EFFECTS ON EATING AND RUMINATION BEHAVIOUR IN SHEEP OF FORMIC ACID AND FORMALDEHYDE TREATMENT AND METHIONINE-SUPPLEMENTATION TO LADINO CLOVER FIBROUS RESIDUE SILAGE

  • The effects of formic acid and formaldehyde treatment and methionine supplementation to ladino clover fibrous residue silage on eating and rumination behaviour were studied in sheep. From the ladino clover fibrous residue, two silage were prepared, either untreated or treated with formic acid and formaldehyde. Four experimental diets: untreated silage, treated silage, untreated silage with supplementation of methionine and treated silage with supplementation of methionine, were offered to four sheep at a restricted level of DM intake (2% of BW/d) twice daily in a two-way layout design. Methionine supplementation with the treated silage significantly (p < 0.05) reduced daily time spent eating, and consequently, markedly increased rate of eating. However, there was little effect of methionine supplementation on the daily time spent eating and eating rate for sheep offered untreated silage. Methionine supplementation with the treated silage reduced daily time spent ruminating, although the same effect was not observed for untreated silage. The rumination index (time spent ruminating/100 g DM eaten) was remarkably smaller (p < 0.05) with methionine supplement in feeding treated silage, although it did not differ for sheep offered untreated silage. There were no clear effect of methionine supplementation on the rumination efficiency (i.e. number of chews/bolus, bolus time and rumination chewing rate) both feeding untreated silage and treated silage.

Residual Characteristics of Insecticides Used for Oriental Tobacco Budworm Control of Paprika (파프리카 재배기간 중 담배나방 방제에 사용되는 살충제의 잔류특성)

  • BACKGROUND: This study was carried out to investigate the residual characteristics of insecticides used for Oriental Tobacco Budworm control and to establish the recommended pre-harvest residue limit leading to contribution in safety of paprika production. METHODS AND RESULTS: The recommended Pre-Harvest Residue Limits (PHRLs) of insecticides during cultivation of paprika were calculated from residue analyses of insecticides in fruits 1, 3, 5, 7, 10, 12, 15, 18 and 21 days after treatment. Paprika samples were extracted with QuEChERS method and cleaned-up with amino propyl SPE cartridge and PSA, and insecticide residues were analyzed either by HPLC/DAD or GLC/ECD. The limits of detection were 0.01 mg/kg for 5 insecticides. Average recoveries were $81.3{\pm}1.62%$-$98.3{\pm}1.58%$ of 5 insecticides at fortification levels of 0.1 and 0.5 mg/kg. The biological half-lives of the insecticides were 8.5 days for bifenthrin, 11.8 days for chlorantraniliprole, 16.8 days for chlorfenapyr, 7.1 days for lamda-cyhalothrin and 31.3 days for methoxyfenozide at recommended dosage, respectively. CONCLUSION(S): The pre-harvest residue limits for 10 days before harvest were recommended 1.05 mg/kg, 1.41 mg/kg, 0.93 mg/kg, 2.06 mg/kg and 1.08 mg/kg as bifenthrin, chlorantraniliprole, chlorfenapyr, lamda-cyhalothrin and methoxyfenozide, respectively. This study can provide good practical measures to produce safe paprika fruit by prevention of products from exceeding of MRLs at pre-harvest stage.

Residues and Exposure Assessment of Carbendazim in Chamnamul on Field Trials for Revising Maximum Residue Limit in Korea (생산단계 참나물의 Carbendazim 잔류특성에 따른 노출평가 및 농약 잔류허용기준 개선)

  • BACKGROUND: The residue dissipation pattern of pesticides for agricultural products during the pre-harvest period after the final application is important to prevent the maximum residue limit (MRL) violations in domestic and export markets. The MRL violations of carbendazim are observed more often in chamnamul by pesticide residue management surveys by the Ministry of Food and Drug Safety. The residue level at the pre-harvest interval (PHI) and the residue dissipation constant from the critical good agricultural practice (cGAP) trials could be estimated to meet the MRL and pose a health risk to consumers. METHODS AND RESULTS: Chamnamuls were harvested at 0, 1, 3, 5, 7, 10, and 14 days after application of carbendazim in accordance with critical GAP. The residue analysis in chamnanul was performed by HPLC-DAD with the C18 column. The limit of quantitation of carbendazim was 0.04 mg/kg, and the recoveries were 74.4 - 95.8% at the two spiked levels (LOQ and 10LOQ) of carbendazim. The dissipation rates in chamnamul were calculated from the residues at the sampling days by statistical method at a 95% confidence level. The biological half-lives of residual carbendazim in the field trials 1 and 2 were 4.9 and 4.4 days, respectively. CONCLUSION(S): In this dissipation study, the residue concentrations at the recommended PHI were higher than the established MRL in Korea. Therefore, the MRL is proposed based on the residue data sets from the trials conducted at the same cGAP and the dietary exposure assessment.

Effects of Application of Rendered Carcass Residue on Greenhouse Gases and Pepper Growth (랜더링된 가축사체 잔류물 시용이 온실가스 및 고추 생육에 미치는 영향)

  • The rendering residue generated by rendering disposal, an eco-friendly livestock carcass disposal method, is a useful agricultural resource. Methods for recycling this are being actively researched, and this study investigated the impact of applying rendered residue directly to soil on crop productivity and the agricultural environment. The chemical properties of the rendering residue were examined. The pH, OM, T-N, T-P, CaO, K2O, and MgO content values were 5.47%, 59.8%, 9.22%, 2.96%, 2.16%, 0.51% and 0.10%, respectively. Treatment conditions were divided into control, inorganic fertilizer, and rendering residue, and rendering residue corresponding to 50, 100, and 200% nitrogen content was applied based on the amount of inorganic fertilizer nitrogen input. Greenhouse gases and ammonia were collected during the cultivation period. Rendering residue increased both the yield and growth of peppers and was effective in improving nutrients such as pH and OM of the soil after harvest. However, compared to inorganic fertilizer treatment, it increased emissions of nitrous oxide and methane as well as ammonia. It is judged that the direct agricultural use of rendering residue is difficult, and a utilization method is needed.

Biological Half-lives of Fungicides in Korean Melon under Greenhouse Condition (시설재배 참외 중 살균제의 생물학적 반감기)

  • This study was conducted to know the biological half-lives and dissipation patterns of fungicides, pyrimethanil, chlorothalonil and tetraconazole in Korean melon under green house condition. The instrument for analyzing pyrimethanil and chlorothalonil was HPLC equipped with UV detector. Initial residue amounts of pyrimethanil were 0.16 mg/kg at recommended rate and 0.28 mg/kg at double recommended rate in Korean melon. The biological half-lives of pyrimethanil were 11.2 days at recommended rate and 10.1 days at double recommended rate in Korean melon. In case of chlorothalonil, initial residue amounts of chlorothalonil were 0.06 mg/kg at recommended and 0.11 mg/kg at double recommended rate in Korean melon. The biological half-lives of chlorothalonil in Korean melon were 3.4 days at recommended rate and 6.6 days at double recommended rate. The instrument for analyzing tetraconazole was GLC equipped with electron capture detector. Initial residue amounts of tetraconazole were 0.14 mg/kg at recommended and 0.22 mg/kg at double recommended rate in Korean melon, respectively. The biological half-lives of tetraconazole were 9.6 days at recommended rate and 18.5 days at double recommended rate in Korean melon.

Problems in the Dietary Exposure Assessment of Pesticide Residues (식품중 잔류농약에 의한 인체피폭 평가에서의 문제점)

  • The demand of safety assessment for pesticide residues in foods is growing in Korea. In order to assess the actual dietary exposure toward pesticide residues, the problems in utilization of pesticide residue data were reviewed and optimization of food factors applicable to Korean population was conducted. The problems and current status in setting pesticide residue tolerance-domestic and foreign-in foods were also argued. The conclusion is summarized as follows: (1) Anticipated residue estimates(ARE) calculated from field trial residue data, the percentage of crop treated, and the effects of cooking or processing should be utilized for dietary exposure assessment in a real sense. (2) Average daily intake of total foods per person in Korea was 1,104g in 1970's and 1,240g in 1980's. Much variation was observed between the two decades by food commodities. (3) Pesticide residue tolerances in Korea have been established at relatively lower levels in comparison with FAO/WHO or western countries. More extensive studies and data accumulation as related to tolerance setting are called for in future for the sake of health protection and smooth interstate trade.

Liquid Chromatographic Determination of Etofenprox Residues in Foods with Mass-Spectrometric Confirmation

  • BACKGROUND: An official analytical method was developed to determine etofenprox residues in agricultural commodities using high-performance liquid chromatography (HPLC). METHODS AND RESULTS: The etofenprox residue was extracted with acetone from representative samples of five raw products which comprised rice grain, apple, mandarin, cabbage, and soybean. The extract was then serially purified by liquid-liquid partition and Florisil column chromatography. For rice and soybean samples, acetonitrile/n-hexane partition was additionally coupled to remove nonpolar lipids. Reversed phase HPLC using an octadecylsilyl column was successfully applied to separate etofenprox from co-extractives. Intact etofenprox was sensitively detected by ultraviolet absorption at 225 nm. Recovery experiment at the quantitation limit validated that the proposed method could apparently determine the etofenprox residue at 0.02 mg/kg. Mean recoveries from five crop samples fortified at three levels in triplicate were in the range of 93.6~106.4%. Relative standard deviations of the analytical method were all less than 10%, irrespective of crop types. A selected-ion monitoring LC/mass spectrometry with positive atmospheric-pressure chemical ionization was also provided to confirm the suspected residue. CONCLUSION(s): The proposed method is simple, rapid and sensitive enough to be employed in routine inspection or monitoring of agricultural products for the etofenprox residue.

Residue safety on ethephon in soybean leaf by drenching and foliar application (에테폰의 관주처리와 엽면살포에 의한 콩 잎 중 잔류 안전성)

  • Ethephon is useful pesticide as ethylene precursor, which is an efficient plant hormone to produce functional secondary metabolites. However, the residual safety of ethephon was not studied on various crops. In here, the dissipation pattern of ethephon residue in soybean leaf was investigated both on the foliar and drenching applications. The biological half-lives of ethephon residues were 26.6, and 21.1 h on the once, and double foliar applications, respectively. Although the residue after three days from the final application was up to $60.6mg\;kg^{-1}$, the residue was below the limit of quantitation on the dried soybean leaf. In addition, drenching application of ethephon could increase the residue up to $36.3mg\;kg^{-1}$ after 20.1 h from the application, however, the treatment would not affect to the total phenol content significantly (p >0.01).

Utilization of Soymilk Residue for Wheat Doenjang (밀가루 고오지에 의한 두유박이용 밀된장 제조)

  • Wheat Doenjang was manufactured by mixing various ratio of the soymilk residue with wheat Koji. As the result, the content of reducing sugar was increased till 50 days and decreased thereafter during aging of the mash. As the combination ratio of soymilk residue was increased, the content of reducing sugar was decreased and amino nitrogen content was increased prominently till 50 days in all testing samples but thereafter the increasing ratio was slowdowned. At the beginning of the aging the content of the amino nitrogen was higher in the testing sample which had higher combination ratio of soymilk residue but at the end of the aging the result was reversed. The content of the total acids was increased prominently till 40 days but thereafter the increasing ratio was slowdowned, and in each testing samples the difference in total acid content was extremely slight. In the color of each testing samples aged 80 days as the combination ratio of soymilk residue was increased, the lightness was increased, and the redness was decreased but the yellowness was increased. From the result of sensory evaluation test of wheat Doenjang aged 90 days, soy sprout Doenjang soup showed the best taste in control and as the combination ratio of soymilk residue was increased, the taste was dropped slightly but no significant differences was obtained among the testing sample A,B and C but between control and testing sample D there was significant difference. In the original taste of Doenjang, the control was the best and as the combination ratio of soymilk residue increased, the point was dropped but there were no significant differences between control and testing sample A and testing sample B and C.

Cleavage-Dependent Activation of ATP-Dependent Protease HslUV from Staphylococcus aureus

  • HslUV is a bacterial heat shock protein complex consisting of the AAA+ ATPase component HslU and the protease component HslV. HslV is a threonine (Thr) protease employing the N-terminal Thr residue in the mature protein as the catalytic residue. To date, HslUV from Gram-negative bacteria has been extensively studied. However, the mechanisms of action and activation of HslUV from Gram-positive bacteria, which have an additional N-terminal sequence before the catalytic Thr residue, remain to be revealed. In this study, we determined the crystal structures of HslV from the Gram-positive bacterium Staphylococcus aureus with and without HslU in the crystallization conditions. The structural comparison suggested that a structural transition to the symmetric form of HslV was triggered by ATP-bound HslU. More importantly, the additional N-terminal sequence was cleaved in the presence of HslU and ATP, exposing the Thr9 residue at the N-terminus and activating the ATP-dependent protease activity. Further biochemical studies demonstrated that the exposed N-terminal Thr residue is critical for catalysis with binding to the symmetric HslU hexamer. Since eukaryotic proteasomes have a similar additional N-terminal sequence, our results will improve our understanding of the common molecular mechanisms for the activation of proteasomes.
