To investigate the optimal conditions for shoot organogenesis in Astragalus sinicus L., hypocotyl explants were cultured in Murashige & Skoog's (MS) medium supplemented with 0.1, 1.0, 2.0, or 4.0 mg/L 2,4-dichlorophenoxy acetic acid (2,4-D) for 6 weeks. 2,4-D concentration significantly effected morphogenesis: some produced calli with adventitious shoots and roots, some produced calli with adventitious roots, some produced only calli, and some produced deep-brownish calli with roots. The formation of calli with shoots and/or roots was observed at lower levels of 2,4-D, whereas calli without shoots or with deep-brownish roots were formed after treatment with higher levels of 2,4-D. Also, a shoot organogenesis ability of callus clones was observed after treatment with medium with 0.1 or 1.0 mg/L 2,4-D grown in MS medium with combinations of benzyl adenine (BA) and 2,4-D for 4 weeks. Medium with a combination of BA and 2,4-D was effective for shoot formation, whereas root organogenesis from calli decreased. The greatest amount of shoot formation was obtained when calli were cultured in MS medium containing 1.0 mg/L 2,4-D and 0.5 mg/L BA. Upon shoot transfer into 1/2 MS basal medium, plantlets developed, and the plantlets grew well in soil in a greenhouse.
Conditions for efficient organogenesis and plant regeneration from Dianthus gratianopol suspension cultured cells were established. Shoot-forming calli of glossy surface, pale green and knobby type were selected from leaf explant-derived calli and were suspension-subcultured every week in CP liquid medium with 1.0 mg/L 2,4-D and 0.5 mg/L BAP. Combinations of 1.0 mg/L 2,4-D and 0.5 mg/L BAP, and 1.5 mg/L 2,4-D and 0.5 mg/L BAP were effective for the induction of regenerative callus from the suspension cultured cell clusters. Multiple shoot primordia were initiated from the green spots of these regenerative callus and formed shoots on MS medium with 1.0 mg/L TDZ and 0.5 mg/L PAA. Shoot regeneration frequency (calli regenerating at least one shoot) was about 87%. For plant regeneration, proliferated shoots were excised and transferred to MS medium with 0.1 mg/L NAA for root initiation after 9 weeks of culture. The regenerants were potted in soil and formed the flowering buds and petals. Also, adventitious shoots were formed from the excised green shoot primordia of regenerative callus and these shoots proliferated successfully and regenerated to whole plants.
This study was carried out to induce plant regeneration via shoot formation from sucker explants of Rubus fruticosus L. To induce adventitious shoots, sucker explants were sterilized in $1.2\%$ NaOCl solution, and cultured on the MS solid medium supplemented with kinetin (0.5, 1.0, 3.0 mg/L) and BA (0.5, 1.0, 3.0 mg/L), respectively. As above, to induce adventitious shoots, sucker explants were cultured on the MS solid medium supplemented with IBA (0, 0.1, 1.0 mg/L) and BA (0, 0.1, 1.0, 2.0 mg/L). After 4 weeks of culture, the highest frquency $(100\%)$ of shoot formation from sucker explants was obtained from the medium with 1.0 mg/L BA. The highest shoot number per explant from in vitro shoot explants was 5.3. After 10 weeks of culture, the number of shoot per explant was increased. The highest frequency $(85\%)$ of root formation was obtained at 0.5 mg/L glycine medium, when the explant with shoot were cultured on the MS medium containing glycine at various concentrations from 0 to 2.0 mg/L. The survival rate of the plantlets after transfer to plastic pots containing sand, soil, and vermiculite (1:1:1, vol.) was $95\%$. The results indicate that micropropagation procedure can be applied for an efficient mass propagation of Rubus fruticosus.
This study was conducted to examine the effect of plant growth regulators on the formation of adventitious bud callus(ABC) and plant regeneration in shoot tip culture of Zantedeschia spp. 'Florex Gold'. Treatment of $0.1mgL^{-1}\;N$-phenyl-N'-1,2,3-thiadiazol-5-ylurea(thidiazuron, TDZ) was more promotive for formation of ABC in shoot tip culture than 6-benzylaminopurine(BA) treatment, and short shoots were developed. Comparing to treatment of BA, mixing treatment of BA and 1-naphthaleneacetic acid(NAA) inhibited the formation of ABC and multiple shoot. The proliferation of ABC derived from sections(0.3 cm) of ABC produced by shoot tip culture in medium supplemented with $0.1mgL^{-1}\;TDZ$ was more effective in medium with $0.1mgL^{-1}\;TDZ$ or $2.0mgL^{-1}\;BA$ than the other treatments. The shoot regeneration and growth from sections of ABC was more promotive in treatment of $0.001mgL^{-1}\;TDZ$. Also, the root growth from sections of ABC was better in medium with $0.001mgL^{-1}\;TDZ$ or $0.2mgL^{-1}\;BA$. Consequently, in vitro mass production of Zantedeschia spp. 'Florex Gold' can be obtained via indirect organogenesis through plant regeneration and proliferation of ABC which was derived from shoot tip culture at $0.1mgL^{-1}\;TDZ$.
Background: The genus Panax in the Araliaceae family has been used as traditional medicinal plants worldwide and is known to biosynthesize ginsenosides and phytosterols. However, genetic variation between Panax species has influenced their biosynthetic pathways is not fully understood. Methods: Simultaneous analysis of transcriptomes and metabolomes obtained from adventitious roots of two tetraploid species (Panax ginseng and P. quinquefolius) and two diploid species (P. notoginseng and P. vietnamensis) revealed the diversity of their metabolites and related gene expression profiles. Results: The transcriptome analysis showed that 2,3-OXIDOSQUALENE CYCLASEs (OSCs) involved in phytosterol biosynthesis are upregulated in the diploid species, while the expression of OSCs contributing to ginsenoside biosynthesis is higher in the tetraploid species. In agreement with these results, the contents of dammarenediol-type ginsenosides were higher in the tetraploid species relative to the diploid species. Conclusion: These results suggest that a whole-genome duplication event has influenced the triterpene biosynthesis pathway in tetraploid Panax species during their evolution or ecological adaptation. This study provides a basis for further efforts to explore the genetic variation of the Panax genus.

Waterlogging strongly affects maize (Zea mays L.) growth. It is necessary to find the screening method of waterlogging tolerant maize lines. This study was to investigate the growth characters at V3 stage of maize, when is very sensitive to waterlogging. Six Korean maize inbred lines were subjected to waterlogging at V3 stage for 30 days. The 30 days waterlogging treatment significantly reduced plant height, number of expanded leaves, and SPAD value, compared with the control plants. SPAD values were significantly different among the six inbred lines, KS140 was the highest. The dry matter accumulation of aerial and root part were significantly decreased by 30 days waterlogging. KS140 was the weightiest among inbred lines. The dry matter of adventitious root showed same trend. Waterlogging treatment significantly reduced to ear length and thickness, grains filling length, grain number per ear, and maize grain. Plant height, SPAD value, and number of fully-expanded leave showed high correlation with maize grain yield, but number of senescent leaves, dry matter of adventitious root and TR ratio did not, suggesting that the former three traits may be good indicator for evaluating 30-day waterlogging tolerance of maize inbred lines. KS164 was the highest yield by increasing of grains filling length and grain number per ear of among waterlogging inbred lines. According to the results, evaluation of maize waterlogging should be consider both early growth characteristics and resilience in the later growth stages.
This research was conducted to investigate the influence of leaching fractions (LF) in each irrigation or fertigation on plant growth and changes in chemical properties of root media during the production of seedling grafts of tomato. Two root media containing Sphagnum peat moss plus vermiculite (5:5, v/v, PV) and coir dust plus vermiculite (5:5, v/v, CV) were formulated and pre-planting fertilizers were incorporated during formulation. Then, each medium was packed into 50 cell (volume 33 cc) and 105 cell (volume 18 cc) trays and the rootstock (cv. J3B Strong) and scion (cv. Sunmyung) were grown, respectively. The seedlings were grafted at 31 days after sowing and then the cut seedling grafts (Sunmyung scion/J3B Strong rootstock) were planted into 50 cell plug trays containing each of the two root media. After induction of the graft union and new adventitious roots for 7 days, the seedling grafts were fed with fertilizer solution once a week containing 4 different N concentrations (0, 50, 100, $200mg{\cdot}L^{-1}$). When determined after 31 days from seed sowing, the highest fresh weights of the root stock seedlings were obtained with 0.75 LF in PV (8.96g/seedling) and CV (7.11g/seedling) mixes. The EC of the both mixes were 0.93 and $1.09dS{\cdot}m^{-1}$, respectively. The fresh weights of the scion seedlings 31 days after seed sowing were 4.29g with 0.50 LF in the PV and 3.13g with 0.50 LF in the CV. The root medium ECs of the two treatments were 0.76 and $1.34dS{\cdot}m^{-1}$, respectively. Fresh weights of the seedling grafts grown for 31 days were greatly influenced by post-planting fertilizer concentrations. The heavier plants were obtained in $100mg{\cdot}L^{-1}$ N treatment than any other treatments in same mixes. The substrate ECs in these two treatments were 0.98 and $1.93dS{\cdot}m^{-1}$, respectively, indicating that the desirable range of soluble salts in soil extracts is higher in the CV mix than the PV mix. Results of this study suggest that optimum EC range is different in each medium and LF need to be adjusted differently for each root medium to produce high quality seedling grafts of tomato.
This study was investigated the physiological properties of auxin-producing bacteria that have plant growth promoting activity and plant pathogen antagonistic ability. Auxin-producing bacteria were isolated from field soils of Gyeongsan, Korea. Selected strains were identified as a Pseudumonas fulva N21 and a Pantoea agglomerans; K35 by morphological and physiological test, and Biolog (Microlog) system. Auxins were determined by Salkowski in vitro test and mungbean adventitious root induction bioassay. Also produced indole-3-acetic acid (IAA) was identified by TLC. During cell growth, auxin production were highest in their idiophase after log phase and $35^{\circ}C$ at pH 7.5.
This experiment was conducted out to investigate the effect of plant growth regulators on callus and shoot formation. The callus formation was effective on 1/2 MS medium containing 2,4-D, while shoot formation was suppressed. Shoot formation and differentiation were the highest in combination 0.1 mg/L of IAA and 0.1 mg/L of BA. The polarity of explants was investigated from cotyledon, which excised 20% of each basal and terminal parts. Formation of shoot was induced from excised ends of the basal part. In excised ends of the basal part, callus was induced vigorously and shoots were produced lately. Root induction was easily achieved in 1/3 MS medium from the adventitious shoot and more than 90% of regenerated plantlets acclimatized successfully and flowered normally.

One of micropropagation methods was investigated by using a multiple-shoots protocol. Multiple shoot formation was obtained from excised axillary buds of Hypericum erectum on half-strength or basal MS medium supplemented with TDZ or BA. The optimal combination of shoot multiplication for the production of more shoots with a suitable size was MS medium supplemented with $0.005\;mg{\cdot}L^{-1}$ TDZ (6.5 adventitious shoots per node). In vitro rooting was carried on half-strength MS medium with $1\;mg{\cdot}L^{-1}\;GA_3\;and\;0.5\;mg{\cdot}L^{-1}$ IBA treatment. In addition, the rooted cuttings were showed a better root growth in the greenhouse and survived in more than 90%. The results show that the species can be micropropagated effectively by the application of axillary bud culture systems.
