A Study on Bacterial Leaching of Low-Grade Copper Mineral(V) (저품위 동광석의 세균침출에 관한 연구 5)

  • It was convinced that with a relatively small capital investment and with a low operating cost, appreciable amounts of cement copper could be produced from low-grade minerals by the application of a bacterial leaching method. For the recovery of cement copper from the impure pregnant solution, direct precipitation of copper with tin plates by a bacterial leaching method was feasible. The results obtained were as follows: 1)In order to remove the cement copper from the reducing metal, aeration and agitation method were more effective and economic than shaking method. 2)The rate of copper recovery from the pregnant solution was accerelated according to increasing quantities of reducing metal. However, the excess of reducing metal reduced the grade of cement copper. 3)Among the comparative experiments of copper recovery at each reaction temperature of $10^{\circ}C$, $20^{\circ}C$, and $30^{\circ}C$, the recovery rate of copper at $30^{\circ}C$from the pregnant solution was highest. 4)Direct cementation method on iron-containing metal was an excellent method for the recovery of cement copper in bacterial leaching.

Effects of Washing of Food Wastes on Aerobic Composting (음식물쓰레기의 세척이 호기성 퇴비화에 미치는 영향)

  • This study was performed to estimate the effects of washing of food wastes on temperature, pH, weight and volume reduction and salinity in aerobic composting of food wastes. Weight ratios of food wastes to water in washing were 1:0(Control), 1:1 (W-1), 1:2(W-2), 1:3(W-3) and :4(W-4), respectively. Ratios of food wastes to wood chips in reactor of Control, W-1, W-2, W-3 and W-4 were 5 kg:5 1, respectively. Reactors were operated for 24 days with 1 hour stirring by 1 rpm and 2 hours aeration per day. The increase in the ratio of food wastes to water used in washing resulted in the decrease of the highest reaction temperature and the elongation of the high temperature reaction period. The lowering of the ratio of food wastes to water used in washing resulted in faster pH increase and the steady state in the weight reduction rate and the volume reduction rate of composts. The final salinities of Control, W-1, W-2, W-3 and W-4 were 0.95%, 0.73%, 0.65%, 0.57% and 0.41%, respectively.

Anaerobic Digestion of Thickened Septage at Municipal Wastewater Treatment Plant (하수(下水) 처리장(處理場)에서 정수조폐액(淨水槽廢液)의 혐기성소화(嫌氣性消化) 처리(處理)에 관한 연구(硏究))

  • Anaerobic Digestion of thickened septage was investigated in this study. Thickening could reduce the volume of septage to be treated to about 40% with 12hr HRT. The VS and BOD removal efficiencies were respectively 28 to 45%, and 75% when digested the thickened septage with 30 day HRT Or $1.4kgVS/m^3/d$. The BOD removal efficiency could be increased to about 90% with subsequent settling tank with about 6 hours HRT. The gas production rate was 0.22 to $0.35m^3gas/kgVSadd$($0.75m^3gas/kgVSrm$), or $1.32m^3gas/kgBOD_{rm}$. In addition, the supernatant of thickener could be returned to the aeration tank treating domestic sewage. In this case, a BOD loading rate of 0.5 to $0.7kgBOD/m^3/d$ or 0.5kgBOD/kgMLVSS/d was proposed for 80% BOD reduction.

Optimization for the DXAMase production from Lipomyces starkeyi using statistically designed experiments

  • The optimal condition for the production of DXAMase, containing the both characteristics of dextranase and amylase, was studied based on different levels of pH, temperature, and aeration rate. Response surface methodology was applied to find the optimatic condition showing the relationship between the fermentation response(dextranase and amylase activity of DXAMase) and the fermentation variables(pH, temperature, and agitation rate). In case of dextranase activity, the condition of pH 4.06, $28.08^{\circ}C$, and 235.14 rpm showed the highest activity, 2.26 U/ml, and for amylase activity, the condition of pH 4.01, $27.96^{\circ}C$, and 212.01 rpm showed the highest activity, 3.52 U/ml. For the production of DXAMase, dextranase and amylase, the optimum condition was pH 4.06, $28.08^{\circ}C$, and 234.80 rpm.

Effects of Stireed Tank Bioreactor Scale-up on Cell Growth and Alkaloids Production in Cell Cultures of Eschscholtzia californica (탱크 교반형 생물반응기의 scale-up이 Eschscholtzia californica 세포생장 및 알칼로이드 생성에 미치는 영향)

  • Studies were made to investigate effects of the scale-up of stirred tank bioreactors on cell growth and alkaloids production for suspension cultures of Eschscholtzia californica. In the 1.5 L STR, cell lysis was observed at 110 rpm or higher agitation speed. The agitation speed of 30 L STR was 43.7 rpm to maintain the same shear stress developed in 1.5 L STR of 100 rpm. As a result of scale-up from 1.5 L to 30 L STR, the specific growth rate was decreased from 0.12 to 0.07 day-1. The alkaloids productivity was also decreased from 0.24 to 0.14 mg/L-day. Changes of mixing performance and oxygen transfer were studied to explain the decrease of cell growth and alkaloids production. Decreased oxygen transfer rate coefficient(KLa) and increased mixing time by the scale-up was observed at various aeration rates.

Effect of seed priming on germination and sprouting vigor of colored rice

  • The study was conducted to select optimal materials for promoting germination rate, high sprouting vigor by priming treatment using PEG 6000 (water potential -0.5 to 2.0 Mpa) and Azolla extracts (0.1, 1.0, and 10.0%) extracted by ethanol, distilled, and/or hot water in colored rice cultivars. Each rice seed (three black rice cultivars and two red rice cultivars) was soaked 24 and 48 hr including untreated control. In black rice, Joseongheugchal rice cultivar, azolla ethanol extract (0.1%) induced highest germination rate, germination speed was taken to 5 days in distilled water and to 3days in Azolla extracts extracted hot water. Otherwise, degree of bacterial inhibition (number of colony, $10^3cfu$) in dry seed, water soaking for 24hr, soaking with fungicide for 24hr and 48hr, soaking with fungicide and aeration for 24hr and 48 hr was 22, 500, 95, and 0.46, respectively. In order to minimize fungal inhibition, a method can be chosen to combination of soaking fungicide and aeration for 48 hr. In seed priming treatments using growth pouch, seed soaking with fungicide did not affect change of germination percentage and germination speed, it delayed only 2 or 4day in the Joseongheugchal rice cultivar. It differs from rice cultivars and priming materials, Azolla extract(0.1 to 1%) promotes seed germination percentage in the Ilpum, Hongjinju, and Joseonghuegchal, in addition, germination in Jeogjinju cultivar was only promoted by PEG solution(10 to 20%), otherwise, it showed much lower or inhibited on the germination in Heugjinju and Sinmyungheugchal rice cultivars. In a paddy field trial, seedling establishment rate by applying PEG6000 and azolla extract did not show significantly statistical difference. When it compared with untreated control, seedling establishment rate was increased over 50% in priming treatments. Interestingly, seedling establishment rate under azolla extract (0.1%) extracted with ethanol was promoted over 2.5 times compared to the control in a black rice, Joseongheugchal and red rice, Jeogjinju.

Comparative Bioreactor Studies in Terms of Oxygen Transfer between Suspended and Immobilized Fungal Systems for Cyclosporin A Fermentation (Cyclosporin A 생산을 위한 액체배양과 고정화배양의 생물반응기에서의 산소전달 비교 연구)

  • In fermentations with a 4-liter stirred tank bioreactor, a better than two-fold enhancement of the gas-liquid mass transfer coefficient$(k_La)$ in the celite-immobilized fungal cultures of Tolypocladium in flatum over the parallel conventional free-cell was observed at identical biomass concentrations, despite the higher specific oxygen uptake rate of the immobilized fungi during exponential growth. As a result oxygen sufficient conditions, i. e., dissolve oxygen(D.O.) concentrations exceeding 75% air saturation, could be maintained throughout exponential growth period of the immobilized culture, in contrast to the suspended fungal culture, whose D.O. levels fell below 50% air saturation. A linear monotonic dependence of $k_La$ upon impeller agitaion rate was found for both immobilized and conventional cultivation modes over a range of 250 to 550rpm, the slope being a function of biomass concentration for the free but not for the immobilized cell system In contrasts oxygen transfer rate was a much weaker function of aeration rate up to about 2.5 vvm for both culture configurations. Above this level, aeration rate had no further effect on the mass transfer. In addition, the immobilized cultures sustained good morphological and physiological states, leading to almost two times higher cyclosporln A (CyA) productivity overt the parallel free cell system. These experiments suggest that the celite-immobilized fungal system in a stirred tank reactor has considerable promise for scaling up cyclosporin A production in terms of high-density cultivation.

  • To reduce energy consumption in wastewater treatment plants (WWTPs), renewable energy applications such as small hydropower, solar energy, and wind energy are popular. However, it should be noticed that energy originated from operation of wastewater treatment process can be reduced through optimized operation based on analysis of factors affecting energy. In this research, the relationship to the various operational variables and influent factors was explored using correlation analysis and decision tree algorithm. Due to the non-linear characteristics of the process, it was difficult to find clear linear patterns through correlation analysis. However, decision tree algorithm showed its usefulness in uncovering hidden patterns that consume energy. As operational factors, influent flowrate, the amount of aeration, nitrate recycling pumping rate, and sludge wasting pumping rate were selected as important factors. For environmental factors associated with influent compositions and removal rate, BOD and T-N removal rate were selected as significant factors.

A Continuous Process for Phosphorus Recovery from Swine Slurry with Forming Struvite (양돈액비에서 Struvite 형성으로 연속적 인의 회수기술)

  • This study was carried out to develop a continuous process for recovering phosphorus in swine slurry. Magnesium chloride ($MgCl_2$) was used in the test as a magnesium source and the pH was regulated by adding NaOH and aerating. The results showed that the recovery rate of soluble phosphorus (SP) has increased with the molar ratios increased. In case of pH regulated with NaOH, the recovery rates of SP with molar ratio of 1:1.5 were over 95% from both farms. The removal of ammonia-nitrogen was at levels of $4{\sim}9%$. With aeration treatment, the SP recovery rate was 66% and the removal rate of ammonia-nitrogen was 15%. The treatment of NaOH to increase pH showed better SP recovery efficiency than the aeation treatment. However, in case of ammonia-nitrogen removal, the treatment of aeration showed better results than the NaOH treatment.

Effects of Dissolved Oxygen Level on Avermectin $B_{1a}$ Production by Streptomyces avermitilis in Computer-Controlled Bioreactor Cultures

  • In order to investigate the effect of dissolved oxygen (DO) level on AVM $B_{1a}$ production by a high yielding mutant of Streptomyces avermitilis, five sets of bioreactor cultures were performed under variously controlled DO levels. Using an online computer control system, the agitation speed and aeration rate were automatically controlled in an adaptive manner, responding timely to the oxygen requirement of the producer microorganism. In the two cultures of DO limitation, the onset of AVM $B_{1a}$ biosynthesis was observed to casually coincide with the fermentation time when oxygen-limited conditions were overcome by the producing microorganism. In contrast, this phenomenon did not occur in the parallel fermentations with DO levels controlled at around 30% and 40% throughout the entire fermentation period, showing an almost growth-associated mode of AVM $B_{1a}$ production: AVM $B_{1a}$ biosynthesis under the environments of high DO levels started much earlier than the corresponding oxygen-limited cultures, leading to a significant enhancement of AVM $B_{1a}$ production during the exponential stage. Consequently, approximately 6-fold and 9-fold increases in the final AVM $B_{1a}$ production were obtained in 30% and 40% DO-controlled fermentations, respectively, especially when compared with the culture of severe DO limitation (the culture with 0% DO level during the exponential phase). The production yield ($Y_{p/x}$), volumetric production rate (Qp), and specific production rate (${\bar{q}}_p$) of the 40% DO-controlled culture were observed to be 14%, 15%, and 15% higher, respectively, than those of the parallel cultures that were performed under an excessive agitation speed (350 rpm) and aeration rate (1 vvm) to maintain sufficiently high DO levels throughout the entire fermentation period. These results suggest that high shear damage of the high-yielding strain due to an excessive agitation speed is the primary reason for the reduction of the AVM $B_{1a}$ biosynthetic capability of the producer. As for the cell growth, exponential growth patterns during the initial 3 days were observed in the fermentations of sufficient DO levels, whereas almost linear patterns of cell growth were observed in the other two cultures of DO limitation during the identical period, resulting in apparently lower amounts of DCW. These results led us to conclude that maintenance of optimum DO levels, but not too high to cause potential shear damage on the producer, was crucial not only for the cell growth, but also for the enhanced production of AVM $B_{1a}$ by the filamentous mycelial cells of Streptomyces avermitilis.
