  • Garlic (Allium sativum) has been used as a general food and a remedy in Oriental for a long time. Since garlic compounds have been also shown to inhibit growth of tumors and to modulate the activity of carcinogenesis, the effects of allicin on growth and survival in human gastric epithelial cells were evaluated by cell viability, cell cycle analysis and DNA fragmentation. Protein levels of cytochrome C, Bcl-xL, Bax and AIF were detected by Western blotting. Effects of recombinant VacA on caspase proteases activity were also determined. (omitted)

Determination of Formaldehyde in Cigarette Smoke and Inhibitory Effect of Plant Volatile Extracts on the Formation of Formaldehyde

  • Formaldehyde (FA) is a carcinogenic compound present in cigarette smoke. In this study, the amount of formaldehyde was analyzed in 5 kinds of cigarettes and the inhibitory effect of plant volatile extracts on the formation of FA was investigated. After extraction of the cigarette sample, FA was converted into its thiazolidine derivatives by reaction with cysteamine, and then measured using a gas chromatography-nitrogen phosphorus detector (GC-NPD). The concentrations of FA in cigarette smoke were found between 138.24 and $217.82{\mu}mol/g$ cigarette smoke. Extracts isolated from Welsh onion (Allium cepa L.), garlic (Allium sativum L.), crown daisy (Chrylsanthemum coronarium L.), green pepper (Capsicum annuum L.), and sesame dropwort (Oenanthe javanica DC) were used for analyzing their inhibitory effects on the formation of FA. The inhibitory effects of extracts of Welsh onion, garlic, crown daisy, green pepper, and sesame dropwort on the formation of FA were 64, 47, 38, 47, and 19%, respectively.

Effects of garlic Allium sativum on the immune responses of olive flounder Paralichthys olivaceus (마늘, Allium sativum이 넙치, Paralichthys olivaceus의 면역반응에 미치는 영향)

  • This study was aimed to investigate the effects of injection of garlic, Allium sativum, extract and immersion in garlic juice on the nonspecific immunity and the resistance against the artificial infection of Streptococcus iniae and Edwardsiella tarda of the olive flounder, Paralichthys olivaceus. The nonspecific immune mechanisms were assessed in terms of lysozyme activity, nitroblue-tetrazolium (NBT) assay and superoxide dismutase (SOD) activity etc. Relative percent survival (RPS) was assessed by the challenge with S. iniae BS10 or E. tarda KE-1. Almost of the garlic extract injected groups showed the enhanced level of the tested nonspecific immune factors. In the challenge test with S. iniae and E. tarda, RPS of 5% garlic extract pre-injected group was much higher than that of any other tested groups, respectively. Almost of the garlic juice immersion tested groups exhibited strengthened nonspecific immune defence factors, lysozyme activity, the number of lymphocytes and neutrophils, NBT reduction and SOD activity in kidney. In the challenge with S. iniae and E. tarda, RPS in the 0.25 g/L of garlic juice immersed group was much higher than any other tested groups, respectively. The results suggest that the garlic extract and juice would be effective to enhance the nonspecific immunity and protective ability of olive flounder against fish disease such as S. iniae and E. tarda.

Changes in Volatile Sulfur Compounds of Garlic (Allium sativum L.) under Various Drying Temperatures (건조 온도에 따른 마늘의 휘발성 함황성분의 변화)

  • The volatile sulfur compounds in raw and dried garlic (Allium sativum L.) under various temperatures (50, 60, 70 and $80^{\circ}C$ ) were separated, identified by GC and GC/MS. In hexane extracts of raw garlic, 8 volatile sulfur compounds were identified and 11 compounds were identified in steam distillation extracts. The volatile sulfur compounds of raw and dried garlic were mostly composed of diallyl disulfide, diallyl trisulfide, allyl methyl trisulfide. According to the increase of drying temperature total amounts of volatile sulfur compounds in the hexane extracts decreased to $40{\sim}75$$, and in the steam distillation extracts decreased to $10{\sim}20%$. The amounts of diallyl disulfide, allyl methyl trisulfide decreased more than other volatile sulfur compounds, but diallyl trisulfide comparatively decreased lower.

Allexivirus Transmitted by Eriophyid Mites in Garlic Plants

  • Viruses in garlic plants (Allium sativum L.) have accumulated and evolved over generations, resulting in serious consequences for the garlic trade around the world. These viral epidemics are also known to be caused by aphids and eriophyid mites (Aceria tulipae) carrying Potyviruses, Carlaviruses, and Allexiviruses. However, little is known about viral epidemics in garlic plants caused by eriophyid mites. Therefore, this study investigated the infection of garlic plants with Allexiviruses by eriophyid mites. When healthy garlic plants were cocultured with eriophyid mites, the leaves of the garlic plants developed yellow mosaic strips and became distorted. In extracts from the eriophyid mites, Allexiviruses were observed using immunosorbent electron microscopy (ISEM). From an immunoblot analysis, coat proteins against an Allexivirus garlic-virus antiserum were clearly identified in purified extracts from collected viral-infected garlic plants, eriophyid mites, and garlic plants infected by eriophyid mites. A new strain of GarV-B was isolated and named GarV-B Korea isolate 1 (GarV-B1). The ORF1 and ORF2 in GarV-B1 contained a typical viral helicase, RNA-directed RNA polymerase (RdRp), and triple gene block protein (TGBp) for viral movement between cells. The newly identified GarV-B1 was phylogenetically grouped with GarV-C and GarV-X in the Allexivirus genus. All the results in this study demonstrated that eriophyid mites are a transmitter insect species for Allexiviruses.

Yield change of seed bulb according to annual field culture after induced meristem culture in garlic (Allium sativum L.) (마늘 생장점 조직배양 후 포장에서의 년차별 생육 및 수량특성 변화)

  • In this research, we surveyed the yield change of garlic cultivation with seed bulbs according to annual field culture after induced meristem culture. From 2007 to 2013, yearly produced seed bulbs from $2^{nd}$ to $8^{th}$ generation were cultivated in the Muan field. Over the generations, the general growth characteristics such as the plant height, leaf number, leaf length and stem diameter were all decreased. When we compared the conventional seed bulbs and TCR ones, the plant height and stem diameter showed bigger difference. And the TCR seed bulbs showed a week late harvest time against the conventional ones. The yield of $2^{nd}$ generation was 72.3 g and 57.1 g for the $8^{th}$ generation. However every generation showed higher yield than conventional seed garlic's ones as 43.9 g. The yield index showed the same trend as $2^{nd}-8^{th}$ to 63% to 21%. After the tissue culture of garlics, the viral infections (OYDV and GSV) of TCR was far less than that of conventional ones. In case of LYSV, the infection ratio was 6.7% at $2^{nd}$ generation and 26% at $8^{th}$. However, the ratio was far less than that (80%) of the conventional ones.

Effects of Storage Gas Concentrations on the Qualities of Garlic(Allium sativum L.) Bulb during CA Storage (통마늘의 CA저장 중 저장기체 농도에 따른 품질변화)

  • This experiment was conducted to measure the physiological qualities and the chemical compositions of garlic bulb (Allium sativum L) stored in air and controlled atmosphere at 0$\^{C}$ for 4months. There were no significant differences in physiological qualities such as soluble solid, dry weight and firmness by O$_2$ and CO$_2$ concentrations during 4 months of storage. The surface color of garlic cloves changed toward more dark direction in Hunter color surface with storage. CA storage inhibited the sprout growth by 50% and had no significant difference in root growth. Garlic stored in CA showed lower level of pungency, especially CA of low oxygen concentration (1%) showed the lowest level of pyruvate. Garlic stored in high CO$_2$ concentration(20%) showed the lowest level of fructan concentrations and the highest free sugar concentrations.

Effect of Garlic Extracts with Extraction Conditions on Antioxidant and Anticancer Activity (추출조건에 따른 마늘 추출물의 항산화 및 항암활성 효과)

  • The purpose of this study was to investigate the effect of garlic(Allium sativum L.) extracts with extraction conditions on antioxidant and anticancer activity. The extracts prepared for garlic by hot temperature extraction (HG), low temperature extraction (LG), UMPM extraction (UG), fermentation (FG) and black garlic hot temperature (BG) method. Content of total polyphenol compound was the BG higher than other extracts. The EDA (electron donating ability) and SOD-like activity was increased in dose-dependent manners, and the activity of BG and UM was significantly higher than LG and FG. We examined cytotoxicity, nitric oxide production of Raw 264.7 cell and inhibition of HT 1080 cell by MTT assay. All extracts does not have any toxic effects in macrophages(Raw 264.7). And UG inhibited the production of nitrite in Raw 264.7 cells activated with LPS. The antitumor effects of LG and UG on HT 1080 cell was indicated a significantly inhibition activity. These results suggested that UG (UMPM extraction of garlic) have activities of antioxidant, anticancer effects.

Effect of Soaking in KNO3 or K3PO4 Solution and Sowing Date of Bulbils on the Seed Bulbs Production of Garlic (Allium sativum L.) (마늘주아(珠芽)에 대한 KNO3및 K3PO4 처리(處理)와 파종시기(播種時期)가 종구(種球) 생산(生産)에 미치는 영향(影響))

  • This study was carried out to investigate seed bulbs production of 'Namdo' garlic (Allium sativum L.) by treating with 200 mM $K_3PO_4$ and 200 mM $KNO_3$ solution to bulbils. The growth characteristics of garlic plants derived from bulbils sown on 3 different dates were compared to those of plants derived from non-treated bulbils. Time required for 50% shoot emergence ($T_{50}$) was shortened as the sowing dates delayed. However, the final emergence rate was increased in the early sowing dates, and was high when treated with $K_3PO_4$ and sown on Oct. 10. The % clove differentiation of bulb was not significantly affected by sowing date and chemicals treatment of the bulbils. In all treatments, % clove differentiation was greater than 85%. The average bulb weight was increased with earlier sowing. Bulbils treated with $K_3PO_4$ and sown on Oct. 10 exhibited the best growth and yield. which was 27% higher compared to the non-treated bulbils.
