, one strain (SS-76) was selected for producing new oligosaccharides in a basal medium containing maltose as the sole source of carbon. Upon morphological characterization using scanning electron microscopy, the cells of strain SS-76 appeared rod-shaped; subsequent 16S rRNA gene sequence analysis revealed that strain SS-76 was phylogenetically close to Bacillus subtilis. The main oligosaccharide fraction B extracted from the culture supernatant of B. subtilis SS-76 was purified by high performance liquid chromatography. Subsequent structural analysis revealed that this oligosaccharide consisted only of glucose, and methylation analysis indicated similar proportions of glucopyranosides in the 6-linkage, 4-linkage, and non-reducing terminal positions. Matrix-assisted laser-induced/ionization time-of-flight/mass spectrometry and electrospray ionization-based liquid chromatography-mass spectrometry/mass spectrometry analyses suggested that this oligosaccharide consisted of a trisaccharide unit with 1,6- and 1,4-glycosidic linkages. The anomeric signals in the $^1H$-nuclear magnetic resonance spectrum corresponded to ${\alpha}$-anomeric configurations, and the trisaccharide was finally identified as panose (${\alpha}$-D-glucopyranosyl-1,6-${\alpha}$-D-glucopyranosyl-1,4-D-glucose). These results suggest that B. subtilis SS-76 converts maltose into panose; strain SS-76 may thus find industrial application in the production of panose.

  • 라일락 꽃 향기 성분의 특성을 고체상 포집 용매추출법과 기체크로마토그래피 이온 포집형 질량분석법으로 연구하였다. 라일락 품종에 따라 향기성분의 조성이 현저한 차이를 보였다. 백색 라일락 꽃의 경우 벤즈알데히드, 페닐 아세트알데히드 및 알파-파네센인데 비하여 보라색 라일락 꽃은 벤즈알데히드, 알파-피넨 및 오시멘이 주된 향기성분으로 발견되었다. 라일락 꽃 향기성분 중 알파-피넨의 거울상 이성질체의 본질을 분석한 결과 ( )형태임을 알 수 있었다.

  • 참쑥(Artemisia lavandulaefolia DC)의 정유를 gas chromatogaphy/mass spectrometry(GC/MS)로 정성분석하고, 저장조건을 달리하여 정유성분의 변화를 살펴본 결과, 다음과 같은 결론을 얻었다. 1. 186 이상의 화합물이 검출되었으며, 그 중 1,8-cineol, camphor, ${\beta}-thujone$, caryophyllene, borneol, coumarin, linalool, terpineol, sabinene, 7-methoxy coumarin ${\alpha}-copaene$, humulene과phytol이 중요성분이었다. Artemisia species에서는 지금까지 밝혀진 바 없는 3,6,6-trimethyl norpinanol, ${\beta}-farnesene$, 7-methoxy coumarin, ${\alpha}-curcumene$ 등이 검출되었다. 2. 참쑥의 저장에 따른 색과 변화를 조사한 결과, 온도에 의해 갈색화와 성분의 증감이 있었다. 즉, 냉암소에 보관된 시료는 변화가 거의 없었으나, 상온방치한 시료는 Air와 질소에 상관없이 모두 변화가 있었다. 1,8-cineol, limonene, 2,2,4-trimethyl 1,3-dioxolane-4-ethanol, terpinene-4-ol 등은 감소하였으며, ${\beta}-phellandrene$, benzyl benzoate, broneol, ${\alpha}-terpineol$, humulene 등은 증가하였다.
