  • The pallet loading pattern and efficiency were analyzed on the packaging boxes for agricultural products. Also, the pallet layout chart was proposed as a convenient tool in selecting the suitable packaging dimensions for them. By researching the layout pattern and maximum number of agricultural products in the packaging box, we can choose the efficient box dimension.

Pesticide Residues Survey on Agricultural Products before Auction at Whole Market in Busan Area during 2006~2008 (2006~2008년 부산지역 농산물도매시장 경매 전 농산물의 잔류농약 조사)

  • This study was conducted to monitor the current status of pesticide residues in agricultural products before auction at whole markets in Busan area from March 2006 to December 2008. Of the 7,237 samples that were analyzed by multiresidue methods, 1,164 samples (16.1%) had pesticide residues and 231 samples (3.2%) exceeded the maximum residue limits (MRLs). 181 samples of the 231 samples exceeding MRLs were applied to the tentative limits because the use of the pesticides for the agricultural products was not established against the MRLs in the Korea Food Code, and it is equivalent to 78.4%. The order of pesticides exceeding MRLs was endosulfan, procymidone, chlorothalonil, ethoprophos, chlorpyrifos, diethofencarb, kresoxim-methyl and EPN. Of 211 samples with two or more pesticides residues, the residues exceeded the MRLs in 80 samples and it is equivalent to 37.9%. This study results suggested that pesticide residues exceeding MRLs could be attributed to the use of inapplicable pesticides for agricultural products and the use of two or more pesticides.

  • This study aims to examine consumers' trust and purchase intention in environment-friendly agricultural products and factors influencing purchase intention degree of environment-friendly agricultural products. In addition, it aims to help seek national health promotion methods through purchase of consumers' environment-friendly agricultural products by providing the government, businesses, consumer organizations, etc. with basic data on them. The survey was performed for 2 months from August 1, 2012 to September 30, 2012, and a total of 350 questionnaires were distributed and 321(91.7%) copies out of them were used as final analysis data. The results of the study are as follows. First, the analysis of the effects of environment-friendly consumption value on trust in environment-friendly agricultural products showed that all the independent variables including environmental conservation value and economic value had a significant influence on trust in environment-friendly agricultural products, and economic value among them was the most influential item. Second, the analysis of the effects of environment-friendly consumption value on purchase intention for environment-friendly agricultural products showed that all the independent variables including environmental conservation value and economic value had a significant influence on purchase intention for environment-friendly agricultural products, and economic value was the most influential item. Third, the analysis of the effects of trust in environment-friendly agricultural products on purchase intention for environment-friendly agricultural products showed that as trust in environment-friendly agricultural products gets higher, consumers' purchase intention for them increased.

Heterologous Expression of a Thermostable α-Galactosidase from Parageobacillus thermoglucosidasius Isolated from the Lignocellulolytic Microbial Consortium TMC7

  • α-Galactosidase is a debranching enzyme widely used in the food, feed, paper, and pharmaceuticals industries and plays an important role in hemicellulose degradation. Here, T26, an aerobic bacterial strain with thermostable α-galactosidase activity, was isolated from laboratory-preserved lignocellulolytic microbial consortium TMC7, and identified as Parageobacillus thermoglucosidasius. The α-galactosidase, called T26GAL and derived from the T26 culture supernatant, exhibited a maximum enzyme activity of 0.4976 IU/ml when cultured at 60℃ and 180 rpm for 2 days. Bioinformatics analysis revealed that the α-galactosidase T26GAL belongs to the GH36 family. Subsequently, the pET-26 vector was used for the heterologous expression of the T26 α-galactosidase gene in Escherichia coli BL21 (DE3). The optimum pH for α-galactosidase T26GAL was determined to be 8.0, while the optimum temperature was 60℃. In addition, T26GAL demonstrated a remarkable thermostability with more than 93% enzyme activity, even at a high temperature of 90℃. Furthermore, Ca2+ and Mg2+ promoted the activity of T26GAL while Zn2+ and Cu2+ inhibited it. The substrate specificity studies revealed that T26GAL efficiently degraded raffinose, stachyose, and guar gum, but not locust bean gum. This study thus facilitated the discovery of an effective heat-resistant α-galactosidase with potent industrial application. Meanwhile, as part of our research on lignocellulose degradation by a microbial consortium, the present work provides an important basis for encouraging further investigation into this enzyme complex.

Metagenomic Insight into Lignocellulose Degradation of the Thermophilic Microbial Consortium TMC7

  • Biodegradation is the key process involved in natural lignocellulose biotransformation and utilization. Microbial consortia represent promising candidates for applications in lignocellulose conversion strategies for biofuel production; however, cooperation among the enzymes and the labor division of microbes in the microbial consortia remains unclear. In this study, metagenomic analysis was performed to reveal the community structure and extremozyme systems of a lignocellulolytic microbial consortium, TMC7. The taxonomic affiliation of TMC7 metagenome included members of the genera Ruminiclostridium (42.85%), Thermoanaerobacterium (18.41%), Geobacillus (10.44%), unclassified_f__Bacillaceae (7.48%), Aeribacillus (2.65%), Symbiobacterium (2.47%), Desulfotomaculum (2.33%), Caldibacillus (1.56%), Clostridium (1.26%), and others (10.55%). The carbohydrate-active enzyme annotation revealed that TMC7 encoded a broad array of enzymes responsible for cellulose and hemicellulose degradation. Ten glycoside hydrolases (GHs) endoglucanase, 4 GHs exoglucanase, and 6 GHs β-glucosidase were identified for cellulose degradation; 6 GHs endo-β-1,4-xylanase, 9 GHs β-xylosidase, and 3 GHs β-mannanase were identified for degradation of the hemicellulose main chain; 6 GHs arabinofuranosidase, 2 GHs α-mannosidase, 11 GHs galactosidase, 3 GHs α-rhamnosidase, and 4 GHs α-fucosidase were identified as xylan debranching enzymes. Furthermore, by introducing a factor named as the contribution coefficient, we found that Ruminiclostridium and Thermoanaerobacterium may be the dominant contributors, whereas Symbiobacterium and Desulfotomaculum may serve as "sugar cheaters" in lignocellulose degradation by TMC7. Our findings provide mechanistic profiles of an array of enzymes that degrade complex lignocellulosic biomass in the microbial consortium TMC7 and provide a promising approach for studying the potential contribution of microbes in microbial consortia.

  • There still exist adhesives that have toxic compounds or consist of those materials in both our life and lots of industries. For instance, formaldehyde adhesive had been still used for woodworking and some medical adhesives had been considered as toxicity problems. In this situation, natural adhesives from raw materials have been suggested as an one of considerable interests. Natural adhesives in recent have been reported fabrication methods via biological materials such as proteins, celluloses, and starches. By-products derived from agricultural something have them richly and each has additional special properties. Using these properties to make natural adhesives, unique adhesives would be attained. In particular, rice-based adhesive is such a good example. Rice-based by-product adhesives have interestingly some pros pertaining to high adhesive strength, excellent water resistance and could dramatically be reduced a formaldehyde a harmful component of plywood. Hence, rice-based adhesive like glues could be applied to lots of industries including agricultural and biological technology. This review paper highlighted some recent development on natural adhesives as a promising biomaterial for agricultural and biological technology fields. Development of Reinforced Bio-filament Composites Composed of Agricultural By-product for 3D Printing Technologies

  • In this study, biocomposite filaments with agricultural by-products can be used in extrusion-based 3D (Three-dimensional) printing. Extrusion-based 3D printing stands as a promising technique owing to its versatility. We hypothesized that bio-filament composite consisted of something derived from agricultural by-products could be used as 3D printing materials that could overcome the drawbacks of PCL (poly-caprolactone). Bio-filament mixed with PCL and agricultural by-products was defined as r-PCL in this study. In order to find it out the optimal mixing ratio of filaments, we had investigated PCL, r-PCL 10%, r-PCL 20%, r-PCL 50% separately. The morphological and chemical characteristics of the filaments were analyzed by FE-SEM (Field emission scanning electron microscope) and EDX (Energy-dispersive X-Ray spectroscopy), and the mechanical properties were evaluated by stress-strain curve, water contact angle, and cytotoxicity analysis. Results of this study have been shown as a promising way to produce eco-friendly bio-filaments composite for FDM (Fused deposition modeling) method based 3D printing technology. Thus, we could establish biomimetic scaffolds based on bio-printer filaments mixed with agricultural by-product.

Relationships among Recognition, Preference, and Purchasing Characteristics for Local Agricultural Products and Festival Satisfaction of Changnyeong Area - Compared by the Gender, Age, and Purchase Experience - (창녕지역 축제만족도와 농특산물 인지도, 선호도, 구매특성과의 관련성 - 성별과 연령, 구매경험을 중심으로 -)

  • The purpose of this study was to investigate the relationships among recognition, preference, and purchasing characteristics for Changnyeong onions and garlic as well as festival satisfaction among participants at agricultural product festivals in the Changnyeong region. Results showed that festival satisfaction of 'place of festival' was highest while 'convenience facility & event contents' earned the lowest scores. Most subjects (90.5%) had purchased Changnyeong agricultural and processed products. A major purchasing type was fresh agricultural products (66.7%). The pathways to recognize Changnyeong agricultural products were mostly 'promotion by related institutions' (22.0%), 'family relatives' (20.8%), 'mass media' (16.6%), and 'festivals and events' (16.1%). The most considered factors for purchasing regional products were 'geographical origin' and 'ingredients'. Changnyeong onion showed higher scores for recognition and preference and rate of purchase experience and intention than for garlic. The correlation coefficients of recognition and preference for onion and galic were 0.603 (p<0.001) and 0.598 (p<0.001), respectively. The explanation power ($R^2$) of related variables for purchase of Changnyeong onions was 0.258. The regression coefficients (${\beta}$) for 'recognition', 'preference' and 'convenient facility & event contents' were positive, whereas the regression coefficient for 'price' was negative. Recognition, preference, and convenient facility & event contents with garlic purchase showed a positive relationship ($R^2$=0.253). The most effective promotion method to increase sales of Changnyeong agricultural products was 'local festivals and events' (27.8%).

Effects of Competitiveness Improvement Measures on Red-pepper and Garlic (고추, 마늘의 경쟁력제고사업 효과 분석)

  • This study attempts to investigate the possible extent that main spicy vegetables' competitiveness can be improved through various measures. More widely open the agricultural market becomes, more important enhancing the competitiveness of domestic agricultural products is especially in high tariff products such as red-pepper and garlic. Up to date, numerous measures have been applied to those products for enhancing the competitiveness by government, regional agencies, and producer's organizations. However, there is few studies about how much the competitiveness can be improved as a whole from implementing the diverse methods. Following the results, the competitiveness of red-pepper and garlic can be improved up to 70% and 50% respectively by reducing production costs.
