Development of Dietary Supplement for the Growth and Exercise Performance Improvement (성장과 운동수행능력 향상에 도움을 주는 식이조성물의 개발)

  • We developed a dietary supplement using natural herbs and nutrients for the growth and exercise performance improvement. It called 50-22 supplement containing distilled extracts of natural herbs to remove bitterness and the addition of Vitamin B1, B6, Ca and xylitol. We investigated on the effect of 10-22 supplementation on the maximal exercise performance, IGF-1 (insulin like growth factor-1) concentration and antioxidant activity in SD rat model. In result, JR-22 supplement group was better than control group about 10% in exercise performance test and increased about 63% of IGF-1 concentration in blood. In addition, the oxidative damage induced by exercise was reduced by JR-22 supplementation. Therefore, we suggested that 50-22 supplementation enhanced effectively exercise performance and IGF-1 concentration and reduced to oxidative stress in muscles. Also, we analysed biochemical factors in blood for the safety of JR-22 supplement. We known that there is no change of blood lactic acid, ammonia, inorganic phosphorous ion and creatine kinase activity. (Korean J Community Nutrition 8(3) : 349∼355, 2003)

Stability of Sweet Potato $\beta$Amylase (I) (고구마 $\beta$아밀라아제의 안정성에 관한 연구 (1))

  • $\beta$-Amylase was purified from sweet potato by acetone fractlonatlon, Sephadex A-50 ion exchange chromatography and Sepgadex G-200 gel chromatographyl The higher enzyme concentration was, the higher heat stability of enzyme became. After 1 hour 30 minute. At 6$0^{\circ}C$ in pH 5, enzyme under concentration of 30$\mu$l/ml lost its activity completely and over the concentration of 100$\mu$g/ml remained 25% of activity. The enzyme was stabilized at range of pH 4~10 and pH stability was increased by glycerol. Five moles of NaCl inhibited completely of the enzyme activity. SDS of 0.05% inhibited the enzyme completely after 12 hours at 37$^{\circ}C$ in pH5. One mole guanidine-HCl and 8M urea inhibited the entire enzyme after 13 hours at 37$^{\circ}C$ in pH 5.

Influence of Garlic and Red Pepper on the Microflora of Kajami Shik-hae

  • Kajami Shik-hae processing consists of the fermentation of low salted Kajami (6% NaCl maximum) coated with a vegetable mixture, composed with cooked millet, red pepper, garlic and ginger. Lactic bacteria are the main component of the microflora. In order to determine their eventual selective role on the microflora the antimicrobial activity of garlic and red pepper was tested with some strain of bacteria and molds isolated from Shik-hae and Shik-hae raw materials. And the influence of their concentration in Kajami Shik-hae on the microflora was also checked. At the concentration of 10% garlic have no inhibitory activity against lactic bacteria but on strains of Bacillus, Micrococcus and Aspergillus niger. At the concentration of 20% red pepper showed a slight inhibitory activity on two strains of Bacillus. These results shows that red pepper and garlic are not only flavoring ingredients but they might play an important role in the control of the microflora growth and composition during Kajami Shik-hae fermentation.

Physical Properties of Nisin-Incorporated Gelatin and Corn Zein Films and Antimicrobial Activity Against Listeria monocytogenes

  • Edible films of gelatin and com zein were prepared by incorporating nisin to the film-forming solutions. Com zein film with nisin of 12,000 IU/ml had an increase of 11.6 MPa in tensile strength compared with the control, whereas gelatin film had a slight increase with the increase of nisin concentration added. Water vapor permeability for both com zein and gelatin films decreased with the increase of nisin concentration, thus providing a better barrier against water. Antimicrobial activity against Listeria monocytogenes increased with the increase of nisin concentration, resulting in 1.4 log cycle reduction for com zein film and 0.6 log cycle reduction for gelatin film at 12,000 IU/ml. These results suggest that incorporation of nisin into com zein and gelatin films improve the physical properties of the films as well as antimicrobial activity against pathogenic bacteria during storage, resulting in extension of the shelf life of food products by providing with antimicrobial edible packaging films.

Accumulation of Selenium and Changes in the Activity of Inulinase and Catalase in the Cells of Kluyveromyces marxianus on Pulsed Electric Field Treatment

  • Pulsed electric field (PEF) of 1Hz, 1.5 kV, and 1ms increased the activities of catalase and inulinase over the whole range of applied Se concentrations compared with the non-treated cultures. A significant effect of selenium concentration (in the range of 5-14 ${\mu}g/ml$) on both intra- and extracellular enzyme activities was noted. At a Se concentration of 10 ${\mu}g/ml$, the activities of intra- and extracellular inulinases and extracellular catalase in the PEF-treated cultures reached the maximum of 71 U/g d.m., 46 U/g d.m., and approx. 8 U/ml, respectively. The maximum activity of intracellular catalase of approx. 6 U/ml (with and without PEF) was recorded at 5 ${\mu}g$ Se/ml. Further increasing of selenium concentration caused a decrease in the activity of the enzymes.

Effects of National Gymnastics and Brain Gymnastics on Frontal Lobe Activity

  • The effects of national gymnastics and brain gymnastics on brain activity were analyzed in 20 healthy university students. The students were assigned to either a national gymnastics group or a brain gymnastics group. Their frontal lobe activity was recorded before and after the exercise, and the resting, attention, and concentration indexes of brain waves were measured. The resting index significantly decreased and concentration index significantly increased(p<.05) after the exercise in both the national and brain gymnastics groups. However, the attention index significantly increased only after brain gymnastics, and, on the contrary, decreased after national gymnastics. These findings suggest that brain gymnastics is effective for improving attention.

A study on the mechanism for reduction of lead-induced toxicity in nervous system by aloe vera (Aloe vera의 연 유도성 신경계 독성 저감 기전에 관한 연구)

  • Effects of water extract of aloe vera on lead-induced neurotoxicity were investigated in sciatic nerve isolated from rat. The mechanism on toxicity reduction by measuring activities of axonal enzymes, metabolism of myo-inositol in nerve, lead concentration in several organs and so on were further examimed. In the lead-treated rats, the transport rate of axonal enzyme, such as acetyl cholinesterase and choline acetyltransferase, was reduced by from 50% to 30% respectively. Reduction in myo-inositol concentration and $Na^+/K^+$ ATPase activity were also observed in sciatic nerve from lead-treated rat. However, the aloe extract administration significantly eliminated the impairment and maintained myo-inositol concentration to about 85% of normal level. Also aloe extract promoted the excretion rate of lead which is accumulated in blood, sciatic nerve and kidney. These results suggest that lead-induced neurotoxicity was significantly reduced by administration of aloe extract and the mechanism might be partly increase in kidney excretion rate of lead and parity normalization of $Na^+/K^+$ ATPase activity which is critical factor in order to keep nerve maintaining normal myo-inositol level.

A study on the whole cell immobilized glucose oxidase from Aspergillus niger

  • Heat treated whole cell of Aspergillus niger containing glucose oxidase-catalase system was entrapped in gelatin matrix crosslinked by glutaral-dehyde. The reaction characteristics of immobilized enzyme was studied in a fludized reactor. Heat treatment enhanced the stability and improved the properties of micellium for the immobilized process. The immobilized enzyme system showed the maximum activity at $35^{\circ}C$ and at pH 5.5. The optimum substrate concentration was 0.04M glucose. The activity of immobilized glucose oxidase was in proportion to the concentration of dissolved oxygen in reaction mixture as other reaction conditions were fixed. It was also demonstrated that the limiting factor for the activity of the immobilized glucose oxidase was the oxygen diffusion resistance which increases proportionally to the glucose concentration.

  • Bacillus subtilis grown on medium containing 0.1% trios fraction, 0.005% sodium deoxycholate, and 0.05% taurocholate respectively was shown to grow as filamentous form and contain 2.3 x 10-5M, 1 x 10-5M, and 2.3${\times}$10-5M of intracellular C-AMP, respectively. The concentration was 3 or 4 times lower than that of the control. But concentrations of extracellula: C-AMP were similar to that of the control. Such decrease in C-AMP concentration was shown to parallel with decrease in autolysin activity indicating 40% and 20% lower activity of Bacillus subtilis grown on medium containing 0.05% and 0. l% triol fraction respectively than that of the control, The activity was also shown to be in inverse proportion with the formation of filament .

Stimulation of Ribosome Activity of Zea mays by Polyamine (Polyamine에 의한 옥수수 Ribosome의 활성 촉진)

  • As a part of the study on the relation between exogenous polyamines and various components necessary for protein biosynthesis in the germinating maize seeds, the effects of the polyamines on protein biosynthesis and irbosome activity were investigated. The protein biosynthesis activity by S-30 containing all components necessary for protein biosynthesis was increased by exogenous polyamines, spermidine, spermine and putrescine. As the concentration of polyamine treated was increased, the optimal Mg2+ concentration of in vitro poly U-dependent protein synthesis system was gradually reduced. However, the optimal Mg2+ concentration of poly U-dependent system containing optimal polyamine was 10 mM regardless of the sort of polyamine. It could be infered that polyamines play an important part in protein biosynthesis in the higher plant, and that the role of polyamines take partially the place of Mg2+ action. The activities of ribosome and S-100 in protein biosynthesis were increased by 46.7% and 17.7% with spermidine, and by 44.1% and 16.2% with spermine, and by 29.1% and 19.3% with putrescine. It could be concluded that the increase of protein biosynthesis by polyamines in mainly owing to the ribosome activation.
