Ensiled Green Tea Waste as Partial Replacement for Soybean Meal and Alfalfa Hay in Lactating Cows

  • The purpose of this study was to evaluate the effects of protein supplementation of green tea waste (GTW) on the performance of lactating cows. Another aim was to increase resource utilization and to eliminate any environmental negative impact from the tea waste. GTW from a beverage company was ensiled at a low pH (<4.0) and high acetic acid and lactic acid concentration, and it contained high crude protein (CP, 34.8%), total extractable tannins (TET, 9.2%) and condensed tannin (CT, 1.7%). Two experiments were conducted to investigate the palatability and performance in lactating cows fed GTW. In the palatability trial, three lactating cows were allocated to three dietary treatments in a 3$\times$3 Latin square design. The animals were offered a total mixed ration (TMR) including GTW at rates of 0, 2.5 and 5.0% on a dry matter (DM) basis. Total DM intake was not different among the treatments. In the performance trial, four lactating cows were used in a 2$\times$2 Latin square design with a 3 week sampling period. GTW was incorporated into TMR at a rate of 5.0% on a DM and 10.0% on a CP basis. Thus GTW replaced alfalfa hay and soybean meal at a level of 25.0% on a DM. DM and CP intake were not affected by the inclusion of GTW, whereas TET and CT intake were significantly increased (p<0.001). Milk production, milk composition and the efficiency of milk production were not altered by the GTW inclusion. Although ruminal pH and VFA, and blood urea nitrogen were not changed, ruminal $NH_{3}-N$ and plasma total cholesterol were relatively low in the GTW group, but not significantly different. The excretion of urinary purine derivatives and estimated MN supply were also not significantly affected by GTW treatment. It is therefore concluded that GTW can be used as a protein source without any detrimental effects on the performance of lactating cows.

  • 동물원에서 사육중이던 8년령의 수컷 그랜트얼룩말이 10일간 침울, 식욕부진, 탈수, 산통의 임상증상을 보이다 폐사하였다. 부검시 하행결장과 직장의 연접부에 무게 1,660 g의 장결석(가로 13.5 cm $\times$ 세로 8 cm)이 발견되었다. 발견된 결석을 주사전자현미경과 에너지 분산형 X선 분광기로 분석한 결과 결석의 중심부는 $SiO_2$ 가 가장 많이 검출되었고, 그 외층은 마그네슘과 인이 가장 많이 검출되었다. 장결석의 생성은 미네랄 성분이 다량 함유된 알팔파 건초의 급여와 깊은 연관이 있는데, 본 예의 장결석증 또한 알팔파 건초의 급여에 기인한 것으로 판단되었다.

섬유소 공급원이 면양의 반추위 pH, Buffering Capacity 및 소화율에 미치는 영향 (Effects of Fiber Sources on Ruminal pH, Buffering Capacity and Digestibility in Sheep)

  • 본 연구는 섬유소공급원 7종, alfalfa hay cube (AHC), corn cob (CC), corn silage (CS), cotton seed hull (CSH), peanut hull (PHL), rice straw(RS), sugar cane bagasse(SCB)의 반추위에서의 반추활동, pH, 완충능력과 전소화기관 내 소화율을 알아보고자 실시하였다. 공시 동물은 평균체중 37.1kg인 Corridale종 수 면양 5두 (3두는 반추위 fistula 부착)를 사용하였으며, 실험기간동안 공시동물에게 급여한 사료는 각각의 조사료원 $45\%$와 corn-based 사료 $55\%$의 비율로 하여 1일 2회 (09:00, 21:00) 총 800g(dry matter basis)을 급여하였다. Ruminal pH는 급여 후 4시간까지는 처리구 간에 차이가 없었지만 8시간 후 타 조사료원$(6.24\~6.61)$과 RS(5.78)간에는 차이가 있었다(p<0.05). 반추위 내 완충능력은 SCB구, CC구와 비교하여 CS구에서 사료급여 후 0시간(p<0.05), 2시간(p<0.05)에 유의한 차이를 나타내었고, 사료급여 후 12시간 경과 시 AHC구를 제외한 CC구, CS구, SCB구와 RS구 보다 PHL구(p<0.01)에서 높게 나타났다. DM 소화율은 CS구에서 $78.19\%$로 타처리보다 유의하게 높았으며(p<0.05), DNF 소화율은 CC구, CS구, CSH구와 RS구가 다른 조사료원 급여구보다 높게 나타났다. (p<0.001).

Effects of Feeding High Forage Diets and Supplemental Fat on Feed Intake and Lactation Performance in Dairy Cows

  • Fifty mid-lactation Holstein cows were used in a six-week feeding trial to study effects of high-forage, high-fat diets on dry matter intake and production performance. Cows were divided into 10 replicates, each consisting of five cows. Each cow was assigned to a control (diet 1) or one of the four experimental diets (high-forage (75%), high-fat (7.5%) (diet 2); high-forage, medium-fat (5%) (diet 3); medium forage (65%), high-fat (diet 4); medium-forage, medium-fat (diet 5)), or a control diet containing about 50% forage and 2% fat. All diets were isonitrogenous (17.7% crude protein). The forage mixture consisted of 20% alfalfa hay, 40% alfalfa haylage, and 40% com silage. Supplemental fat included 80% rumen-protected fat and 20% yellow grease. Dry matter intake was decreased (p<0.01) in cows fed experimental diets (18.4, 20.9, 19.9, and 22.6 kg for cows fed diets 1-4, respectively vs. 27.5 kg for cows fed the control diet). Daily milk production was lower (p<0.05) for cows consuming experimental diets (30.5, 31.3, 31.0, and 32.5 kg for cows fed greater for cows consuming experimental diets (1.74, 1.55, 1.60, and 1.53 kg milk/kg dry matter intake for cows fed diets 1-4, respectively, vs. 1.26 kg milk/kg dry matter intake for cows fed the control diet).

Effects of Forage Sources on Rumen Fermentation Characteristics, Performance, and Microbial Protein Synthesis in Midlactation Cows

  • Eight multiparous Holstein cows ($632{\pm}12$ kg BW; $135{\pm}16$ DIM) were used in a replicated $4{\times}4$ Latin square design to evaluate the effects of forage sources on rumen fermentation characteristics, performance, and microbial protein (MCP) synthesis. The forage portion of the diets contained alfalfa hay (AH), oat hay (OH), Leymus chinensis (LC), or rice straw (RS) as the primary source of fiber. Diets were isonitrogenous and isocaloric, and cows were fed four corn silages based total mixed rations with equivalent nonfiber carbohydrate (NFC) and forage neutral detergent fiber (NDF). Dry matter intake was not affected by the source of dietary forages, ranging from 18.83 to 19.20 kg/d, consequently, milk yield was similar among diets. Because of the numerical differences in milk fat and milk protein concentrations, 4% FCM and ECM yields were unchanged (p>0.05). Mean rumen pH, NH3-N content, and concentrations of volatile fatty acids in the rumen fluid were not affected by the treatments (p>0.05). Dietary treatments did not affect the total tract apparent digestibility of dry matter, organic matter, and crude protein (p>0.05); however, digestibility of NDF and acid detergent fiber in RS diet was higher compared with AH, OH, and LC diets (p<0.05). Total purine derivative excretion was higher in cows fed AH, OH, and LC diets compared with those fed RS diet (p<0.05), consequently, estimated MCP synthesis was 124.35 g/d higher in cows fed AH diet compared with those fed RS diet (p<0.05). The results indicated that cows fed AH, OH, LC, and RS diets with an equivalent forage NDF and NFC have no unfavourable effect on the ruminal fermentation and productive parameters.

Effects of microbial enzymes on starch and hemicellulose degradation in total mixed ration silages

  • Objective: This study investigated the association of enzyme-producing microbes and their enzymes with starch and hemicellulose degradation during fermentation of total mixed ration (TMR) silage. Methods: The TMRs were prepared with soybean curd residue, alfalfa hay (ATMR) or Leymus chinensis hay (LTMR), corn meal, soybean meal, vitamin-mineral supplements, and salt at a ratio of 25:40:30:4:0.5:0.5 on a dry matter basis. Laboratory-scale bag silos were randomly opened after 1, 3, 7, 14, 28, and 56 days of ensiling and subjected to analyses of fermentation quality, carbohydrates loss, microbial amylase and hemicellulase activities, succession of dominant amylolytic or hemicellulolytic microbes, and their microbial and enzymatic properties. Results: Both ATMR and LTMR silages were well preserved, with low pH and high lactic acid concentrations. In addition to the substantial loss of water soluble carbohydrates, loss of starch and hemicellulose was also observed in both TMR silages with prolonged ensiling. The microbial amylase activity remained detectable throughout the ensiling in both TMR silages, whereas the microbial hemicellulase activity progressively decreased until it was inactive at day 14 post-ensiling in both TMR silages. During the early stage of fermentation, the main amylase-producing microbes were Bacillus amyloliquefaciens (B. amyloliquefaciens), B. cereus, B. licheniformis, and B. subtilis in ATMR silage and B. flexus, B. licheniformis, and Paenibacillus xylanexedens (P. xylanexedens) in LTMR silage, whereas Enterococcus faecium was closely associated with starch hydrolysis at the later stage of fermentation in both TMR silages. B. amyloliquefaciens, B. licheniformis, and B. subtilis and B. licheniformis, B. pumilus, and P. xylanexedens were the main source of microbial hemicellulase during the early stage of fermentation in ATMR and LTMR silages, respectively. Conclusion: The microbial amylase contributes to starch hydrolysis during the ensiling process in both TMR silages, whereas the microbial hemicellulase participates in the hemicellulose degradation only at the early stage of ensiling.

조사료 자원의 단백질 분획 및 Buffer 추출이 In Vitro 발효 성상, 분해율 및 Gas 생성량에 미치는 효과 (Effect of Protein Fractionation and Buffer Solubility of Forage Sources on In Vitro Fermentation Characteristics, Degradability and Gas Production)

  • 본 시험에서 건초(티머시, 알팔파 및 클라인)와 짚류(톨페스큐 및 볏짚)의 buffer 용해도와 단백질 분획이 실시되었으며, 조사료 자원의 buffer 추출이 $In$ $vitro$ 발효 성상, 분해율 및 가스($CO_2$$CH_4$) 생성량에 미치는 효과를 조사하였다. 다른 조사료에 비해 총 단백질 중 buffer 가용성 조단백질과 A fraction은 알팔파 건초에서 각각 61% 및 41.77%로 가장 높았으며 볏짚에서 가장 낮았다(각각 42.8% 및 19.78%). 총단백질 중 B1 fraction은 조사된 조사료간 비교적 큰 차이를 보이지 않았으나 B2 fraction에서는 다른 조사료(6.34~8.85%)에 비하여 톨페스큐짚(10.05%) 및 클라인 건초(12.34)%에서 다소 높은 수준을 보였다. 총 단백질 중 B3 fraction이 차지하는 비율은 톨페스큐짚에서 38.49%로 가장 높았으나 다른 조사료 자원 간에는 큰 차이가 없었으며, C fraction의 경우 볏짚에서 가장 높은 비율(15.05%)을 보였다. 모든 사료에서 배양 개시 후 3시간(P<0.01) 및 6시간(P<0.05)에서 buffer 추출 전에 비해 추출 후 배양액의 pH가 증가되었으며, 배양 6시간(P<0.05) 및 12시간(P<0.001)에서 다른 사료에 비해 티모시 건초 및 알팔파 건초로부터의 pH가 낮았다. 배양액의 암모니아 농도는 모든 배양시간에서 가용성 물질의 추출 전 후에 다른조사료에 비해 알팔파 건초에서 가장 높았으나 모든 사료의 추출효과는 배양 3시간(P<0.01)에서만 나타났다. 배양액의 총 VFA 농도는 배양 24시간까지 알팔파 건초에서 가장 높았던 반면 톨페스큐짚과 볏짚에서 가장 낮았다. 또한 모든 조사료에서 buffer 추출 전에 비하여 추출후에 총 VFA 농도가 감소되었다(P<0.01~P<0.001). Acetic acid ($C_2$)의 조성 비율에서는 배양 6시간까지 추출 전에 더 높았으나(P<0.001) 사료 간 차이는 없었다. Propionic acid ($C_3$) 조성 비율 역시 배양 개시 후 3, 24 및 48시간(P<0.001)에서 추출 전에 더 높았으며, 6 및 12 시간의 배양액에서 대부분 건초(티모시, 알팔파 및 클라인)와 짚류(톨페스큐짚 및 볏짚) 간 차이가 있는 것으로 조사되었다(P<0.05). 그러나 butyric acid ($C_4$) 조성비율의 경우 대부분의 배양시간에서 사료 간 차이는 없었다. 건물에서의 분해율 관련 parameter 중 a 값은 조사된 전체 조사료에서 buffer 추출 전이 추출 후에 비해서 높았으며(P<0.001), 다른 조사료에 비해 톨페스큐짚과 볏짚에서 크게 낮았다(P<0.05). 또한 b 값의 경우 역시 추출 전에 비해 추출 후에서 현저히 낮았으나(P<0.001) 사료 간 차이는 없었다. 볏짚을 제외한 조사료에서 추출 후에 비해 추출 전의 건물 유효분해율(EDDM)이 더 높았다(P<0.001). 조단백질에서의 a, b 및 c 값은 추출 전에 비해 추출 후에서 현저히 낮았으나(P<0.05) 사료 간 차이는 없었다. 조단백질 유효분해율(EDCP)에서는 다른 조사료 종류에 비해 톨페스큐짚과 볏짚에서 낮았다(P<0.05). 한편, NDF의 경우 a 값과 b 값(P<0.01) 및 NDF 유효분해율(EDNDF, P<0.001)은 추출 후에 비해 추출 전에 더 높았으나(P<0.01) 사료 간 차이는 보이지 않았다. 반추위미생물에 의해 사료분해과정 중 생성되는 $CO_2$ 량도 24시간 배양까지는 추출 전에 더 많았으며(P<0.05~P<0.001), 톨페스큐짚과 볏짚에 비해 건초 형태의 조사료로부터의 $CO_2$ 생성량이 더 많았다(P<0.05~P<0.01). 메탄가스($CH_4$) 생성량 역시 모든 배양시간에서 추출 전에 비해 추출 후에 크게 감소되었으며(P<0.01~P<0.001), 12~24시간을 제외하고는 짚류에 비해 건초에서 현저히 높은(P<0.05) 것으로 나타났다. 본 시험의 결과를 종합하면, 조사료 자원에 대한 buffer 용해도와 단백질의 분획이 $In$ $vitro$ VFA 농도와 분해율 및 gas ($CO_2$$CH_4$) 발생량 간 상호 밀접한 관계를 보이는 것으로 여겨진다. 이에 따라 조사료 이용 효율 개선을 위해 조사료자원에 대한 buffer 용해도와 단백질 분획을 반추동물 TMR 조제에 활용할 필요가 있는 것으로 여겨진다.

Comparison of In vivo and In vitro Techniques for Methane Production from Ruminant Diets

  • This study was conducted to compare the methane ($CH_4$) production estimated by in vivo (sulfur hexafluoride tracer technique ($SF_6$)) with that of two in vitro rumen simulation (RUSITEC) and gas production (IVGPT)) techniques. Four adult dry Holstein cows, aged $7.4{\pm}3.0$ years and weighing $697{\pm}70$ kg, were used for measuring methane production from five diets by the $SF_6$ technique. The experimental diets were alfalfa hay ($D_1$), corn silage + soybean meal (SBM) (910: 90, $D_2$), Italian rye grass hay +SBM (920: 80, $D_3$), rice straw +SBM (910: 90, $D_4$) and Sudan grass hay +SBM (920: 80, $D_5$). Each diet was individually fed to all 4 cows and 5 feeding studies of 17 d each were conducted to measure the methane production. In the RUSITEC, methane production was measured from triplicate vessels for each diet .In vitro gas production was measured for each of the diets in triplicate syringes. The gas produced after 24 and 48 h was recorded and gas samples were collected in vacuum vials and the methane production was calculated after correction for standard temperature and pressure (STP). Compared to the $SF_6$ technique, estimates of methane production using the RUSITEC were lower for all diets. Methane production estimated from 24 h in vitro gas production was higher (p<0.001) on $D_1$ as compared to that measured by $SF_6$, whereas on $D_2$ to $D_5$ it was lower. Compared to $SF_6$, methane production estimated from 48 h in vitro gas production was higher on all diets. However, methane estimated from the mean of the two measurement intervals (24+48 h/2) in IVGPT was very close to that of $SF_6$ (correlation 0.98), except on $D_1$. The results of our study confirmed that IVGPT is reflective of in vivo conditions, so that it could be used to generate a database on methane production potential of various ruminant diets and to examine strategies to modify methane emissions by ruminants.

Effect of Exogenous Fibrolytic Enzyme Application on the Microbial Attachment and Digestion of Barley Straw In vitro

  • The effects of exogenous fibrolytic enzymes (EFE; a mixture of two preparations from Trichoderma spp., with predominant xylanase and ${\beta}$-glucanase activities, respectively) on colonization and digestion of ground barley straw and alfalfa hay by Fibrobacter succinogenes S85 and Ruminococcus flavefaciens FD1 were studied in vitro. The two levels (28 and 280 ${\mu}g$/ml) of EFE tested and both bacteria were effective at digesting NDF of hay and straw. With both substrates, more NDF hydrolysis (p<0.01) was achieved with EFE alone at 280 than at 28 ${\mu}g$/ml. A synergistic effect (p<0.01) of F. succinogenes S85 and EFE on straw digestion was observed at 28 but not 280 ${\mu}g$/ml of EFE. Strain R. flavefaciens FD1 digested more (p<0.01) hay and straw with higher EFE than with lower or no EFE, but the effect was additive rather than synergistic. Included in the incubation medium, EFE showed potential to improve fibre digestion by cellulolytic ruminal bacteria. In a second batch culture experiment using mixed rumen microbes, DM disappearance (DMD), gas production and incorporation of $^{15}N$ into particle-associated microbial N ($^{15}N$-PAMN) were higher (p<0.001) with ammoniated (5% w/w; AS) than with native (S) ground barley straw. Application of EFE to the straws increased (p<0.001) DMD and gas production at 4 and 12 h, but not at 48 h of the incubation. EFE applied onto S increased (p<0.01) $^{15}N$-PAMN at 4 h only, but EFE on AS increased (p<0.001) $^{15}N$-PAMN at all time points. Prehydrolysis increased (p<0.01) DMD from both S and AS at 4 and 12 h, but reduced (p<0.01) $^{15}N$-PAMN in the early stage (4 h) of the incubation, as compared to non-prehydrolyzed samples. Application of EFE to barley straw increased rumen bacterial colonization of the substrate, but excessive hydrolytic action of EFE prior to incubation decreased it.

Use of Awamori-pressed Lees and Tofu Lees as Feed Ingredients for Growing Male Goats

  • Awamori is produced by fermenting steamed indica rice. Awamori-pressed lees is a by-product of the Awamori production process. Tofu lees is a by-product of the Tofu production process. Research was conducted to test if dried Awamori-pressed lees and Tofu lees can be used as a mixed feed ingredient for raising male goats. Eighteen male kids were divided into three groups of six animals (control feed group (CFG), Awamori-pressed lees mixed feed group (AMFG), Tofu lees mixed feed group (TMFG)). The CFG used feed containing 20% soybean meal as the main protein source, while the AMFG and TMFG used feed mixed with 20% dried Awamori-pressed lees or dried Tofu lees. The groups were fed mixed feed (volume to provide 100 g/d increase in body weight) and alfalfa hay cubes (2.0 kg/d) twice a day (10:00, 16:00). Klein grass hay and water was given ad libitum. Hay intake was measured at 10:00 and 16:00. Body weight and size measurements were taken once a month. At the end of the experiment, a blood sample was drawn from the jugular vein of each animal and the carcass characteristics, the physical and chemical characteristics of loin were analyzed. DCP and TDN intakes in AMFG and TMFG showed no significant difference to the CFG. Cumulative measurements of growth in body weight and size over the 10 mo period in the AMFG and TMFG were similar to the CFG. Blood parameter values were similar to those in normal goats. Dressing carcass weight and percentages, and total weight of meat in the AMFG were similar to that in the CFG, but smaller in the TMFG. The compressed meat juice ratio was higher in both the TMFG and AMFG than the CFG. While the fat in corn, Awamori-pressed lees, and Tofu lees contains more than 50% linoleic acid, the loin fat in both the AMFG and TMFG was very low in linoleic acid due to the increase in the content of oleic acid, stearic acid, and palmitic acid. This indicates that feeding on AMF and TMF does not inhibit hydrogenation by ruminal microorganisms. As in the CFG, the total essential and non-essential amino acids in the loin of the AMFG and TMFG were well balanced. Compared to the CFG, the AMFG and TMFG were high in taurine and carnosine. The results indicate dried Awamori-pressed lees and Tofu lees can be used as a feed ingredient for raising male goats.
