Caenorhabditis elegans를 이용한 phenol류의 독성 연구 (Toxicity of Phenols to the Nematode Caenorhabditis elegans)

  • Caenorhabditis elegans(C. elegans) is a free-living soil nematode that commonly used as a biological model and recently, much work has been done using C. elegans as a toxicity model. To evaluate the acute toxicity of phenols to C. elegans, worms were subsequently exposed to nine different xenobiotics. This study described lethal toxicity, reproductive toxicity and movement inhibition using 2-propylphenol, 4-propylphenol, 2-tert-butylphenol, 3-tert-butylphenol, 4-tert-butylphenol, 2-phenylphenol, 4-phenylphenol, nonylphenol and 4-dodecylphenol to C. elegans for 24 hr or 72 hr. We found that phenols used in this study were very toxic to C. elegans. The order of lethal toxicity, reproductive toxicity and movement inhibition is as follows. 4-propylphenol > 2-phenylphenol > 2-tert-butylphenol > 2-propylphenol > nonylphenol > B-tert-butylphenol > 4-dodefylphenol > 4-tert-butylphenol > 4-phenylphenol.

털목이버섯의 독성(毒性)에 관한 연구(硏究) (Studies on Toxic Components of Auricularia polytricha)

  • To screen biologically active components of the higher fungi of Korea, the dried carpophores of Auricularia polytricha were extracted with water. The extract was examined for acute toxicity in ICR mice. A low molecular weight toxin of this fungus was purified by acetone precipitation followed by cellulose, silica gel and LH-20 Sephadex column chromatography. Major symptoms of this toxin were eye extrusion, hair erection, trembling of head, paralysis, rapid running or moving before death and depression of respiration. The median lethal doses of the total extract were 1.25 g/kg and 4.18 g/kg by i.p. and p.o. administrations, respectively. The amounts of one mouse lethal unit (MLU) of the total extract and final fraction that killed a 20-g mouse within 30 minutes were 28.5 mg/mouse and 12 mg/mouse, respectively.

ACUTE MAMMALIAN TOXICITY OF O-CHLOROBENZYLIDENE MALONONITRILE(CS)

  • Acute inhalation intoxication of CS (O-chlorobenzylidene malononitrile) occurred among the 192 animals in confined animal cages of farm as the result of prolonged exposure. A total of 8 animals (3 silver foxes, 3 fitches and 2 minks) died in 15 hours after the exposure. Distinct evidences of pulmonary atelectasis were observed as with hepatorenal damages. The lethal toxicity of CS was considered to be due to early severelung damages leading to asphyxia, accompanying acute toxic hepatitis and nephritis.

농약의 노출시간에 따른 급성어독성의 변화 (The changes of acute toxicity of pesticides depending on the exposure time to killifish, Oryzias latipes.)

  • 담수산 어류인 송사리(Oryzias latipes)에 대한 6개 농약의 급성독성을 7일동안 시간의 경과에 따라 측정, toxicity curve를 작성하여 각 농약의 시간의 경과에 따른 급성독성 발현의 현상을 평가하고 48시간 및 96시간 LC50값과 lethal threshold 농도와의 관계를 분석하여 얻은 결과는 다음과 같다. 농약의 toxicity curve를 중심으로 시간의 경과에 따른 급성독성값의 변화를 보면 butachlor가 120시간째, fenobucarb가 48시간째, chlorothalonil이 48시간째 lethalthreshold 농도를 나타내어, 각각 120시간, 48시간만에 급성독성현상이 더이상 시간의 경과에 따라 발현되지 않으나, 나머지 3개의 농약인 alachlor, diazinon, iprobenfos는 노출 7일 이후에도 계속 급성독성현상을 나타냄을 알 수 있었다. 이로써 6개의 농약에 대한 lethal threshold 농도는 butachlor 0.53 mg/l(0.49-0.57 mg/1), alfachlor 1.15 mg/1>, fenobucarb 10.3 mg/1(9.3-11.4 mg/1), diazinon 6.70mg/1>, chlorothalonil 0.085 mg/1(0.082-0.087 mg/1), iprobenfos 3.42 mg/1>이었다. 결국 노출시간 96시간이내에 독성현상이 발현되어 더이상 치사율이 증가하지 않은 농약은 6개중 2종류뿐이며 나머지 4개중 3개에서는 노출후 7일이 경과하여도 계속하여 독성이 발현됨을 알 수 있었다.

백부자-대체 가능 한약재의 계종버섯에 대한 급성독성시험과 안전성등급화 (Acute toxicity test and safety classification for Termitomyces albuminosus containing pharmacologically similar ingredient of Aconitum koreanum)

  • Objectives : Termitomyces albuminosus (Berk.) Heim is one of the famous wild edible mushrooms in the southern part of China. It is known that Termitomyces albuminosus, like Aconitum koreanum used in Korean traditional medicine, contains a kind of cerebroside, termitomycesphin, causing a pharmacologic effect on the neuron system. The pharmacologic effect of Termitomyces albuminosus can be used to possibly replace Aconitum koreanum. However, It needs to be certified as safe before it can be used. Here, a single-oral toxicity test and safety classification was conducted to obtain acute information of the toxicity of dried-Termitomyces albuminosus powder and to secure its safety in clinical applications. Methods : In order to calculate approximate lethal dose(ALD), test substance was orally administered to male and female SD-rat at dose levels of 5,000 and 0 (vehicle control) mg/kg (body weight). Based on the result of this toxicity, also the estimation of safety classification was calculated using the HED-based (human equivalent dose) MOS (margin of safety). Results : There were no mortalities, test substances treatment-related clinical signs, no changes in the body or organ weights, and no gross or histopathological findings at 14 days after treatment with test substance. Thus, the approximate lethal dose of dried-Termitomyces albuminosus powder was considered over 5,000 mg/kg in both female and male mice. Conclusions : Based on the limit dose, 5000 mg/kg, it was estimated that dried-Termitomyces albuminosus powder is classified as "Specified class B" indicating that clinical dose is not limited to patients as safe as food.

Toxicity characteristics of sewage treatment effluents and potential contribution of micropollutant residuals

  • Background: A typical sewage treatment plant is designed for organic and nutrient removal from municipal sewage water and not targeted to eliminate micropollutants such as pesticides, pharmaceuticals, and nano-sized metals which become a big concern for sustainable human and ecological system and are mainly discharged from sewage treatment plant. Therefore, despite contaminant removal by wastewater treatment processes, there are still remaining environmental risks by untreated pollutants in STP (sewage treatment plant) effluents. This study performed aquatic toxicity tests of raw wastewater and treated effluents in two sewage treatment plants to evaluate toxicity reduction by wastewater treatment process and analyze concentration of contaminants to reveal potential toxic factors in STP effluents. Methods: Water samples were collected from each treatment steps of two STPs, and acute and chronic toxicity tests were conducted following USEPA (United States Environmental Protection Agency) and OECD (Organization for Economic Cooperation and Development) guidelines. Endpoints were immobility for mortality and reproduction effect for estrogenicity. Results: Acute $EC_{50}s$ (median effective concentration) of influents for Seungki (SK) and Jungnang (JN) STPs are $54.13{\pm}32.64%$ and $30.38{\pm}24.96%$, respectively, and reduced to $96.49{\pm}7.84%$ and 100%. Acute toxicity reduction was clearly correlated with SS (suspended solids) concentration because of filter feeding characteristics of test organisms. Chronic toxicity tests revealed that lethal effect was reduced and low concentration of influents showed higher number of neonates. However, toxicity reduction was not related to nutrient removal. Fecundity effect positively increased in treated wastewater compared to that in raw wastewater, and no significant differences were observed compared to the control group in JN final effluent implying potential effects of estrogenic compounds in the STP effluents. Conclusions: Conventional wastewater treatment process reduced some organics and nutritional compounds from wastewater, and it results in toxicity reduction in lethal effect and positive reproductive effect but not showing correlation. Unknown estrogenic compounds could be a reason causing the increase of brood size. This study suggests that pharmaceutical residues and nanoparticles in STP effluents are one of the major micropollutants and underline as one of estrogenic effect factors.

미세조류와 패류의 성장에 미치는 Triorganotin의 급성 독성영향 및 만성독성 실험을 위한 Chemostat System의 설계 (The Acute Toxicity Effect of Triotganotin on the Growth of Microalgae and Shellfish and A Design of A Chemostat System for the Chronic Toxicity Experiment)

  • The acute toxicity effect of triorganotin of trioganotin on the growth of microalgae and shellfish was investigated through flask culture. The value of 120 hr-LC$_{50}$ that is the median lethal concentration of TBTO on the shellfish (R. philipinarum) was found to be 6 $\mu$g/L. The acute toxicity effect of TBTO on T. suecica was obviously shown even at the concentration of 0.5 $\mu$g/L, and the effect diminished as the initial cell density increased. The effect also diminished less in the experiment done under aeration than in that done under non-aeration. To design a chemostat system for the test of chronic toxicity, the culture of T. suecica was executed in photobioreactor. In batch culture, the profiles of chlorophyII a and D.C.W. showed the growth of T. suecica very well, and the maximum specific growth rate was estimated to be 0.54 d$^{-1}$. with this value, as a dilution rate in contimuous culture, pH was nicely maintained between 7 and 9 when air was supplied with 3% CO$_{2}$. From all results and the natural environment of clam, a novel chemostat system was invented. Through this system, we can observe each independent toxicity effect of TBTO and plankton and combined toxicity effect as well.

흰쥐에서 GLP 기준에 의한 한약처방 77종의 단회 투여 급성 독성 연구 (Acute toxicity study of 77 herbal formulas according to GLP guidelines in rats)

  • Objectives: This study aimed to investigate the acute oral toxicity of 77 herbal formulas and performed in male and female Sprague-Dawley (SD) rats as per the guidelines mentioned in Ministry of Food and Drug Safety. Methods: Each sex of SD rat were administered a single dose (2000 or 5000 mg/kg) of 77 herbal formulas via oral gavage; the control group received vehicle only. After administration, the mortality, clinical signs, gross findings, and body weight were followed up for 15 days. Results: After administration of 77 herbal formulas, mortality, clinical signs, body weight changes, and gross findings related to the test substances were not observed in both male and female groups. Conclusion: Our results demonstrate the single-dose oral administration of 77 herbal formulas produced no mortality indicating the approximate lethal dose is greater than 2000 or 5000 mg/kg body weight.

포소화약제의 어류급성독성 시험을 통한 환경독성 검증 (The Verify of Environmental Toxicity of Foam Extinguishing Agents by Fish-Acute Toxicity Test)

  • There are various studies on the fire suppression process but the study on second pollution from fire products is not enough yet. Therefor, in this study verify that environmentally-friendly properties($LC_{50}$) of foam extinguishing agent with increases its amount used through with Fish-Acute Toxicity Test using a fish named Misgurnus anguillicaudatus that is appointed by OECD Test Guideline. In conclusion, proven that environmentally friendly properties of the agent of hoseo university through 16 times of LC50 than that of market.

오령산 열수추출물의 단회투여 급성독성 연구 (Acute Toxicity Study on Oryeong-san in Mice)

  • Objectives : Traditional medicine Oryeong-san (ORS) has been prescribed for a long time to treat light fever, thirst, dysuria, and accompanying edema. However, the acute toxicity and safety were not reported. In this study, we evaluated the potent acute toxicity and safety of ORS. Methods : ICR mice were used to evaluate acute toxicity and safety by oral administration of 0, 500, 1,000, 2,000mg/kg of ORS. Mortality, body weight, and clinical symptoms were observed, and organ weight and blood biochemical parameters were analyzed after necropsy. Results : We found no mortality and no toxic or abnormal clinical symptoms by administration of ORS. Comparing with control group, no significant alterations in organ weight and blood biochemical parameters were observed. Conclusions : ORS recognized as safe and non-toxic medicinal material, and median lethal dose considered to be over 2,000 mg/kg in both male and female ICR mice.
