Phytochemical and Biological Studies on Verbascum sinaiticum Growing in Egypt

  • The aerial parts of Verbascum sinaiticum Benth. (Scrophulariaceae) yielded two iridoids that were idenified as ajugol 3 and aucubin 4. Also, investigation of the flavonoid constituents revealed the isolation and identification of luteolin 1 and chrysoeriol-7-glucoside 2. All the isolated compounds were identifid by spectroscopicmethods (UV, MS, $^{1}H-NMR$ and $^{13}C-NMR$) and in comparison with the literature data. Both alcohol and methylene chloride/methanol extracts (1 : 1) exhibited a hepatoprotective effect by $CCl_{4}challenge$ test.

  • In order to verify the efficacy of Lespedeza cuneata G. Don which have been used as a folk traditional herb medicine from ancient times, the test of antioxidant activity and the analysis of constituents were carried out. Methanol extracts from L. cuneata G. Don showed a radical scavenging activity corresponding to that of (+)-catechin as positive control at 250 ${\mu}g$/mL (ca 90% inhibition), and inhibited more than 30% of DPPH radical at low concentration of 10${\mu}g$/mL. In the present study, as a fundamental study for the specification of antioxidant components from L. cuneata G. Don, the analysis for constituents for L. cuneata with HPLC was preformed although there was no results to identify functional flavonoids such as quercetin.

  • A putative protein encoded by Arabidopsis AtMTN3 gene, a homologue of Medicago truncatula MTN3, consists of 285 amino acid residues, and has a predicted molecular mass of 31.5 kDa and a calculated pI of 9.1. Primary amino acid sequence analyses have revealed that the protein contains seven putative transmembrane regions with N-terminus oriented to the outside of the membrane. The AtMTN3 protein shows overall 16.4% of amino acid identity with the rat GALR3 protein, known to be a G-protein-coupled receptor. The gene is present as a single copy in the Arabidopsis genome, and expressed in aerial parts but not in roots of Arabidopsis. Therefore, AtMTN3 appears not to be specifically involved in Rhizobium-induced nodule development, as was predicted for the MTN3 gene. These proteins possibly mediate signal transmission through G-protein-coupled pathways during general interactions between plants and symbiotic or pathogenic microbes.

  • The chemical composition of essential oils obtained from aerial parts in spearmint, apple mint and chocolate mint, was investigated by gas chromatography/mass spectrometry analyses. (-)-Carvone (33.0%) was quantitatively major compound in spearmint, followed by R-(+)-limonene (11.7%) and ${\beta}$-phellandrene (9.7%); (-)-carvone (37.4%) and germacrene D (11.9%) in apple mint; and (-)-menthol (34.3%), p-menthone (18.4%) and menthofuran (9.8%) in chocolate mint. Hierarchical cluster analysis and principle components analysis showed the clear difference in chemical composition of the three mint oils.

  • From the aerial parts of Saururus chinensis growing in Korea, four flavonoids (hyperin, isoquercitrin, quercitrin and rutin) were isolated and identified by high performance liquid chromatography, gas chromatography and thin layer chromatography. However, avicularin, which was known as one of the main constituents of flavonoid, was not detected.

  • Four antioxidative flavonoids were isolated from the aerial parts of Hypericum erectum. Their structures were identified as quercitrin (I), isoquercitrin (Ⅱ), hyperoside (Ⅲ) and orientin (IV) on the basis of spectroscopic means. Antioxidative activities for flavonoids $I{\sim}IV$ were determined by measuring lipid peroxide using 2-thiobarbituric acid (TBA) method and by evaluation the radical scavenging activity on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. Compound IV, orientin, was found to have strong antioxidative potency.

  • White needle crystalline compound, mp.\;78{\sim}79$, was isolated from the petroleum ether fraction of the aerial parts of Chenopodium acuminatum. This compound was supposed to be cycloalkane by chemical properties, spectral data and elemental analysis. After extraction with petroleum ether, the methanolic extract of this plant found to contain aspartic acid, threonine, serine, glutamic acid, glycine, cystine, valine, isoleucine, tyrosine, and phenylalanine. Inorganic elements contained in this plant are as following: Mg 4.84%, Ca 9.602%, Zn 1.11%, K 29.104%, Mn 5.124%, Fe 27.039%, Co 0.31%, Pb 0.995%, Na 0.039% and Cu, 0.0076%.

  • The crude drug 'CheonKun' is derived from the underground portions of Rubia species. However, the aerial parts of the plants are also sold under the 'GgogDuSeoNiIp' in Korea. To clarify the botanical origin of 'GgogDuSeoNiIp' the morphological and anatomical characteristics of the leaves of Rubia species growing in Korea, i.e. Rubia akane, R. chinensis var. glabrescens, R. cordifolia var. pratensis were compared. As a result, it was determined that 'GgogDuSeoNiIp' was the leaf of Rubia akane and R. cordifolia var. pratensis.

  • Cissus quadrangularis L. variant I (Family: Vitaceae), the common variant with square stem is widely used for peptic ulcer disorders (PUD) in traditional medicine. Aerial parts were collected during flowering and vegetative seasons and analysed. Aqueous (hot and cold) and solvent extracts (acetone, chloroform and ethanol) were screened for their anti-Helicobacter pylori (Hp) activities. Among them chloroform extract was observed to recover bioactive principles markedly with low minimal inhibitory concentration (MIC) and minimal lethal concentration (MLC). MIC was $30\;{\mu}g$ in both samples and MLC was $35\;{\mu}g$ for vegetative and $30\;{\mu}g$ for flowering seasons, respectively. Extracts from samples collected during flowering season were better than thse of vegetative season.

  • The effects of various fractions from the aerial parts of Echinosophora koreensis Nakai on the antimutagenic and the immuno-regulating activities were evaluated by in vivo bone-marrow micronucleus test and HA titer reaction. No significant suppressive effects of these extracts and echinoisosophoranone were shown on cyclophosphamide-induced micronuclei, but HA titers were significantly enhanced in ether and BuOH extract treated-group. Tetracosanol and docosanol were isolated from the ether extract of this plant.
