Control of Algal Blooms in Eutrophic Water Using Porous Dolomite Granules

  • The use of aluminum-based coagulants in water pretreatment is being carefully considered because aluminum exposure is a risk factor for the onset of Alzheimer's disease. Lightly burned-dolomite kiln dust (LB-DKD) was evaluated as an alternative coagulant because it contains high levels of the healthful minerals calcium and magnesium. An organic pore forming agent (OPFA) was incorporated to prepare porous granules after OPFA removal through a thermal decomposition process. A spray drying method was used to produce uniform and reproducible spherical granules with low density, since fine dolomite particles have irregular agglomeration behavior in the hydration reaction. The use of fine dolomite powder and different porosity granules led to a visible color change in raw algae (RA) containing water, from dark green to transparent colorlessness. Also, dolomite powders and granules exhibited a mean removal efficiency of 48.3% in total nitrogen (T-N), a gradual increase in the removal efficiency of total phosphorus (T-P) as granule porosity increased. We demonstrate that porous dolomite granules can improve the settling time and water quality in summer seasons for the emergent treatment of excessive algal blooms in eutrophic water.

연못을 이용한 동절기 인공습지 오수처리수의 추가 처리 (Pond System for Further Polishing of Constructed Wetland Effluent during Winter Season)

  • Pilot study was performed to examine the feasibility of the pond system for further polishing of treatment wetland effluent from December 2000 to June 2001. The wetland system used for the experiment was highly effective to treat the sewage during the growing season, but it was less effective and its effluent was still high to discharge to the receiving water body. Therefore, the wetland effluent may need further treatment to prevent water quality degradation. Pond system could be used to hold and further polish the wetland effluent during the winter season and ots feasibility was evaluated in this study. Additional water quality improvement was apparent in the pond system during winter season, and the pond effluent could be good enough to meet the effluent water quality standards if it is properly managed. Timing of the pond effluent discharge appears to be critical for pond system management because it is a closed system and whole water quality constituents are affected by physical, chemical, and biological pond environments. Once algae started to grow in mid-April, constituents in the pond water column interact each other actively and its control becomes more complicated. Therefore, upper layer of the pond water column which is clearer than the lower layer my need be discharged in March right after ice cover melted. In the experiment, water quality of the upper water column was markedly clear in March than ant other times probably because of freezing-thawing effect. The remaining lower water column could be further treated by natural purification as temperature goes up or diluted with better quality of wetland effluent for appropriate water uses. This study demonstrated the feasibility of pond system for subsequent management of wetland effluent during the winter season, however, more study is needed for field application.

Isolation of Anagonistic Fungi Associated with the Lichens Distributed in Southern Parts of Korea

  • Lichen-forming (LFF) or lichenicolous fungi (LCF) were isolated from the lichens collected at‘Backwoon’mountain area,‘Chiri’mountain area and‘Sorok’island in the southern regions of Korea and were screened for antagonistic efficacy against several phyto-pathogenic fungi. Symbiotic algae-free LFF and LCF were isolated by the following methods: I) discharged spores (ascospores), II) macerated thallus suspension and III) direct use of thallus fragments. Among 58 isolates obtained from 34 lichens, 8 isolates showed antifungal activity against Rhizoctonia solani. Antifungal activities of the strongest antagonistic isolate (LB9810) originated from the thallus of Parmelia quercina lichen were evaluated against 15 phyto-pathogenic fungi. When crude methanol extract of mycelia of the LB8910 isolate was employed at the rate of 0.5% (v/w), fungal growth of Magnaporthe grisea and Rhizoctonia solani was severly and Rhizoctonia solani was severly inhibited as much as approximately 60% compared to control. Growth of various food-borne same extract. The extract was successively partitioned with n-hexane, ethyl acetate and n-butanol. n-Hexane fraction displayed the strongest antifungal activities against R. solani. The LB9810 isolate was finally identified as Fusarium equiseti (Corda) Sacc., which has not been reported as LFF or LCF yet. Therefore, it is very likely that F. equiseti isolated it the study was originated from the contaminants associated with thallus fragments rather than from LFF or LCF.

Anti-Inflammatory Effect of 3-Bromo-4,5-Dihydroxybenzaldehyde, a Component of Polysiphonia morrowii, In Vivo and In Vitro

  • 3-Bromo-4,5-dihydroxybenzaldehyde (BDB) is a natural bromophenol compound that is most commonly isolated from red algae. The present study was designed to investigate the anti-inflammatory properties of BDB on atopic dermatitis (AD) in mice induced by 2,4-dinitrochlorobenzene (DNCB) and on lipopolysaccharide (LPS)-stimulated murine macrophages. BDB treatment (100 mg/kg) resulted in suppression of the development of AD symptoms compared with the control treatment (induction-only), as demonstrated by reduced immunoglobulin E levels in serum, smaller lymph nodes with reduced thickness and length, a decrease in ear edema, and reduced levels of inflammatory cell infiltration in the ears. In RAW 264.7 murine macrophages, BDB (12.5, 25, 50, and $100{\mu}M$) suppressed the production of interleukin-6, a proinflammatory cytokine, in a dose-dependent manner. BDB also had an inhibitory effect on the phosphorylation of nuclear factor kappa-light-chain-enhancer of activated B cells (NF-${\kappa}B$) and signal transducer and activator of transcription 1 (STAT1; Tyr 701), two major signaling molecules involved in cellular inflammation. Taken together, the results show that BDB treatment alleviates inflammatory responses in an atopic dermatitis mouse model and RAW 264.7 macrophages. These results suggest that BDB may be a useful therapeutic strategy for treating conditions involving allergic inflammation such as atopic dermatitis.

Changes in Phosphorus and Sediment Oxygen Demand in Coastal Sediments Promoted by Functionalized Oyster Shell Powder as an Oxygen Release Compound

  • In this study, we performed a sediment elution experiment to evaluate water quality in terms of phosphorus, as influenced by the dissolved oxygen consumed by sediments. Three separate model column treatments, namely, raw, calcined, and sonicated oyster shell powders, were used in this experiment. Essential phosphorus fractions were examined to verify their roles in nutrient release from sediment based on correlation analyses. When treated with calcined or sonicated oyster shell powder, the sediment-water interface became "less anaerobic," thereby producing conditions conducive to partial oxidation and activities of aerobic bacteria. Sediment Oxygen Demand (SOD) was found to be closely correlated with the growth of algae, which confirmed an intermittent input of organic biomass at the sediment surface. SOD was positively correlated with exchangeable and loosely adsorbed phosphorus and organic phosphorus, owing to the accumulation of unbound algal biomass-derived phosphates in sediment, whereas it was negatively correlated with ferric iron-bound phosphorus or calcium fluorapatite-bound phosphorus, which were present in the form of "insoluble" complexes, thereby facilitating the free migration of sulfate-reducing bacteria or limiting the release from complexes, depending on applied local conditions. PCR-denaturing gradient gel electrophoresis revealed that iron-reducing bacteria were the dominant species in control and non-calcined oyster shell columns, whereas certain sulfur-oxidizing bacteria were identified in the column treated with calcined oyster powder.

Impact of Ecklonia stolonifera extract on in vitro ruminal fermentation characteristics, methanogenesis, and microbial populations

  • Objective: This study was conducted to evaluate the effects of Ecklonia stolonifera (E. stolonifera) extract addition on in vitro ruminal fermentation characteristics, methanogenesis and microbial populations. Methods: One cannulated Holstein cow ($450{\pm}30kg$) consuming timothy hay and a commercial concentrate (60:40, w/w) twice daily (09:00 and 17:00) at 2% of body weight with free access to water and mineral block were used as rumen fluid donors. In vitro fermentation experiment, with timothy hay as substrate, was conducted for up to 72 h, with E. stolonifera extract added to achieve final concentration 1%, 3%, and 5% on timothy hay basis. Results: Administration of E. stolonifera extract to a ruminant fluid-artificial saliva mixture in vitro increased the total gas production. Unexpectedly, E. stolonifera extracts appeared to increase both methane emissions and hydrogen production, which is contrasts with previous observations with brown algae extracts used under in vitro fermentation conditions. Interestingly, real-time polymerase chain reaction indicated that as compared with the untreated control the ciliate-associated methanogen and Fibrobacter succinogenes populations decreased, whereas the Ruminococcus flavefaciens population increased as a result of E. stolonifera extract supplementation. Conclusion: E. stolonifera showed no detrimental effect on rumen fermentation characteristics and microbial population. Through these results E. stolonifera has potential as a viable feed supplement to ruminants.

Effects of Ecklonia cava Extract on Neuronal Damage and Apoptosis in PC-12 Cells against Oxidative Stress

  • Marine algae (seaweed) encompass numerous groups of multicellular organisms with various shapes, sizes, and colors, and serve as important sources of natural bioactive substances. The brown alga Ecklonia cava Kjellman, an edible seaweed, contains many bioactives such as phlorotannins and fucoidans. Here, we evaluated the antioxidative, neuroprotective, and anti-apoptotic effects of E. cava extract (ECE), E. cava phlorotannin-rich extract (ECPE), and the phlorotannin dieckol on neuronal PC-12 cells. The antioxidant capacities of ECPE and ECE were 1,711.5 and 1,050.4 mg vitamin C equivalents/g in the ABTS assay and 704.0 and 474.6 mg vitamin C equivalents/g in the DPPH assay, respectively. The dieckol content of ECPE (58.99 mg/g) was approximately 60% higher than that of ECE (36.97 mg/g). Treatment of PC-12 cells with ECPE and ECE increased cell viability in a dose-dependent manner. Intracellular oxidative stress in PC-12 cells due to ECPE and ECE decreased dose-independently by up to 63% and 47%, respectively, compared with the stress control (323%). ECPE reduced the production of the pro-apoptotic proteins Bax and caspase-3 more effectively than ECE. Early and late apoptosis in PC-12 cells were more effectively decreased by ECPE than ECE treatments. From the results obtained in this study, we concluded that ECPE, which is rich in phlorotannins, including the marker compound dieckol, may be applied to the development of functional materials for improving cognition and memory.

수문기상예측자료를 활용한 대청호 Chl-a 3개월 선행예측연구 (A Study on the 3-month Prior Prediction of Chl-a Concentraion in the Daechong Lake using Hydrometeorological Forecasting Data)

  • 최근 반복되고 있는 녹조는 수질관리에 가장 큰 과제로서 대두되고 있다. 현재 환경부에서는 7일 단위의 선행수질예측을 통한 수질예보를 수행하고 있으나, 선제적인 조치를 위해서 좀 더 장기간의 수질예측이 필요한 시점이다. 이에 본 연구에서는 수질예보의 보완자료로서 대청호의 Chl-a 농도를 3개월 선행예측하기 위한 방법론을 제안하고 그 적용성을 검토하고자 한다. 이를 위하여 대청호의 수질자동측정망 자료와 ECMWF의 수문기상예측자료를 수집하였으며 각 시계열 자료의 특성을 분석하였다. 대청호의 Chl-a 농도와의 상관 및 웨이블릿 분석을 바탕으로 수문기상입력인자를 결정하고 지연시간을 가지는 NARX모형을 이용하여 대청호의 Chl-a에 대한 3개월 선행예측 모형을 구축하였으며, 결과에 대한 비교분석을 통하여 모형의 적용성을 제시하였다.

Assessment of the potential of algae phycobiliprotein nanoliposome for extending the shelf life of common carp burgers during refrigerated storage

  • This study is focused on the effect of phycobiliprotein extraction of Gracilaria on the quality of common carp burgers, and the application of nanoliposomes containing pigment in the improvement of its antimicrobial and antioxidant activity of burgers during refrigerated storage in 18 days. Burgers were incorporated with phycobiliprotein and liposomal phycobiliprotein (2.5% and 5% w/w), and their chemical and microbial changes in terms of pH, peroxide value (PV), thiobarbituric acid (TBA), total volatile basic nitrogen (TVB-N), total viable counts (TVC), psychrotrophic bacterial counts (PTC), and sensory characteristics were evaluated. Results presented a nanoliposome size of about 515.5 nm with capable encapsulation efficiency (83.98%). Our results showed non-encapsulated phycobiliprotein could delay the deterioration of common carp burgers, as a reduction in PV, TBA, and TVB-N, TVC, and PTC values in burgers treated with free and nano encapsulated phycobiliprotein. Moreover, the potential of phycobiliprotein was improved when it was encapsulated into chitosan coated liposomes. Burgers treated with 5% nanoliposomes displayed the lowest amount of lipid oxidation and microbial deterioration in comparison to others during storage. According to chemical, microbial and sensory evaluation, the shelf life of common carp burgers was increased in samples treated with encapsulated phycobiliprotein at 2.5% and 5%, as compared to the control (p ≤ 0.05).

다시마 추출물의 항산화와 염증 조절 효과 (Antioxidant and Immunomodulatory Effects of Laminaria japonica Water Extract)

  • Laminaria japonica is a type of brown algae widely consumed in Asian countries and contains many essential nutrients and exhibits anti-obesity, antioxidant, and anti-inflammatory effects. In this study, the antioxidant and immunomodulatory effects of a Laminaria japonica water extract (LJE) were investigated using an in vitro model. Mean total polyphenol content of LJE was 2.16±0.11 ㎍ GAE/mg, and LJE dose-dependently inhibited ABTS radical activity but did not scavenge DPPH radicals. In addition, LJE enhanced nitric oxide (NO) production and upregulated the mRNA expressions of proinflammatory cytokines (i.e., tumor necrosis factor-α and interleukin-6) in RAW 264.7 cells. On the other hand, LJE inhibited NO production and downregulated proinflammatory cytokine mRNA levels in endotoxin-stimulated RAW 264.7 cells. Thus, our data show that LJE has moderate antioxidant activity and biphasic immunomodulatory effects on RAW 264.7 cells. In summary, the study indicates that LJE has potential therapeutic use as a novel biphasic immuno-modulator.
