Anti-inflammatory effects of Coptidis Rhizoma in chronic cold stress-exposed mice (만성 저온 스트레스 동물모델에서의 황련(黃連)의 항염증 효능 연구)

  • Objectives : The aim of this study was to investigate whether the extract of Coptidis Rhizoma inhibits inflammation in chronic cold stress (CCS)-exposed mice or not. Methods : Coptidis Rhizoma extract (CRE) was made by reflux with distilled water. Male ICR mice (7 weeks old) were divided randomly into 5 groups: (1) control, (2) CCS, (3) CCS+CRE 100 mg/kg, (4) CCS+CRE 300 mg/kg, (5) CCS+CRE 1,000 mg/kg groups. Mice were orally administered once a day for 14 days starting from 1 day before CCS. Group (2)-(5) were exposed to CCS conditions that maintained at $4^{\circ}C$ for 2 h once a day for 14 days. The levels of serum cortisol and hypothalamic prostaglandin E1 (PGE1) and PGE2 were measured by enzyme-linked immunosorbent assay kit. The expression levels of several pro-inflammatory factors like heat shock protein 70 (HSP70), c-fos, and nuclear factor kappa-light-chain-enhancer of activated B cells (NF-kB) were measured by western blot analysis in mouse hypothalamus. Results : Oral administration of CRE 1,000 mg/kg significantly suppressed the increase of serum cortisol levels in mice exposed to CCS. CCS-exposed mice had significantly increased the expression of HSP70, c-fos, and NF-kB in hypothalamus, while CRE treatment significantly attenuated the elevation of these pro-inflammatory factors. The ratio of PGE2/PGE1 was also higher in CCS-exposed mice than control group. CRE treatment significantly reduced the increase of PGE2/PGE1 ratio induced by CCS. Conclusion : These findings suggest that Coptidis Rhizoma may work as a potential agent to modulate inflammatory responses under the condition of cold adaptation formed by CCS.

Whole-genome Transcriptional Responses to Hypoxia in Respiration-proficient and Respiration-deficient Yeasts: Implication of the Mitochondrial Respiratory Chain in Oxygen-regulated Gene Expression (저산소 환경에 대한 전체 유전자 발현 반응에서 미토콘드리아 호흡계의 연루)

  • Cells sense, respond, and adapt to a low oxygen environment called hypoxia, which is widely involved in a variety of human diseases. Adaptation to low oxygen concentrations includes gene expression changes by inducing hypoxic genes and reducing aerobic genes. Recently, the mitochondrial respiratory chain has been implicated in the control of these oxygen-regulated genes when cells experience hypoxia. In order to obtain an insight into an effect of the mitochondrial respiratory chain on cellular response to hyxpoxia, we here examined whole genome transcript signatures of respiration-proficient and respiration-deficient budding yeasts exposed to hypoxia using DNA microarrays. By comparing whole transcriptomes to hypoxia in respiration-proficient and respiration-deficient yeasts, we found that there are several classes of oxygen-regulated genes. Some of them require the mitochondrial respiratory chain for their expression under hypoxia while others do not. We found that the majority of hypoxic genes and aerobic genes need the mitochondrial respiratory chain for their expression under hypoxia. However, we also found that there are some hypoxic and aerobic genes whose expression under hypoxia is independent of the mitochondrial respiratory chain. These results indicate a key involvement of the mitochondrial respiratory chain in oxygen-regulated gene expression and multiple mechanisms for controlling oxygen-regulated gene expression. In addition, we provided gene ontology analyses and computational promoter analyses for hypoxic genes identified in the study. Together with differentially regulated genes under hypoxia, these post-analysis data will be useful resources for understanding the biology of response to hypoxia.

Analysis of biodiversity change trend on urban development project - Focusing on terrestrial species in Environmental Impact Assessment - (도시의 개발 사업에 따른 생물다양성 변화 추세 분석 - 환경영향평가의 육상 동물종을 중심으로 -)

  • The Environmental Impact Assessment (EIA) plays a pivotal role in predicting the potential environmental impacts of proposed developments and planning appropriate mitigation measures to minimize effects on species. However, as concerns over biodiversity loss rise, there's ongoing debate about the efficacy of these mitigation plans. In this study, we utilized data from EIAs and post-environmental impact surveys to understand the trends in biodiversity during construction and operation phases. By examining 30 urban development projects, we categorized species richness indices of mammals, birds, amphibians, and reptiles into pre-construction, during construction, and post-construction operational stages. The biodiversity trends were analyzed based on the rate of change in these indices. The results revealed three distinct biodiversity change patterns: (A) An initial increase in biodiversity indices post-development, followed by a gradual decline over time; (B) a sustained increase in biodiversity as a result of mitigation measures; and (C) a continuous decline in biodiversity post-development. Furthermore, all species exhibited a higher rate of biodiversity decline during the construction phase compared to the operational phase, with mammals showing the most significant rate of change. Notably, the biodiversity change rate during operation was generally lower than during construction. In particular, mammals seemed to be most influenced by mitigation measures, displaying the smallest rate of change. This study provides empirical evidence on the efficacy of mitigation measures and deliberates on ways to enhance their effectiveness in minimizing the adverse impacts of urban development on biodiversity. These findings can serve as foundational data for addressing terrestrial biodiversity reduction.

Improvement Effect of Stachys sieboldii MIQ. According to Mixing Ratio of Calcium on Memory Impairment in Scopolamine-induced Dementia Rats (칼슘 배합 비율에 따른 초석잠의 scopolamine 치매유도 흰쥐에 대한 기억손상 개선 효과)

  • The aim of this study was to investigate the anti-amnesic effect of Stachys sieboldii MIQ. according to the mixing ratio of calcium on scopolamine-induced learning and memory impairment, in vivo. At the end of the adaptation period, SD rats were divided into a normal group (N), a control group (C: scopolamine), a positive control group (PC: scopolamine + tacrine), and a sample group (S: scopolamine + Stachys sieboldii MIQ., 1CS: scopolamine + low calcium-mixed Stachys sieboldii MIQ., 5CS: scopolamine + high calcium-mixed Stachys sieboldii MIQ.), and were tested with learning and memory tests. The C and CS groups were found to have a decreased scopolamine-induced memory deficit in the Y-maze and water maze tests. Brain tissue analysis showed that the CS group decreased acetylcholinesterase (AChE) activity and increased acetylcholine (Ach) content, both of which are indicative of neuronal cell activity. From a light microscopy study, the nucleus of neurons in the hippocampus of the brain was more shrunken or condensed in the C group compared to the CS group. In the CS group, the damage to the neurons in the hippocampus of the brain was suppressed. These results suggest that Stachys sieboldii MIQ. according to the mixing ratio of calcium provides a significant anti-amnesic effect against scopolamine-induced cholinergic system deficits and cognitive impairment.

A Study on Dress Design with Application of Spiral Form (나선형(螺旋形) 모티브를 응용(應用)한 복식(服飾) 디자인 연구(硏究))

  • The spiral form, which comes from the organic form of natural phenomenon such as growth of creatures, has been used as a factor of formative shape in various fields of art until now. In conjunction herewith, this study intends to discover and express the life force and the formative beauty of natural substances with the organic spiral forms into clothing design, using various creating methods and materials such as Korean traditional paper. The natural substances with the spiral structure, such as seashells, land snails, passion flower, curled flower, growing chart of plants, Impatiens textori was used as subjects of the designs. The seven pieces of work were completed with attempts to develop aesthetic forms through the presentation techniques and methods via restructuring process of simplification, partial transformation and consolidation. Through such process the conclusion of this study is as follows: First, the nature, with unlimited possibilities, could be subject of human formative activities, leading to the creative world of formative art for designers. Second, adaptation of the spiral organic forms of nature into the contemporary clothing designs proved the motif as a source of inspiration of diverse subject, in recognition with its innate formative beauty as well as external shape. Third, design expressions via restructuring process of simplification, partial transformation and consolidation with designer's subjective point of view were adequate for the creations of contemporary fashion designs. Fourth, the Korean traditional paper, as a fine material for various shape according to the handling method, could be used appropriately in the contemporary clothing designs, expressing our aesthetic senses. Fifth and finally, expansion of the realm of formative expression of clothing through the development of possibilities of expression in contemporary clothing would enhance the creative possibilities of clothing design as formative art. In conclusion, the expression of clothing design as formative art was developed on the emphasis of re-creation of natural objects of the organic spiral form. For the future study, the applications of spiral form into everyday clothing designs, consolidating artistic senses and practical senses, are expected as opportunity of proposing developmental possibilities of the contemporary clothing designs.

The Effects of Treadmill Training on Neurotrophins and Immediately Early Protein in Obese Rats (트레드밀 트레이닝이 비만 쥐의 neurotrophins와 초기발현 단백질에 미치는 영향)

  • The purpose of this study was to investigate the biological effect of obesity-induced oxidative damage on neurogenesis and early protein expression. Obesity was induced I thirty 4-week old male Sprague-Dawley rats through a high fat diet for 15 weeks. After one week of environmental adaptation, the rats were divided into 2 groups: high fat diet sedentary group (HDS, n=15) and high fat diet training group (HDT, n=15). Exercise training was performed 5 times a week for 8 weeks, with mild-intensity treadmill running for weeks 1-4 and moderate-intensity treadmill running for weeks 5-8. After the 8 week training period, we analyzed lipid profiles, serum 8-hydroxyguanosine (8-OHdG), liver tissue malondialdehyde (MDA) related to oxidative damage factors, nerve growth factor (NGF), brain derived neurotrophic factor (BDNF), c-fos, c-jun, and extracellular signal regulated kinase (Erk) in the hippocampus. The results of this study are as follows. There were differences between HDS and HDT in triglyceride (TG) and total cholesterol (TC) (p<0.05). In high density lipoprotein (HDL-c), the HDT was higher than HDS after treadmill training (p<0.05). In 8-OHdG, the HDT was lower than HDS after treadmill training (p<0.05). Genetic expressions of c-jun, BDNF and MDA in the HDT were higher than in the HDS after treadmill training in hippocampus (p<0.05). Therefore, we conclude that 8 weeks of treadmill training can improve imbalanced lipid profiles, reduce oxidative damage, and activate neurogenesis in obese rats.

A unique genetic lineage at the southern coast of China in the agar-producing Gracilaria vermiculophylla (Gracilariales, Florideophyceae)

  • Ocean warming can have significant negative impacts on population genetic diversity, local endemism and geographical distribution of a wide range of marine organisms. Thus, the identification of conservation units with high risk of extinction becomes an imperative task to assess, monitor, and manage marine biodiversity for policy-makers. Here, we surveyed population structure and genetic variation of the red seaweed Gracilaria vermiculophylla along the coast of China using genome-based amplified fragment length polymorphism (AFLP) scanning. Regardless of analysis methods used, AFLP consistently revealed a south to north genetic isolation. Populations at the southern coast of China showed unique genetic variation and much greater allelic richness, heterozygosity, and average genetic diversity than the northern. In particular, we identified a geographical barrier that may hinder genetic exchange between the two lineages. Consequently, the characterized genetic lineage at the southern coast of China likely resulted from the interplay of post-glacial persistence of ancestral diversity, geographical isolation and local adaptation. In particular, the southern populations are indispensable components to explore evolutionary genetics and historical biogeography of G. vermiculophylla in the northwestern Pacific, and the unique diversity also has important conservation value in terms of projected climate warming.

Analysis of Ecological Variation after Creation of the Eco-pond (생태연못 조성공법 적용후의 자연생태 변화분석)

  • The purpose of this study is to evaluate the creation techniques of eco-pond, one of biotopes to promote biodiversity in urban residence area. Investigation were classified out plant, mammals, amphibia, reptiles, birds, fishes and insects. The results were summarized as follows: Around the eco-pond shows simple vegetation structurs, consisted of Pinus densoflora S et Z. and Robinia pseudoacacia under competition. In case of shrub, consisted of 4 species but plant growing appearence diversely by seasons. The evaluation of vegetation of eco-pond, there are found 4 species of aquatic plants. Inside the revetment of pond, Echinochloa crus-galli, Persicaria hydropiper, Digiaria sanguinalis, Cyperus microiria and Bidens frondosa L. are mainly distributed. Near the revetment, Trifolium repens L. and Digiaria sanguinalis are prevailed. And in its background, Erigeron canadensis, Erigeron annuus and vines are begins to make their appearances. When evaluation animals in eco-pond and contrast plot, it show simple species and numbers of mammals. It seemed to be resulted from its isolation and outside intervention by users In eco-pond, Pica pica and Streptopelia orientalis are mainly found and in contrast plot of Columba livia, which are so strong adaptation to city life environment. In case of amphibia and reptiles, none is observed in contrast plot, but in ecological pond, Rana nigromaculata and Hyla japonica are constantly observed. In case of insects, more species are found in eco-pond than contrast plot. And in eco-pond, more dragonflies are visibly increased one year after its construction. In floral zone inside of pond revetment, grasshopper and Locusta migratoria are frequently observed. In case of butterflies, they are mainly found in log fence and willow(salix) around eco-pond. In case of fishes inside of eco-pond, the species and its density are remarkable increased one year after the construction. With above evaluation results, we have identify the increase effect of biodiversity after construction of the eco-pond.

  • Musculoskeletal neck dysfunction syndromes are common in outpatient musculoskeletal pain practice. The underlying musculoskeletal and neurologic causes of pain are variable. In the management of these patients, it is important to accurately identify and treat these pain generators to optimize patient outcome. It is the purpose of this review to discuss three main categories of functional anatomy, the role of superficial/deep muscular system and the scientific evidence for optimal physical therapy intervention for cervical dysfunction. Specifically there is evidence of lowered microcirculation in the upper trapezius muscle, morphological signs of disturbed mitochondrial function which appears to be limited to type I fibers and an increased cross-sectional area of type I muscle fibers despite a lower capillary to fiber area ratio. In acute neck pain syndrome, changes in muscle activity of painful muscles may result from segmental and supraspinal inhibitory effects. Muscle activation is closely related to the control of joint movements and postures and it is difficult to separate the influence of the two components. Both the altered muscle recruitment patterns and altered kinematics appear to be a poor adaptation for pain of the head - neck region, as they are likely to result in increased compressive loading in the cervical spine, affecting muscles, articular structures such as zygapophyseal joints, connective tissues and neural tissues which are all peripheral generators of referred pain. The rectus capitus posterior minor muscle shows that it is one of the most important muscles of the suboccipital region. In this article, i reviewed the anatomy, neurophysiology, function and dysfunction as well as the treatment of cervical dysfunction.

Freezing Injury of Evergreen Broad-Leaved Trees in Warm-Temperature in the Southern Region in Korea -A Case of Machilus Thunbergii Siebold & Zucc. on the Jungma-dong, Gwangyang-city, Jeollanam Province- (한국 남부지방 난온대성 상록활엽수의 동해피해 -전라남도 광양시 중마동의 후박나무를 중심으로-)

  • This study was carried out to clarify the climatic factors of the freezing injury for the judgement on the adaptation areas of evergreen broad-leaved trees. We surveyed and analyzed the climatic factors of the freezing injury to Machilus thunbergii Siebold & Zucc. on the streets with the analyzation of planting grounds, soil conditions and the surrounding buildings. This study showed that only the minimum air temperature factor out of the other climate elements, which were the annual precipitation; the average annual temperature; the average monthly temperature of january; the average monthly minimum temperature of January; the average temperature of the coldest month; the warmth index and the coldness index, was matched up with the previous theories and reports on the freezing damages on the evergreen broad leaved trees and Machilus thunbergii Siebold & Zucc. The freezing injury of Machilus thunbergii Siebold & Zucc was occurred when the mean minimum temperature of the coldest month(TMC) in winter season fell down below $-4.1^{\circ}C$ and the temperature fell down below $-9.2^{\circ}C$. The freezing damage on Machilus thunbergii Siebold & Zucc surrounded by high buildings were less than those surrounded by low buildings or at non buildings.
