Bacillus cereus 1-1 균주의 5-Aminolevulinic Acid (ALA) 생산 (Production of 5-Aminolevulinic Acid (ALA) by Bacillus cereus 1-1)

  • Bacillus cereus 1-1 균주는 광이 없는 호기적 환경에서 levulinic acid와 같은 저해제 처리 없이도 5-aminolevulinic acid (ALA)를2 mM까지 생산하였다. B. cereus 1-1 균주는 TCY 배지에서 전 배양과 본 배양을 18시간 동안 지속하고, 배지의 pH가 6.8에 도달하는 대수기 후기에 acetic acid를 비롯한 유기산들을 16 mM 첨가하였을 때 많은 ALA를 생산하였으며, 본 배양 시작시 0.3% glucose를 첨가하는 것이 효과적이었다. Acetic acid 대신 glutamic acid를 첨가하였을 때 ALA 생산이 8시간 이상 지속되었고, $40\;{\mu}M$의 gabaculine을 첨가하면 생산이 현저히 저해되는 것으로 보아 B. cereus 1-1 균의 ALA 생산은 C-5 경로에 의함을 알 수 있었다.

Manganese Superoxide Dismutase (MnSOD Val-9Ala) Gene Polymorphism and Susceptibility to Gastric Cancer

  • Background: Oxidative stress caused by the generation of reactive oxygen species plays an important role in human carcinogenesis. Manganese superoxide dismutase (MnSOD) Val-9Ala in the mitochondrial target sequence is the best known polymorphism of this enzyme. The purpose of the current research was to assess the association of MnSOD Val-9Ala genotypes with the risk of gastric cancer. Materials and Methods: This case-control study covered 54 gastric cancer patients compared to 100 cancer free subjects as controls. Extraction of DNA was performed on bioptic samples and genotypes were identified with a polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. Results: The frequencies of MnSOD Ala/Ala, Ala/Val and Val/Val genotypes in healthy individuals were 24.3, 66.7 and 9%, respectively. However, in gastric cancer patients, Ala/Ala, Ala/Val and Val/Val were observed in 24.0, 48.0 and 28.0% (p=0.01). In patients the frequency of MnSOD Val allele was higher (52%) compared to that in controls (42%). Conclusions: The results of this study show a positive association between MnSOD Val-9Ala gene polymorphism and risk of gastric cancer disease in Iranian population.

Effect of Alpha-Linolenic Acid on Oocyte Maturation and Embryo Development in Pigs

  • The aim of this study was to determine the effect of additional alpha-linolenic acid (ALA) supplementation during in vitro maturation (IVM) and culture (IVC) on nucleic maturation and embryo development of pigs. Cumulus-oocyte complexes (COCs) were incubated in IVM medium containing different concentration of ALA (25, 50 and $100{\mu}M$) for 44 h. After in vitro maturation, nuclear maturation of oocytes were evaluated by aceto-orcein stain. Mature oocytes with $50{\mu}M$ ALA were fertilized and cultured in IVC medium with ALA (25, 50 and $100{\mu}M$) during early-embryogenesis (48 hours after fertilization). Then, embryos were cultured with $25{\mu}M$ ALA during early embryogenesis and/or late embryogenesis (120 hours after early-embryogenesis). In results, oocyte maturation were significantly increased by $50{\mu}M$ ALA treatment groups compared with control groups (p<0.05). Treatment of $25{\mu}M$ ALA during early-embryogenesis enhanced cleavage rate of embryo compared with other groups (p<0.05), whereas formation and total cell number of blastocyst had no significant difference. Similarly, cleavage rate of embryos were increased by $25{\mu}M$ ALA supplement during early- or late-embryogenesis than ALA treatment both stage of embryogenesis (p<0.05), but did not influence to blastocyst formation. Interestingly, total cell number of blastocyst were enhanced in ALA treatment group during early-embryogenesis. These findings indicated that ALA supplement enhance the nuclear maturation of oocyte and embryo development, however, excessive ALA could negatively influence. Therefore, we suggest that ALA is used for improvement of in vitro production of mammalian embryo and further study regarding with functional mechanism of ALA is needed.

Effect of Alpha-Linolenic Acid with Bovine Serum Albumin or Methyl-Beta-Cyclodextrin on Membrane Integrity and Oxidative Stress of Frozen-Thawed Boar Sperm

  • The study was conducted to investigate the effects of alpha-linolenic acid (ALA) combined with bovine serum albumin (BSA) or methyl-beta-cyclodextrin (MBCD) on plasma and acrosomal membrane damages, mitochondrial activity, morphological abnormality, motility, and oxidative stress in frozen-thawed boar sperm. In previous our study, 3 ng/mL ALA had been shown protective effect during freezing process of boar sperm. Therefore, we used 3 ng/mL ALA in present study and ALA was combined with same molar ratio of BSA or MBCD (ALA+BSA and ALA+MBCD, respectively). To confirm the effect of two carrier proteins, same volume of BSA and MBCD without ALA were added during cryopreservation. Membrane damage, mitochondrial activity, reactive oxygen species (ROS) and lipid peroxidation (LPO) levels were measured using flow cytometry, and movement of sperm tail as motility parameter and morphological abnormality were observed under light microscope. In results, all of sperm parameters were enhanced by ALA combined with BSA or MBCD compared to control groups (p<0.05). Mitochondrial activity, morphological abnormality, ROS and LPO levels in ALA+BSA or MBCD groups were no significant difference compared with ALA, BSA and MBCD treatment groups. On the other hand, plasma and acrosomal membrane intact, and sperm motility in ALA+MBCD group were higher than single treatment groups (p<0.05), whereas ALA+BSA did not differ. Our findings indicate that carrier proteins such as BSA and MBCD could improve the effect of ALA during cryopreservation of boar sperm, and treatment of ALA with carrier proteins enhance membrane integrity, mitochondrial activity through reduction of ROS-induced LPO.

포유류 세포주에서 δ-Aminolevulinic acid (ALA)의 세포증식과 사멸에 미치는 영향분석 (Analysis of the effects of δ-Aminolevulinic acid on the proliferation and apoptosis of mammalian cells)

  • ${\delta}$-Aminolevulinic acid (ALA)는 생물권내에 폭넓게 존재하는 화합물이고, 포유류에서 헴(heme)과 식물에서 엽록소 생성을 하게하는 경로에서 만들어진 테트라피롤의 중간물질로써 생체내 중요한 역할을 한다. ALA는 생분해성 매개자, 성장 조절자, 헴단백질의 전구체 그리고 암 치료에서 사용되는 효과적인 물질로써 관심이 있다. 최근에는 ALA가 피부 치료의 좋은 효과를 가지고 있어 피부과학에서 빈번히 사용된다고 보고되어 있는데, 하지만 지난 몇 십 년 동안, 많은 연구가 ALA 메커니즘의 설명과 치료적 활성 개선에 초점을 맞춰왔지만, 세포 기능과 세포생장에 대한 ALA의 효과는 아직 불분명하다. 본 연구에서는 ALA의 약물학적 효과가 HEK293T 뿐만 아니라, HaCaT와 HeLa 세포에서 세포분열을 억제하는 것으로 확인을 하였다. 또한, ALA가 처리된 세포에서는 세포예정사가 유도되는 것을 확인하였다. 이러한 결과들은 특정 세포에 ALA가 처리되면 세포증식을 억제하며, 특히 인간의 암세포의 죽임을 유발하는 효과적인 약물 중 하나로써 ALA를 개선된 치료 전략으로 가능성을 제시한다.

가물치(Channa argus)로부터 평활근 수축활성 펩타이드의 정제 (Isolation of a Novel Neuropeptide with Contractile Activity on the Smooth Muscle from the Snakehead Channa argus)

  • A novel neuropeptide was isolated from the skin of the snakehead Channa argus using the dorsal retractor muscle (DRM) of a starfish Asterina pectinifera as a bioassay system. The amino acid sequence of the purified peptide was analyzed using automated sequencing and MALDI-TOF mass spectrophotometry. The primary structure of the purified peptide was determined to be Pro-Ala-Leu-Ala-Leu. To investigate the complete primary structure of this peptide, Pro- Ala-Leu-Ala-Leu-OH and Pro-Ala-Leu-Ala-Leu-NH2 were synthesized. The chemical and pharmacological properties of the synthetic peptides were compared with those of the native peptide. Both the native peptide and synthetic Pro-Ala- Leu-Ala-Leu-OH had identical behaviors on the reverse-phase and cation-exchange HPLC chromatograms. Synthetic Pro-Ala-Leu-Ala-Leu-OH showed contractile activity on the DRM, and the threshold concentration of this peptide was approximately $10^{-8}$ M. The maximal contractile effect ($E_{max}$) of this peptide was $294{\pm}45.4$% at $10^{-5}$ M.

Rhodocyclus gelatinosus KUP-74의 고정화균체로부터 ${\delta}-Aminolevulinate$의 연속생산 (Continuous Production of ${\delta}-Aminolevulinate$ from Immobilized Cells of Rhodocyclus gelatinosus KUP-74)

  • Rhodocyclus gelatinosus KUP-74의 고농도 휴지균체로부터 ${\delta}-aminolevulinate$(ALA)를 연속적으로 생산하기 위한 최적조건을 검토하였다. 균체외 ALA양은 20 mg cells/ml의 균체농도까지는 농도 증가에 따른 rectangular hyperbola의 증가양식을 보였으나, 그 이상의 균체농도는 ALA 생산에 전혀 증가효과를 보이지 않았다. 20 mg cells/ml의 반응계에서, 균체외 ALA의 효과적 생산을 위한 첨가물질의 최저농도는 levulinate 4 mM과 L-glutamate 5 mM로 나타났으며, 배양 3시간후 ALA 양의 최대치를 보였다. 한편, Ca-alginate를 이용한 포괄법으로 고정화시킨 균체로부터 효과적으로 ALA를 생산하기 위한 첨가물질의 최적농도는 levulinate 6 mM과 L-glutamate 10 mM로 나타났으며, 배양 6시간째에 균체외 ALA 양의 최대치를 보였다. 또한, 이 조건 하에서 고정화균체와 고농도의 휴지균체에 의한 ALA의 연속생산성을 검토한 결과, 이들 균체에 의한 50% ALA 생산성은 각각 185시간과 100시간에서 나타났으며, 생산된 균체외 ALA의 회수를 위해서는 균체를 고정화시키는 방법이 효과적이었다.

Rhodobacter sphaeroides에서 5-aminolevulinic acid 생산에 대한 850 nm 근적외선 발광다이오드 조사 효과 (Effect of 850 nm near-infrared light emitting diode irradiation on the production of 5-aminolevulinic acid in Rhodobacter sphaeroides)

  • 광감각제를 이용한 광역학 치료는 필요한 특정 부위에만 빛을 조사하여 치료 효과를 나타내는 부작용이 적은 방법이다. 5-aminolevulinic acid (ALA)는 다양한 생물체에서 합성되는 대표적 광감제로 암진단과 치료를 포함하는 다양한 분야에서 사용되고 있다. 본 연구에서는 다양한 파장의 LED, 유기산 전구체 및 glucose 농도 변화를 통한 Rhodobacter sphaeroides의 최적 성장 조건과 ALA 생산 조건을 확립하기 위한 실험을 진행하였다. 백열등과 동일한 광도 아래에서 Rhodobacter sphaeroides에 850 nm LED 빛을 조사하면 대조군 대비 균주의 성장과 ALA의 생산 농도를 각각 1.5배 및 1.8배 증가시킬 수 있고, 전구체로 pyruvic acid를 첨가한 경우 850 nm 파장의 LED만 조사한 경우 보다 ALA의 생산 농도를 약 2.8배 증가 시켰으며 동일 배양 조건에 40 mM glucose를 첨가하여 배양한 결과 Rhodobacter sphaeroides의 성장은 850 nm 파장의 LED 조사와 pyruvic acid를 첨가한 것에 비해 약 2.9배, ALA의 생산 농도는 약 3.4배 (20 mM) 증가되었다. 건조체 질량당 ALA의 생산은 20 mM과 40 mM glucose에서 대조군 대비 각각 약 1.4배 높은 결과를 나타냈다. 결론적으로 다양한 파장의 LED 중 850 nm 파장의 LED가 Rhodobacter sphaeroides의 성장률 및 ALA의 생산을 최대로 높였으며, 5 mM pyruvic acid와 40 mM glucose의 농도에서 최적의 Rhodobacter sphaeroides 성장과 ALA 생산을 확인하였다.

Enhancing Skin Delivery of 5-Aminolevulinic Acid with Transferosome Using Lyso-Phospholipid and Surfactant

  • In order to enhance the clinical efficacy of 5-aminolevulinic acid-induced photodynamic therapy (ALA-PDT), liposomal formulations using bulk hydrogenated phospholipids from soybean were introduced. Three types of lipids, S75-3, S100-3, and SL80-3 were used for formulating ALA. The pH of all the liposomal ALA is 4.5~5.5 and the size is 50~200 nm. All the liposomal formulations gave better ex vivo ALA skin penetration using nude mice skin in Franz cell than free ALA did. Among them, SL80-3 including 22% of lyso-phosphocholine achieved excellent ALA penetration when compared with those of S75-3 and S100-3 which have only 1~2% of lyso-phospholipids. S100-3 showed a little better results than S75-3 did. Addition of humectants (glycerine, propylene glycol, butylene glycol, betaine) in liposomal ALA formulated with SL80-3 produced little enhancing effect in ALA penetration. On the other hand, addition of surfactants (Tween 20, 60, Brij 72, 76, 78) in same liposomal system produced significant increase in ALA penetration. Among them, transferosomal system of lyso-phospholipid, SL80-3 and the surfactant, Brij76 showed the highest ALA penetration. Furthermore, this system also established the highest in vivo PpIX biosynthesis in hairy mice skin of C57BL/6. These results concluded that the transferosome of SL80-3 and Brij76 produced the best results in both ALA penetration and PpIX biosynthesis, and proved good correlation between them.

SOME FACTORS INFLUENCING TRI-L-ALANINE DISAPPEARANCE AND RUMEN BACTERIAL GROWTH YIELD IN VITRO

  • A series of in vitro incubation studies with washed rumen bacteria were conducted to determine the influence of incubation time and concentrations of peptides, alanine, ammonia nitrogen and carbohydrate on the rate of peptide disappearance and on bacterial growth. Disappearance rate of tri-alanine (ala3) under various conditions was between 30.6 and $58.2mg\;hr^-$ per gram bacterial dry matter. Ala3 was removed from the incubation medium in an almost linear fashion as incubation time and ala3 concentration was increased. Washed rumen bacteria utilized ala3 faster than di-l-alanine (ala2) at all concentrations. Adding 9mM carbohydrate significantly increased ala3 disappearance, but level of ammonia nitrogen had no influence on ala3 disappearance. The presence of alanine in the medium significantly lowered ala3 utilization by rumen bacteria. Bacterial dry matter and nitrogen growth yield were not influenced by alanine and peptides when incubation medium already contained a sufficient level of ammonia nitrogen. Increased ammonia nitrogen in the presence of ala3 did not stimulate bacterial growth. Carbohydrate significantly increased bacterial dry matter and nitrogen growth as expected. Results indicate that the rate of peptide utilization by rumen bacteria may be altered by type and concentration of peptides, and energy supply, and this may be mediated through changes in numbers and type of bacteria.
