Anti-inflammatory activity of methanol extract isolated from stem bark of Albizia julibrissin

  • Albizia julibrissin (AJ) has been used widely as a traditional medicine. In macrophages nitric oxide (NO) is released as an inflammatory mediator and has been proposed to be an important modulator of many pathophysiological conditions including inflammation and carcinogenesis. In this study we have examined the NO inhibition effect of 85% methanol extracts of AJ in mouse macrophage. Lipopolysaccharide (LPS) has been reported to induce production of NO. Extracts of AJ (1, 10, $100{\mu}g/ml$) suppressed nitric oxide production in LPS-stimulated ($100{\mu}g/ml$) mouse (C57BL/6) macrophages and analyzed by ELISA. In addition, it also attenuated the expression of inflammatory products like Interleukin-6 (IL-6), cyclooxygenase-2 (COX-2), and inducible NOS (iNOS) as assessed by immunoblotting with specific antibodies. These results suggest that 85% methanol extracts of AJ would be useful in inflammatory diseases.

Effects of Albizia julibrissin Durazz through Suppression of Mitochondrial Fission and Apoptosis in Cisplatin-induced Acute Kidney Injury

  • Albizia julibrissin Durazz. (AJ; family Minosaceae) is widely distributed worldwide, and its stem bark has been used as a traditional herbal medicine. Acute kidney injury (AKI) is a clinical syndrome that results in sudden loss of renal function. This study aimed to investigate the effects of AJ against cisplatin-induced AKI using a human kidney proximal tubule epithelial cell line (HK-2) and cisplatin-treated mice. In vitro, cisplatin treatment increased apoptosis in HK-2 cells. However, AJ treatment decreased apoptosis of cisplatin-treated HK-2 cells. In vivo, cisplatin treatment accelerated renal injury by increasing the levels of renal injury markers, such as blood urea nitrogen, creatinine, kidney injury molecule 1, and neutrophil gelatinase-associated lipocalin, which were reversed by AJ treatment. Histopathologically, AJ treatment resulted in decreased renal damage with less tubular necrosis and brush border desquamation compared with the AKI group. Additionally, cisplatin treatment upregulated mitochondrial fission, a pathological characteristic of AKI, which was downregulated by AJ treatment. Along with increased mitochondrial fission, AJ treatment also reduced cisplatin-induced apoptosis. These results suggest that AJ may be a potential therapeutic agent for cisplatin-induced AKI.

Inhibitory Effects of the Stem Bark of Albizia julibrissin on Catecholamine Biosynthesis in PC12 Cells

  • The methanol extract of Albizzia julibrissin Durazz. (Leguminosae) was successively partitioned into dichloromethane, ethylacetate, butanol (BuOH) and water fractions, and the effects of the each solvent extract on catecholamine biosynthesis in PC12 cells were investigated. Among them, the BuOH fraction $(5{\mu}g/ml\;medium)$ showed 68.8% and 63.6% inhibition on dopamine and norepinephrine content in PC12 cells, respectively. Tyrosine hydroxylase (TH) activity was also reduced markedly by treatment of the BuOH fraction (41.8% inhibition at $5{\mu}g/ml$ in the medium). Each solvent fraction did not show cytotoxicity towards PC12 cells by trypan blue exclusion test. This result suggests that the BuOH fraction has an inhibitory effect on catecholamine biosynthesis by reducing TH activity in PC12 cells.

자귀나무(A. julibrissin) 수피 추출물의 생리활성 (The Physiological Activities of Bark Extract of Albizia julibrissin)

  • 한방생약재로 사용되고 있는 자귀나무 수피를 천연 항산화제 및 기능성 식품이나 제품을 개발하기 위한 연구의 일환으로 자귀나무 수피를 물을 용매로 $80^{\circ}C$$110^{\circ}C$의 고온에서 추출된 물 추출물(AW)과 열수 추출물(AHW) 그리고 70% 에탄올을 용매로 $60^{\circ}C$에서 추출된 에탄올 추출물(AE)에 대한 생리활성 물질의 함량과 효과를 측정하였다. 자귀나무 수피의 열수 추출물인 AHW에서 가장 많은 8.57 mg/g의 플라보노이드와 108.67mg/g의 폴리페놀 화합물을 함유하였다. SOD 유사활성은 1.0 mg/mL의 농도에서 10.46~16.73%였으며, 아질산염 소거능은 pH 1.2의 조건에서 49.86~60.82%로 AHW에서 가장 높은 소거효과를 보였으며 $IC_{50}$$770.18\;{\mu}g$/mL였다. DPPH를 이용한 전자공여능 측정 결과에서는 0.3 mg/mL의 농도에서 AHW가 92.30%로 가장 우수한 활성을 보였으나, $IC_{50}$은 AW($31.31\;{\mu}g$/mL)와 AHW($36.22\;{\mu}g$/mL)가 ascorbic acid ($39.10\;{\mu}g$/mL)보다 낮은 농도에서 50%의 전자공여능을 나타내었다. XO에 대한 저해효과를 측정한 결과에서도 AHW가 94.05%의 저해율와 127.78 ug/mL의 $IC_{50}$ 농도를 나타내었다. 이상의 결과 합환피 한약재명으로 사용되고 자귀나무 수피는 물을 용매로 $110^{\circ}C$에서 추출된 열수 추출물(AHW)이 세가지 추출물 중 가장 많은 플라보노이드와 폴리페놀 화합물을 함유하며, 생리활성 효과에서도 우수하며 특히 전자공여능은 낮은 농도에서도 ascorbic acid보다 높은 활성을 나타내어 자귀나무 수피는 천연 생리활성 물질로서 식품과 기능성 제품에 활용 가능성이 있는 약용식물자원으로 생각된다.

전북 지역사회에서 천렵에 이용된 식물들의 전통지식에 관한 연구 (Traditional Knowledge of Plants Used for River Fishing in Local Communities of North Jeolla Province, Korea)

  • This study investigates traditional knowledge of plants used for river fishing in local communities of North Jeolla Province, Korea. Data were collected using the participatory rural appraisal method based on interviews, informal meetings, open and group discussions, and overt observations with semi-structured questionnaires. Field investigations were conducted at 19 sites from July 2008 to April 2010. Interviews included 37 key informants (16 men and 21 women) who lived more than 50 years in the area. The average age of the informants was 83 (range = 60 to 96). The analysis recorded seven species, namely Albizia julibrissin Durazz., Juglans mandshurica Maxim., Perilla frutescens var. japonica (Hassk.) H. Hara, Nelumbo nucifera Gaertn., Persicaria hydropiper (L.) Spach, Zanthoxylum piperitum (L.) DC., and Styrax japonicus Siebold and Zucc..

Neuroprotective Effects of Medicinal Herbs in the Transient Focal Ischemia

  • It is thought that highly reactive oxygen radicals generated at the ischemia-reperfusion in case of strokes play an important role in damaging the brain. We examined the neuroprotective effects from the several medicinal herbs in the transient ischemic rat model and compared their effects with the free radical scavenging activities. Transient ischemia was induced by intraluminal occusion of the right middle cerebral artety for 120 min and reperfusion was continued for 22 h in rats. The free radical scavenging properties of medicinal herbs were examined in vitro by determination of the interaction with the 1,l-diphenyl-2-picrylhydrazyl (DPPH) stable free radical. Aqueous extracts of 11 medicinal herbs (200 mg/kg) were orally administered, promptly prior to reperfusion and 2 h after reperfusion. Total infarction volume in the ipsilateral hemisphere of ischemia reperfusion rats was significantly lowered by the treatment of 7 medicinal herbs (Sophora flavescens, Lycopus lucidus, Sanguisorba officinalis, Caesalpinia sappan, Albizia julibrissin, Rubia akane, Psoralea corylifolia, Prunella vulgaris). However, all of these medicinal herbs did not show antioxidative activities. These results suggest that neuroprotective effects of several drugs are not always correlated with their antioxidative properties.

전라북도 임실군 병아리꽃나무 자생지의 생태적 특성 (Ecological Characteristics of Rhodotypos scandens Habitat in Imsil-gun, Jeollabuk-do, Korea)

  • 본 연구는 병아리꽃나무 자생지의 식물상, 식생구조 및 토양과 같은 생태적 특성을 조사 및 분석하여 자생지 보전과 관리를 위한 기초 자료를 제공하는데 목적이 있다. 식물상은 57과 99속 107종 1아종 17변종 6품종 등 총 131분류군이며, 생활형은 교목(MM) 21분류군, 아교목(M) 24분류군, 관목(N) 30분류군, 지표식물(Ch) 6분류군, 반지중식물(H) 20분류군, 지중식물(G) 17분류군, 일년생식물(Th) 12분류군, 수생식물(HH) 1분류군이었다. 군락유형은 느티나무군락, 갈참나무군락, 참느릅나무군락, 까마귀베개군락, 밤나무군락, 자귀나무군락, 팽나무군락, 아까시나무군락 등 8개이다. 층위별 우점종의 경우 교목층은 느티나무, 갈참나무, 참느릅나무, 밤나무, 자귀나무, 팽나무, 아까시나무, 아교목층은 까마귀베개, 갈참나무, 느티나무, 자귀나무, 참느릅나무, 닥나무, 관목층은 병아리꽃나무로 나타났다. 종다양도 분석 결과, 종다양성지수는 1.691~2.610, 최대종다양성지수는 2.197~3.466, 균재도는 0.646~0.903, 우점도는 0.097~0.354로 나타났다. 토양 분석 결과, 평균 산도는 5.6, 평균 전기전도도 0.5ds/m, 평균 유효인산 59.1mg/kg, 평균 유기물함량 49.7%, 평균 치환성칼륨 $0.4cmol^+/kg$, 평균 치환성칼슘 $13.5cmol^+/kg$, 평균 치환성마그네슘 $3.3cmol^+/kg$으로 분석되었다.

일시적 국부 뇌허혈 동물모델에서 한약재의 신경보호 효과 (Neuroprotective Effects of Medicinal Herbs in the Transient Focal Ischemia in Rats)

Aqueous extracts of 8 medicinal herbs (200 mg/kg) were orally administered twice to transient ischemic rat prior to reperfusion and 2 hr after reperfusion. Total infarction volume in the hemisphere ipsilateral to the ischemia-reperfusion was significantly decreased in 7 groups treated with Sophora flavescens, Lycopus lucidus, Sanguisorba officinalis, Caesalpinia sappan, Albizia julibrissin, Rubia akane, Psoralea corylifolia, or Prunella vulgaris. However, neuroprotective effects of these medicinal herbs were not correlated with their antioxidative activities. These results suggest that these medicinal herbs exert neuroprotection via antioxidative as well as unknown mechanism.

국내 수집 두과식물의 부위별 pinitol 함량 변이 (Variation of Pinitol Content for Domestic Legume Species in Korea)

  • 본 연구에서는 경상북도 일원에서 수집한 자귀나부(Albizia julibrissin)를 포함한 두과식물 17종에서 인체의 혈당저하제의 원료로 사용되는 pinitol 함량을 식물체 부위별로 조사하였는 바, 결과를 요약하면 아래와 같다. 1. 수집한 17개 두과식물 종간에는 pinitol 함량에서 유의적인 차이가 인정되었으며, 종별 총 pinitol 함량에서는 비수리(Lespedeza cuneata)가 가장 높은 값을 보였다. 식물체 부위별로 pinitol 함량을 살펴보면 대체적으로 잎, 줄기, 꼬투리, 종자, 뿌리의 순으로 많았다. 2. 잎에서 비교적 많은 pinitol 함량을 보인 식물은 골담초, 족제비싸리, 돌동부 및 비수리로 각각 59.9 mg/g, 62.2 mg/g, 69.6 mg/g, 65.4 mg/g으로 약 60 mg/g이상을 나타내었고, 줄기의 경우는 황기가 34.0 mg/g으로 비교적 높게 나타났으며, 뿌리에서는 칡이 24.6 mg/g으로 가장 높았으며, 감초와 골담초가 16.9 mg/g, 16.5 mg/g으로 그 다음으로 높았다. 그러나 꼬투리나 종자에서는 제비콩(52.5 mg/g)과 비수리(24.9 mg/g)를 제외하고 pinitol 함량은 잎이나 줄기에 비해 모든 조사식물에서 상대적으로 낮았다. 3. 조사대상 식물체의 pinitol 함량과 수확 가능한 식물체량을 고려할 때, 실제 약용으로 이용 가능성이 가장 높은 식물은 비수리이었다. 비수리의 경우 전국적인 수집을 통해 pinitol 함량이 높은 계통을 선발한다면 이용가능성 이 높은 pinitol 추출용 계통의 선발이나 보다 이용가치가 높은 새로운 품종의 육성도 가능할 것으로 생각된다.
