We evaluated the presence of uniconazole residual pesticide in agricultural products by using multiclass pesticide multiresidue methods. Samples were collected from January to August, 2012. The pesticide was detected in 14 samples among the 3,632 samples tested. Amount of the uniconazole pesticide ranged from 0.098 to 2.2 mg/kg in the 14 samples. This method was described for the simultaneous determination of uniconazole by using gas chromatography with an electron capture detector (GC-ECD) and mass spectrometry (MS). For evaluating the GC-ECD method, uniconazole was spiked into gyeojachae at a level of 0.05, 0.5 mg/kg. The recoveries of uniconazole with the GC-ECD method ranged from 98.9-109.4%. The results indicate that our method of simultaneous analysis is applicable to uniconazole analysis.
The use of microwave-assisted extraction and an acid-base clean-up process to determine the amount of methylmercury (MeHg) in marine products was suggested in order to improve the complicated sample preparation process. The optimal conditions for microwave-assisted extraction was developed by using a 10% NaCl solution as an extraction solution, setting the extraction temperature at $50^{\circ}C$, and holding for 15 minutes to extract the MeHg in marine products. A NaOH solution was selected as a clean-up substitute instead of L-cysteine solution. Overall, 670 samples of marine products were analyzed for total mercury (Hg). Detection levels were in the range of $0.0006{\sim}0.3801{\mu}g/kg$. MeHg was analyzed and compared using the current food code and the proposed method for 49 samples which contained above 0.1 mg/kg of Hg. Detection ranges of methylmercury followed by the Korea Food Code and the proposed method were $75.25(ND{\sim}516.93){\mu}g/kg$ and $142.07(100.14{\sim}244.55){\mu}g/kg$, respectively. The total analytical time of proposed method was reduced by more than 25% compared with the current food code method.
Rancidity changes were examined for 6 herbal medicines, namely Persicae Semen, Armeniacae Semen, Lini Semen, Trichosanthis Semen, Arecae Semen, Myristicae Semen known to have relatively high fat content. In order to reduce rancidity of herbal medicines, samples were stored at 3 different conditions of room, refrigerating and freezing temperatures, and the rancidity was measured for 10 months with every 2 month interval. Fat content was extracted by using ethyl ether, and acid values and peroxide values, which are generally accepted indicators of fat rancidity, were measured. When storing Persicae Semen, Lini Semen and Arecae Semen at room temperature, the acid values increased as the storage period increased, and it was higher than when stored in refrigeration or freezing. The measurement of peroxide value showed more significantly higher initial degree of rancidity when Persicae Semen, Trichosanthis Semen, Arecae Semen and Myristicae Semen were stored at room temperature. It was observed that storing herbal medicines in refrigeration or freezing inhibited their rancidity compared to storing them at room temperature. To investigate the quality changes according to rancidity, the analysis of aflatoxins and indicator components showed that aflatoxins B1 and B2 were detected in Armeniacae Semen, Arecae Semen and Myristicae Semen, and the amount of amygdalin was well maintained within the specification standard.

Polycyclic aromatic hydrocarbons(PAHs) contents were analyzed by measuring benz(a)anthracene(BaA), chrysene(Chr), benzo(b)fluoranthene(BbF) and benzo(a)pyrene(BaP), and the related risk characterization was conducted for 113 samples out of 14 different agricultural products used for food and medicine. Detection rate of PAHs was 90.3% as a whole, and the highest one was 80.5% for BaP. The detection rate of BaP exceeding the maximum permitted concentration of Rehmanniae Radix Preparata and Rehmanniae Radix, 5.0 ㎍/kg was 1.8%, and the detection rates of BaA, Chr and BbF were within the range of 2.7~10.6%. The highest average concentration of BaA was 3.41 ㎍/kg detected from Lycii Fructus, while those of Chr, BbF, BaP and PAH4(sum of detected BaA, Chr, BbF and BaP) were 5.00, 1.79, 2.36, 12.36 ㎍/kg, respectively, detected from Rehmanniae Radix Preparata. As for the risk characterization on PAHs, the overall MOE(Margin of Exposure) values were measured within the range of 105~107, which is unlikely to cause direct health concerns, but the worring values of MOE were measured 6.57×104 for BaP and 6.10×104 for PAH4 from Rehmanniae Radix Preparata, which may require an improvement plan to reduce BaP contents.
This study was performed to investigate the contaminated status of endocrine disruptor-suspected pesticides in agricultural products distributed in northern Seoul in 20l0. We analyzed 33 kinds of pesticides by multiresidue analysis method in 3,081 samples. Twelve pesticidcs were detected 600 times and violated 22 times from the agricultural products. The highest frequency of detection was procymidone and that of violation was endosulfan. The highest rates of detection divided violation times as diazinon. In agricultural products, 466 samples had pesticide residues and 22 samples violated the maximum residue limits (MRLs). Agricultural products were also classified by type and red pepper powder belonged to the others was the highest detection rate at 70.7%, fruiting vegetables 35.3%, tea leaves 23.6%, stalk and stem vegetables 21.2%, fruits 14.6% and leafy vegetables 11.2%. The rates of violation sample were stalk and stem vegetables 1.1%, leafy vegetables 1.0% and roots and tubers 0.4%. Leek was to be managed primarily because it had the high detection and violation ratio and was detected together several pesticides.
This study was conducted to monitor 284 pesticides residue level in 2,914 commercial agricultural products in the northern area of Seoul in 2011 by the multi class pesticide multiresidue analysis methods in Korea Food Code using GC, HPLC, GC-MSD and, LC-MSD. The detection rate of pesticide residues were 14.8% (431/2,914). The order of agricultural products in which the pesticide residues were detected was perilla leaves 40.0% (28/70), chamnamul 35.5% (11/31), amaranth 30.0% (3/10) and spinach 27.7% (38/137) etc. The percentage of products that exceeded the MRLs (maximum residue limits) were 1.0% (31/431). Those products that exceeded MRLs were ginseng (6), perilla leaves (4), leek (4), welsh onion (3) and sedeum (3) etc. The 59 kinds of the pesticides were detected on this study, 21 pesticides of them were detected over MRLs. Detection rate of fungicides (56.4%) was higher than that of insecticides (42.1%). And procymidone was detected with considerable high frequency. Additionally, residual residual violates pesticides were in the order of tolclofos-methyl, endosulfan, dimethomorph, diniconazole and fludioxonil. According to the agricultural marketing channels, detection and excess rate of them were monitored. Detection and excess rate of samples circulated in agricultural traditional market were highest. The estimated daily intakes (EDI) of the pesticides were compared to personal acceptable daily intakes (PADI) in order to risk assessment by food consumption. Diazinon in Korean cabbage showed the highest %ADI, 2.9901 and others showed below 3.0 %ADI. Overall, these results indicate that residue levels of pesticides detected were evaluated as safe.

This work was conducted to apply the multi class pesticide multiresidue method for determining the use of hexaconazole in the agricultural products using GC-NPD. The multi class pesticide multiresidue method results were validated for the assay of hexaconazole by using linearity, accuracy, precision, limit of detection and quantitation. The linearity in the concentration ranged from 0.025 to 5.0 mg/L ($R^2$ > 0.999). Lettuce recoveries ranged from 89.42% to 94.15% with relative standard deviations below 7.78%, for spiking levels from 0.04 to 4.0 mg/kg. The limit of detection was 0.04 mg/kg, and the limit of quantitation was 0.11 mg/kg. The intra- and inter-day precisions were 2.42~3.49% and 4.90~7.78%, respectively. We suggested that the multi class pesticide multiresidue method for determining hexaconazole was highly accurate and reproducible, and it will be used as a routine analysis in agricultural products.

The modified QuEChERS method was evaluated for rapid determination of pesticide residues in spinach by gas chromatography-nitrogen phosphorous detector (NPD) and electron capture detector (ECD). Fifty GC-amenable pesticides which were most frequently detected in monitoring were selected in the current study. Matrix-matched calibration was performed. The detector response for all pesticides was linear with determination coefficients higher than 0.995. LODs for most compounds ranged between 0.001 and 0.1 ug/g, and about 90% of the compounds had LODs of less than 0.05 ug/g. LOQs ranged from 0.001 to 0.32 ug/g, which were well below the MRLs permitted for most of pesticides. In the majority of cases, the recoveries (80-120%) and relative standard deviations (RSDs) (less than 20%) were within acceptable levels except for dichlorvos, propamocarb, chlorothalonil, dichlofluanid, cyhalothrin and fenvalerate. Also, this method which is applied to routine samples showed good results when comparing with traditional multi-residue method.
The objective of this study was to examine the relationship between meat quality attributes and the changes of sarcoplasmic protein acetylation and myofibrillar protein acetylation in lamb longissimus thoracis et lumborum muscles at different postmortem phases. Protein acetylation, color, pH, shear force, myofibril fragmentation index and cooking loss were measured. The total level of acetylated sarcoplasmic proteins showed a negative relation with pH, a positive relation with a*, b* and cooking loss at the pre-rigor phase. Sarcoplasmic proteins acetylation affected postmortem pH by regulating glycolysis, which in turn affects color and cooking loss. The total level of acetylated myofibrillar proteins showed a positive relation with shear force at the pre-rigor phase. Myofibrillar proteins acetylation affected meat tenderness by regulating muscle contraction. This study indicated that acetylation played a regulatory role of meat color, water-holding capacity, and tenderization process at early postmortem.
This study investigated the residual characteristics of bifenthrin and chlorothalonil in crown daisy and suggested pre-harvest residue limits (PHRLs) based on their dissipation patterns and biological half-lives. The samples for residue analysis were harvested at 0 (3 hr), 1, 3, 5, 7, 9, 11, 13, 15, 18, 22 and 26 days after treatment, and analyzed by $GC/{\mu}-ECD$ and TOF/MS. The limit of quantitation (LOQs) of bifenthrin and chlorothalonil were 0.0046 mg/kg and 0.0007 mg/kg, respectively. Recoveries ranged from $88.67{\pm}7.97%$ and $99.90{\pm}16.03%$, showing that this method is appropriate for the analysis of the pesticide residues in crown daisy. Being well within first order kinetics, the biological half-lives of the pesticide residues in crown daisy were 9.63 days for bifenthrin and 6.54 days for chlorothalonil. The PHRLs of bifenthrin and chlorothalonil were recommended as 11.70 mg/kg and 24.10 mg/kg for 26 days before harvest, respectively.
