Improvement of Antigen Blotting in a Tissue Blot Immunobinding Assay for the Detection of Two Chili Pepper Viruses

  • The tissue blot immunobinding assay (TBIA) is widely used for the detection and localization of plant viruses in various plant tissues. The basic experimental procedures of TBIA sampling and blotting were simplified using commercially available micropipette tips. This method was termed the ring-blot immunobinding assay (R-BIA), as the blot on the membrane forms a ring shape. The detection efficacy of R-BIA was tested for two chili pepper viruses, pepper mild mottle tobamovirus (PMMoV) and pepper mottle potyvirus (PepMoV), following the optimized serological procedures of TBIA (length of the incubation period and BSA concentration, and primary and secondary antibodies). Sensitivity of the R-BIA was about 1 ng/ml of purified PMMoV in pepper leaf sap from a healthy pepper plant. R-BIA also showed high specificity in the detection of PMMoV and PepMoV. Moreover, the modified sampling and blotting procedures were simpler and more reliable than other TBIA methods (such as whole-leaf blotting and crushed-leaf blotting), suggesting that the R-BIA may be used for medium- to large-scale detection of plant viruses in laboratories with minimal facilities.

  • It is necessary to evaluate the location suitability for protected horticulture to plan new protected horticultural facilities in rural areas. For objective and systematic evaluation of location suitability for protected horticulture, three different evaluation models were constructed based on grading, proportion, and analytic hierarchy process(AHP). The three methods were tested to evaluate the location suitability for protected horticulture in three selected regions; suburban, plain, mountainous. The results indicated that the grading method underestimated the difference of importance among grouping categories, compared to the proportion and AHP methods. Based on the results, the AHP method was founded to be better than the other two methods in terms of evaluation process, reproductivity and reliability of the results.

A Study on the Patterns of Medical Utilization among Inhabitants in Ulnung Island (울릉도 주민들의 의료이용 형태)

  • Authors surveyed the inhabitants in Ulnung Island to assess the patterns of medical utilization. One hundred eighty six population(65 male and 121 female) were surveyed with formed questionnaire from Aug. 16 to Aug. 19 in 1994. Results are as follows. 1. The prevalence rate of acute diseases was 19.3%. 2. The prevalence rate of chronic diseases was 35.0%. In classification of chronic disease, the disease of musculoscletal system was the highest(33.9%) and that of digestive system in next order. 3. The first-visit medical facility when disease developed was community health center mainly. The admission care was taken in 37.6%. The 80.0% among location of medical facility for admission care was out of island. The surgical operative care were taken in 19.9%. The 86.5% among location of medical facility for surgical operative care was out of island. 4. Among the contents of dissatisfaction for medical service within island, 'Insufficient equipment' was the highest(35.8%), and 'Insufficient traffic networks' in next order. The results of this study suggest that public health facilities and medical personnel be strengthened and emergency transfer system be secured in Ulnung Island.

Comparison of Perceptions of Local Residents and Public Health Center Staffs on the Performance of Medical Service Improvement Plan in Rural Areas (농어촌 의료서비스 개선사업 성과에 대한 지역주민과 사업담당자간의 인식 비교)

  • Objectives: The purpose of this study is to compare the perceptions of the rural healthcare service improvement project' performance and reorganization of public health centers between project staffs and local residents. Methods: Data collection from this study was performed in 141 project areas using structured questionnaires. Data analysis was used in SPSS 22.0 version. Results: The public health center staffs were more positive about the items for improving health facilities than the local residents. Residents in the Si area generally perceived performance as more positive than residents in the Gun area, while public health center staffs in the Gun area perceived performance as more positive than public health center staffs in the Si area. Local residents expressed negative opinions about the reduction in the number of branches of public health clinics and health medical clinics. Conclusions: In conclusion, careful improvement projects for rural health care and the establishment of health care systems will be necessary, reflecting the opinions of local residents, along with a variety of regional characteristics.

  • This study aims to identify limitations and pending problems after reviewing the overall policies and status of rural Newtown projects, and to seek solutions to its problems. This study targeted the villages of 5 districts, which were developed as rural new-towns after 2009 and included the basic status and progress of the pilot districts. This study conducted a literature review to examine the basic status and progress of the pilot districts, and based on this, analyzed the demand prediction, site selection, project implementation, and housing and amenity facilities of the pilot districts. The study methods included literature reviews, on-site surveys, interviews with village representatives, and consultations with experts. According to the analysis results, a low occupancy rate of the Newtown project districts is because the prediction of occupancy demand was incorrectly completed before implementing the projects. Also, the eligibility for occupancy, such as age restriction and mandatory farming was too strict. Other problems included an absence of income generation support policies for rural returnees, a housing supply policy in disregard of agricultural characteristics, and a lack of understanding of maintenance of communal space, etc.

  • Transformation of housing space is the most accurate physical trail to understand main needs from residents. The scope of the study targeted the 7 si & gun districts in Gyeongsangnam-do, this study is designed to consider the characteristics of space transformation in rural apartment houses and provide basic materials for the development of rural apartment houses. The results of the study are as follow. First, such spaces as corridor and stairway are transformed into storages and outside the houses are installed tap water facility, bicycle storage. and toilet. Second, in autumn seasons when crops and grains are harvested, parking lot and rooftop are used as the space drying such agricultural products. Third, space transformation within the houses has been different depending on family type and main businesses in terms of function and time to seek efficiency and stability of residence. Fourth, rural residents concentrate on a simple change of space usage and replacement of ageing or dilapidated equipment and facilities rather than space transformation incurring more financial burden.

Physical Properties of Rice Husk (왕겨의 물리적 성질)

  • Kinetic friction coefficient, bulk density, dynamic and static angle of repose, and terminal velocity of rice husk at the moisture range 7 to $23\%$ w.b. were determined. It could lead to better design and operation of the processing machinery and handling facilities. Friction coefficient was determined from the horizontal traction force measured by pulling the container holding a mass of rice husk on various plate materials. Dynamic angle of repose was calculated from the photos of bulk samples piled by gravity flow on a circular platform. Static angle of repose was determined by measuring the side angle of the bulk material which was left in a cylindrical container after natural discharge of the bulk sample through a circular hole in the bottom plate. Kinetic friction coefficients of rice husk were in the range of $0.254\~0.410,\;0.205\~0.520,\;0.229\~0.400,\;and 0.133\~0.420$ on PVC, mild steel, galvanized steel, and stainless steel, respectively. Bulk density, dynamic and static angle of repose, and terminal velocity were in the range of $91.7\~98.3$ $kg/m^3$, $40.2\~47.6^{\circ},\;52.8\~83.7^{\circ},$ and $1.36\~1.73$ m/s, respectively. These physical properties of rice husk increased linearly as the moisture content increased.

Some Physical Properties of Chopped Rice Straw (절단 볏짚의 물리적 성질)

  • This study was performed to determine the kinetic friction coefficient bulk density, dynamic and static angle of repose, and terminal velocity of the chopped rice straw in the moisture range of 8~23%, which could be used for better design and operation of the processing machinery and handling facilities. Friction coefficient was determined from the horizontal traction force measured by pulling the container holding the mass of rice straw on the various plate materials. Bulk density was measured with an apparatus consisting of a filling funnel and a receiving vessel. Dynamic angle of repose was calculated from the photos of bulk samples piled by gravity flow on a circular platform. Static angle of repose was determined by measuring the side angle of the bulk material which was left in the cylindrical container after natural discharge of the bulk sample through a circular hole in the bottom plate. Kinetic friction coefficients of rice straw on the PVC, mild steel, stainless steel, and galvanized steel were in the range of 0.303~0.434, 0.222~0.439, 0.204~0.448, and 0.206~0.407, respectively. and indicated linear increase with moisture content. The effects of moisture change on the friction coefficients were in the order of PVC, mild steel, galvanized steel, and stainless steel. Bulk density, dynamic and static angle of repose, and terminal velocity were in the range of 56.8~60.3 kg/m$^3$, 41.4~45.9$^{\circ}$, 94.4~100.8$^{\circ}$, and 1.07~4.48 m/s, respectively, and were increased linearly with the moisture content.

Survey and Economic Analysis of Food Industry Residues for Biomass-to-energy Conversion in Merced and Stanislaus Counties, California, USA (바이오에너지로의 전환을 위한 캘리포니아 식품가공공장 오.폐수 특성 조사 및 경제성 분석)

  • This research expands investigations into the biomass resource potential associated with California's food processing industry by surveying industries within a two county region in the San Joaquin Valley, California, USA. A previous survey conducted in 2005 for the Sacramento Municipal Utility District (SMUD) quantified residue and waste generation from food processors and food preparation businesses in the Sacramento region. The present survey investigates residue and waste streams from food processors located in Merced and Stanislaus Counties. Sixty food processors were identified to participate in the survey, of which 49 responded (82%) and data were acquired for 38 (63%) (6 facilities closed or moved, 8 decided not to participate). Within the two counties, total annual waste among survey respondents amounted to 24,044 dry tons of high moisture (${\geq}$60%) food residuals, 5,358 dry tons of low moisture (<60%) food residuals; and 23.7 million $m^3$ of wastewater containing 38,814 tons of biochemical oxygen demand ($BOD_5$). The total potential electric power generation from these food residues was estimated at approximately $7\;MW_e$. Total solid waste resource included in the survey response was estimated at about 10% of statewide residue generation for processors falling within the Standard Industrial Classification (SIC) System Major Group 20 (Food and Kindred Products) categories.

Urbanized land-use / landscape patterns in the city's countryside (도시근교농촌의 토지이용 및 경관의 변화)

  • The city's countryside is adjusting to change by urbanization. The recent changes in rural landuse and damages of local landscape characteristics are very serious. But, few studies address the relationship between land-use and quality of landscape for these area. The aim of this study is to clarify causes and problems of the change through investigating changes of landscape as a function of land-use. The results are as follows 1) The change of landscape can be seen as a functional of land-use. Landscapes are characterized by changes of regional environments. These are the urban-rural fringe area which need special consideration for urban development and also for rural landscape quality. 2) 11 types of landscape patterns are found for landscape management. these are Regional commercial area, Apartment development, Recreation & seisure, Golfs, Housing complex, Industrial complex, Vinyl house, Rural village, Evaded facilities, Rivers, Cultural heritages. These landscape types are explained as 'stimuli-responses model'in Bryant's Forces of urbanization. 3) The policy implications of these study are as follows : First is the necessity of landscape management in Grown Management Zone and Natural Reserved Zone in the Metropolitan level, Second is the necessity of development control in semi- agricultural area. The last is the necessary of long range management plan in the urban-rural fringe area.
