Phospholipid Production by Pseudomonas sp.CH-414 under Various Culture Conditions (배양 조건에 따른 Pseudomonas sp.CH-414의 Phospholipid 생산능의 변화)

  • Using Pseudomonas sp. CH-414, the optimum culture conditions were investigated for the cell growth and the phospholipid production in batch culture by varying pH and aeration rate. With starting the cultivation under the conditions of pH 7.0 and 1vvm, pH was controlled to 6 or 8 at 30 hours of culture time. In the case of changing into pH 6.0, the phospholipid production was increased by ca. 20% with comparison to the case of pH 7.0. However, the biomass and the phospholipld concentration were rapidly decreased after 30 hours of culture time when pH was controlled to 8.0. As the aeration rate was increased, the biomass was increased while the phospholipid concentration was considerably varied and unstable. Especially, the concentration of phospholipid was rapidly decreased with 3vvm of aeration rate. Finally, under the culture conditions of pH 7.0 and 3vvm until 30 hours for the cell growth, which were controlled to pH 6.0 and 1vvm for the stable production of phospholipid beyond that time, the dry cell weight was $18.5g/\ell$ and the phospholipid concentration was $\0.83g/ell$ (45mg/g cell).

  • This study was done to improve the effectiveness of nitrification and denitrification using the aeration-anoxic combination method using CFSTR(continuous-flow stirred-tank reactor) attached with an anoxic reactor filled with a media. In order to calculate the concentration of nitric acid within the aeration tank proportional to the anoxic rate within the reactor, supernatant within the inflow and precipitation tanks were influxed into the anoxic reactor. The rate of nitrogen removal was calculated using the concentration of inflow and flow of returned supernatant. From the results of this experiment, the carbon source needed in the anoxic reactor came from the inflow so that anoxification was achieved completely using the inflow source without the introduction of an external carbon source. However, as the ratio of nitric acid becomes large in inflow and nitric acid flow, the carbon source within the input source decreases so that the concentration of carbon source is important.

Simulations of a System Dynamics Model for Operations and Maintenance of Activated-Sludge Wastewater Treatment Plants (활성슬러지 하수처리시설 운영 및 유지관리를 위한 시스템다이내믹스 모델의 모의에 관한 연구)

  • In this paper, simulation methods of the system dynamics model developed by Das et al. (1997) for activated-sludge wastewater treatment plants are illustrated in an attempt to determine the operating rules and the policies related to capacity expansion of an activated-sludge wastewater treatment plant. For existing conditions, the analyses were performed by varying activated-sludge return rate to observe changes in effluent water quality and treatment efficiency. The effluent water quality is also analyzed for various average daily inflow conditions and activated-sludge return rates. As a result, without expanding the aeration tank, maximum average daily inflow that can satisfy the effluent water quality standard of BOD $0.02kg/m^3$ was determined as $2,840m^3/hr$, subject to 100% of activated-sludge return rate while other factors remain constant. When the activated-sludge return rate is less than 100%, expansion of the aeration tank is necessary and minimum sizes of the aeration tank to satisfy the effluent water quality standard were determined for various activated-sludge return rates. In addition, the total operating and maintenance as well as unit treatment cost regression equations for activated-sludge wastewater treatment plants are suggested by using the cost data that are obtained from Water and Wastewater Division, Ministry of Environment. The regression analyses showed that the economies of scale phenomena exist in the operating and maintenance costs of activated-sludge wastewater treatment plants.

The design of an ejector type microbubble generator for aeration tanks

  • The ejector type microbubble generator, which is the method to supply air to water by using cavitation in the nozzle, does not require any air supplier so it is an effective and economical. Also, the distribution of the size of bubbles is diverse. Especially, the size of bubbles is smaller than the bubbles from a conventional air diffuser and bigger than the bubbles from a pressurized dissolution type microbubble generator so it could be applied to the aeration tank for wastewater treatment. However, the performance of the ejector type microbubble generator was affected by hydraulic pressure and MLSS(Mixed Liquor Suspended Solid) concentration so many factors should be considered to apply the generator to aeration tank. Therefore, this study was performed to verify effects of hydraulic pressure and MLSS concentration on oxygen transfer of the ejector type microbubble generator. In the tests, the quantity of sucked air in the nozzle, dissolved oxygen(DO) concentration, oxygen uptake rate(OUR), oxygen transfer coefficient were measured and calculated by using experimental results. In case of the MLSS, the experiments were performed in the condition of MLSS concentration of 0, 2,000, 4,000, 8,000 mg/L. The hydraulic pressure was considered up to $2.0mH_2O$. In the results of experiments, oxygen transfer coefficient was decreased with the increase of MLSS concentration and hydraulic pressure due to the increased viscosity and density of wastewater and decreased air flow rate. Also, by using statistical analysis, when the ejector type microbubble generator was used to supply air to wasterwater, the model equation of DO concentration was suggested to predict DO concentration in wastewater.

Growth Characteristics of in Vitro Mass Propagated Plantlets of Ever-bearing Strawberry 'Goha' according to Aeration Rate in Bioreactor (사계성 딸기 '고하' 조직배양묘의 대량증식 시 생물반응기 내 공기주입량에 따른 생육 특성)

  • This study was conducted to determine the optimal aeration rate for mass propagation of ever-bearing strawberry by bioreactor culture. The aeration rate was treated in four levels: 0.1 vvm (air volume/medium volume/min), 0.2 vvm, 0.3 vvm, and 0.4 vvm. In 0.2 vvm conditions, shoot length was the longest at 9.03 cm in bioreactor culture, leave numbers were 40.4 ea and fresh weight was 6,106 mg. Plant growth rate at 0.2 vvm condition was faster than other treatments. In the aeration condition, 0.2 vvm was most effective to increase aerial part growth and to decrease medium consumption. As the culture periods increased, the fresh weight also increased rapidly. After six weeks of cultivation, shoots were emerged with 10.4 ea per plantlet, resulting in developing a complete plant. As a result, the bioreactor culture system for mass propagation of strawberry is required to continuously supply the air by 0.2 vvm speed and cultivate at least for six weeks.

The Optimum Design of Suspended Growth Systems (부유성 미생물을 이용한 생물처리법의 최적 설계)

  • The purpose of this study is to examine the basis of design condition of existing treatment processes, namely, Extended Aeration Process, Conventional Activated Process, High Rate Aeration Process and Modified Aeration Process, by clarifying the correlations of influent wastewater concentration, hydraulic retention time, food-to-microorganism ratio and sedimentation of sludge, as well as to ascertain the feasibility of design, regardless of the existing design condition. In particular, this study made a priority investigation of hydraulic retention time and sludge sedimentation, because sludge sedimentation is the main factor$^{1)}$ which determines the operating conditions of existing treatment processes. Therefore, it is generally known that in case exceeds the sphere of design presented for each treatment, sludge bulking may occur. The results of Lesperance's test$^{1)}$, which formed the basis of design, showed the sphere of loading without security of sludge sedimentation, as in Fig. 1. The reason for sludge bulking in a certain condition, as above, is due to failure in application of optimum loading corresponding to each retention time by employing a few operating condition, which proved to be consecutively workable after years of trials and failures by Lesperance, for test conditions. However, the result of this test showed that in case of proper maintenance of loading. sludge sedimentation can be ensured under 120 SVI. Therefore, this study suggested hydraulic retention time and its corresponding optimum loading, and identified the hydraulic retention time as a determinant of sludge sedimentation. And. on the basis of these findings, this study suggested the feasiblity of UHR(Ultra High Rate), a new operating process, exceeding several times the applicable loading value of High Rate Aeration Process under one hour retention time which has not yet applied to the existing treatment processes.

A Novel Approach to the Production of Hyaluronic Acid by Streptococcus zooepidemicus

  • It has been shown that the initial conditions of bacterial cultivation are extremely important for the successful production of hyaluronic acid (HA) by fermentation. We investigated several parameters that affect cell growth rate and the productivity and molecular weight of hyaluronic acid--i.e., agitation speed, aeration rate, culture temperature, pH, and pressure--to determine how to optimize the production of HA by Streptococcus zooepidemicus on an industrial scale. Using a 30-1 jar fermentor under laboratory conditions, we achieved maximum HA productivity and biomass when the agitation speed and aeration rate were increased simultaneously. By shifting the temperature downward from 35$^{\circ}C$ to 32$^{\circ}C$ at key levels of cell growth during the fermentation process, we were able to obtain HA with a molecular weight of $2.8{\times}10^6$ at a productivity of 5.3 g/l. Moreover, we reproduced these optimized conditions successfully in three 30-1 jar fermentors. By reproducing these conditions in a 3-$m^3$ fermentor, we were able to produce HA with a molecular weight of $2.9{\times}10^6$ at a productivity of 5.4 g/l under large-scale conditions.

Effect of Aeration Rates on Ammonia Emissions during Composting of Livestock Manure (축분(畜糞) 퇴비화시(堆肥化時) 공기주입율(空氣注入率)이 암모니아 배출(排出)에 미치는 영향(影響))

  • This experiment was conducted to find out the optimum condition of aeration rates for removal of malodor and to improve the compost quality. The aspect of ammonia emission and amounts of volatilization were investigated in the enclosed composting reactor of 242 liters piled with mixed materials of dairy manure and rice straw, which adjusted to 65% of initial moisture content and controlled by four different aeration rates. Mature temperature increased suddenly in initial composting time and decreased with Increasing aeration rates. The treatment of $1.79l\;min^{-1}kg\;dry-solids^{-1}$ results in overcooling and rapid drying of composting materials because of too much aeration. The average concentration of ammonia emitted from composting for 24 days was the range of 25.3 to $239.8mg\;l^{-1}$ and was highest in the treatment of $0.09l\;min^{-1}kg\;dry-solids^{-1}$, followed by 0.90. 0.18 and $1.79l\;min^{-1}kg\;dry-solids^{-1}$. The range of maximum concentration by different aeration rates was $335{\sim}2279mg\;l^{-1}$ and it wan highest in the treatment of $0.09l\;min^{-1}kg\;dry-solids^{-1}$, followed by 0.18, 0.09 and $1.79l\;min^{-1}kg\;dry-solids^{-1}$. Relationship between the ammonia concentration emitted and temperature matured under different aeration rates showed an exponential positive correlation with 1% significance and had a trend of clear increase in ammonia concentration with increasing temperature over $50^{\circ}C$. Most of ammonia volatilized within plays after composting. The volatilization rate of ammonia ranged from 0.056 to 0.453 per dry solids of materials and it was highest in the treatment of $0.09l\;min^{-1}kg\;dry-solids^{-1}$, followed by 0.18, 0.09 and $1.79l\;min^{-1}kg\;dry-solids^{-1}$. Amounts of ammonia volatilized under composting condition of this experiment was estimated to be highest in the aeration range of 0.9 to $1.0l\;min^{-1}kg\;dry-solids^{-1}$.

Quality Improvement of Retort Oyster Food by the Coating Method (보호피막 처리에 의한 굴 레토르트 식품의 품질 개선)

  • Removing shup from oyster by aeration method and coating with edible coating materials were investigated to improve oyster quality for the retort processing. Aeration rate and pore size of nozzle were critical factors to remove shup by aeration method. Optimum aeration rate and nozzle size were 45 L/min and 0.4 mm, respectively, when aeration was peformed two times and each operation maintained for 1 minute. Sodium alginate (SA) was used for oyster as the basal coating material. However, SA affected mouth-feel when it was applied at high concentrations. Sub-coating materials including skim milk, waxy corn starch, Purity CSC, white corn starch and carboxymethylcellulose (CMC) in decreasing order showed a positive effect on improving coatability and mouth-feel. Therefore, it is suggested that 1.5% SA and 0.9% skim milk should be the optimum composition of coating materials for oyster. Browning and syneresis of the porridge containing the coated oyster were considerably inhibited as compared with the sample without the coated oyster.

A Study on Treatment Characteristics and Design of Ultra High Rate Method (Ultra High Rate(UHR) 법의 처리특성 및 설계에 관한 연구)

  • For the treatment of wastewater being discharged in large quantities as in these modern times, qualitative and quantitative concepts should be introduced in consideration of the recycling. In view of a qualitative concept, high rate aeration process is known as the most effective process up to now. However, Lee suggested UHR process showing the superiority over high rate process in treatment rate and quantity. Therefore, this study was performed to investigate the basic sphere of design and treatment quality, such as applicable $BOD_5$-loading and influent wastewater concentration of UHR process, based on Lee's suggestions1). Consequently, this process showed applicable $BOD_5$-loading of $2.2{\sim}7.0kg-BOD_5/kg-MLSS{\cdot}day$, exceeding three times or more of high rate process with maximum applicable loading of $2.25kg-BOD_5/kg-MLSS{\cdot}day$. Meanwhile, with the range of influent wastewater concentration from 200 to 450 mg/L. it showed the treatment rate from 94.7 to 97.3%, it indicated very good condition. In view of quantitative concept of treatment, UHR process is considered an epoch-making treatment process being superior to existing ones.
