Dietary Aloe Vera Gel Powder and Extract Inhibit Azoxymethane-induced Colorectal Aberrant Crypt Foci in Mice Fed a High-fat Diet

  • Aloe vera gel exhibits protective effects against insulin resistance as well as lipid-lowering and anti-diabetic effects. The anti-diabetic compounds in this gel were identified as Aloe-sterols. Aloe vera gel extract (AVGE) containing Aloe-sterols has recently been produced using a new procedure. We previously reported that AVGE reduced large-sized intestinal polyps in Apc-deficient Min mice fed a high fat diet (HFD), suggesting that Aloe vera gel may protect against colorectal cancer. In the present study, we examined the effects of Aloe vera gel powder (AVGP) and AVGE on azoxymethane-induced colorectal preneoplastic aberrant crypt foci (ACF) in mice fed a HFD. Male C57BL/6J mice were given a normal diet (ND), HFD, HFD containing 0.5% carboxymethyl cellulose solution, which was used as a solvent for AVGE (HFDC), HFD containing 3% or 1% AVGP, and HFDC containing 0.0125% (H-) or 0.00375% (L-) AVGE. The number of ACF was significantly lower in mice given 3% AVGP and H-AVGE than in those given HFD or HFDC alone. Moreover, 3% AVGP, H-AVGE and L-AVGE significantly decreased the mean Ki-67 labeling index, assessed as a measure of cell proliferation in the colonic mucosa. In addition, hepatic phase II enzyme glutathione S-transferase mRNA levels were higher in the H-AVGE group than in the HFDC group. These results suggest that both AVGP and AVGE may have chemopreventive effects on colorectal carcinogenesis under the HFD condition. Furthermore, the concentration of Aloe-sterols was similar between 3% AVGP and H-AVGE, suggesting that Aloe-sterols were the main active ingredients in this experiment.

한외여과 알로에 농축액의 Urease 저해 및 무기물 응집 활성 (Urease Inhibition and Flocculating Activity of Concentrated Aloe vera Gel by Using Ultrafiltration Process)

  • For physiological function of aloe concentrate by ultrafiltration (UF) process, jack bean urease inhibitory activity and bentonite flocculating activity of UF aloe concentrate was investigated and compared with fresh aloe gel. Urease inhibitory activity of UF aloe concentrate ranged from 87 to 90% in 1 mL sample. Also, urease inhibitory activity of UF aloe concentrate increased about 10% by heat treatment showing the heat stability. From Lineweaver-Burk plot for UF aloe concentrate, urease inhibition pattern indicated general non-competitive inhibition. From flocculation test of UF aloe concentrate about 1% (w/v) bentonite suspension, maximum flocclulating activity of 97% was obtained at 0.5 mL addition of UF aloe concentrate/ 5 ml bentonite suspension. However, flocculating activity of 81% was obtained at 1 mL addition of UF aloe concentrate/ 5 mL bentonite suspension, which was typical flocculating behavior of polymers with re-dispersion at overdose area. FT-IR spectra of UF aloe concentrate showed the characteristic patterns of $\beta$-binding polysaccharide and less deacetylation indicating higher level of bioactive polysaccharide content.

복수암 생쥐와 인체 암세포에 대한 알로에의 항암 작용 (Anticancer Effects of Aloe on Sarcoma 180 in ICR Mouse and on Human Cancer Cell Lines)

  • Anticancer effects of Aloe on sarcoma 180 in ICR mouse or human cancer cells were determined. Sarcoma 180 cells were inoculated subcutaneously into male ICR mouse to determine effect of Aloe on tumor gowth, or inoculated intraperitoneally into male ICR mouse to determine effect of Aloe on life span prolongation, followed by oral administration of Aloe vera(10 mg/kg/day, 50 mg/kg/day) or Aloe arborescens(10 mg/kg/day, 100 mg/kg/day) once a day for 14 days. The administration of Aloe vera or Aloe arborescens did not suppress tumor growh. However the life span of ICR mouse was prolonged to 19%(p<0.05), 22%(p<0.05) and 32%(p<0.05) by administration of Aloe vera 10 mg/kg/day, Aloe vera 50 mg/kg/day, and Aloe arborescens 100 mg/kg/day, respectively. To determine anticancer effect of Aloe in vitro, Aloe extract was added to the culture of human gastric cancer cells(SNU-1) and colorectal cancer cells(SNU-C2A), and concentration of Aloe to inhibit cancer cell growth was determined using MTT(3-[ 4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) cytotoxicity assay. High $ID_{50}$ values of Aloe vera and Aloe arborescens against gastric cancer cell line(SNU-1) and colorectal cancer cell line(SNU-C2A) suggest that Aloe gel does not have anticancer effect on these specific human cancer cells although high concentration of Aloe inhibited growth of human cancer cells significantly.

삼투탈수 알로에 건조제품의 구조적 및 물리화학적 특성 (Structural and Physicochemical Properties of Dried Aloe Vera Gel Using DIS (Dewatering & Impregnation Soaking) Process)

  • The structural and physicochemical properties of dried aloe vera gel by DIS (dewatering impregnation soaking) process under optimum conditions were investigated. FT-IR spectra for dried samples of DIS aloes showed the typical patterns of standard aloe polysaccharide, and surface structures by SEM (scanning electron microscopy) were similar to a gel-like structure. In case of physicochemical properties of dried aloe samples by DIS process, solubilities and swelling powers of control (not osmotic treated aloe), DIS (S) and DIS (G), samples treated by osmotic solution of 60% sucrose/0.25% NaCl and 50% glucose/0.5% NaCl, were 48.3-57.3% and 8.3-11.7%, respectively, showing no significant differences among samples, but swelling power of DIS (PEG), sample treated by using 50% polyethylene glycol as an osmotic agent was about 5 times higher that of control. Also, water holding capacities of control, DIS (S) and DIS (G) were similar to each other, but that of DIS (PEG) was about 5 times higher that of control. Oil holding capacities of control and DIS aloes maintained the 50.9-86.4% levels of water holding capacities showing no significant differences among samples. Rehydration ratio of DIS (PEG) aloes were significantly dependent on the temperature of rehydrated solvent (water), and rehydration ratio of not-fileted aloe was about two folds higher than that of fileted aloe.

An Optimized Methodology to Observe Internal Microstructures of Aloe vera by Cryo-Scanning Electron Microscope

  • Aloe vera has been used in the pharmaceutical, food and cosmetic industry for its therapeutic properties. However, there are not many current studies on the microstructure of A. vera compared to studies on the chemical constituents and health efficacy of A. vera. Therefore, we compared the morphology of an A. vera leaf using an optical microscope, a conventional scanning electron microscope (SEM) and a cryo-SEM. Especially, this study focused on observing the gel in the inner leaf of A. vera, which is challenging using standard imaging techniques. We found that cryo-SEM is most suitable method for the observation of highly hydrated biomaterials such as A. vera without removing moisture in samples. In addition, we found the optimal analytical conditions of cryo-SEM. The sublimation conditions of $-100^{\circ}C$ and 10 minutes possibly enable the surface of the inner leaf of A. vera to be observed in their "near life-like" state with retaining moisture. The experiment was repeated with A. arborescens and A. saponaria to confirm the feasibility of the conditions. The results of this study can be applied towards the basic research of aloe and further extend previous knowledge about the surface structures of the various succulent plants.

Extraction and Characterization of Aloe Glucomannan: Assessing Its Flocculation Capability

  • Acemannan, a highly acetylated glucomannan, was extracted from fresh Aloe vera leaves by ethanol fractionation, resulting in a concentration increase of more than threefold. The presence of acemannan was confirmed using FTIR and 1H NMR analysis, revealing an average molecular weight of 780 kDa. The flocculating activity of the fractionated aloe gel polysaccharide was assessed through settling tests in a 1% (w/v) bentonite suspension. The results demonstrated that the aloe polysaccharide exhibited remarkable stability within a temperature range of 20~70 ℃. The maximal flocculation rate at different pH levels ranged from 93% to 97%, with an optimal dose for maximum flocculation rate between 0.25 mg/mL. Notably, the minimum dose required for flocculation was achieved at a pH of 3, attributed to the compression of electrostatic repulsion on the surface of bentonite particles. However, the flocs obtained under acidic conditions were less dense and compact, exhibiting lower sedimentation velocity compared to those formed under neutral and alkaline pH conditions. Additionally, the addition of salt showed a slight synergistic effect on flocculation, significantly enhancing the sedimentation velocity. This investigation highlights the potential of Aloe vera polysaccharide as a natural and edible flocculant, offering promising applications in various industries.

알로에 베라 겔 가공부산물로서의 섬유질 분획의 성분 및 물리화학적 특성 (Physicochemical Properties of Fibrous Material Fraction from By-product of Aloe vera Gel Processing)

  • The fibrous material fraction as a by-product from the commercial aloe vera gel processing was obtained and freeze dried. The physicochemical characteristics such as the proximate composition, crystalline/surface structures and several physical functionalities including the water holding capacity (WHC), swelling capacity (SW), oil holding capacity (OHC), emulsion/foam properties and viscosity properties of this powdered sample (100 mesh) were investigated and analyzed by comparison with commercial $\alpha$-cellulose as a reference sample. The total dietary fiber content of powdered sample was very high as much as 87.5%, and the insoluble dietary and soluble dietary fiber content ratios were 77.6 and 22.4%, respectively. The FT-IR spectrum of powdered sample showed a typical polysaccharide property and exhibited a x-ray diffraction pattern for cellulose III and IV like structure. SW (8.24${\pm}$0.15 mL/g), WHC(6.40${\pm}$0.19 g water/g solid) and OHC(10.32${\pm}$0.29 g oil/g solid) of freeze dried aloe cellulose were about 3.3, 1.4 and 2 times higher than those of commercial $\alpha$-cellulose, respectively. Aloe cellulose (~2%, w/v) alone had no foam capacity while improved the foam stability of protein solution (1% albumin+0.5% $CaCl_{2}$) by factor of 300%. Emulsion capacity of 2%(w/v) aloe cellulose was about 70% level of 0.5%(w/v) xanthan gum, but its emulsion stability was about 1.2 times higher than that of xanthan gum. Also, aloe cellulose containing CMC (carboxyl methyl cellulose) of 0.3%(w/v) showed a very good dispersity. Aloe cellulose dispersion of above 1%(w/v) exhibited higher pseudoplasticity and concentration dependence than those of $\alpha$-cellulose dispersion, indicating the viscosity properties for new potential usage such as an excellent thickening agent.

Aloe vera peel 추출물에 의한 구강염증 저해 효과의 효소학적 평가 (Enzymological Evaluation of Oral Inflammation inhibitory activity by Aloe vera peel extract)

  • Aloe vera 껍질로부터 구내염등 항염증제재를 개발하기 위해 항염증 활성이 있는 분획을 분리, 동정하고, 염증과 관련된 효소활성에 대한 저해효과를 조사하였다. Aloe vera 껍질의 추출물을 용매에 따라 분획하였고, 활성이 우수한 ethylacetate층을 silica gel column chromatography 및 preparative thin layer chromatography를 통해 두 가지 활성물질을 정제하였다. 정제된 두 가지 물질은 UV spectrum과 FT-IR, $^1H-NMR$, 및 Mass spectrum 분석을 통해 aloe-emodin과 barbaloin으로 동정하였다. Aloe vera 분획물의 hyaluronidase 활성저해를 측정한 결과는 aloe-emodin은 $40\;{\mu}g/mL$, barbaloin은 $70\;{\mu}g/mL$$IC_{50}$값을 나타내었다. Leucko-cyte elastase 활성 저해를 측정한 결과는 aloe-emodin은 $50\;{\mu}g/mL$, barbaolin은 $60\;{\mu}g/mL$$IC_{50}$값을 나타내었다. Collagenase 활성 저해를 측정한 결과는 aloe-emodin은 $40\;{\mu}g/mL$, barbaloin은 $60\;{\mu}g/mL$$IC_{50}$값을 나타내었다. 또한 prostaglandin endoperoxide synthase 활성 저해를 측정한 결과는 aloe-emodin은 $40\;{\mu}g/mL$, barbaloin은 $70\;{\mu}g/mL$$IC_{50}$값을 나타내었다. 쥐를 이용한 carrageenan족 부종 억제효과는 aloe-emodin의 경우 100mg/kg 투여량으로 52.9%의 억제효과를 나타내었으며, 이 억제효과는 indomethacin의 약 1/10이었으며, aspirin과는 유사한 활성을 보였다. 반면 barbaloin의 경우 100mg/kg의 투여량으로 74.9%의 억제효과를 나타내었으며 이 억제효과는 indomethacin의 약 1/5이었으며, aspirin의 약 1.5배 효과를 나타내어 우수한 항염증 활성을 갖는 것으로 판단된다. 인체치은세포에 대한 독성은 aloe-emodin보다 barbaloin이 적게 나타났으며 이 두 가지 물질은 1 및 $5\;{\mu}g/mL$농도에서는 독성이 전혀 나타나지 않으나 10 및 $20\;{\mu}g/mL$에서는 독성이 있는 것으로 나타났다. 그러나 상용되고 있는chlorhexidin에 비해서는 유의적으로 독성이 적은 것으로 나타났다. 이상의 결과로 보아 aloe-emodin 과 barbaloin은 항우식 및 항염증 활성이 우수할 뿐만 아니라 치은세포의 독성이 비교적 낮아 구강병 예방제재로의 개발 가능성이 우수하다고 사료된다.

알로에 첨가 식빵의 품질 특성 (Quality Characteristics of Bread with Added Aloe(Aloe vera Linne))

  • The quality characteristics of breads made by adding levels of 30%, 60%, and 100% aloe gel were investigated. The 60% aloe gel sample had a larger dough volume than the control dough. Also, the loaf volume and specific volume of the bread made with 60% aloe gel were larger than those of the control bread. The pasting temperature increased gradually with the increasing amounts of substituted aloe gel as compared to wheat flour with water added$(66.5{\pm}12^{\circ}C)$; the wheat flour with 100% aloe gel had a pasting temperature of $90.7{\pm}1.1^{\circ}C$. Peak viscosity decreased gradually with increasing amounts of aloe gel when compared to the wheat flour with water added. Setback also was decreased gradually with the increasing amounts of aloe gel as compared to the wheat flour with added water. From the setback decrease it is suggested that the aloe controlled retrogradation of the bread during short-term storage. The crumb color of the bread made with the aloe gel was not significantly different to that of the control bread, and the color of the crumb was yellow-white. Bread hardness decreased gradually with increasing amounts of the substituted aloe gel. However, bread gumminess and chewiness increased gradually with increasing amounts of aloe gel. The springiness of the bread made with 100% aloe gel was lower than that of the control bread. With regard to flavor and taste, the bread made with 100% aloe gel produced a green-like odor and had a bitter taste. The mouth feel of the breads made with aloe gel was considered soft and moist. The overall acceptabilities of the breads made with 30% and 60% aloe gel were not significantly different from the control bread, but the bread made with 100% aloe gel had a green smell and bitter taste. Yet accordingly, the results indicate that functional and health products with improved quality could be developed by adding aloe gel to breads and cakes.

In vitro Angiogenic Activity of Aloe vera Gel on Calf Pulmonary Artery Endothelial (CPAE) Cells

  • Angiogenic activity of Aloe vera gel was investigated by in vitro assay. We obtained the most active fraction from dichloromethane extract of Aloe vera gel by partitioning between hexane and 90% aqueous methanol. The most active fraction (F3) increased the proliferation of calf pulmonary artery endothelial (CPAE) cells. In addition, F3 fraction induced CPAE cells to invade type I collagen gel and form capillary-like tube through in vitro angiogenesis assay, and increased the invasion of CPAE cells into matrigel through in vitro invasion assay. Furthermore, the effect on the MRNA expression of proteolytic enzymes which are key participants in the regulation of extracellular matrix degradation was investigated by northern blot analysis. F3 fraction enhanced mRNA expression of urokinase-type plasminogen activator (u-PA), matrix metalloproteinase-2 (MMP-2), and membrane-type MMP (MT-MMP) in CPAE cells whereas the expression of plasminogen activator inhibitory (PAl-1) mRNA was not changed.
