Isolation and Identification of Pathogenic Bacteria of Grapevine Crown Gall in Korea (우리나라 포도나무 줄기혹병 병원세균의 분리 및 동정)

  • 우리나라 포도재배 지역인 포천, 남양주 및 수원의 포도나무 줄기혹병에 이병된 포장으로부터 채집한 줄기혹(gall), 수액(sap) 및 근권토양으로부터 56균주의 유사 Agrobacterium를 분리하였으며, 이들 중 7균주가 포도나무 유묘의 줄기에 혹ㅇ르 형성시키는 병원성 Agrobacterium biovar의 선택배지에서의 생장, oxidase의 생성, Na L-tartrate로부터 알칼리의 성장, 2% NaCl 첨가 배지에서 생장, 3-ketolactose의 비생산, mesoerythritol과 ethanol로부터 산을 생성하지 못하는 등 Agrobacterium biovar 3인 A. vitis로 동정되었다.

Characterization and biovar. cetermination of agrobacterium tumefaciens T7 isolated in Korea (한국에서 분리한 agrobacterium tumefaciens T7의 특성과 biovar.결정)

  • For the purpose of securing of strains which can be usefully utilized to study symbiosis between Rhizobium and legume plant, A. tumefaciens T7 was isolated and characterized and then subgroup biovar was determined. A. tumefaciens T7 induced smooth tumor like nopaline type one and did not grow at $37^{\circ}C$ and in the presence of 2% NaCl on yeast extract mannitol medium. The strain was able to grow on the New and Kerr selective media and utilize erythritol but not phenylalanine, tryptophan, and tartarate as a sole carbon source. Negative results were obtained from 3-keto-lactose production and oxidase test. The strain produced alkalifrom malonate and citrate and showed acid litmus milk reaction At least two large plasmids were detected in the cell lysate. According to all of these results, it could be concluded that subdivision of isolated strain was biovar 2.

  • Agrobacterium vitis causes a crown gall disease in grapevine and that is one of the major hindrances for the wide cultivation and production of grapevine. We studied the possibility of biological control using selected biological control agent. One isolate from the infected soil, named as strain 27, was able to inhibit the biovar 1; A. tumefaciens C58 and Ach5, biovar 2; A. rhizogenes 13264, and biovar 3; A. vitis, in vitro and in vivo test. The putative biological control agent, A. radiobacter strain 27 was carrying the plasmid and the size of isolated plasmid was very similar to that of pAgK84 of A. radiobacter K84.

Distribution of Agrobacterium tumefaciens Biovars in Jordan and Variation of Virulence

  • One hundred and nine Agrobacterium isolates were recovered from 68 samples(51 plant tumor and 17 soil) that were collected from different habitats in Northern Jordan. The isolated cultures were grouped into 3 biovars based on their biochemical characteristics and biovar I, II, and III comprised a total number of 46, 41, and 22 isolates, respectively. Isolates of biovar I were obtained primarily from the diseased peach, oak and rose plants, whereas isolates of biovar II and ill were obtained mostly from apple and grape plants, respectively. Twenty-nine isolates were found to be virulent to at least one of the tested hosts such as carrots, chickpeas, garden peas and tomato plants with a response of tumor formation or tumor with roots induction. Our result suggested that A. tumefaciens strains from tumor of various plants and soil of Jordan were diverse and they have a variation in their virulence.

Identification of Agrobacterium tumefaciens from Soil and Transformation of Maize (토양으로부터의 Agrobacterium tumefaciens의 분리, 동정 및 옥수수의 형질전환에 이용)

  • Several strains of Agrobacterium tumefaciens were isolated from soil in the Taegu area and characterized to develop some useful vector systems for higher plant genetic engineering. The selected colonies had a unique form, and strains from the colonies were capable of tumor formation on the sunflower leaf surface. They had a large plasmid. The restriction analysis showed that they were another kinds of Ti plasmic compared with C58 and Ach5. The isolated strains were identified as the nopaline type and also as biovar 1 A. tumefaciens, according to their tumor morphology, blophyslcal and biochemical characteristics. One of the isolated strains, AK204 was transformed with binary vector (pGA642), having selectable marker (Kmr, Tcr). Furthermore, maize tissue cells were transformed by cocultivation with AK204/pGA642, and the transformants were selected on the selective medium and identified using PAGE patterns of their soluble proteins.

  • Curdlan is a natural $\beta$-1,3-glucan produced by Agrobacterium biovar 1. In this study, the anticoagulant activity of sulfoalkyl derivatives of curdlan was investigated by carrying out activated partial thromboplastin time (APTT) assay and compared with that of o-sulfonated curdlan. Approximately 100-fold higher concentration of o-sulfonated curdlan than heparin was required to obtain the same level of the clotting time. Anticoagulant activity of curdlan derivatives was dependent on the degree of sulfation in prolonging the clotting time. However, the chain length of the substituent did not play a role in prolonging the clotting time. The curdlan derivatives enhanced thrombin inhibition by mediating through antithrombin III. The inhibition of thrombin by o-sulfonated curdlan was found to be approximately 10-fold weaker than that by heparin.
