Shoot Regeneration from Cambial Tissue Culture of European Larch (Larix decidua) (유럽낙엽송의 형성층조직 배양으로부터 줄기의 재분화)

  • Adventitious shoots were induced from cambial tissue cultures of 3-year-old seedlings using BLG mineral salts medium supplemented with 10 mM glutamine and 30 mM sucrose. The optimum growth regulator level for bud induction was 4,5 $\mu$M BA which produced average 25.5 shoots per cambium segment. Induced buds were elongated on GD medium supplemented with 30 mM sucrose followed by LMG medium supplemented with 30 mM sucrose for further shoot elongation. Elongated shoots were rooted on half-strength GD medium containing $0.54 ;\mu\textrm{M}$ NAA with the frequency of 20%. This system proved the high morphogenic potential of cambial tissue in larch.

Adventitious Shoot and Plant Regeneration from Anther Culture of Hypericum ascyron L. (물레나물 약배양에 의한 부정 신초 및 식물체 재분화)

  • In order to investigate the effects of low temperature pretreatment of floral bud and plant growth regulators on anther-derived callus and shoot differentiation, anthers were cultured on 1/2 MS medium supplemented with 2,4-D, NAA, BA and TDZ. This plant depends on the plant growth regulators, for these anthers couldn't respond on 1/2 MS medium without plant growth regulators. 2,4-D was a prerequisite substance in this experiment, especially 52.6% of callus formation on MS medium with 2.0mg/L 2,4-D alone. However, the optimum medium was on 1/2 MS medium with 0.1 mg/L 2,4-D and 1.0mg/L BA for continuous growth and shoot differentiation from the anther. Calli derived from on MS medium with 2.0mg/L 2,4-D transferred to the 1/2MS medium with TDZ and BA. TDZ were less superior to BA, only one anther could produce shoot on MS media with 1.0mg/L TDZ. On the other hand, when the calli transferred to the medium with 3.0mg/L BA, adventitious shoots were proliferated, subsequently, regenerated shoots elongated from the embryogenic calli. After floral buds of one week before anthesis were incubated at $5^{\circ}C$ refrigerator for eight or fifteen days, anthers seperated from floral buds were cultured on 1/2MS medium supplemented with 0.1mg/L 2,4-D and 1.0mg/L BA. Callusing and shoot differentiation on anthers from treated at $5^{\circ}C$ for eight days were more effective than those of fifteen days or control.

In Vitro Micropropagation of Polygonatum odoratum cv. Gungangbeaksea (둥굴레 '건강백세'의 기내 대량번식)

  • The Polygonatum odoratum cv. Gungangbeaksea, bred in Gyeongsangnam-Do Agricutural Research & Extension Service, was cultured in vitro for micropropagate rapidly through the culture of rhizome explants ($5{\times}5mm$). The $7{\times}7mm$ explants of adventitious multi-bud clusters (AMC), obtained through the culture of rhizome explants (MS + 3.0 mg/L BA) were cultured on MS media with BA and TDZ. The shoot multiplication was favorable on the MS medium containing 3.0 mg/L TDZ with 2.8 in shoot number. But the formation of AMC was low in all media tested. The explants of AMC were cultured on MS media containing 1.0~5.0 mg/L TDZ and NAA to multiplicate AMC more. The formation of AMC was a little more stimulated on combined MS media of TDZ and NAA, than that with TDZ alone. The multiplication of shoots and AMC was favorable on MS media with 3.0 mg/L TDZ and 5.0 mg/L NAA, and 5.0 mg/L TDZ and 3.0 mg/L NAA. As the concentration of MS salts increased, the formation of AMC was decreased. But the formation of AMC was more stimulated, as the concentration of sucrose increased to 7%. Therefore, the multiplication of shoots and AMC was suitable on media containing 3.0~5.0 mg/L TDZ and NAA, and 7% of sucrose. The explants of AMC were rooted on media with 3.0 mg/L IBA, or 2.0 mg/L NAA with more than 80% in rooting ratio. The plantlets were treated at $5^{\circ}C$ for 8 weeks, and cultured ex vitro for 8 weeks. The survival ratio of plantlets were 100% in vermiculite, and the mixed soil with perlite 1 volumn and vermiculite 1 volumn.

Freezing Resistance of Cryptomeria japonica - Its clonal and Seasonal Differences - (삼나무의 내한성(耐寒性) - 품종별(品種別) 채취시기별(採取時期別) 차이(差異) -)

  • This study aimed to know difference in freezing resistance among different clonal seedlings or different seed source seedlings of Cryptomeria japonica which has been selected where extreme cold prevails in Korea and Japan. The freezing resistance of three 12-50 year old trees was also measured in the experiment. The freezing resistance was measured in different tissue parts: mainly leaf, cambiam and xylem, at three different collection dates in two different collection places during the winter of 1977-1978. The following results and discussions were made: 1. The clonal difference in freezing resistance of Cryptomeria japonica was $9^{\circ}$ to $15^{\circ}C$ in maximum according to the collection place. However, the clonal difference in freezing resistance was not related to the difference in climatic conditions where the parent tree have been growing. This impiled that the natural selection of cold resistant genes in Cryptomeria japonica has not reached its evolutional equilibrium yet since most of the Cryptomeria forest has been established by artificial regeneration. 2. The difference in freezing resistance among leaf, cambium and xylem was not apparent except that leaf of several clones showed higher freezing resistance than cambium or xylem when they collected at mid-winter. The least freezing resistant tissue part, thought its freezing resistance was not measure in all clones and all temperatures were appeared in the apical buds. The new shoot growth was observed in the next spring with being replaced by its dormant or adventitious bud growth when the apical bud was injured dy cold during winter. 3. The freezing resistance of leaf, cambium and xylem was shown high enough so that freezing resistance Cryptomeria clones in this experiment were supposed to be able to survive in cold winter conditions at the middle part of Korea. However, it was reported that the most susceptible tissue part to winter injury was the basal stem, but of which freezing resistance was not-measured in this experiment. Several silvicultural methods for prevention of Cryptomeria seedlings from cold damage were discussed in literature.

Plant regeneration and soil acclimatization through photoautotrophic culture from leaf explant of a rare species in Sedum tosaense Makino (희귀수종인 주걱비름(Sedum tosaense Makino)의 잎절편으로부터 기내 식물체 재분화 및 광독립배양을 통한 토양순화)

  • The aim of this study was to establish plant regeneration from leaf explants of Sedum tosaense Makino, which is globally rare and endangered species. The leaf explants of S. tosaense were cultured on the MS medium supplemented with different concentration of BA and NAA for callus induction. Callus induction was showed the highest (100%) on MS medium containing $2.0mg{\cdot}L^{-1}$ BA and $1.0mg{\cdot}L^{-1}$ NAA. The highest number of shoots were regenerated when callus were cultured on MS medium containing $2.0mg{\cdot}L^{-1}$ BA and $1.0mg{\cdot}L^{-1}$ NAA for 5 weeks. The axillary bud were cultured on the MS media supplemented with combination of BA and NAA for in vitro propagation. The highest number of adventitious shoot (7.9 per explants) formed at $1.0mg{\cdot}L^{-1}$ NAA and $2.0mg{\cdot}L^{-1}$ BA. For rooting, MS medium supplemented with or without $2.0g{\cdot}L^{-1}$ activated charcoal was tested. The optimal results were observed using MS medium supplemented with $2.0g{\cdot}L^{-1}$ activated charcoal, on which 85.7 (No. of root), 4.6 cm (length of root). 1,200 ppm $CO_2$ and 350 ppm $CO_2$ were supplied for make certain the effects of $CO_2$ on pre-acclimatization by photoautotrophic culture. 1,200 ppm $CO_2$ treatment was established higher than 350 ppm $CO_2$ treatment. Soil acclimatization of in vitro plantlets was the best in mixture soil consisted of peat moss and perlite with 100% survival rate and they showed the maximum growth.

Influences of Air Pollution on the Growth of Ornamental Trees - With Particular Reference to SO2 - (대기오염(大氣汚染)이 조경수목(造景樹木)의 생육(生育)에 미치는 영향(影響) - 아황산(亞黃酸)가스에 대(對)하여 -)

  • For the purpose of detecting the capability of the trees to resist air pollution and of determining the tree species best suited for purification of polluted air, particularly with regard to $SO_2$ contamination, six following ornamental tree species were selected as experimental materials: i.e., Hibiscus syriacus L., Ginkgo biloba L., Forsythia koreana Nak., Syringa dilatata Nak., Larix leptolepis Gordon, and Pinus rigida Miller. The susceptiblities of the trees were observed and analyzed on the basis of area ratio of smoke injury spots to the total leaf area. The results of the experiments are as follows: I. The Susceptibilities to Sulfur Dioxide. (1) The decreasing order of tolerance to $SO_2$ by species was as follows: 1. Hibiscus syriacus 2. Ginkgo biloba, 3. Forsythia koreana, 4. Syringa dilatata, 5. Larix leptolepis, and 6. Pinus rigida. In general, Hibiscus syriacus and Ginkgo biloba can be grouped as the most resistant and Larix leptolepis and Pinus rigida as the least resistant and Forsythia koreana and Syringa dilatata as of intermediate resistance. (2) The sulfur content of the leaves treated by $SO_2$ increased in proportion to the increase of the concentration of the fumigation. The content in the coniferous species proved to be less than that of the broad-leaved species, but Ginkgo biloba proved to contain as much sulfur as broad-leaved species. (3) The earlier-stage leaves fumigated in June with the $SO_2$ concentration up-to-l-ppm showed that sulfur content increases in proportion to the increase of the concentration of the fumigation, but the difference between concentration was not so significant. (4) The later-stage leaves fumigated in October showed higher sulfur content than the earlier stage leaves, and a wider range of difference in sulfur content was detected among different concentrations. The limit of fumigation resulting in culmination of sulfur absoption in broad-leaved species, such as Syringa dilatata, Hibiscus syriacus, and Forsythia koreana proved to be around 0.6 ppm. (5) Due to the sprouting ability and the adventitious bud formation, the recovery from $SO_2$ fumigation was prominent in Hibiscus syriacus, Syringa dilatata, and Forsythia koreana. (6) The differences in the smoke spot color were recognized by species: namely, dirt-brown in Syringa dilatata, brilliant yellowish-brown in Pinus rigida and Ginkgo biloba, whitish-yellow in Hibiscus syriacus and reddish-brown in Forsythia koreana. (7) The leaf margins proved to be most susceptible, and the leaf bases of the mid-rib most tolerant. In both Ginkgo biloba and Larix leptolepis, the younger leaves were more resistant to $SO_2$ than the older ones. II. The ulfur Content of the Leaves of the Ornamental Trees Growing in the City of Seoul. (1) The sulfur contents in the leaves of the Seoul City ornamental trees showed a remarkably higher value than those of the leaves in the non-polluted areas. The sulfur content of the leaves in the non-polluted area proved to be in the following descending order: Salix pseudo-lasiogyne Leveille, Ginkgo biloba L., Alianthus altissima swingle, Platanus orientalis L., and Populus deltoides Marsh. (2) In respect to the sulfur contents in the leaves of the ornamental trees in the city of Seoul, the air pollution proved to be the worst in the areas of Seoul Railroad Station, the Ahyun Pass, and the Entrance to Ewha Womans University. The areas of Deogsu Palace, Gyeongbog Palace, Changdeog Palace, Changgyeong Park and the Hyehwa Intersection were least polluted, and the areas of the East Gate, the Ulchi Intersection and the Seodaemun Intersection are in the intermediate state.
