국내 주택에서 물 피해 유형에 따른 부유곰팡이 농도 수준 평가 및 동정 분석 (Assessment of the level and identification of airborne molds by the type of water damage in housing in Korea)

  • Mold grows more easily when humidity is higher in indoor spaces, and as such is found more often on wetted areas in housing such as walls, toilets, kitchens, and poorly managed spaces. However, there have been few studies that have specifically assessed the level of mold in the indoor spaces of water-damaged housing in the Republic of Korea. We investigated the levels of airborne mold according to the characteristics of water damage types and explored the correlation between the distribution of mold genera and the characteristics of households. Samplings were performed from January 2016 to June 2018 in 97 housing units with water leakage or condensation, or a history of flooding, and in 61 general housing units in the metropolitan and Busan area, respectively. Airborne mold was collected on MEA (Malt extract agar) at flow rate of 100 L/min for 1 min. After collection, the samples were incubated at $25^{\circ}C$ for 120 hours. The cultured samples were counted and corrected using a positive hole conversion table. The samples were then analyzed by single colony culture, DNA extraction, gene amplification, and sequencing. By type of housing, concentrations of airborne mold were highest in flooded housing, followed by water-leaked or highly condensed housings, and then general housing. In more than 50% of water-damaged housing, the level of airborne mold exceeded the guideline of Korea's Ministry of Environment ($500CFU/m^3$). Of particular concern was the fact that the I/O ratio of water-damaged housing was greater than 1, which could indicate that mold damage may occur indoors. The distribution patterns of the fungal species were as follows: Penicillium spp., Cladosporium spp. (14%), Aspergillus spp. (13%) and Alternaria spp. (3%), but significant differences of their levels in indoor spaces were not found. Our findings indicate that high levels of mold damage were found in housing with water damage, and Aspergillus flavus and Penicillium brevicompactum were more dominant in housing with high water activity. Comprehensive management of flooded or water-damaged housing is necessary to reduce fungal exposure.

어린이활동공간에서의 바이오에어로졸 포집 전략 (A Suggested Air Sampling Strategy for Bioaerosols in Daycare Center Settings)

  • Objectives: We aimed to compare the sampling performance of different flow-based impactor samplers for collecting fungal spores and bacteria and to explore the association of the level of bioaerosols with activity patterns of occupants in daycare center settings. Methods: For comparison of sampling performance, two different flow-based samplers (greater than 100 L/min or not) were selected; a low flow-based sampler (one-stage Andersen sampler) and two high flow-based samplers (DUO SAS SUPER 360 sampler, BUCK bio-culture sampler). We collected airborne mold and bacteria in 30 daycare centers with various levels of contaminated air. Three repeat samplings per each sampler were performed. Mold and bacteria were grown for 96 hours at $25{\pm}1^{\circ}C$ and 48 hours at $35{\pm}1^{\circ}C$, respectively. The Andersen and SAS samplers were used for investigating the association between the level of bioaerosols and the activity patterns of occupants in daycares. Particular matters 10($PM_{10}$), temperature, and relative humidity were monitored as well. Samplings were carried out with one-hour interval from 9 to 5 O'clock. For statistical comparisons, Kruskal-Wallis test, Wilcoxon's signed rank test, and multiple regression analysis were carried out. Results: The airborne level of molds by the low flow-based sampler were significantly higher than that of high flow-based samplers (indoor, P=0.037; outdoor, P=0.041). However, no statistical difference was observed in the airborne level of bacteria by each sampler. Also the level of bioaerosols varied by the time, particularly with different activity patterns in daycare centers. The higher level of mold and bacteria were observed in play time in indoor. Similarly, the concentrations of $PM_{10}$ were significantly associated with the level of bioaerosols (P<0.05). Conclusions: Our findings indicate that the flow rate of sampler, rather than total air volume, could be able to affect the results of sampling. Also, the level of airborne mold and bacteria vary behavior patterns of occupants in indoor of daycare settings. Therefore, different samplers with other flow rate may be selected for mold or bacteria sampling, and activity patterns should be considered for bioaerosol sampling as well.

DNA 기반 곰팡이 평가기법을 활용한 주택의 실내 곰팡이 노출수준 평가 (Evaluation of Indoor Mold Exposure Level in dwelling Using DNA-Based Mold Assessment Method)

  • Objective: Allergic diseases such as asthma due to fungal exposure in houses have increased, and proper management is urgent. Mold can grow in the air, floor, walls, and other areas according to environmental conditions, and there are many limitations to the conventional methodology for examining fungal exposure. For this reason, the degree of fungal contamination is being evaluated by ERMI (Environmental Relative Moldiness Index), a quantitative analysis method proposed by the EPA. In this study, we compared ERMI values between water-damaged dwellings and non-damaged ones to evaluate the effectiveness of Korean ERMI values. We also explored the association of ERMI values with the level of airborne mold and characteristics of dwellings. Methods: Floor dust was collected after installing a Dustream collector on the suction port of a vacuum cleaner. The collected samples were filtered to remove only 5 mg of dust, and DNA was extracted using the FastDNA SPIN KIT protocol. Results: The ERMI values were found to be 19.6 (-6.9-58.8) for flooded houses, 7.5 (-29.2-48.3) for leaks/condensation, and 0.8 (-29.2-37.9) for non-damaged dwellings. The airborne concentration of mold for flooded, leakage or condensed, and non-damaged houses were $684CFU/m^3$, $566CFU/m^3$, and $378CFU/m^3$, respectively. The correlation between ERMI values and the levels of airborne mold was low (R = 0.038), but a weakly significant association of the ERMI values with the concentration of particulate matter ($PM_{10}$) was observed as well(R=0.231,P<0.05). Conclusions: Our findings show that the reference value using ERMI can be used to distinguish water-damaged and non-damaged dwellings. It is believed that ERMI values could be a promising tool for assessing long-term fungal exposure in dwellings.

Evaluation of Airborne Bacteria and Fungi in Surgical Areas at the Animal Hospital

  • Studies on the concentration of airborne microorganisms in human medicine as a part of a study on the nosocomial infections have been conducted properly, but in veterinary medicine, there has been rarely performed in Korea to the best of study's knowledge. The purpose of this study was to evaluate the distribution of airborne microorganisms and to identify their species in different places in the animal hospital to alert the necessity of thorough cleanliness management. This study evaluated the concentrations of airborne bacteria and fungi in hospital areas, such as patient waiting room, internal medicine ward, surgical ward and radiological diagnostic ward. The concentration of bacteria and fungi was significantly lower (p < 0.05) in two operating rooms and higher in the patient waiting room. The dominant species of bacteria were Micrococcus spp., Staphylococcus spp., and fungi were Penicillium spp., Dermatophyte mold. Animal hospitals need to perform proper procedures for disinfection, sterilization, and environmental cleaning as well as appropriate employee training and monitoring in order to the maximum prevention of the risk of nosocomial and surgical infections.

전국 다중이용시설의 실내공기 부유 곰팡이의 현황 및 특성 분석 (An Analysis of the Current Status and Characteristics of Airborne Fungi in Indoor Air in Multi-Use Facilities Nationwide)

  • Background: Airborne fungi are ubiquitous in the air and exposure to an airborne fungus can be a significant risk factor. The composition of fungi has been potentially important for human health, especially for respiratory diseases like asthma and atopic dermatitis. Therefore, we attempted to ascertain what kind of airborne fungi affect human health at a nationwide level. Objectives: This study was carried out to provide information on indoor fungi distribution at multi-use facilities throughout South Korea. Methods: We classified our data by region and public facility after collection, cultivation, and identification via the sequencing of the ITS (internal transcribed spacer) region. We investigated whether or not the proliferation of HaCaT cells was affected by the identified airborne fungi. Results: In our data, the most isolated airborne fungi by region were Penicillium spp (Seoul, Daegu), Periconia sp (Gyeonggi-do), Iprex sp (Gangwon-do), Phanerochaete sp (Busan), Bjerkandera sp (Gwangju), and Aspergillus sp (Jeju-do). In the public facilities, the most detected fungi were Cladosporium sp (public transport), Penicillium sp (apartment house, retail market, financial institution, karaoke room), Bjerokandera sp (underground parking lot, public toilet, medical institution), Periconia sp (retail store), and Fusarium sp (general restaurant). Next, we selected twenty airborne fungi to examine their cytotoxicity and proliferation of human skin cells. In this experiment, the proliferation of the cells was influenced by most of the identified fungi. In case of the cytotoxicity test, most genera except for Rhodotorula sp and Moesziomyces sp showed cytotoxicity in HaCaT cells. Conclusions: The distribution of mold in the indoor air in multi-use facilities in South Korea differs from region to region, and this is an indicator that should be considered in future health impact studies. In addition, as a result of culturing about 20 types of bacteria dominant in indoor air, it was found that most (90%) inhibit the growth of skin cells, which can be harmful to health. An in-depth study of the health effects of floating fungi is needed.

Evaluation of a Fungal Spore Transportation in a Building under Uncertainty

  • A fungal spore transportation model that accounts for the concentration of airborne indoor spores and the amount of spores deposited on interior surfaces has been developed by extending the current aerosol model. This model is intended to be used for a building with a mechanical ventilation system, and considers HVAC filter efficiency and ventilation rate. The model also includes a surface-cleaning efficiency and frequency that removes a portion of spores deposited on surfaces. The developed model predicts indoor fungal spore concentration and provides an indoor/outdoor ratio that may increase or decrease mold growth risks in real, in-use building cases. To get a more useful outcome from the model simulation, an uncertainty analysis has been conducted in a real building case. By including uncertainties associated with the parameters in the spore transportation model, the simulation results provide probable ranges of indoor concentration and indoor/outdoor ratio. This paper describes the uncertainty quantification of each parameter that is specific to fungal spores, and uncertainty propagation using an appropriate statistical technique. The outcome of the uncertainty analysis showed an agreement with the results from the field measurement with air sampling in a real building.

Isolation and Characterization of Airborne Mushroom Damaging Trichoderma spp. from Indoor Air of Cultivation Houses Used for Oak Wood Mushroom Production Using Sawdust Media

  • Some species of the Trichoderma genus are reported as the major problem in oak wood mushroom production in Korea. In spite of economic loss by the fungi, scientific information on airborne Trichoderma species is not much available. To generate information for disease management development we analyzed airborne Trichoderma. A total of 1,063 fungal isolates were purely obtained from indoor air sampling of cultivation houses used for oak wood mushroom using sawdust media. Among the obtained isolates, 248 isolates were identified as Trichoderma fungi including T. harzianum, T. atroviride, T. citrinoviride, and T. pseudokoningii, by morphological and molecular analysis. T. harzianum was dominant among the four identified species. All the four Trichoderma species grew fast on solid nutrient media tested (potato dextrose agar [PDA], malt extract agar [MEA], Czapek's Dox + yeast extract agar [CYA] and cornmeal dextrose agar). Compact mycelia growth and mass spore production were better on PDA and CYA. In addition, T. harzianum and T. citrinoviride formed greenish and yellowish mycelium and spores on PDA and CYA. Greenish and yellowish pigment was saturated into PDA only by T. pseudokoningii. These four Trichoderma species could produce extracellular enzymes of sawdust substrate degradation such as β-glucosidase, avicelase, CM-cellulase, amylase, pectinase, xylanase, and protease. Their mycelia inhibited the growth of oak wood mushroom mycelia of two tested cultivars on dual culture assay. Among of eleven antifungal agents tested, benomyl was the best to inhibit the growth of the four Trichoderma species. Our results demonstrate that the airborne Trichoderma fungi need to be properly managed in the cultivation houses for safe mushroom production.

  • Objectives: Much scientific evidence indicate a positive association between moldy environments and respiratory illnesses and/or symptoms. However, few comprehensive assessments of mold have been performed for such settings. Spore counts or microscopic enumeration only may not be sufficient for evaluating fungal exposure. Recently, Mold Specific QPCR technology developed by the US EPA (Environmental Relative Moldiness Index, ERMI) has been widely used worldwide and great performance for assessing fungal exposure has been shown. Methods: We aimed to develop a Korean version of ERMI suitable for the distribution of fungal flora in Korea. Thirty dwellings in the Seoul and Incheon area were selected for sampling, and each was classified as 'Flooded, 'Water-damaged' or 'Non-water-damaged'. Results: Dust on the floor and airborne sampling were collected using an MAS100 and a 'Dustream' collector. Samples were analyzed by quantitative polymerase chain reaction(QPCR) for the 36 molds belonging to ERMI. Student t-test and ANOVA tests were carried out using SAS software. The median ERMI values of flooded, water damaged, and non-water damaged dwellings were 8.24(range: -5.6 to 27.9), 5.47(-25. 4 to 32.7), and -15.30(-24.6 to 14.8), respectively. Significant differences were observed between flooded and non-water damaged dwellings (P=0.001) and between water-damaged and non-water damaged dwellings (P=0.032). Conclusion: Our findings indicate that ERMI values attributed to dust samples in Korea could be applicable for the identification of flooded or water damaged buildings. However, much data is needed for continuously developing the Korean version of ERMI values.

지르코니아의 거칠기 증가를 위한 다양한 표면처리방법이 레진 시멘트와의 전단결합강도에 미치는 영향 (Effects of various zirconia surface treatments for roughness on shear bond strength with resin cement)

  • Purpose: The purpose of this study was to evaluate the effects of various zirconia surface treatment methods on shear bond strength with resin cements. Methods: We prepared 120 cylindrical zirconia specimens (⌀10 mm×10 mm) using computer-aided design/computer-aided manufacturing (CAD/CAM). Each specimen was randomly subjected to one of four surface treatment conditions: (1) no treatment (control), (2) airborne-particle abrasion with 50 ㎛ of Al2O3 (A50), (3) airborne-particle abrasion with 125 ㎛ of Al2O3 (A125), and (4) ZrO2 slurry (ZA). Using a polytetrafluoroethylene mold (⌀6 mm×3 mm), we applied three resin cements (Panavia F 2.0, Super-Bond C&B, and Variolink N) to each specimen. The shear bond strength tests were performed in a universal testing machine. The surfaces of representative specimens of each group were evaluated under scanning electron microscope. We used one-way analysis of variance (ANOVA), two-way ANOVA, and post hoc Tukey honest significant difference test to analyze the data. Results: In the surface treatment method, the A50 group showed the highest bond strength, followed by A125, ZA, and control groups; however, no significant difference was observed between A50 and A125, A125 and ZA, and ZA and control (p>0.05). Among the resin cements, Super-Bond C&B showed the highest shear bond strength, followed by Panavia F 2.0 and Variolink N (p<0.05). Conclusion: Within the limitations of this study, application of airborne-particle abrasion and ZrO2 slurry improved the shear bond strength of resin cement on zirconia.

드레싱 제조업체의 HACCP 시스템 적용을 위한 미생물학적 위해도 평가 (Evaluation of the HACCP System on Microbiological Hazard during Dressing Production)

  • Dressing 가공공정에 대한 HACCP(Hazard Analysis Critical Control Point) 시스템 구축을 위하여 위해요소 분석을 실시하였다. 위해요소 분석은 주원료, 용수, 미생물 평가, 작업장별 공중 낙하균과 종업원에 대한 병원성 미생물 검사를 하였다. 충청북도 진천 소재의 SJ 회사에서 2012년 4월 1일부터 30일까지 30일 동안 수행하였다. 그 결과 원료의 미생물은 5회 동안 검출되지 않았으며, 작업장 I과 작업장 II, 포장실, 세척용수, 검사실에서 사용된 용수의 총균수는 $3.0{\times}10^1$ CFU/mL 이하로 검출되었다. 드레싱 생산과정 중 열처리 및 혼합 과정에서 총균수는 평균 $3{\times}10$ CFU/mL가 검출되었으나 대장균과 진균류, 병원성 미생물은 검출되지 않았다. 공중 낙하균(총균수와 진균류)은 각각의 작업장에서 법적 허용치 이하로 검출되었다. 작업자들의 총균수 미생물 평가에서 모두 양성반응이었으나 대장균과 황색포도상구균은 검출되지 않았다. 결론적으로 위생관리 기준은 HACCP 시스템을 위한 미생물의 한계기준과 일반세균과 병원성 미생물과 같은 위해요소를 예방 및 감소시키기 위하여 설정하여야 한다.
