Detection of transgene in early developmental stage by GFP monitoring enhances the efficiency of genetic transformation of pepper

  • In order to establish a reliable and highly efficient method for genetic transformation of pepper, a monitoring system featuring GFP (green fluorescent protein) as a report marker was applied to Agrobacteriummediated transformation. A callus-induced transformation (CIT) system was used to transform the GFP gene. GFP expression was observed in all tissues of $T_0$, $T_1$ and $T_2$ peppers, constituting the first instance in which the whole pepper plant has exhibited GFP fluorescence. A total of 38 T0 peppers were obtained from 4,200 explants. The transformation rate ranged from 0.47 to 1.83% depending on the genotype, which was higher than that obtained by CIT without the GFP monitoring system. This technique could enhance selection power by monitoring GFP expression at the early stage of callus in vitro. The detection of GFP expression in the callus led to successful identification of the shoot that contained the transgene. Thus, this technique saved lots of time and money for conducting the genetic transformation process of pepper. In addition, a co-transformation technique was applied to the target transgene, CaCS (encoding capsaicinoid synthetase of Capsicum) along with GFP. Paprika varieties were transformed by the CaCS::GFP construct, and GFP expression in callus tissues of paprika was monitored to select the right transformant.

Induction of RNA-mediated Resistance to Papaya Ringspot Virus Type W

  • Transformation of cantaloupes with the coat protein (cp) gene of papaya ringspot virus type W (PRSV-W), Thai isolate, was used to introduce virus resistance. Binary vectors containing either the full length coat protein coding region under control of the 35S CaMV promoter(pSA1175), or the inverted-repeat of a coat protein coding region (pSA1304), were constructed and used for Agrobacteriummediated transformation of cotyledonary explants of the cantaloupe cultivar Sun Lady. Four independent transgenic lines were obtained using pSA1304 and one using pSA1175. Integration of the PRSV-W cp gene into the genome of these transgenic lines was verified by PCR amplification, GUS assays and Southern blot hybridization. In vitro inoculation of these lines with PRSV-W revealed that whereas the line containing pSA1175 remained sensitive, the four lines containing pSA1304 were resistant. The presence of small RNA species, presumably siRNA, corresponding to regions of the viral cp gene in transgenic lines resistant to PRSV-W supports the involvement of post-transcriptional gene silencing in the establishment of resistance.

Effect of Introducing Chitinase Gene on the Resistance of Tuber Mustard against White Mold

  • The objective of this research was introduction of chit42 to tuber mustard plants through Agrobacteriummediated transformation against white mold caused by Sclerotinia sclerotiorum. The binary plasmid pGisPEC1 was used in this study. Polymerase chain reaction analysis detected the transgene in 27 transformants with a transformation efficiency of 6.9%. Southern blot test was used to assess the copy number of transgene in tuber mustard plants. One, two, two, and two chit42-related bands were observed in the transformed lines TMB4, TMB7, TMB12, and TMB18, respectively. Enzymatic tests showed a significant increase in the activity of endochitinase in protein isolated from leaf tissues of chit42 transgenic 75-day tuber mustard lines. The pathogenicity of three pathogen isolates was tested on the leaves of transformed plans. The results of current study showed that expression of the gene chit42 in tuber mustard plants markedly reduced infection radius on the leaves 7 days after inoculation with the fungus.

Metabolic engineering of Lilium ${\times}$ formolongi using multiple genes of the carotenoid biosynthesis pathway

  • Lilium ${\times}$ formolongi was genetically engineered by Agrobacterium-mediated transformation with the plasmid pCrtZW-N8idi-crtEBIY, which contains seven enzyme genes under the regulation of the CaMV 35S promoter. In the transformants, ketocarotenoids were detected in both calli and leaves, which showed a strong orange color. In transgenic calli, the total amount of carotenoids [133.3 ${\mu}g/g$ fresh weight (FW)] was 26.1-fold higher than in wild-type calli. The chlorophyll content and photosynthetic efficiency in transgenic orange plantlets were significantly lowered; however, after several months of subculture, they had turned into plantlets with green leaves that showed significant increases in chlorophyll and photosynthetic efficiency. The total carotenoid contents in leaves of transgenic orange and green plantlets were quantified at 102.9 and 135.2 ${\mu}g/g$ FW, respectively, corresponding to 5.6- and 7.4-fold increases over the levels in the wild-type. Ketocarotenoids such as echinenone, canthaxanthin, 3'-hydroxyechinenone, 3-hydroxyechinenone, and astaxanthin were detected in both transgenic calli and orange leaves. A significant change in the type and composition of ketocarotenoids was observed during the transition from orange transgenic plantlets to green plantlets. Although 3'-hydroxyechinenone, 3-hydroxyechinenone, astaxanthin, and adonirubin were absent, and echinenone and canthaxanthin were present at lower levels, interestingly, the upregulation of carotenoid biosynthesis led to an increase in the total carotenoid concentration (+31.4%) in leaves of the transgenic green plantlets.

ACC oxidase 발현 억제 식물체에서 폴리아민 생합성 증가에 의한 스트레스 저항성 증강 (Enhanced tolerance through increasing polyamine contents in transgenic tobacco plants with antisense expression of ACC oxidase gene)

  • 에틸렌 생합성 효소인 ACC oxidase 유전자인 CAO cDNA가 antisense 방향으로 도입된 형질전환 담배 식물체는 endogenous한 NtACO의 발현 및 에틸렌 생합성이 모두 억제되었다. 또한 폴리아민 생합성 효소인 SAMDC의 유전자 발현 및 효소 활성이 증가하였고 spermidine 함량이 증가하였다. 이러한 결과는 에틸렌 생합성의 감소로 인하여 세포 내 폴리아민 함량의 변화가 유도되는 경쟁적 관계로 spermidine이 증가하였고 이것은 SAMDC의 발현 및 활성 증가를 유도하였다고 여겨진다. 또한 이들 형질전환 식물체는 담배 역병균에 대한 저항 효과 뿐만 아니라 PR 단백질의 유전자들의 발현양이 증가하였는데 이는 에틸렌 감소에 유도되는 폴리아민 중에서도 spermidine의 증가가 PR 단백질의 유전자들의 발현을 촉진시키고 그 결과 병원균 감염에 대한 저항성을 유도하는 기작이 작동한 것으로 여겨진다. Spermidine을 합성하는데 중요하게 작용하는 SAMDC의 활성을 억제하는 MGBG를 처리한 경우에는 PR1a 발현이 억제된 경우에서 이 기작의 가능성을 확인하였다. 결론적으로 병원균 감염에 대한 과정에서 spermidine의 생합성이 증가하게 되면 PR 단백질 등 스트레스 저항성 관련 단백질들이 유도되어 스트레스 저항성을 높일 수 있을 것으로 사료된다.
