A Study on Evaluation of Culture and Leisure Resources of Rural Areas (농촌의 문화여가자원 평가를 위한 척도 개발 연구)

  • The purpose of this study is to develop a culture and leisure resources scale and examine the validity of the scale. The culture and Leisure resources scale was developed and identified its validity by exploratory factor analysis, confirmatory factor analysis, and correlation analysis. The culture and leisure resources scale consisted of four factors, which were 'personal resources', 'support staff resources', 'facility resources', and 'program resources'. Those four factors indicated the reasonable fit indices from confirmatory factor analysis. In addition, this scale showed convergent validity with the satisfaction of cultural and leisure environment and the satisfaction of one's cultural and leisure life. The result of scale score comparison between rural and urban areas showed that culture and leisure resources in rural areas were less abundant than those in urban areas. Furthermore, while the variables which affected the satisfaction of urban residents' were mainly 'personal resources', the variables in case of rural residents' were mainly 'program resources' and 'facility Resources'. The culture and leisure resources scale can be used for policymakers practicably to evaluate regional level of culture and leisure resources, to compare between regions and to find policy priorities for improving the quality of leisure, especially for disadvantaged areas with less resources.

Efficient Production of Ginger (Zingiber officinale Roscoe) Rhizome by Shoot-Tip Culture

  • High productivity of ginger (Zingiber officinale Roscoe) was obtained from the rhizome produced by shoot-tip culture with Korean native variety, Seosanjong. Seed rhizomes induced by shoot-tip culture were successfully established in the field. The rhizomes induced by both plant or rhizome were higher in emergence rate and faster in days to emergence than those of home seed production. The seed rhizome production induced by shoot-tip culture was two times heavier than that of home seed production. These results suggest that shoot-tip culture might be one of mass propagation methods in seed rhizome of ginger plant.

Studies on Simple Hydroponic Culture in Cultivation of Grafted Cactus for Export (수출용 접목선인장 재배시 간이양액재배 연구)

  • This study was carried out to investigate the effect of simple hydroponic culture in cultivation of grafted cactus. The results obtained are as follows: Growth indexes of grafted cactus cultivars, such as scion diameter, fresh weight, and scion hardness were improved in simple hydroponic culture compared with soil culture regardless of cultivars. Stem rot and rate of non-graft take were lower in simple hydroponic culture than in soil culture, but no significant difference was monitored between cultivars. Time for harvesting was fast in simple hydroponic culture compared with soil culture. Marketable yield increased in simple hydroponic culture.

First Report of Penicillium brasilianum and R. daleae Isolated from Soil in Korea

  • In this study, a total of 300 isolates of Penicillium and related teleomorphic genera were collected from soils of 17 locations in Korea from April to May, 2004. Ninety four isolates were identified as the species of Penicillium subgenus Furcatum based on cultural and morphological characteristics and ${\beta}-tubulin$ gene sequences. Among the specie's, Korean isolates of P. brasilianum Bat. and P. daleae K. M. Zalessky were phylogenetically identical to the reference species based on DNA sequence of the ${\beta}-tubulin$ gene. Here we described and illustrated P. brasilianum and P. daleae that are new in Korea.

Bottle Cultivation of Pleurotus ostreatus, Agrocybe aegerita and Ganoderma lucidum using Rice hull media

  • Rice hull was used as a additive in order to find the effect for incresing of mushroom growth and yield in Chungnam Provincial techinical institution. 1 Treatment of 80% rice hull in small Neutaribeosut mycelial grow duration is shorter about 11 days and yield increased about 7% than conventional culture. 2. In case of Chongpung Neutaribeosut bottle culture, mycelial growth duration is shorter about two to three days in additive of 30 to 80% rice hull compared to conventional but yield similar to conventional. 3. Treatment of 30% rice hull in Agrocybe aegerita bottle culture, mycelial growth and yield increased 6days and 6% than convrntional, respectively 4 Treatment additived of 30% to 40% rice hull in Ganoderma lucidum bottle culture, similar to 454ays demand in mycelial grow duration and 38g yield/bottle in conventioal culture methods.

Plant Regeneration through Leaf Explant Culture of Boxthorn (Lycium chinense Mill.)

  • This study was conducted to investigate the optimal plant growth regulator level for the shoot formation of Lycium chinense Mill. In vitro plant propagation was developed for leaf explants of boxthorn. Leaf explants were cultured on MS medium supplemented with cytokinins (BA and 2-iP) alone. Plants were successfully regenerated through in vitro culture by using leaf explants of boxthorn grown in the field. After 4 weeks of culture, 58% of shoot formation had developed from the leaf explants. The shoot formation rate of 'Jangmyeong' was highest followed by 'Myeongan', ;Cheondae', and 'Bullo'. The use of 0.2mg/L BA was critical for enhanced production of shoot formation and resulted in 58% of the culture producing shoot formations. Regenerated plantlets transplanted to pots were developed and successfully acclimatized to greenhouse.
