In this research, a 2-stage intermittently aerated activated sludge system(IA) and intermittently aerated dynamic flow activated sludge system(DF) were investigated for the removal of nutrients in domestic wastewater. Wastewater was characterized by low C/N( organics/nitrogen) ratio. $COD_{cr}$, $BOD_s$, TKN and TP concentrations of domestic wastewater were 235, 47, 32 and 5.4 mg/L, respectively. Three sets of IA and one set of DF were operated. Three of four systems were added with fermented settled sludge taken from primary settling tank as an external electron donor and the other(IA) for control reactor was operated without addition of electron donor. All systems were operated at same sludge retention time of 20 days and hydraulic retention time of 12hrs. The supplemental electron donor was supplied into the anoxic mode. A higher denitrification rate was observed from the reactors with fermented settled sludge as an electron donor for denitrification compared to that of without addition of organic source. The result of this study indicates that the settled primary sludge, if the fermented at the acid stage, was an excellent electron donor for denitrification. 81 % of TN and 80% of TP were removed from the systems with the supplemental organic source added. However, the control reactor without addition of electron donor showed only 39% of TN and 43% of TP.

The effects of inorganic salts, inoculum concentration, aeration rate and shaking speed on insoluble phosphate solubilization by Pseudomonas fluorescens RAF15 were investigated. Soluble phosphate production was dependent on the presence of $MgCl_2{\cdot}6H_2O$ and $MgSO_4{\cdot}7H_2O$ in the medium. Supplementation of medium with 0.01% $CaCl_2{\cdot}2H_2O$ and 0.01% NaCl slightly increased soluble phosphate production. The optimal medium compositions for the solubilization of insoluble phosphate by P. fluorescens RAF15 were 1.5% glucose, 0.005% urea, 0.3% $MgCl_2{\cdot}6H_2O$, 0.01% $MgSO_4{\cdot}7H_2O$, 0.01% $CaCl_2{\cdot}2H_2O$ and 0.01% NaCl, respectively. Optimal inoculum concentration was 2.0%(v/v). Maximum soluble phosphate production was obtained with 20-50 ml/250-ml flask and 200 rpm of shaking speed, respectively. The addition of EDTA decreased cell growth and soluble phosphate production.
The effects of aliphatic hydrocarbons (n-hexadecane and n-dodecane) on the volumetric oxygen mass transfer coefficient $(k_L\;a)$ were studied in flat alveolar airlift reactor and continuous stirred tank reactors (CSTRs). In the flat alveolar airlift reactor, high aeration rates (>2vvm) were required in order to obtain efficient organic-aqueous phase dispersion and reliable $k_La$ measurements. Addition of 1% (v/v) n-hexadecane or n-dodecane increased the $k_La$ 1.55- and 1.33-fold, respectively, compared to the control (superficial velocity: $25.8{\times}10^{-3}m/s$, sparger orifice diameter: 0.5 mm). Analysis of the gas-liquid interfacial area a and the liquid film mass transfer coefficient $k_L$ suggests that the observed $k_La$ increase was a function of the media's liquid film mass transfer. Addition of 1% (v/v) n-hexadecane or n-dodecane to analogous setups using CSTRs led to a $k_La$ increase by a factor of 1.68 and 1.36, respectively (superficial velocity: $2.1{\times}10^{-3}m/s$, stirring rate: 250 rpm). These results propose that low-concentration addition of oxygen-vectors to aerobic microbial cultures has additional benefit relative to incubation in purely aqueous media.
The effect of dissolved oxygen(DO) on microbial transglutaminase(mTG) production by Streptoverticillium morbaraense was studied in on-line computer controlled fermentation system. In order to control dissolved oxygen during fermentation, the agitation speed and aeration rate of 2.5 L fermenter ranged from 260 to 360 rpm and 0.3 to 3.9 L/min, respectively. The maximum microbial transglutaminase production was obtained at controlled 20% of dissolved oxygen among the various dissolved oxygen controlled batch cultures tested. The production of microbial transglutaminase at controlled 20% of dissolved oxygen was about 2.12 U/mL which was 1.1 times higher than that obtained in batch culture without control of dissolved oxygen. Also, the highest microbial transglutaminase production was obtained in fed-batch cultures in which dissolved oxygen was controlled at 20%, and it was improved almost 1.3 times in comparison with that without control of dissolved oxygen. Maximal dry cell weight and microbial transglutaminase production were 13.2 g/L and 2.6 U/mL, respectively. Finally, it was also found that fed-batch fermentation at controlled 20% of dissolved oxygen showed a good performance for the microbial transglutaminase production by on-line computer controlled fermentation system which may be generally applicable to other microbial cultures.

High quality seed of proso millet that has high germination percentage, germination speed, and uniformity demanded to increases rates of mechanization in cereal crop cultivation. In order to improve germination characteristics, proso millet seeds were treated with red light, ultrasonication and liquid smoke (LS) solution that generated from hickory wood. All treatments were performed in seed priming solution with 100mM $NH_4NO_3$ at $15^{\circ}C$ for 24hrs under aeration condition. Seeds were exposed under light intensity of 2000 lux for 15m, 30m, 60m, and 120m in priming solution. Ultrasonic treatment was performed at 60%, 80%, and 100% intensity of 21.6 KHz for 5m, 10m, and 20m in priming solution. For LS treatment seed were soaked in 0%, 0.5%, 1.0%, 5.0% and 10.0% of diluted solution with $dH_2O$ or 100mM $NH_4NO_3$ solution. The effect of seed treatment was evaluated with germination percentage (GP), mean germination time (MGT), germination index (GI), germination rate (GR), Germination uniformity (GU) and heath seed percentage (HS). Our results demonstrate that red light (15min) or ultrasonication (21.6kHz, 5min) treatment improved MGT, GI, GR, and GU comparing to untreated control. Importantly, we show that LS treatments have significant effect on the health seedling and germination characteristics. Proso millet seeds that treated with 5% LS solution for 24hrs improves HS up to 97% that similar results obtained in 100mM $NH_4NO_3$ priming for 24hrs. The combined treatment with LS solution and 100mM $NH_4NO_3$ priming were not effective in all treatments. Our results demonstrate that treating seeds with LS or 100mM $NH_4NO_3$ priming or ultrasonication improves germination characteristics. The methods described here will help advance research using this species by increasing the germination performance at which successive seed pellet process.

This study was carried out to analyze effects of soil, water level and shading on growth of sweet flag(Acorus calamus var. angustatus). Three types of soil were used, which included sandy, silty loam and paddy loam soil. Three levels of shading were applied in the experiment: no shading, 55% shading and 75% shading. The water levels were also adjusted to three levels in the experiment. The results are summarized as follows; 1. The cultivation of sweet flag in sandy soil with low water level resulted in decreased fresh weight compared to that at planting. This result indicates that the water level should be maintained higher than the soil surface for sweet flag growth in sandy soil. 2. 5 out of 72 sweet flags died in paddy loam soil. Water saturation of soil easily reduced paddy loam soil, and root growth of sweet flags in reduced soil condition were restricted, resulting in the dead plants. 3. The growth of sweet flag in paddy loam soil was worse than those in silty loam, indicating that reduced soil conditions in paddy loam is harmful to root growth. In planting sweet flags in paddy loam, improved soil aeration in paddy loam soil is necessary for good growth of sweet flag. 4. The maintaining of high water levels is better than that of low water levels in sweet flag cultivation. During winter, soil near the water surface froze and sweet flags in frozen soil were stressed physiologically. Maintaining high water levels prevents soil from being frozen which is good for the growth of sweet flags. 5. There was not significant difference in the growth of the sweet flag between non-shading and 55% shading. It thus appears that sweet flags can grow soundly under shading rate lower than 55%.
A culture system for lactating rat mammary acini was evaluated, where the primary indicator of performance was lactose secretion, measured by a sensitive bioluminescence assay. Lactose secretion was reduced by half (p<0.01) over the first 6 h of culture by overnight feed withdrawal (FW) from tissue donors but was sensitive to increased glucose concentration in the culture media (p<0.001) up to 30 mM. Lactose production of cells from fed donors over the first 6 h in culture in 30 mM glucose was 8.9 fmol/cell/h - a rate calculated to be about half that in vivo. No significant difference was shown in lactose secretion by cells from fed or FW rats over 6-24 h. Lactose secretion was 3.6 fmol/cell/h by cells from fed animals in 40 mM glucose concentration media over the 6-24 h culture period. Addition of insulin to the culture media had no effect on rates of lactose secretion while addition of prolactin and hydrocortisone, with or without insulin, significantly (p<0.001) decreased lactose production over both 0-6 h and 6-24 h culture periods. Lactose synthesis in vitro was significantly enhanced by aeration of the media during collagenase digestion of mammary tissue (p<0.05). No improvement in lactose secretion was effected by shaking of cells during culture, Matrigel coating of culture dishes or change in cell density over a range up to 2.5 million cells per ml.
A membrane-coupled sequencing batch reactor (MSBR) was used for the advanced treatment of rural village sewage which is very low C/N ratio. The effect of powdered activated carbon, aeration rate, and external organic material loadings on the treatment efficiency and filtration performance were investigated in sequencing batch reactor, in which a flat-sheet type microfiltration membrane with a pore size of $0.4{\mu}m$ was submerged. At the initial operation (within 54 days) MLSS concentration, and the removal efficiencies of COD, T-N, and T-P were increased with the increase of C/N ratio. After 89 days the removal efficiencies of COD, T-N, and T-P were 97.1%, 75.0%, and 48.3%, respectively. Suspended solid-free effluent was obtained by membrane filtration. The T-P removal was relatively low because of depending on the amount of excess sludge wasting. During the operation of MSBR with powdered activated carbon, the particle size of the sludge reduced by the increase of collision frequency and mixing intensity. In comparison with MSBR without powdered activated carbon, TMP of MSBR with that was significantly elevated.

Arachidonic acid is a polyunsaturated fatty acid(PUFA) containing twenty carbon atoms with four double bonds. The family of w-6 PUFA, including arachidonic acid as well as r-linoleic acid, was served as intermediates in the formation of several key prostaglandin and leukotrienes. Several fungal strains of the genus Mortierella accumulate high amounts of arachidonic acid. In this study experiments were carried out to optimize the culture conditions for the mass production of fungus Mortierella alpina DSA -12 and lipid production with high proportion of polyunsaturated fatty acids, especially arachidonic acid. The batch culture was carried out in 500 L fermenter containing 50 g/L glucose, 18 g/L corn-steep powder and 100 mg/L MnS04 under $25^{\circ}C$, aeration rate of 0.5 vvm and agitation speed of 200 rpm without pH control. As a result, we could be obtained 22 g/L of cell mass with high contents of lipid 12.1 g/L) and arachidonic acid (5.1 g/L) The intermittent fed-batch culture was performed in the medium containing 20 g/L glucose and 10 g/L corn-steep powder. The final glucose concentration was 170 g/L and pH was maintained at 5.5 ${\sim}$ 6.0 by adding 14% ammonia solution. It was shown relatively high cell concentration (70.5 g/L) with high contents of lipid (45.8 g/L) and arachidonic acid 08.3 g/L). Therefore, when compared to batch cultures, the high concentration of arachidonic acid could be obtained by fed-batch culture using M. alpina DSA -12. These results imply that the fed-batch culture of M. alpina DSA -12 was feasible in industrial purpose and could be employed in the commercial production of arachidonic acid.
The green colonial algae Botryococcus braunii is characterized by unusual high hydrocarbon contents, ranging from 15 to 75% of dry weight, as long-chain unsaturated hydrocarbons. In two-phase bubble column using various organic solvents, poor recovery 08 - 32%) of hydrocarbon seems to be caused by insufficient mixing between two phases, which was operated using only aeration on the narrow interface between hydrophobic solvent and cell suspension. In addition, hydrocarbon was entrapped tightly in cell-matrix (formed by exopolysaccharide) of algal colony, which make difficult to extract using two-phase system. To improve recovery efficiency, mixed-solvent of extractive solvent (octane) and biocompatible solvent (octane) was tested in two-phase column for in situ extraction. In two-phase extraction culture using mixed-solvent, the algal growth was intensely inhibited even at low concentration of polar octanol solvent. the hydrocarbon recovery in two-stage cell-recycle extraction showed a 2.9 fold increase (57%) over that in two-phase extraction. Up to 60 % of hydrocarbon could be recovered without serious cell-damage in the case of downstream separation for 6 h at the high recycle flow rate using this process after batch culture.
