Effects of Growth Regulators and Culture Environment on ex vitro Rooting and Acclimatization of Apple Rootstock in vitro Propagated (기내배양 사과 대목의 기외 삽목 시 발근과 순화에 미치는 배양조건 및 생장조절물질의 효과)

  • Growth of M.9 (Malus domestica Bark. cv) and M.26 (Malus domestica Bark. cv) of dwarf apple rootstock, cultured on MS agar medium in a vessel with ventilating stopper (VS) and then in vivo rooting and acclimatization under combined-treatment by some materials with IBA, were investigated. Concentration of $CO_2$ and ethylene in the vessel with VS was lower then in the vessel with non-VS. Change of temperature and humidity in the vessel with VS was repeated by light condition. Stomatal pares of tissue in the vessel with VS were immediately closed after plantlets were exposed to room humidity but those in the vessel with non-VS were opened after 20 minutes exposure to room humidity. Leaf area and chloroplast index of tissue in the vessel with VS was higher then in the vessel with non-VS. In vivo rooting ratio and acclimatization ratio of M.9 and M.26 was highest in 300mg/L IBA+3% sucrose dip-treatment among other combined- treatments.

Effects of Pre-sowing Seed Soaking and Planting Depth on Dormancy Breaking and Seedling Emergence of Pearl Millet (Pennisetum americanum(L.) Leeke) (진주조의 파종전 침종 및 파종심도가 휴면타파와 출아에 미치는 영향)

  • One of the most important cultural techniques of pearl millet (Pennisetum americanum (L.) Leeke) is to encourage rapid and uniform emergence of seedlings to establish good stand and to let them grow well. Thus the objectives of the study were to investigate the effects of pre-sowing seed soaking and planting depth on dormancy breaking, germination and emergence of the seedlings, and to estimate the optimum planting season of pearl millet in Suwon, Korea. The seeds with dormancy germinated 99 to 100 percent when soaked in the H$_2$O$_2$ 1% solution for 24 hours and rinsed with pure water, but germinated only 38% and 83% when soaked in pure water for 24 hours just after harvest and drying, and one month later from the harvest time, respectively. The seeds of Australia inbred line did not germinate at the constant 10$^{\circ}C$, but germinated at the constant 11$^{\circ}C$. It also was possible to estimate the optimum planting season by applying minimum temperature 11$^{\circ}C$ for germination. The minimum air temperature reached from late April in Suwon, Korea in regular years but fluctuated from late April to early May in 1986 and 1987. Thus, the safe planting season was mid-May for rapid and uniform germination of pearl millet seed. The optimum depth of planting was 2∼4cm under the optimum soil moisture condition, and 4 to 6 cm under the drier soil moisture condition. Subcoleoptile internode(mesocotyle) length increased according to increased depth of planting. Seedling crown placement also became deeper due to deeper planting of the seeds. The subcoleoptile internode length and seedling crown depth were positively correlated with actual planting depth, indicating that deeper planting would be not good for appropriate adventitious root and tiller development.

Effective Micropropagation of Pulsatilla cernua var. koreana through Apical Meristem Culture (할미꽃 정단 분열조직 배양을 통한 효율적 미세번식)

  • In order to investigate the effect of plant growth regulators on effective in vitro micropropagation, apical meristems of Pulsatilla cernua var. koreana were cultured on Murashige and Skoog's (MS) medium with 2,4-D, NAA, TDZ and BA. Media containing 2,4-D and kinetin, 2,4-D and TDZ, NAA and TDZ were not effective on callus induction. However, embryogenic or organogenic callus was obtained on media containing NAA and BA. Especially, on MS medium with 0.5mg/L NAA and 1.0mg/L BA was optimal for a high frequency (62%) of shoot or shoot bud obtained from callus. Callus proliferation, shoot multiplication and elongation were significantly increased by adding 10% coconut water on MS media with 0.5mg/L NAA and 1.0mg/L BA. Repeated subculturing of in vitro grown shoots resulted in propagation rate of 12.9 shoots per explant every 30 days. Root formation from the adventitious shoots was not easily achieved. However, roots were only produced through callus on MS medium with 2.0mg/L NAA alone or 0.5mg/L NAA and 1.0mg/L BA. These roots were used materials for callus and shoot production repetitively.

Micropropagation of Cassava by Suspension Culture Derived from its Nodal Explants (마디 절편의 현탁배양에 의한 카사바의 미세증식)

  • For the micropropagation, node explants of cassava were cultured in liquid MS medium with various concentrations of cytokinins on a rotary shaker (100 rpm) for 2 weeks. The adventitious roots and shoots from the explants were differentiated more efficiently in liquid medium than in solid. But root formation was not inhibited in medium with BAP and kinetin at low concentration (>0.05 mg 1/sup -1/), while in medium added with BAP and zeatin at high level (<0.25 mg 1/sup -1/), it was inhibited by callus forming on cut end of the cuttings. However, all of plantlets grown in liquid medium for more than 2 weeks showed symptoms of hyperhydricity. The plantlets grown in liquid medium were transferred into culture bottles filled with fine sand or artificial soil (pitmoss:perlite:vermiculite, 1:1:1 v/v) wetted with half strength of Knop's solution. After transplanted to culture bottles, some of vitriscent leaves were defoliated and new leaves were normally formed from shoot apex. Most of plantlets (>95%) were hardened-off successfully only in culture bottles with fine sand, and grew into 3-5 cm seedlings possessing 4-6 nodes after 4 weeks. Thus, the mass propagation of cassava on medium containing cytokinin could be established based on the suspension culture using node explants.

Plant regeneration and soil acclimatization through photoautotrophic culture from leaf explant of a rare species in Sedum tosaense Makino (희귀수종인 주걱비름(Sedum tosaense Makino)의 잎절편으로부터 기내 식물체 재분화 및 광독립배양을 통한 토양순화)

  • The aim of this study was to establish plant regeneration from leaf explants of Sedum tosaense Makino, which is globally rare and endangered species. The leaf explants of S. tosaense were cultured on the MS medium supplemented with different concentration of BA and NAA for callus induction. Callus induction was showed the highest (100%) on MS medium containing $2.0mg{\cdot}L^{-1}$ BA and $1.0mg{\cdot}L^{-1}$ NAA. The highest number of shoots were regenerated when callus were cultured on MS medium containing $2.0mg{\cdot}L^{-1}$ BA and $1.0mg{\cdot}L^{-1}$ NAA for 5 weeks. The axillary bud were cultured on the MS media supplemented with combination of BA and NAA for in vitro propagation. The highest number of adventitious shoot (7.9 per explants) formed at $1.0mg{\cdot}L^{-1}$ NAA and $2.0mg{\cdot}L^{-1}$ BA. For rooting, MS medium supplemented with or without $2.0g{\cdot}L^{-1}$ activated charcoal was tested. The optimal results were observed using MS medium supplemented with $2.0g{\cdot}L^{-1}$ activated charcoal, on which 85.7 (No. of root), 4.6 cm (length of root). 1,200 ppm $CO_2$ and 350 ppm $CO_2$ were supplied for make certain the effects of $CO_2$ on pre-acclimatization by photoautotrophic culture. 1,200 ppm $CO_2$ treatment was established higher than 350 ppm $CO_2$ treatment. Soil acclimatization of in vitro plantlets was the best in mixture soil consisted of peat moss and perlite with 100% survival rate and they showed the maximum growth.

Effects of Depth and Duration of Flooding on Growth and Yield at Flowering Stage in Tomato(Lycopersicon esculentum). (토마토(Lycopersicon esculentum)의 개화기 침수 처리에 따른 생육 반응)

  • Tomatoes are flooded differently 0, 5, 10 and 15cm, according to the developing stages such as flowering stage under the condition of greenhouse. Along with this, they are treated according to the time condition such as 6, 12, 24, 48 and 120 hours. The results obtained are summarized as follows. Plant height decreased in the depth of $0{\sim}10cm$ for over 48 hours, in the depth of 15cm for over 24 hours. Number of leaves was the same as in control, and it decreased over. Number of flowers and fruit setting of individuals decreased conspicuously according as the depth and the hours got greater and longer. Adventitious root occurred remarkably in the depth of $0{\sim}10cm$, for over 24 hours and in the depth of 15cm, 12 hours. Epinastic curvature increased greatly as the depth and the hours got greater and longer. Diffusion resistance of stomata cell increased as the depth and the hours got greater and longer. Diseases occurred conspicuously as the hours of flooding got longer rather than as the depth greater. The preventing of diseases caused by insecticide was observed, but it was not greater than in the seedling and transplanting stage. Fertilization was effective in the case of increasing the weight of shoot. Number of fruits per plant did not decrease in the depth of 0cm up to 24 hours, but decreased on the deeper level of flooding and increased as the hours got longer. Moreover with the exception of 120 hours per respective depth of the treatment, average weight of a fruit got greater as the depth and the hours got greater and longer. In the case of epinastic curvature and diffusion resistance, there was negative correlation between all the other investigated characters and positive correlation between weight of a fruits and average weight of a fruit.

Effects of Inoculation with Mycorrhizal Fungi, Pisolithus tinctorius and Glomus sp. on the Rooting of Quercus acutissima Carr. Cuttings at Various Ortet Ages (모래밭 버섯균(菌)과 Glomus 균근균(菌根菌)의 인공접종(人工接種)이 연령(年齡)이 다른 상수리나무에서 채취(採取)한 삽수(揷穗)의 삽목발근(揷木發根)에 미치는 영향(影響))

  • This study was conducted to determine the effects of mycorrhizal inoculation and ortet ages on the rooting of Quercus acutissima cuttings. The cuttings taken from 12-week-, 2-year-, 4-year-, and 20-year-old plants were rooted in mid summer in the rooting medium (vermiculite 2 : peatmoss 1 by volume) with or without Pisolithus tinctorius (Pt) and Glomus sp. inocula and IBA under intermittent misting system in an open shed. The average percentages of rooting were about 82%, 49%, 29%, and 13% for cuttings taken from 12-week-, 2-year-. 4-year-, and 20-year-old seedlings, respectively. Pt inoculation, enhanced rooting of cuttings at all age classes, except 12-week-old seedlings, with the highest enhancement(22%) observed in cuttings taken from 20-year-old trees. The highest percentage of rooting in each age group eras 88.9% in 12-week-old seedlings treated with Pt plus 3.000ppm IBA, 75% in 2-Year-old plants with 1,000ppm IBA, 58.3% in 4-year-old plants with 3.000ppm IBA and 22% in 20-year-old plants. The addition of Glomus sp. fungus inoculum failed to enhance rooting. Pt mycorrhizal inoculation enhanced root dry weight, length, and diamter of adventitious roots at cuttings taken from 12-week- and 20-year-old trees, except the cuttings taken from 4-year-old seedlings. Rooted cuttings had more total nitrogen content in the leaves than unrooted cuttings, and the greater rooting response was associated with the higher phosphorus content in the leaves.

The Rooting Ability of Selected Clones of Populus alba×glandulosa Hybrid (Populus alba×glandulosa의 Clone간(間) 발근력(發根力)의 차이(差異))

  • In order to investigate the difference of rootability between 15 clones of Populus alba${\times}$glandulosa selected based on the growth performance, rooting of cutting experiments with these 15 clones were conducted at the nursery for six years from 1970 to 1975. Cutting experiments in a temperature controlled incubator in which the temperature of the cutting bed were set to $10^{\circ}C$, $15^{\circ}C$, $20^{\circ}C$, $25^{\circ}C$ and $30^{\circ}C$ were also performed. Along with these experiments air layering experiments were performed to compare with the rootabilities obtained from nursery trial. The results obtained so far could be summarized as follows. 1. The best rooting clones were 65-22-4 and 65-22-11, and the average rooting percentages of these two clones for six years were 76.7%, and 72.9% respectively. The poorest rooting clone was 66-6-8 showing average rooting percentage of 45.8%. 2. The middle class of rooting percentage was ocuppied by the clones; 66-14-29, 66-14-93, 66-25-5 and 67-6-3, and the range of their rooting percentage was 60~69% on average. 3. The rooting performances observed through the nursery, the incubator and the air layering experiments were almost the same with exception of few clones. 4. P. alba${\times}$glandulosa showed the best rooting percentage at the cutting bed of $20^{\circ}C$ 5. The most roots, i.e. 78.5% of root per cutting were developed from the bottom part of the cutting shoot. 6. Adventitious and call use roots could observe in the cuttings.

The Establishment of Nitrogen Fixation by Cultured Cell-Rhizobium Association Through Tissue Culture Technique in Soybean (대두조직배양세포(大豆組織培養細胞) - Rhizobium에 의(依)한 질소고정력(窒素固定力))

  • This experiment was carried out to elucidate the factor of nitrogenase formation and to establish the nitrogen fixation system in mixed culture of cultured cells and rhizobia through tissue culture technique using three soybean varieties, Hwangkeum, Namcheon and D 68-0099 as host plants. The results obtained were as follows; The callus was induced in embryo and radicle, but not in hypocotyl. The most favorable callus induction was caused by the individual application of 2,4-D and NAA at the concentration of 2mg/1 and 4mg/1, respectively, but in case of treating both 2,4-D and kinetin, that was done at the concentration of 0.2mg(2,4-D)/0.05mg(kinetin)per liter. The growth of cultured cell was good at the concentration of 2.0mg(2,4-D)/1 and 0.2mg(2,4-D)/0.05mg(kinetin)per liter. When cultured cells were inoculated with R. japonicum 019 and 011, their growthes were considerably inhibited. The addition of single amino acid inhibited the growth of cultured cells. Hwangkeum was inhibited considerably by methionine and leucine. The inhibition of growth by single amino acid can be abolished by the addition of certain amino acids. The differentiation of adventitious root was good at the concentration of 2.0mg 2,4-D and 0.2mg 2,4-D/0.05mg kinetin per liter. Of three host plants tested with 25 R. japonicum strains, Hwangkeum had affinity for 10 strains, Namcheon for 7 strains and D68-0099 for none. The nitrogen fixing abilities of Hwangkeum and Namcheon caused by cultured cell-Rhizobium association were high in strain 019, 007, and in 007 mixed with 119, respectively.
