An Integrated Genomic Resource Based on Korean Cattle (Hanwoo) Transcripts

  • We have created a Bovine Genome Database, an integrated genomic resource for Bos taurus, by merging bovine data from various databases and our own data. We produced 55,213 Korean cattle (Hanwoo) ESTs from cDNA libraries from three tissues. We concentrated on genomic information based on Hanwoo transcripts and provided user-friendly search interfaces within the Bovine Genome Database. The genome browser supported alignment results for the various types of data: Hanwoo EST, consensus sequence, human gene, and predicted bovine genes. The database also provides transcript data information, gene annotation, genomic location, sequence and tissue distribution. Users can also explore bovine disease genes based on comparative mapping of homologous genes and can conduct searches centered on genes within user-selected quantitative trait loci (QTL) regions. The Bovine Genome Database can be accessed at http://bgd.nabc.go.kr.

A Study on the Influx of Population by Rural Tourism in Rural Village (농촌마을의 농촌관광 시행에 따른 인구유입효과에 관한 연구)

  • Settlement problems in rural area are gradually becoming more intense due to decreasing agricultural income led by market globalization. Rural tourism is considered as one of alternatives for complementing agricultural income. In this study, we analyzed the relation of rural development project and the influx of population on green tourism villages. Results show that the influx of population in rural area is related with project cost, tourism income, visiting numbers, and ratio of labor population. With these results, we modified rural population model for estimating future rural population. Adapted result to Buraemi village by modified model showed that estimation error can be decreased from 7.23% to 0.95%.

  • The purpose of this study is to develop a estimation model of migration with only population data using the cohort-survival model which has been used for forecasting future population. The fluctuation of population can be bisected to the natural change which can be occurred by birth and death and the social change which means migration. The factors of the social change are usually very important for establishing rural policies. However, researches using migration data has limitations because the usage of them are restricted. For verifying a estimation model of migration, comparing estimated population in 2000 year and migration quantity between 1996 and 2000 of 25 gu with real values, using population data and death ratio from 1995 to 2000 of the 25 gu in Seoul. Result shows a reliable data that R-square of forecating population model is 0.9755 and migration is 0.9180. So these model are worth to estimate a population and migration quantity to restricted migration data.

  • The purpose of this study is to analyze the factors affecting residential mobility between urban and rural. After classifying urban and rural region based on discriminatory attributes of the regions, we applied a multinomial logistic model, using the sample data of 2020 Korea Population and Housing Census. The major findings are as follows. The young highly educated in cities avoided rural. The young less educated in rural engaged in 2, 3th industries as well as agricultural industry, but remained in low-paying and unstable jobs. In addition, various classes moved to rural and rising house prices in cities pushed people to rural. Therefore, it is necessary to develop diversified regional industry models and provide opportunities for high quality and stable jobs in rural by linking industrial demand, education and jobs. Also, preserving the rural environment, settlement conditions and residential environment are needed for satisfying various needs of urban residents who migrate to rural areas. While regional policies so far have focused on maintaining the population size and promoting a population influx, rural development and population policies should be established in a way that responds to diverse population classes in an era of population decline.

Genetic Diversity and Population Structure of Mongolian Wheat Based on SSR Markers: Implications for Conservation and Management

  • Production of spring wheat, the major crop in Mongolia, accounts for 98% of the cultivated area. Understanding genetic variability in existing gene bank accessions is critical for collection, conservation and use of wheat germplasms. To determine genetic diversity and population structure among a representative collection of Mongolian local wheat cultivars and lines, 200 wheat accessions were analyzed with 15 SSR markers distributed throughout the wheat genome. A total of 85 alleles were detected, with three to five alleles per locus and a mean genetic richness of 5.66. Average genetic diversity index was 0.69, with values ranging from 0.37-0.80. The 200 Mongolian wheat accessions were mainly divided into two subgroups based on structure and phylogenetic analyses, and some phenotypes were divergent by the subgroups. Results from this study will provide valuable information for conservation and sustainable use of Mongolian wheat genetic resources.

  • The purpose of this study was to interpret the transformation of Cooperative Groups for Agricultural Production(CGAP) with the change of the Agricultural Productive Force. The specific objectives were; 1) To investigate the change of agricultural labour-power, 2) To investigate the change of agricultural mechanization and arable land, 3) To interpret the transformation and content of CLAP. The population of farmhouseholds has decreased continuously since the late 1960s. Especially, with the move-outs of youth ages of twenties to forties, the condition of agricultural labour-power has been more serious. The processing of agricultural mechanization was a small scale step in the 1970s, but after the 1980s there was a spread of middle-large machines. However the usage rate of agricultural machines was constrained by the bad conditions of arable land. From the 1970s to now, the CGAP have bean processed by many kinds of patterns. In the 1970s, the lack of labour-power caused the creation of the Co-Working Team. After the late of 1970s, the wage of agricultural employees was raised, because the working population of agriculture was cut down. Also, the induction of agricultural machine was promoted. As a result, in the 1980s, the Machine-Using Team occurred due to these conditions of agricultural productive force. In the late of 1980s, the population decreased more rapidly, and the use of large machines were spread. Than farmhouseholds laking labour-power gave a trust to other farmhouseholds and Teams which had machines. In 1990, Given-Trust Cooperations were enacted by law, and in order to overcome the lack of labour-power, and solve the problem of the successors of agriculture, Cooperative Organizations were also enacted by law. Finally, in Korea from the 1970s to now, as the agricultural productive force has barn changed, the Co-Working Team was transformed into the Machine-Using Team, and the Machine-Using Team was transformed into the Given-Trust Cooperation, and the Cooperative Organization.

Genetic diversity and population structure in five Inner Mongolia cashmere goat populations using whole-genome genotyping

  • Objective: As a charismatic species, cashmere goats have rich genetic resources. In the Inner Mongolia Autonomous Region, there are three cashmere goat varieties named and approved by the state. These goats are renowned for their high cashmere production and superior cashmere quality. Therefore, it is vitally important to protect their genetic resources as they will serve as breeding material for developing new varieties in the future. Methods: Three breeds including Inner Mongolia cashmere goats (IMCG), Hanshan White cashmere goats (HS), and Ujimqin white cashmere goats (WZMQ) were studied. IMCG were of three types: Aerbas (AEBS), Erlangshan (ELS), and Alashan (ALS). Nine DNA samples were collected for each population, and they were genomically re-sequenced to obtain high-depth data. The genetic diversity parameters of each population were estimated to determine selection intensity. Principal component analysis, phylogenetic tree construction and genetic differentiation parameter estimation were performed to determine genetic relationships among populations. Results: Samples from the 45 individuals from the five goat populations were sequenced, and 30,601,671 raw single nucleotide polymorphisms (SNPs) obtained. Then, variant calling was conducted using the reference genome, and 17,214,526 SNPs were retained after quality control. Individual sequencing depth of individuals ranged from 21.13× to 46.18×, with an average of 28.5×. In the AEBS, locus polymorphism (79.28) and expected heterozygosity (0.2554) proportions were the lowest, and the homologous consistency ratio (0.1021) and average inbreeding coefficient (0.1348) were the highest, indicating that this population had strong selection intensity. Conversely, ALS and WZMQ selection intensity was relatively low. Genetic distance between HS and the other four populations was relatively high, and genetic exchange existed among the other four populations. Conclusion: The Inner Mongolia cashmere goat (AEBS type) population has a relatively high selection intensity and a low genetic diversity. The IMCG (ALS type) and WZMQ populations had relatively low selection intensity and high genetic diversity. The genetic distance between HS and the other four populations was relatively high, with a moderate degree of differentiation. Overall, these genetic variations provide a solid foundation for resource identification of Inner Mongolia Autonomous Region cashmere goats in the future.

Development and characterization of eleven microsatellite markers for a popular pet stag beetle, Dorcus hopei (Coleoptera, Lucanidae) using paired-end Illumina shotgun sequencing

  • Eleven polymorphic microsatellite loci were developed and characterized for Dorcus hopei in this study. The number of alleles varied from 2 to 21. The observed heterozygosity and expected heterozygosity ranged from 0.1058 to 0.9744 and 0.0997 to 0.8941, respectively. Two loci showed low polymorphism, while the rest were highly polymorphic. Six loci deviated from Hardy-Weinberg Equilibrium. The set of markers will provide effective tools for examining the population genetic structures and be helpful for managing wild population in D. hopei.

Variation in Population Size of Mudfish by Agricultural Practices in Paddy Fields (논 생태계에서 영농방법에 따른 미꾸라지개체군의 변동 요인 분석)

  • BACKGROUND: This study was carried out to compare population size of mudfish (Misgurnus mizolepis) between the agricultural practices and to investigate the causes of its differences. We also provided basic information for sustainable use of mudfish population in paddy fields. METHODS AND RESULTS: Mudfish and benthic invertebrates which are diet of mudifish were investigated from 8 sites of organic and conventional rice paddy fields in South Korea. Total number of mudfish were 1,882 individuals in survey sites. Mudfish population were 2.4 times larger in organic paddy fields (1,333 individuals) than in conventional paddy fields (549 individuals). The population size of mudfish was larger in Mungyeong-si, Gyeongsangbuk-do and Anseong-si, Gyeonggi-do with relatively better environmental conditions than the other 5 sites including Suwon-si, Gyeonggi-do, Cheongyang-gun, Chungcheongnam-do, Gunsan-si, Jeollabuk-do, Gimje-si, Jeollabuk-do, Hwasun-gun, Jeollanam-do, Hamyang-gun, Gyeongsangnam-do. Benthic invertebrates collected from survey sites were 74 species, 68 genera, 46 families, 19 orders, 6 classes and 3 phyla. According to agricultural practices, benthic invertebrates were identified 66 species, 62 genera, 41 families, 17 orders, 6 classes and 3 phyla in organic paddy fields while there were 66 species, 60 genera, 42 families, 18 orders, 6 classes and 3 phyla in conventional paddy fields. Dominant invertebrates were Chironomidae sp., Branchiopoda sp., Ostracoda sp., and Copepoda sp. There were no differences in dominant species between organic and conventional paddy fields. Population size of mudfish tended to increase with the population size of Chironomidae sp., Branchiopoda sp., Ostracoda sp., and Copepoda sp. But, only population of Chironomidae sp. and Copepoda sp. statistically related to population size of mudfish. The number of individuals of mudfish (Misgurnus mizolepis) was higher at the low rate of urban area than any other surveyed region and was affected by appearance ratio of main preys such as Chironomidae sp. and Ostracoda sp. CONCLUSION(S): The population size of mudfish in rice paddy fields could be affected by environmental conditions and agricultural practices such as organic and conventional methods.
