Evaluation of Pharmacological Effect of Soybean Trypsin Inhibitor-Chondroitin Sulfate Conjugates (Soybean Trypsin Inhibitor와 황산 콘드로이친 포합체의 약리 효과 평가)

  • Kunitz-type soybean trypsin inhibitor (SBTI) and chondroitin sulfate (A, and C type) were conjugated using sodium periodate method. And the physicochemical, pharmacokinetic properties and immunogenecity of the conjugates (Chon-A-SBTI or Chon-C-SBTI) were characterized. We expected the conjugation using chondroitin sulfate to reduce the immunogenecity and to improve the pharmacological effect. As the results, the mean molecular weight of the conjugate highly increased. After I.V. injection of the radiolabeled conjugates or native SBTI into mice, it was found that native SBTI showed rapid elimination from plasma, whereas Chon-A-SBTI and Chon-C-SBTI were slowly eliminated. Organ distribution of the two agents at 30 min after I.V. injection was different : Chon-A-SBTI or Chon-C-SBTI accumulated to a large extent in the liver (13% in Chon-A-SBTI and 16% in Chon-C-SBTI), whereas native SBTI was taken up more rapidly by the kidney (107% dose/g of tissue) and excreated into the urine (26%). In addition we evaluated the therapeutic value of the conjugates by using the sublethal septic shock model caused by pseudomonal elastase and tested the immunogenecity by passive cutaneous anaphylaxis shock (PCA). The conjugates were more effective than native SBTI against pseudomonal elastase induced septic shock in guinea pig. In case of the conjugates, the pharmacological and therapeutic effect lasted over 3 hours long. In immunogenecity test, both of the conjugates showed the reduction of their immunogenecity, especially Chon-A-SBTI looked most effective.

  • A stereoselective synthesis of 3-phenoxybenzyl (${\pm}$)-cis and trans-3-(2,2-dichlorovinyl)-2,2-dimethyl-1-cyclopropanecarboxylic acid starting from readily available 2-methyl-3-buten-2-ol($\underline{2}$) is described. Allylic rearrangement of 2-methyl-3-buten-2-ol, in the presence of acetic acid and acetic anhydride gave 3-methyl-2-butenyl acetate($\underline{3}$). The [3,3] sigmatropic rearrangement of the allyl acetate($\underline{3}$), as the silylketene acetal, produced the ${\gamma},\;{\delta}$-unsaturated acid($\underline{4}$). Treatment of 3,3-dimethyl-4-pentenoic acid($\underline{4}$) with SOCl2 followed by esterification with 3-phenoxybenzyl alcohol yielded 3, 3-dimethyl-4-pentenoic acid ester($\underline{5}$). Addition of carbon tetrachloride to the olefin ester($\underline{6}$) furnished 4,6,6,6-tetrachloro-3,3-dimethylhexanoic acid ester ($\underline{7}$). Cyclization with potassium t-butoxide and elimination of hydrogen chloride afforded 3-phenoxybenzyl (${\pm}$) cis- and trans-3-(2,2-dichlorovinyl)-2,2-dimethyl-1-cyclopropanecarboxylic acid.

Prognostic factors for outcome of surgical treatment in medication-related osteonecrosis of the jaw

  • Objectives: The number of patients with medication-related osteonecrosis of the jaw (MRONJ) is increasing, but treatment remains controversial. Published papers and systematic reviews have suggested that surgical treatment is effective in patients with MRONJ. The purpose of this study was to determine whether preoperative University of Connecticut Osteonecrosis Numerical Scale (UCONNS), other serologic biomarkers, and size of necrosis are prognostic factors for outcome of surgical treatment in MRONJ. Materials and Methods: From January 2008 to December 2016, 65 patients diagnosed with MRONJ at the Department of Oral and Maxillofacial Surgery in College of Dentistry, Dankook University who required hospitalization and surgical treatment were investigated. Patient information, systemic factors, and UCONNS were investigated. In addition, several serologic values were examined through blood tests one week before surgery. The size of osteolysis was measured by panoramic view and cone-beam computed tomography in all patients. With this information, multivariate logistic regression analysis with backward elimination was used to examine factors affecting postoperative outcome. Results: In multivariate logistic analysis, higher UCONNS, higher C-reactive protein (CRP), larger size of osteolysis, and lower serum alkaline phosphate were associated with higher incidence of incomplete recovery after operation. This shows that UCONNS, CRP, serum alkaline phosphate, and size of osteolysis were statistically significant as factors for predicting postoperative prognosis. Conclusion: This study demonstrated that CRP, UCONNS, serum alkaline phosphate, and size of osteolysis were statistically significant factors in predicting the prognosis of surgical outcome of MRONJ. Among these factors, UCONNS can predict the prognosis of MRONJ surgery as a scale that includes various influencing factors, and UCONNS should be used first as a predictor. More aggressive surgical treatment and more definite surgical margins are needed when the prognosis is poor.

  • As the demand of digital image processing devices has been rapidly increased recently, the excellent image quality is required. However, degradation can be occurred with multiple causes during transmission and processing process. Therefore, the needs to eliminate the noise are increased and the noise elimination technology became the major study area. Therefore, image restoration algorithm was suggested to apply the filter differently by edge and non-edge areas, using modified edge detection with preprocessing process so as to relieve the effect of additive white Gaussian noise(AWGN) which is added in the image, in this article. In addition, it was compared with the existing methods using peak signal to noise ratio(PSNR) as the objective determination standard of the improvement effect.

  • A new high efficiency phase shifted full bridge (PSFB) converter for sustaining power module of plasma display panel (PDP) is proposed in this paper .The proposed converter employs the rectifier of voltage doubler type without output inductor. Since it has no output inductor, the voltage stresses of the secondary rectifier diodes can be clamped at the level of the output voltage. Therefore, no dissipative resistor-capacitor (RC) snubber for rectifier diodes is needed and a high efficiency as well as low noise cutout voltage can be realized. In addition, due to elimination of the large output inductor, it features a simple structure, lower cost, less mass, and lighter weight. Furthermore, the proposed converter has wide zero voltage switching (ZVS ) ranges with low current stresses of the primary switches. Also the resonance between the leakage inductor of the transformer and the capacitor of the voltage doubler cell makes the current stresses of the primary switches and rectifier diodes reduced. In this paper, the operational principles, analysis of the proposed converter, and the experimental results are presented.

  • This paper introduces the generalized finite element method with global-local enrichment functions using the preconditioned conjugate gradient method. The proposed methodology is able to generate enrichment functions for problems where limited a-priori knowledge on the solution is available and to utilize a preconditioner and initial guess of good quality with only small addition of computational cost. Thus, it is very effective to analyze problems where a complex behavior is locally exhibited. Several numerical experiments are performed to confirm its effectiveness and show that it is computationally more efficient than the analysis utilizing direct solvers such as Gauss elimination method.

  • In this study, the flow characteristics of an injection nozzle installed in a high-pressure holder for improving productivity were determined. The inlet velocity, nozzle inflow angle, and nozzle outlet diameter were selected as design factors having an influence on the flow characteristics, and numerical analysis was conducted for these factors. As the inlet velocity is high and the nozzle outlet diameter is small, the pressure and velocity of the injected flow are high. In the case of the nozzle inflow angle, the variation of flow characteristics according to angle was slight, but the highest pressure and velocity were found at $15^{\circ}$. In addition, the possibility of chip elimination by the injected flow was analyzed on the basis of the numerical results.

  • The endoplasmic reticulum (ER) is an important intracellular organelle for folding and maturation of newly synthesized transmembrane and secretory proteins. The ER provides stringent quality control systems to ensure that only correctly folded proteins exit the ER and unfolded or misfolded proteins are retained and ultimately degraded. The ER has evolved stress response both signaling pathways the unfolded protein response (UPR) to cope with the accmulation of unfolded or misfolded proteins and ER overload response (EOR). Accumulating evidence suggests that, in addition to responsibility for protein processing, ER is also an important signaling compartment and a sensor of cellular stress. In this respect, production of bio-functional recombinant-proteins requires efficient functioning of the ER secretory pathway in host cells. This review briefly summarizes our understanding of the ER signaling developed in the recent years to help of the secretion capacities of recombinant cells.

Sintering Behavior and Mechanical Property of B4C Ceramics Fabricated by Spark Plasma Sintering (방전플라즈마 소결법에 의한 탄화 붕소 세라믹스의 소결 거동 및 기계적 특성)

  • [ $B_4C$ ] ceramics were fabricated by spark plasma sintering process and their sintering behavior, microstructure and mechanical properties were evaluated. Relative density of $B_4C$ ceramics could be achieved by spark plasma sintering method reached as high as 99% at lower temperature than conventional sintering method, in addition, without any sintering additives. The mechanical properties of $B_4C$ ceramics could be improved by the heat treatment at $1300^{\circ}C$ during sintering process which can be removed $B_2O_3$ phase from a $B_4C$ powder surface. This improvement results from the formation of a fine and homogeneous microstructure because the grain coarsening was suppressed by the elimination of $B_2O_3$ phase. Particularly, mechanical properties of the specimen experienced the $B_2O_3$ removing process improved over 30% compared with the specimen without that process.

A study on the prices trends of landscape woody plants(1985-1994) in Korea (조경수목 가격의 10년간(1985-1994) 변화에 관한 연구)

  • This study was conduced out to analyze the price trends of landscape woody plants during the past ten years. The method of this study was applied to secendary data analysis and the data were collected from "Government Specified Monthly Price Book" and "Monthly Price Book" The results of this study were as follows : 1. The annual average increasing rate of the GSP price was 3.9% while the KPRC price was 8.9% for the past ten years. The ratio of the KPRC price by GSP price was 94.7% in 1985 and it increased by 147.3% in 1994. This increase indicates a large price margin between two prices. The GSP price should be readjusted to a realistic level. 2. For the same period, the prices of native Korean tree were raised by 3.8% and 9.5% in annual average rate. Meanwhile, the exotic tree prices were raised by 3.6% for and 7.8% for in annual average rate. The prices of native Korean tree were raised more than those the exotic tree. 3. The annual average increasing rates of the twenty for species prices which were high ranked among all species prices were 12% for and 21.5% for . These top twenty species drove to markup trend of tree prices. 4. The annual average price increasing rate of major species which are used frequently for landscaping was higher than total increasing rate of all species. This result shows that the prices of the major species markup more than others. 5. From 1985 to 1994, the prices of 29 new species were listed on "The government specified monthly price book" and the prices of 40 species were newly listed on "Monthly price book". On the contrary, 3 species from "The government specified Monthly Book" and 10 species from "Monthly Price Book" were eliminated. The number of new listed on the native Korean species were twice as many as the exotic species. In addition, there is a need to study and explain reaon of the elimination.
