A study of the Body Growth and Development in Albino Rats Fed by the Cereals and Legumes Proteins (곡류 및 두류 단백질이 흰쥐의 성장 발육에 미치는 영향)

  • This study was designed to compare the Biological effects of the cereals and legumes Proteins on albino rats. Fifty weaning albino rats weighing $49\;gr{\pm}2$ in both sexes were divided into 5 groups and fed corn crude protein diet, wheat crude protein diet, soybean protein diet, yeast diet and 30% sugar-casein diet for the control group. The protein contains same levels with isocaloric values each diet. After 12 weeks the rats were sacrificed for chemical analysis and the results are as follow ; 1. The highest food consumption was found in the groups of yeast and soybean, and the lowest was found in the groups of wheat diet and corn diet. 2. The groups of yeast & soybean protein showed the highest body weight increase, while the groups of wheat diet and corn diet showed the lowest. 3. Highly significant difference was found between the standard group and the groups of corn protein and soybean protein in Food Efficiency Ratio. (F. E. R.) (p<0.01). 4. Protein Efficiency Ratio showed a similar pattern as the F.E.R., however, there was no significant differences among the groups. 5. The kind of diets did not influence the hematology of the subjects. 6. The rate of nitrogen retention of male standard group was lower compared with all the experimental groups (p<0.01), and that of female soybean group was higher than any other groups (p<0.01). 7. Female corn diet group had the lowest organ weights, as found in the growth rate, which was significant (p<0.01). In the male corn diet group only the kidney showed significantly low in weight (p<0.01), and the spleen of male yeast group was also shown low with significance (p<0.01). 8. Nitrogen retentions of the liver and muscle in male corn diet group were lower than any other groups (p<0.05), and the brain nitrogen content of female standard group was high with significance (p<0.01). According to the results above, yeast and soybean protein can be regarded as a nutritious and also inexpensive protein sources.

Radiomodifying Potential of Panax ginseng in Liver of Swiss Albino Mice against Gamma Radiation

  • Panax ginseng occupies an important role in the folk medicine of China, Korea and Japan. The present study was undertaken to determine the radioprotective efficacy of ginseng root extract in the liver of Swiss albino mice. The animals were divided into 4 groups. Group I-Only vehicle was administered. Group II-The animals received 10 mg/kg body weight ginseng root extract i.p. for 4 consecutive days. Group III-Animals were irradiated with 8Gy gamma radiation at the dose rate of 1.69 Gy/min at the distance of 80 ems. Group IV-Animals were given by ginseng root extract (10 mg/kg body weight) continuously for 4 days and on 4th day they were irradiated with 8 Gy gamma radiation after 30 min. The animals from above groups were autopsied on 1,3,7,14 and 30 days. Biochemical estimations of phosphatases (acid & alkaline), LDH (lactate dehydrogenase), LPO (lipid peroxidation) and GSH (reduced glutathione) in liver and SGOT (serum glutamate oxaloacetate transaminase), SGPT (serum glutamate pyruvate transaminase) and alkaline phosphatase in serum were done. In ginseng treated group acid phosphatase (ACP), alkaline phosphatase (ALP), LPO and LDH in liver and SGOT, SGPT and alkaline phosphatase in serum did not show any significant alteration. However, a significant increase in GSH content in liver was recorded. In irradiated group there was a significant increase in ACP, ALP and LPO content in liver and SGOT & SGPT in serum was noted. Whereas, a significant decrease was recorded in GSH and LDH activity in liver and alkaline phosphatase activity in serum. Pretreatment of ginseng with radiation significantly alters the biochemical parameters in liver and serum. A significant decline in ACP, ALP activity and LPO content in liver and SGOT and SGPT activity in serum was observed. However, a significant increase in GSH content and LDH activity in liver and ALP activity in serum was estimated. The present study suggests that pretreatment of ginseng before irradiation significantly protects the liver and maintains the enzyme activity.

Effect of Heated Soybean on the Cholesterol Contents in Brain and Aorta of the Albino Rat (熱處理大豆가 흰쥐腦와 大動脈의 Cholesterol 含量에 미치는 영향)

  • The effect of heat treatment (autoclaving at $100^{\circ}C$ for 30 minutes) of soybean on the cholesterol contents in the brain and aorta of the albino rat was studied. The subjects were 72 young male rats and divided into 12 groups by diet and periods. Experimented periods were 5, 15 and 30 days, diet standard are raw soybean, soybean diets heated at $100^{\circ}C$ and $120^{\circ}C$ respectively, and each diet was prepared to contain 14g protein and 400 Kcal per diet 100g. After corresponding treatment, the cholesterols in the brain and aorta were determined. The results are summarized as follows: In the brain, total, free and ester cholesterol contents of each group are not remarkedly different from those of control group, in the experimental periods. In the aorta total and free cholesterol at $100^{\circ}C$ and $120^{\circ}C$ than those of control group, but there is no significant difference among groups fed for 15 or 30 days, and the ester cholesterol has no regular tendency. It seems that the heat treatment at $100^{\circ}C$ and $120^{\circ}C$ of solybean has no effect on the cholesterol contents of the brain and aorta.

  • cis-Dichlorodiammineplatinum (II) (cis-Platin) inhibits the proliferation and growth of the tumor cells by way of inhibiting DNA and protein synthesis of the cancer cells. Although cis-Platin is very effective antitumor drug, it also produces many other side effects. Thus the author has studied the effects of cis-Platin on the proximal epiphyseal plate in the tibia of the rat. The results were as follows: In the chondrocyte of the proliferative zone, sacculated, and fragmented cisternae of rough endoplasmic reticulum, some mitochondria with disorganized mitochondrial cristae and distorted procollagens were observed, and in the matrix some large matrix granules and dispersed collagen fibrils were revealed on the 1st, 3rd day and 1st week group of cis-Platin treated rats. In the chondrocyte of the proliferative zone of cis-Platin treated rats on the 2nd and 3rd week group, parallely arranged rough endoplasmic reticulum and many procollagens were shown, and in the matrix a number of large matrix granules and many small matrical granules as well as collagen fibrils were revealed. Consequently it is suggested that though cis-Platin induces the degenerative changes of the chondrocyte resulting in components of the cartilagenous matrix, these toxic effects are regressed with time.

Management of Radiation Injuries by Panax ginseng Extract

  • Chemical radiation protection is an important strategy to protect living beings against the deleterious effects of radiation. In the present study, the radioprotective effect of hydro-alcoholic extract of Panax ginseng extract (PGR-HAE) was studied on radiation-induced deleterious alterations in Swiss albino mice. Oral administration of such extract (25 mg/kg b wt/day/animal) for 5 consecutive days, half an h. before whole-body exposure to 6 Gy gamma radiation, enhanced the 30 days survival and also inhibited the radiogenic sickness, weight loss and life shortening. PGR-HAE ameliorated radiation induced depletion in blood constituents at different necropsy intervals between 12 h to 30 d, and significantly increased the number of femoral spleen colony forming units that survived after irradiation. Furthermore, it checked depletion of glutathione and antioxidant enzymes (superoxide dismutase, catalase, and glutathione S-transferase) as well as elevation of lipid peroxidation (LPO) level in blood and liver. The significant reduction in the yield of LPO demonstrates that PGR-HAE protects the membranes against radiation-induced oxidative damage. These findings conclude that such plant extract provides significant radioprotection, and it may be potentially valuable in the prevention of injuries caused during planned and unplanned radiation exposure.

Central nervous system activity of the methanol extracts of Caesalpinia bonducella and Bauhinia racemosa (Caesalpinaceae) in experimental animal model

  • The aim of the present study is to investigate central nervous system (CNS) activity of the methanol extracts of leaves of Caesalpinia bonducella (MECB) and stem bark of Bauhinia racemosa (MEBR) (Caesalpinaceae) in Swiss albino mice and Wistar albino rats. General behavior, exploratory behavior, muscle relaxant activity and phenobarbitone sodium-induced sleeping time were studied. The results revealed that the methanol extracts of leaves of Caesalpinia bonducella at 100 - 200 mg/kg and stem bark of Bauhinia racemosa 100 - 200 mg/kg caused a significant reduction in the spontaneous activity (general behavioral profile), remarkable decrease in exploratory behavioral pattern (Y-maze and head dip test), a reduction in muscle relaxant activity (rotarod and traction tests), and also significantly potentiated phenobarbitone sodium-induced sleeping time. The results suggest that MECB and MEBR exhibit CNS depressant activity in tested animal models.

THE EFFECT OF TETRACYCLINE ON DENTINOGENESIS OF DEVELOPING ALBINO RATS (Tetracycline이 백서치질형성기(白鼠齒質形成期)에 미치는 영향(影響)에 관(關)한 실험적(實驗的) 연구(硏究))

  • The study is about the effect of tetracycline-HCl on the amelogenesis and the dentinogenesis of the albino rats by means of histochemistry and fluorescence microscopy. Females in oestrus were mated overnight and examined the next morning for evidence of copulation. The mothers were intraperitonealy injected with a single dose of tetracycline-HCl from the eighth to tenth day of gestation. The heads of new born rats were fixed in Carnoy's solution and 10% formalin solution. The staining methods were alizarin red S stain, PAS reaction, colloidal iron reaction, Morin's stain and hematoxylin-eosin stain, The results were as follows: 1. By the single injection of tetracycline, the matrix formation of enamel and dentin were disturbed, and the shape and arrangement of ameloblast and odontoblast were distorted. 2. It seemed that, with the higher dose of tetracycline, the positive materials of PAS reaction were increased in the disturbed enamel and dentin matrix, but those of alizarin red S stain and colloidal iron reaction were decreased. 3. The fluorescence intensity in the disturbed enamel and dentin matrix were higher than the other areas and appeared to increase gradually with the higher dose of tetracycline.

  • The effects of dietary cellulose on the lipid metabolism in albino rats were observed, especially the effects of dietary cellulose from three different sources; cereal, vegetable and pure-cellulose. The results were summarized as follows. 1. Dietary cellulose from rice bran and vegetable resulted in lowering of reserved lipid in the body and the effect of rice bran was remarkable as compare to that of vegetable. 2. But the fatty acids composition in serum, liver, gastrocnemius, small intestine and epididymal fat pad was unchanged by dietary cellulose. This means the effect of dietary cellulose was only to change the body fat in quantify and not in quality. 3. Cellulose from rice bran increased serum cholesterol content and maintained medium level of serum total lipid. Cellulose from vegetable decreased serum total lipid and cholesterol content. Pure-cellulose decreased also serum total lipid and cholesterol content but the effect was not as low as vegetable cellulose. 4. It need to be further investigated on the different effects of cellulose from three sources on serum cholesterol content, particularly the increasing effects of rice bran.

  • Susceptibility of some vertebrates was examined to the early third-stage larvae (EL3) of Gnathostomn hispidum. The larvae collected from the Chinese loaches were infected to 4 silk carps, 3 snake heads, 3 bullfrogs. 5 mice and 9 albino rats. No worms were detected in fish. silk carps and snake heads. In 3 bullfrogs fed 30 larvae, a total of 9 EL3 was recovered in the gastrointestinal tract (8 larvae) and liver (ones). In 5 mice inferred with 50 larvae, a tolal of 37 (74.0%) advanced third-stage larvae (AdL3) was recovered from the muscle (31 larvae), liver (5 larvae) and kidney at 4 weeks after infection. In 9 albino rats iilfected with 115 larvae, a total of 40 (34.8%) AdLa was found in the muscle. The Inammalian hosts were found susceptible to the EL3 of G. hispinum from Chinese loaches.

  • The therapeutic effects of praziquantel on Fibricola seoulensis infection were studied by experimental infection of albino rats with the metacercariae obtained from the snakes. Total 51 albino rats were infected each with 500 or 1, 000 meta cercariae in number through intragastric tubes. One or four weeks la er the rats were treated with 1, 5, 10 or 20 mg/kg single dose of prazipuantel and sacrificed after 3 days or 10-120 minutes to search for the worms in their small intestine. The worm recovery rate at 3 days after the treatment was 10.7 % in 1 mg/kg dose group and o. 03~0.2% in 5, 10 and 20 mg/kg groups, while that of untreated control was 51. 3%. The minimum effective dose to treat F. seoulensis infection in the rats is considered to be 5 mg/kg in single dose. By observing the distribution pattern of worms in the small intestine after the treatment, dislodgement of the worms from the duodenum, their normal habitat, to the lower portions was recognizable within as early as 10 minutes. The majority of worms was found to have descended to the ileo-caecal portion after 60-120 minutes.
