Rice variety IPB3S and IPB prima production technology to support food self-sufficiency in Indonesia

  • Dissemination of IPB3S rice variety combined with cultivation technology named IPB Prima was aimed to introduce IPB research product particularly for IPB rice variety with high-yield character that is IPB3S. The rice variety IPB3S and IPB Prima cultivation technology was expected to be one of solution to improve rice productivity and accelerate to food self-sufficiency in Indonesia. Research sctivity was consist of three main research unit i.e. (1) Dissemination of IPB3S rice variety and IPB Prima production technology; (2) The development of Information and management web-based system (IMS) for planning and monitoring IPB3S and IPB Prima application distribution; and (3) The development of High-capacity grain drying system in Fluidized-bed drying ang in-store drying system. The objective of main research i.e. to introduce IPB high-yield rice variety, to accelerate rice productivity to support self-sufficiency, to develop integrated system model through fluidized and in-store drying, and to develop web-based management-information system in result analyzing IPB3S and IPB Prima distribution and technology application. The dissemination activities was arranged in two location. The first location was in Banyuwangi, East Java with total area 10.87 ha, consist of 8.91 ha planting area for IPB3S and 1.96 ha planting area for Ciherang. The second location is in Tegal, Middle Java with total planting area in 5 ha. The experiment was arranged in different treatment of varieties and cultivation method. The experiment consist of (1) rice variety Ciherang with conventional cultivation technology (P0); (2) rice variety Ciherang with IPB Prima cultivation technology (P1); (3) rice variety IPB3S with conventional cultivation technology (P2); (4) rice variety IPB3S with IPB Prima cultivation technology (P3). Planting distance for twin rows system is $50cm{\times}25cm{\times}12.5cm$. Planting distance for single row system is $25cm{\times}25{\times}cm$. The research result elucidated that productivity result in two location has different grades in similar trend. Experiment in Tegal resulted P0 result is $6.18ton\;ha^{-1}$, P1 result is $630ton\;ha^{-1}$, P2 result is $6.82ton\;ha^{-1}$, P3 result is $7.31ton\;ha^{-1}$. Experiment in Banyuwangi resulted optimum production of IPB3S variety productivity number are 7.29 ton ha-1, while Ciherang are $6.73ton\;ha^{-1}$.

  • The purpose of this study was carried out to determine the possibility of in vitro maturation of tiger oocytes. Immature oocytes were recovered from a pairs of ovaries. A total of 78 oocytes were collected, of which forty threes were identified as compact cumulus cells and uniform cytoplasm. 43 COCs were in vitro matured at 39℃, 5% CO₂ in air atmosphere for 48 h in a IVM medium (TCM-199 supplement with 10% FBS, 0.6 mM cysteine, 0.2 mM pyruvic acid and 10 IU/㎖ HMG).   • Rhizobium tropici CIAT 899 is capable of synthesizing inactive apo-glucose dehydrogenase (GDH). To become an active holo enzyme, the GDH requires a cofactor, PQQ. When R. tropici CIAT 899 was grown in a broth culture medium containing hydroxyapatite and pyrrolo quinoline quinone (PQQ), pH decreased while the concentration of soluble P increased. The solubilization of hydroxyapatite was associated with the production of gluconic acid and 2-ketogluconic acids. The organic acid production and P solubilization were greatly enhanced when the bacterium was grown with air supply. Effect of R. tropici CIAT 899 with (CI+PQQ) and without PQQ (CI) on the common bean growth was examined. Shoot and root weight, and N and P contents in CI+PQQ treatment, were significantly higher than those in control and CI treatment. Nodule weight and acetylene reducing activities were also significantly higher in CI+PQQ treatment than in other treatments.

Cacao bean husk: an applicable bedding material in dairy free-stall barns

  • Objective: The objectives of the study were to assess the effect of cacao bean husk as bedding material in free-stall barn on the behavior, productivity, and udder health of dairy cattle, and on the ammonia concentrations in the barn. Methods: Four different stall surfaces (no bedding, cacao bean husk, sawdust, and chopped wheat straw) were each continuously tested for a period of 1 week to determine their effects on nine lactating Holstein cows housed in the free-stall barn with rubber matting. The lying time and the milk yield were measured between d 4 and d 7. Blood samples for plasma cortisol concentration and teat swabs for bacterial counts were obtained prior to morning milking on d 7. The time-averaged gas-phase ammonia concentrations in the barn were measured between d 2 and d 7. Results: The cows spent approximately 2 h more per day lying in the stalls when bedding was available than without bedding. The milk yield increased in the experimental periods when cows had access to bedding materials as compared to the period without bedding. The lying time was positively correlated with the milk yield. Bacterial counts on the teat ends recorded for cows housed on cacao bean husk were significantly lower than those recorded for cows housed without bedding. Ammonia concentration under cacao bean husk bedding decreased by 6%, 15%, and 21% as compared to no bedding, sawdust, and chopped wheat straw, respectively. The cortisol concentration was lowest in the period when cacao bean husk bedding was used. We observed a positive correlation between the ammonia concentrations in the barn and the plasma cortisol concentrations. Conclusion: Cacao bean husk is a potential alternative of conventional bedding material, such as sawdust or chopped wheat straw, with beneficial effects on udder health and ammonia concentrations in the barns.

Status of Plasma Technology Applied to Agriculture and Foods (플라즈마 기술의 농식품 분야 활용)

  • Recently, plasma technology has been increasingly applied to agriculture and foods. Owing to the three important plasma characteristics which are activation, sterilization, and catalysis, the plasma technology can be properly applied to agriculture, in the other word, to the whole cycle of agriculture from farm to table: seed germination, plant growth, harvest and storage, washing, packaging, transport, in store, household, cooking, garbage, etc. Some representative case studies for plasma activation, sterilization, and catalysis show well that plasma technology can be successfully applied to the whole cycle of agriculture.

New Evidence of Alleles (V199I and G52S) at the PRKAG3 (RN) Locus Affecting Pork Meat Quality

  • The porcine PRKAG3 (RN) gene encodes the regulatory gamma subunit of adenosine monophosphate-activated protein kinase (AMPK), which is a good candidate gene affecting meat quality. In this study, the effects of two missense mutations A595G (Ile199Val) and G154A (Gly52Ser) in porcine PRKAG3 gene on meat quality traits were studied in M. Longissimus dorsi (LD), M. Semispinalis capitis (SC) and M. Biceps femoris (BF) from different populations of 326 pigs. The PRKAG3 alleles 199I, 199IV, 52S and 52G were identified with PCR-RFLPs and all genotypes - 199I/199I, 199I/199V, 199V/199V, 52S/52S, 52S/52G and 52G/52G - were found. The frequency of V allele was larger than that of I allele in all populations. I allele frequency was zero in Chinese Meishan pigs (population D) especially. G allele frequency was larger than that of S allele in all populations except Large White (population A). Both variations at the PRKAG3 locus significantly affected these meat quality traits. The pork meat quality has not previously been established in Meishan or crosses thereof. The results suggested that generally pH of LD, SC and BF was higher in Meishan pigs than that in other populations. Moreover, Meishan pigs showed higher water-holding capacity and intramuscular fat (IMF), lower water content and water loss percentage compared to other populations in terms of the two variations. The results present here supply new evidence that alleles V199I and G52S at the PRKAG3 locus affect pork meat quality and provide useful information on pork production.
