In this study, Artemisia capillaries, which has been used as a folk remedy, was investigated for its antimicrobial activity. First, the Aremisia capillaris was extracted with methanol at room temperature, and fractionation of the methanol extracts from Artemisia capillaris was carried out using petroleum ether, chloroform, and ethyl acetate. Second, the antimicrobial activity of the Artemisia capillaris extracts was determined using a paper disc method and minimum inhibitory concentration of ethyl acetate extracts from Artemisia capillaris against food-borne pathogens and food spoilage bacteria was measured. Finally, the growth inhibition curve was determined using ethyl acetate extracts of Artemisia capillaris against Staphylococcus aureus and Salmonella typhimurium. The ethyl acetate extract of Artemisia capillaris showed strong antimicrobial activity against S. typhimurium at a concentration of 1,000 ppm. The 3,000 ppm of ethyl acetate extract from Artemisia capillaris retarded the growth of S. aureus and S. typhimurium for up to 6 hours.
Membranes prepared from Bacillus cereus KCTC 3674, grown aerobically on a complex medium, oxidized NADH exclusively, whereas deamino-NADH was little oxidized. The respiratory chain-linked NADH oxidase exhibited an apparent $K_m$ value of approximately $65\;{\mu}m$ for NADH. The maximum activity of the NADH oxidase was obtained at about pH 8.5 in the presence of 0.1 M KCl (or NaCl). Respiratory chain inhibitor 2-heptyl-4-hydroxyquinoline-N-oxide (HQNO) inhibited the activity of the NADH oxidase by about 90% at a concentration of $40\;{\mu}m$. Interestingly, rotenone and capsaicin inhibited the activity of the NADH oxidase by about 60% at a concentration of $40\;{\mu}m$ and the activity was also highly sensitive to $Ag^+$.
Acetylcarnitine, a metabilite of carnitine, has been porven to be a potent inhibitor of ethanol oxidation in hepatocytes. It inhibits the activity of alcohol dehydrognase (ADH), but not the microsomal ethanol oxidizing system. which was significatly inhibited by acetylcarnitine at NAD ; acetylcarnitine $\leq$1. the main objectives of his study were to ascertain the interaction between acetylcarnitine and NAD on ADH activity and to elucidate whether different species have different effects. Tehpost-mocrosomal supernatant (PMS) was prepared from normal rat, guinea pig, mouse and broilers by differential centrifugation . Horse and yeast ADH were purchased from the Sigma Chemical Co. Prepared and purchased ADH are used for determination of ADH activity in the presence or absence of carnitine and acetylcar- nitine. Binding studies showed that acetylcarnitine did bind to ADH in a dose realted manner when low NAD ; acetylcar- nitine ratio was provided. It was found that the inhibitionof ADH activity occurred only when NAD concentration was less than the inhibitor concentration . Crystalline and crude ADH preparation from different vertebrate species wer inhibited by acetylcarnitine, whereas the yeast ADH was not affected by acetylcarnitine.
Polysaccharide (PS) was fractionated from hot-water extract of Artemisia iwayomogi Kitamura stems. PS showed considerably higher hydroxyl radical scavenging activity than caffeic acid and glutathione. PS showed lower superoxide anion radical scavenging activity than hydroquinone and ascorbic acid. The scavenging activity of PS on the reactive oxygen species (ROS) induced by human neutrophils with zymosan was determined by the lucigenin-enhanced chemiluminescence assay. The scavenging effect of the PS on ROS as determined by the chemiluminescence assay was about 2-fold stronger than that of ascorbic acid at the same concentration. PS significantly decreased protein carbonyl and malonaldehyde contents in UVB irradiated skin homogenates, which was comparable to glutathione at the same concentration. This result suggested that PS derived from A. iwayomogi Kitamura stems may be a potent candidate as functional compound for the protection on UVB induced skin damage in cosmetics.
The activity of photosystem II in isolated chloroplast form the leaves of Sedum sarmentosum was measured. The photoreduction rate of DCPIP by photosystem II showed the circadian rhythm with a peak at near midday sample for a continuing fine day and at near afternoon between nidday and sunset sample for a continuing cloudy day in summer. The optimum light intensity of photoreduction by photosystem II in the chloroplast preparation was about 5~9$\times$$10^4$ lux. The saturated light intensity was over 9$\times$$10^1$lux. Photosystem II activity was inhibited by even the lowest concentration of lead. When Pi and ATP of the same concentration as Pb were added to the reaction mixture containing Tris buffer lacking of Pi prior to Pb incubation, photosystem II activity was protected from Pb-inhibiting effect by the pretreament of Pi and ATP. It was assumed that Pb inhibiiton was probably due to one, P-depriving by the precipitates of $Pb_3$$($Pb_4$)_2$ in the reaction mixture and the other, partially Pb-combing with Pi groups of the active site of photosystem II.
The antifungal activities of propolis on Cryptococcus neoformans and Candida albicans were evaluated. In microbroth culture assay, the MIC(minimum inhibitory concentration) of propolis for C. neoformans and C. albicans were 2 and 16 mg/ml, respectively. In propolis-included solid medium assay, the MIC of propolis for C. neoformans and C. albicans were 4 and 16 mg/ml, respectively. Propolis showed fungicidal activity against C. neoformans, whereas propolis possesed fungistatic activity against C. albicans. The MFC(minimum fungicidal concentration) for C. neoformans was 8 mg/ml. Cell morphology of C. neoformans was affected by treatment of propolis. In scanning electron microscope, the appearance of cell rupture was observed.
Extracts from sawdust of Pinus densiflora were showed antifungal activity against Lentinula edodes. It was extracted by hot water and then successively extracted by n-hexane, ethyl acetate, and methanol. The yields of the n-hexane-soluble, ethyl acetatesoluble, methanol-soluble and methanol-insoluble fractions of water extracts were 8.2%, 10.6%, 32.0%, and 49.2%, respectively. The ethyl acetate-soluble fraction showed the greatest antifungal activity against L. edodes: 41.5% inhibition at 1,000 ppm. However, there were not significant differences of antifungal activities between n-hexane-soluble fraction and methanol-soluble fraction at a concentration of 1,000 ppm. The hot water extracts showed 23.5% of antifungal activity against L. edodes at a concentration of 1000 ppm. The four antifungal compounds were separated from ethyl acetate fraction by thin layer chromatography.
The butanol extract of paulownia tomentosa stem showed antibacterial activity against staphyl ococcus aureus (SG511, 285 and 503), Streptococcus pyogenes (A308 and A77) and Streptococcus farcium MD8b etc. The most active compound of the extractg was identified to be campneoside I, which had a minimal inhibitory concentration(MIC) of $150{\;}{\mu}g/ml$ against Strptococcus and Staphylococcus species. From such antibacterial activity, the methoxy group of campneoside I was posulated to be the essential element for the antibacterial activity.
As part of our search for less toxic antimicrobial substances from natural resources, naringin was isolated from feels of Citri fructus and then hydrolyzed to naringenin. The antimicrobial activity of naringenin was examined by measuring the minimal inhibitory concentration(MIC) against fourteen species of bacteria. The antimicrobial activity of narngenin in combination with rutin or hesperetin was evaluated by checkerboard method. Among fourteen species tested, the antimicrobial activity of naringenin was the most prominant against Staphylococcus aureus and Shigella boydii showing MIC of $100\;{\mu}g/ml$ for both species. Combinations of naringenin with rutin or hesperetin showed synergism against several species of bacteria, but no antagonism was observed.
The effect of polyamine and Ca2+ on D-glucose-6-phosphate cyclohydrolase activity was studied in Daucus carota root. The enzyme activity was reduced in response to increase in concentration of Ca2+, not the Ca2+-calmodulin complex. The inhibition effect due to Ca2+ was reversed by polyamine, especially remarkable at low concentration of Ca2+. The effect of the Ca2+ on the enzyme seemed to compete with polyamine according to the Lineweaver-Burk plot. The enzyme activity from carrot root protoplast cultured in the prescence of verapamil was higher than that of the control. Such cumulative results suggest that the inhibition by the Ca2+ and enhancement or reversal by polyamine could regulate the biosynthesis of pectin and hemicellulose to some extent.
