Effects of Aloe on Liver Function and Lipid Metabolism in Alcohol-Consuming Rats (Aloe가 알코올을 섭취한 흰쥐의 간 기능 및 지질대사에 미치는 영향)

  • The effects of aloe on liver function and lipid metabolic disorders induced by alcohol consumption were studied in rats using aloe power (0.25%, 0.5%, 1%) and 10% ethanol. 35 Sprague-Dawley (male, 4 weeks old) rats were divided into five groups and fed experimental diets for six weeks. Body weights of rats tended to be lower in all alcohol supplemented groups than in the control. Food intakes and dry feces per day were significantly lower in all alcohol supplemented groups than in the control. Atherogenic indices (AI) were highest in the alcohol group and decreased in proportion with aloe amount. Serum triglyceride level was significantly higher in the alcohol group than in the control, but tended to be lower in the aloe supplemented groups. In relation to liver function, glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), and alkaline phosphatase (ALP) activities tended to be higher in the alcohol groups than in the control, but lower in the aloe groups, especially in the alcohol+0.5% AO group. The levels of liver cholesterol were significantly lower in the alcohol group than in the control and aloe supplemented groups. In the histochemical evaluation, fat droplets appeared extensively on the liver-lobule in the alcohol group, whereas they decreased slightly in the alcohol+0.25% AO group and apparently disappeared in the alcohol +0.5% AO. On the other hand, fat droplets appeared again on the liver-lobule in the alcohol+1% AO group, but were reduced compared with the alcohol group. Regarding the fatty acid composition of adipose tissue triglycerides, the level of linoleic acid (18:2) was significantly higher in the aloe supplemented group. Regarding the fatty acid composition of liver phosphatidylcholine (PC), the level of linoleic acid was higher in the alcohol group and alcohol+1% AO group than the other groups. In contrast, the level of arachidonic acid was significantly lower in the alcohol group. As a result, arachidonic / linoleic acid ratios were significantly lower in the alcohol group compared to the control group, whereas the ratios of the aloe supplemented groups were similar to that of the control group. Therefore, aloe had some beneficial effects on lipid metabolic disorders induced by alcohol and affected desaturation of fatty acids.

  • This study was carried out to estimate beneficial effects of medicinal plant [Oenanthe javanica (MNR), Coicis lachryma-jobi L. var. (YM), Plantaginis asiatica L. (CJJ)] water extracts on activities of key enzymes such as lipoprotein lipase (LPL), acyl-CoA synthetase (ACS) and carnitine acetyltransferase (CAT) on lipid metabolism. LPL and ACS were extracted from the epididymal adipose tissue and liver of Zucker lean rats (lean) and Zucker fatty rats (fa/fa). MNR or YM water extract treatment significantly reduced the activity of lean and fa/fa LPL. When 10000 ppm of MNR, YM, and CJJ water extracts were tested, they decreased fa/fa LPL activity by 32.5%, 30.1% and 22.8%, respectively. The lean ACS activity was significantly higher in YM water extract treatment compared to the control and the MNR water extract treatment significantly increased the activity of fa/fa ACS, compared to the activity in the control (p<0.05). MNR water extract activated fa/fa ACS activity by 12-fold compared with control at 10000 ppm concentration. CAT activity was significantly higher in 10000 ppm and 20000 ppm CJJ water extract treatment than in the control. Thus, the MNR, YM and CJJ water extracts may have beneficial effects due to activities of enzymes related with fat metabolism in obese humans.

Effect of 8 Weeks of Low Calorie Diet on Body Composition and Blood Parameters in the People with BMI Greater than $23\;kg/m^2$ (체질량지수 $23\;kg/m^2$ 이상 남녀에서 8 주간의 의학 영양 치료에 의한 저열량 식이 섭취가 체격지수 및 혈액학적 지표에 미치는 영향)

  • The principal objective of this study was to assess the effects of low-calorie diets(LCD) for 8 weeks of medical nutrition therapy in individuals with BMI in excess of $23\;kg/m^2$. 41 over-weight or obese individuals (male=14, female=27, age $28.6{\pm}8.9\;yrs$), none of whom were diagnosed with a medical disease, were administered MNT 4 times over the study period. Approximately $1,100{\sim}1,300\;kcal/day$ were prescribed by a dietitian, via individualized counseling. Anthropometric parameters, daily nutrient intake, and blood levels of leptin, insulin, and lipid profiles were measured prior to and after the 8 weeks of the intervention period. We noted significant reductions in body weight, body mass index (BMI), fat mass (FM), visceral adipose tissue (VAT), abdominal skin-fold thickness, and waist circumference in both men and women. The mean body weight losses in men and women were $3.2{\pm}0.6\;kg$/8 weeks and $2.8{\pm}0.6\;kg$/8 weeks, respectively. However, the % of lean body mass (LBM) in men and women was shown to increase significantly (p<0.05). The daily intake of calorie, fat, protein, vitamin E, folate, and iron were significantly reduced during the LCD period. Blood levels of lipids and glucose were in normal range, and evidenced no changes after LCD. However, the serum levels of leptin in female subjects were significantly reduced (p<0.00l) from $8.9{\pm}4.8\;ng/mL$ to $6.9{\pm}4.8\;ng/mL$. In conclusion, 8 weeks of LCD with individualized counseling by a dietitian effectively reduced body fat and visceral fat in both men and women with BMI in excess of $23\;kg/m^2$.

Effect of Curcuma longa L. on the Obesity and Insulin Resistance in Sprague-Dawley Rats and db/db Mice (SD계 랫트와 db/db 마우스에서 Curcuma longa L.가 비만과 인슐린저항성에 미치는 영향)

  • In this study, the effect of Curcuma longa L. on obesity and insulin resistance was investigated in animals fed a moderate high fat diet. The animals used in this study were normal weight Spargue-Dawley (SD) rats and type 2 diabetic obese db/db mice. Accumulation of abdominal adipose tissue and weight gain were inhibited in the animals fed the C. longa extract. C. longa decreased fasting insulin and HOMA-IR in the SD rats, and effectively decreased blood glucose and hemoglobin A1c in db/db mice. C. longa decreased serum free fatty acid (FFA) level in the SD rats. FFA in db/db mice fed C. longa tended to decrease. C. longa significantly decreased serum triglyceride level. Our results collectively represent that C. longa prevented fat accumulation and insulin resistance in both normal weight SD rats and type 2 diabetic obese db/db mice fed a moderate high fat diet.

Anti-Obesitic Effects of Cathepsin S Inhibitory Fraction Derived from Paecilomyces tenuipes in Mice Fed a High-Fat Diet (고지방식이를 섭취한 쥐에서 눈꽃동충하초 유래 cathepsin S 저해 분획의 비만 억제 효과)

  • In this study, cathepsin S (CTSS) inhibitory fraction was isolated from Paecilomyces tenuipes and anti-obesitic effects of the fraction were evaluated in mice, fed a high-fat diet. Hot water extract of P. tenuipes (DHW) was divided into 2 fractions, water eluate fraction (DHP1) and methanol eluate fraction (DHP2) using Diaion HP-20. $IC_{50}$ values for DHW, DHP1 and DHP2 against CTSS were 108.7, 890.3 and 2.3 ${\mu}g$/mL, respectively. To evaluate anti-obesitic effects of the fractions, each fraction was administrated orally to C57BL/6 mice for 4 weeks with a high-fat diet. DHP2 had the highest inhibitory effect on CTSS activity, causing serious reduction in weight gain, a reduction in the amount of adipose tissue and in serum lipids levels. These results suggest that the inhibition of CTSS by compounds derived from P. tenuipes may be effective in preventing and in ameliorating obesity.

Resveratrol Ameliorates High-fat-induced Metabolic Complications by Changing the Expression of Inflammasome Markers and Macrophage M1 and M2 Markers in Obese Mice (라스베라트롤 투여가 고지방식이 비만쥐의 지방조직에서의 inflammasome과 대식세포 마커에 미치는 영향)

  • The purpose of this study was to investigate the effects of resveratrol supplementation on inflammasome, inflammation, and macrophage markers in subcutaneous adipose tissue of high-fat-diet-induced obese mice. C57BL/6 mice were randomly assigned to three groups: normal diet control (NC; n=10), high-fat diet control (HC; n=10), or high fat with resveratrol (HRE; n=10) group. The mice were fed a high-fat diet (60% of calories from fat) or normal diet (18% of calories from fat). Resveratrol dissolved in a 0.1ml solution of dimethyl sulfoxide was supplemented orally at 25 mg/kg body weight. After 15 weeks, the body weight was significantly higher in the high-fat diet group than in the normal diet group. The inflammasome markers NLRP3, ASC, and caspase1 were significantly lower in the HRE group than in the HC group. The levels of an inflammation marker, IL-18, were also significantly lower in the HRE group than in the NC and HC groups. The levels of macrophage markers F480 and CD86 were significantly lower in the HRE group than in the HC group. The levels of the M2 macrophage marker CD206 were significantly decreased in the HC and HRE groups. Resveratrol had a positive effect on ameliorating the complications of high fat diet-induced obesity by reducing inflammasome and M1 macrophage gene expressions. However, resveratrol supplementation did not reduce inflammation gene expression.

Effects of Phyto-Extract Mixture on Adiposity and Serum Lipid Levels in Obese Mice Induced by High Fat Diet (고지방식이로 유도된 비만쥐에서 식물혼합 추출물의 체지방 형성 및 혈청 지질에 미치는 영향)

  • In this study, we investigated the inhibition of phyto-extract mixture (PEM) in several digestive enzymes ($\alpha$-amylase, $\alpha$-glucosidase and lipase) for anti-obesity. The current study also examined the effects of PEM on adiposity and serum lipid levels in obese mice fed with high fat diet. ICR male mice weighing $33{\pm}1.1\;g$ were randomly divided into three groups, one normal diet group (control, ND group) and two high fat diet groups with or without PEM supplement (HFD group and PEM group). The mice were fed the PEM experimental for 6 weeks and then they were sacrificed. The results showed that the final weight, weight gain, food efficiency ratio and body fat were decreased by the addition of PEM compared to those of HFD group. White adipose tissue weights of epididymal, mesenteric and retroperitoneal areas in the PEM group were reduced to 31.2%, 8.8%, and 37.8%, respectively, compared to the HFD group. The levels of serum triglyceride, total cholesterol, LDL-cholesterol in the PEM group were significantly lower than those of HFD group. The body weight gain and food efficiency ratio of PEM group were significantly lower compared with those of HFD group. From the above results, the PEM may be effective material for anti-obesity through reducing serum triglyceride and body fats as well as decreasing body weight.

Antioxidant Enzyme Activity and Anti-Adipogenic Effects of (-)-Epigallocatechin-3-Gallate in 3T3-L1 Cells ((-)-Epigallocatechin-3-Gallate의 3T3-L1 세포에서 항산화 효소 활성 및 지방세포 분화 억제 효과)

  • Obesity contributes to the development of diseases, such as type II diabetes, hypertension, coronary heart disease, and cancer. In addition, oxidative stress caused by reactive oxygen species (ROS) is recognized widely as a contributing factor in the development of chronic diseases. This study was examined the antioxidant and anti-adipogenic activities of epigallocatechin-3-gallate (EGCG) in 3T3-L1 preadipocytes. 3T3-L1 cells were differentiated with or without EGCG for 6 days. The production of glutathione (GSH) and the activities of the antioxidant enzymes, such as glutathione reductase (GR), glutathione peroxidase (GPx), catalase (CAT), and superoxide dismutase (SOD) were measured. EGCG inhibited significantly the lipid accumulation and the expression of adipogenic specific proteins including CCAAT/enhancer binding protein ${\alpha}$ and adipocyte fatty acid binding protein. The production of intracellular ROS was decreased significantly by EGCG in 3T3-L1 cells. EGCG increased the GSH production and the activities of GPx, GR, CAT, and SOD. Moreover, EGCG increased the protein expression of glutamate-cysteine ligase and heme oxygenase-1 in 3T3-L1 cells. These results suggest that EGCG increased the activity and expression of antioxidant enzymes and suppressed the lipid accumulation in 3T3-L1 cells. Therefore, the use of phytochemicals that can maintain the GSH redox balance in adipose tissue could be promising for reducing obesity.

Effects of Phytoplant Diets on Body Weight, Feces Production, Body Fat, and Serum Lipid Levels in High-fat Diet-induced Hyperlipidemic Rats (식물성 식이조성물이 고지혈증을 유발시킨 흰쥐의 체중, 배변량, 체지방 및 혈청 지질농도에 미치는 영향)

  • This study was performed to evaluate the anti-obesity and lipid-lowering effects of phytoplant diets in rats fed with a high-fat/cholesterol diet (HFCD). Experimental diet formulae contained various phytoplants such as brown rice, barley, soybean, germinated brown rice, malt, black bean, sea tangle, and/or dietary fibers including polydextrose, garcinia combogia, glucomannan, ${\small}L$-carnitine, and chitosan. Male Sprague-Dawley rats were fed with a HFCD for 6 weeks and then fed with a HFCD with/without phytoplants for another 6 weeks. Rats fed with phytoplant diets showed lower body weights, liver weights, visceral fat levels, and blood lipid levels compared to those of rats fed with HFCD alone. In addition, rats administered phytoplant diets showed increased daily feces production during the second experimental phase. These results suggest that phytoplant diets improve body weight, feces production, adipose tissue weight, and lipid metabolism.

The +1316 T/T Genotype in the Exon 3 of Uncoupling Protein Gene is Associated with Daily Percent Lay in Korean Native Chicken (한국 재래 닭의 Uncoupling Protein 유전자 Exon 3에서의 +1316 T/T 유전자형이 산란율에 미치는 효과 분석)

  • Uncoupling protein(UCP) is expressed exclusively in brown adipose tissue(BAT). It is blown to uncouple phosphorylation from oxidation and hence to be involved in energy metabolism and heat production, especially under cold exposure. In the present study, we identified single nucleotide polymorphism(SNP) in exon 3 of avUCP gene in Korean native chicken(KNC) population. It was detected a SNP T+1316C in exon 3 of avUCP gene by sequence analysis in KNC population. For PCR-RFLP analysis of the SNP T+1316C, used by AP III restriction enzyme. The result of PCR-RFLP analysis showed that allele T has two fragments of 255 bp and 86 bp, and allele C has only one fragment of 341 bp. The genotype frequencies were TT type, 0.7875; TC type, 0.1875 and CC type, 0.025; and the frequencies of allele T and C were 0.881 and 0.119, respectively in KNC population. Next study was conducted to investigate the effect of the SNP in avUCP gene on economic traits in the KNC population. The TT genotype had a significant higher daily percent lay(84.61) than CC genotype(p<0.05) in KNC population. This study may be useful for genetic studies of avCUP gene and selection on daily percent lay of KNC.
