방아(Agastache rugosa O. Kuntze)로 동전된 estragole과 방아추출물의 향균효과 (Estragole Identified and Extracts from Agastache rugosa O. Kuntze Inhibited Bacterial Growth)

  • Water extract, and methanol extract, its chloroform and hexane fractions, and estragole from Agastache rugosa O. Kuntze were tested to find the inhibition effect on the growth of several microorganisms. The organisms used were Escherichia coli ATCC 1129, Staphylococcus aureus IAM 1011, Vibrio parahaemolyticus WP, Bacillus subtilis ATCC6633, Aspergillus oryzae KFCC 890, Aspergillus niger KCCM 11240. Water and methanol extracts at the concentration of 0.5%, and chloroform and hexane fraction at the concentration of 0.05% inhibited the growth of microorganisms from 1/5 to 2/3 of the control group. Estragole identified from the hexane faction as a major component, its authentic compound completely inhibited the growth of Vibrio parahaemolyticus completely at the concentration of 0.03%, and the other bacteria were at 0.05%.

Agrobacterium rhizogenes strains이 배초향 모상근 유도와 Rosmarinic acid 생산에 미치는 영향 (Influence of Different Strains of Agrobacterium rhizogenes on Hairy Root Induction and Rosmarinic Acid Production in Agastache rugosa Kuntze)

  • rosmarinic acid 생산에 미치는 영향을 알아보기 위하여 5계통의 strain (13333, 15834, R1000, R1200, R1601)을 이용하여 조사한 결과 A. rhizogenes R1601이 가장 높은 72.9%의 모상근 유도율을 보였다. 모상근 유도시 모상근 발생 수와 길이 신장에 미치는 영향을 조사한 결과도 A. rhizogenes R1601으로 감염시킨 잎 조직에서 평균 5.4개의 모상근이 발생하였으며, 평균 2.0 cm의 길이 신장으로 가장 좋은 결과를 나타냈다. 다섯 계통의 A. rhizogenes감염으로부터 유도된 각각의 모상근의 생육은 서러 다른 양상을 보였으나 경향은 모상근유도 결과와 유사하였다. A. rhizogenes R1601 감염으로 유도된 모상근의 생육은 배양 2주후 건물중을 조사한 결과 13.5 g/l로 다른 계통에서 유도된 모상근 보다 생육이 양호하였으며, Rosmarinic acid 생산량도 22.6 mg/g D.W.로 다른 계통에 비하여 가장 높게 나타났다. A. rhizogenes R1601이 배초향 모상근 유도, 생육과 rosmarinic acid 생산에 가장 좋은 적합한 계통이었음을 알 수 있었다.

진균류의 증식과 대사에 미치는 방아(Agastache rugosa)추출물과 Estragole의 효과 (The Effect of Estragole Identified and Extracts from Agastache rugosa O. Kuntze on the Fungal Growth and Metabolism)

  • The extracts from Agastache rugosa O. Kuntze, their chloroform and hexane fractions, and estragole identified from hexane fraction were tested to investigate the effects on the growth and metabolic activities of several true fungi. The fungi used were: Aspergillus oryzae KFCC 890, Aspergillus niger KCCM 11240, Saccharomyces cerevisiae IAM 4597, Saccharomyces ellipsoideus PNU 2215. The growth of S. Cerevisiae by treatment of water extract(1%), hexane fraction (0.05%), and estragole (0.05%) were inhibited 93%, 50%, and 33% respectively, and S. ellipsoideus was also inhibited markedly with delaying the alg phase maximum 12 hrs. The growth of A. oryzae was inhibited by treatment of extracts and fractions. The echanol production by S. cerevisiae was increased more than two times in the highest value around 42 hrs incubation by water extract, but chloroform fraction inhibited its production. The glucoamylase actibities by A. niger were strongly inhibited by hexane and chloroform fractions (0.05%). The invertase activity by S. cerevisiae using estragole (0.05%) reached to 57.5% of control group. S. cerevisiae treated with the estragole was damaged the cell wall and cell membrane, leaked the protoplasm, and observed broken pieces of cell.

Anti-oxidant Effect of Agastache rugosa on Oxidative Damage Induced by $H_2O_2$ in NIH 3T3 Cell

  • The plant Agastache rugosa Kuntze has various physiological and pharmacological activities. Especially, it has been regarded as a valuable source for the treatment of anti-inflammatory and oxidative stress-induced disorders. However, little has been known about the functional role of it on oxidative damage in mammalian cells by ROS. In this study, we investigated the DPPH radical, hydroxyl radical, hydrogen peroxide and intracellular ROS scavenging capacity, and $Fe^{2+}$ chelating activity of the extracts from Agastache rugosa. In addition, we evaluated whether the extract can be capable of reducing $H_2O_2$-induced DNA and cell damage in NIH 3T3 cells. These extracts showed a dose-dependent free radical scavenging capacity and a protective effect on DNA damage and the lipid peroxidation causing the cell damage by $H_2O_2$. Therefore, these results suggest that Agastache rugosa is useful as a herbal medicine for the chemoprevention against oxidative carcinogenesis.

전자코를 이용한 배초향 추출물의 향기패턴 분석 (Aroma Pattern Analysis of Various Extracts of Agastache rugosa O. Kuntze by Electronic Nose)

  • 본 연구에서는 전자코를 이용하여 추출용매, 추출온도 등의 추출조건에 따른 배초향의 부위별 추출물들에 대한 향기성분의 차이를 측정하고 그 데이터를 관능검사와 비교 분석하였다. 같은 배초향 부위라도 추출온도가 다르면 추출물들간의 향기 차이가 있었으며, 배초향 부위에 따라서도 추출물들의 향기 차이가 있는 것으로 나타났다. 동일한 부위에 대한 에탄올추출물과 다른 용매추출과는 향기차이가 있는 것처럼 보이지만 그 차이는 에탄을 자체의 향에 의한 차이로서 실질적으로 같은 배초향 부위에 대해서는 추출용매에 따른 추출물간의 향기 차이는 별로 없는 것으로 판단된다. 전체적으로 배초향 추출물들의 관능적 특성은 박하냄새, 풀냄새, 박하맛, 한약맛, 단맛 등이 대표적으로 강하게 느껴지며, 추출용매, 추출온도, 추출부위에 따라 관능적 특성이 다르게 나타났다. 종합적으로 판단하면 전자코에 의한 향기성분의 차이를 분석한 결과를 관능검사 결과와 연계하여 비교 분식해 보면 전자코에서 차이가 나는 향기성분의 실체가 박하냄새, 단맛 등과 같이 구체적으로 어떤 맛이나 냄새가 더 강하고 약한지를 분성하게 알 수 있었다.

  • A new diterpene, mp $178{\sim}180^{\circ}$, $C_{21}H_{28}O_5$, was isolated from the root of Agastache rugosa O. Kuntze in the family of Labiatae. The structure of this compound was postulated as $19(4{\rightarrow}3)-abeo-12,14,15-trihydroxy-11-methoxy-abiet-4(18)$, 8,11,13-tetraen-7-one by spectroscopic methods.

방아(Agastache rugosa O. Kuntze)의 향기성분 (Volatile Flavor Components of Bangah (Agastache rugosa O. Kuntze) Herb)

  • 방아의 휘발성 향기성분을 수증기 증류장치로 추출한 후 GC 및 GC/MS에 의하여 비교 분석하였다. 확인된 성분을 기능기 별로 보면 hydrocarbon류 14종, aldehyde류 6종, ketone류 5종, alcohol류 3종, phenol류 5종, ester류 2종, 기타 1종으로 총 36종의 향기성분이 확인 되었다. 이중에서 estragole이 90% 이상으로 가장 많이 함유되었고 그밖에 limonene($0.7{\sim}4.7%$), caryophyllene($0.9{\sim}1.1%$), eugenol($1.4{\sim}1.9%$), anethol($0.2{\sim}0.3%$)등이 주요성분이었다. 그러고 이들 향기성분의 종류 및 함량을 채취시기 별로 보면 어린잎보다 성숙된 잎에서 높게 나타났고, 부위별로는 꽃대에서 가장 높았고 잎, 줄기의 순으로 남아졌다.

  • The experiment was carried out to study the influence of planting densities and on some agronomic characters and yield of rresh weight in Agastache rugoso. The results obtained are summarized as fol-lows; stem length was long in dense planing and short in spacious Planting. Stem diameter was thickIn spacious planting, and was thin in dense planting. Yield was higher in dense planting densities (20x20cm : 2spiants/m, 30$\times$20cm : 17plants/m).

배초향 에센셜오일의 화학적 조성과 생리활성 특성 (Chemical Composition and Biological Activity of Essential Oil of Agastache rugosa (Fisch. & C. A. Mey.) O. Kuntze)

  • Background: In Korea, Agastache rugosa (Fisch. & C. A. Mey.) O. Kuntze is one of the well-known perennial plants belonging to Lamiaceae. This mint-fragranced plant has long been used for the treatment of abdominal pain, congestion, chills, and diarrhea since the Goryeo Dynasty. Although this plant has various medicinal properties, it is only used as a spice and for landscape purposes. Methods and Results: The objective of this paper was to review the chemical composition and biological properties of the essential oil of A. rugosa. Several studies reported that the essential oil contains more than 60 different chemical components of monoterpene and sesquiterpene hydrocarbons and oxygenated hydrocarbons. The major component is methyl chavicol (estragole), accounting for 64% - 88% of the oil. The chemical composition of this essential oil vaired widely according to the planting time, environmental conditions, planting distance, fertilizer application, and harvesting time. Conclusions: The essential oil of A. rugosa possesses various pharmacological properties such as antioxidant, antibacterial, anticancer, antiviral, nematicidal, antifungal, insecticidal, wrinkle improver, stress reliever, and Alzheimer's disease alleviator. Hence, the essential oil from A. rugosa could be used for the development of high value-added industrial products in the near future.

Agastache rugosa Kuntze Attenuates UVB-Induced Photoaging in Hairless Mice through the Regulation of MAPK/AP-1 and TGF-β/Smad Pathways

  • Chronic exposure to ultraviolet (UV) radiation, regarded as a major cause of extrinsic aging or photoaging characterized by wrinkle formation and skin dehydration, exerts adverse effects on skin by causing the overproduction of reactive oxygen species. Agastache rugosa Kuntze, known as Korean mint, possesses a wide spectrum of biological properties including anti-oxidation, anti-inflammation, and anti-atherosclerosis. Previous studies have reported that A. rugosa protected human keratinocytes against UVB irradiation by restoring the anti-oxidant defense system. However, the anti-photoaging effect of A. rugosa extract (ARE) in animal models has not yet been evaluated. ARE was orally administered to hairless mice at doses of 100 or 250 mg/kg/day along with UVB exposure for 12 weeks. ARE histologically improved UVB-induced wrinkle formation, epidermal thickening, erythema, and hyperpigmentation. In addition, ARE recovered skin moisture by improving skin hydration and transepidermal water loss (TEWL). Along with this, ARE increased hyaluronic acid levels by upregulating HA synthase genes. ARE markedly increased the density of collagen and the amounts of hydroxypoline via two pathways. First, ARE significantly downregulated the mRNA expression of matrix metalloproteinases responsible for collagen degradation by inactivating the mitogen-activated protein kinase/activator protein 1 pathway. Second, ARE stimulated the transforming growth factor beta/Smad signaling, consequently raising the mRNA levels of collagen-related genes. In addition, ARE not only increased the mRNA expression of anti-oxidant enzymes but also decreased inflammatory cytokines by blocking the protein expression of nuclear factor kappa B. Collectively, our findings suggest that A. rugosa may be a potential preventive and therapeutic agent for photoaging.
