Comparison of Physical Activity Questions in the Korea National Health and Nutrition Examination Surveys (국민건강영양조사 신체활동 설문 문항 비교)

  • Objectives: This study aims to compare the contents of physical activity questions of Korean National Health and Nutrition Examination Survey(KNHANES), and to propose suggestions for improvement. Methods: Questions measuring physical activity prevalence of adults in the KNHANES from KNHANES I(1998) to the first year of KNHANES V(2010) were compared. Results: The questions used in each survey showed some problems in consistency and appropriateness. Major issues identified are as follows. First, the questions in each survey were not consistent, and would be inappropriate to make time series comparisons. Second, the purpose of physical activity was not clear. Third, the examples of physical activities suggested in the questions were not appropriate. Fourth, the intensity of physical activity were not adequately described. Fifth, by excluding walking from the moderate-intensity physical activity category, the prevalence of moderate-intensity physical activity could be underestimated. Sixth, it is difficult to measure the total amount of physical activity performed in one day because answers from the other domains cannot be clearly differentiated. Conclusions: The physical activity questions in the KNHANES questionnaire should be improved so that they are consistent with the purpose of survey, the respondents can easily understand the contents of the questions, and the questions can measure all physical activity performed in the different domains.

Effect of Five Korean Native Taraxacum on Antioxidant Activity and Nitric Oxide Production Inhibitory Activity (국내 자생 민들레 5종의 항산화 활성 및 Nitric Oxide 생성억제 활성)

  • The effect on the antioxidant activity and Nitric Oxide activity production inhibitory activity of Taraxacum has not been known. Therefore, phenolics and flavonoid contents were investigated from the ethanol extracts of five different Taraxacum species. The results showed that, among the five Taraxacum, T. hallaisanensis contains the highest total phenolic and flavonoid contents. When the antioxidant activity was measured by DPPH, $ABTS^+$ and reducing power activity, the free radical scavenging activity of T. hallaisanensis was also the highest among five Taraxacum species. However, measurement by CCK-8 assay in Raw264.7 cells indicated that the extracts of Taraxacum species have no effect on cell viability. Moreover, we also investigated the effect of Taraxacum species on NO scavenging activity in lipopolysaccharide (LPS)-stimulated Raw264.7 cells. The results clearly showed that Taraxacum species inhibited NO production, and the inhibitory effect of T. hallaisanensis was the strongest. The above results suggested that Taraxacum species affected the antioxidant and NO scavenging activity, and among the five species, antioxidant and NO scavenging activity assay of T. hallaisanensis was significantly higher than those of other four Taraxacum species. Therefore, T. hallaisanensis could be used as a potential drug with anti-oxidant and anti-inflammatory effect.

Antioxidative and Antibacterial Activity and Tyrosinase Inhibitory Activity of the Extract and Fractions from Taraxacum coreanum Nakai (흰민들레 에탄올 추출물 및 분획물의 항산화 및 항균활성과 Tyrosinase 저해활성)

  • In this study, we investigated antioxidative activity, antibacterial activity against pathogenic strains including methicillin resistant Staphylococcus aureus (MRSA), and tyrosinase inhibitory activity in 75% ethanol extract of Taraxacum coreanum and its fractions. The total polyphenol and flavonoid contents of the extract were 238.59mg/g and 33.18mg/g and the total polyphenol and flavonoid contents of the ethyl acetate fraction were 427.81mg/g and 148.90mg/g as the highest content of fractions. In DPPH radical scavenging ability, $SC_{50}$ values of the ethyl acetate and butanol fraction were 38.40${\mu}g/ml$ and 82.28 ${\mu}g/ml$, respectively. In antibacterial activity by the disc diffusion assay against S. aureus, S. epidermidis and MRSA, the ethyl acetate fraction showed stronger antibacterial activity than other fractions and the extract. Especially, the ethyl acetate fraction was exhibited effective antibacterial activity against MRSA. In the cytotoxicity measurement by MTT assay, the extract and fractions were exhibited Raw 264.7 cell viabilities of 96.32~143.21% as nontoxic result in concentration of 5~100 ${\mu}g/ml$. As a result, the ethyl acetate fraction of the 75% ethanol extract from T. coreanum could be applicable to functional materials for related fields.

Antioxidant and QR Inductive Activities of Novel Functional Soybean 'Agakong3'

  • In order to evaluate the bioactivity of 'Agakong3', which was newly bred, quinone reductase (QR) inductive activity and antioxidant activity were both assessed. The methanol extract of 'Agakong3' showed a significantly stronger QR inductive activity than other soybeans. The methanol extract of 'Agakong3' also showed a significantly stronger cytotoxicity on hepa1c1c7 than other soybeans. 'Agakong3' exhibited the most potent antioxidant activity in the Trolox equivalent antioxidant capacity (TEAC) assay whereas it showed significantly weak antioxidant in the DPPH assay. In total phenol and flavonoid contents, 'Agakong3' showed the highest contents regarding phenol and flavonoid compounds. Major isoflavones such as daidzein and genistein were quantitified by high performance liquid chromatography. 'Agakong3' also showed the highest total isoflavone contents. Results of correlation analysis showed that there were high correlation coefficients between the contents of isoflavone and TEAC and the contents of isoflavone and QR inductive activity, respectively. These results suggest that 'Agakong3' will be a promising and functional food material.

Nutritional Composition and Antioxidative Activity of Different Parts of Taraxacum coreanum and Taraxacum officinale (흰민들레와 서양민들레의 부위별 영양성분 및 항산화효과)

  • This study compared the nutritional composition and antioxidative activities of different parts (aerial part and root) of Taraxacum coreanum and Taraxacum officinale. To identify the nutrient composition of different parts of Daedelion species, nutrient contents were analyzed. Crude ash, protein, and fat contents of T. coreanum and T. officinale were higher in the aerial part compared to the root. There was no significant difference in crude ash, protein, or carbohydrate content of the aerial part depending on the Daedelion species. Aerial part contents of vitamin A, E, and C were significantly higher in T. coreanum than T. officinale. The K content of the aerial part of T. coreanum and T. officinale was highest, although there was no significant difference among Daedelion species. Contents of oxalic acid, citric acid, and acetic acid in the aerial part of T. coreanum were highest. Contents of total polyphenols, total flavonoids, and DPPH radical scavenging activity of the aerial part were significantly higher in T. coreanum than T. officinale. In conclusion, the aerial part of T. coreanum and T. officinale could be used as antioxidative functional food sources.

Cell Proliferation and Antioxidative Effects of Ultrasonic Coffee Extracts

  • Recently, coffee is the most popular beverage for modern people. A great number of substances are found in coffee beans and have been studied for many years such as aliphatic and aromatic compounds. However, studies on the physiological activity of coffee extracts are insufficient. This study was performed to determine the contents of caffeine and chlorogenic acid in coffee extracts according to the solvent and to investigate the physiological activity of coffee extracts. Coffee extracts were extracted by ultrasonication method with various types of solvents including distilled water, ethanol, and other organic solvents under $50^{\circ}C$ and $80^{\circ}C$. The contents of caffeine and chlorogenic acid in coffee extracts were determined by Liquid Chromatography-Mass Spectrometry (LC-MS). Also, cytotoxic and antioxidative effects of coffee extracts were evaluated with MTT and DPPH assays to analyze the physiological activity. As a result, it was confirmed that caffeine and chlorogenic acid contents were extracted in distilled water with the highest rate. Antioxidative activity was observed below 10-fold dilute of coffee extracts, however cytotoxicity was not observed. In conclusion, distilled water was the best solvent for extracting caffeine and chlorogenic acids from coffee bean with ultrasonication and these coffee extracts are less cytotoxic in human skin cell lines and have antioxidant effect.

Purification and Characterization of Soybean Cotyledonary Spermidine Dehydrogenase

  • Decrease in the amount of cotyledonary spermidine in Glycine max under anaerobic conditions related to an increase in spermidine dehydrogenase. Under the same conditions, no enzymatic activity of diamine oxidase was observed. Exposure of Glycine max both to spermidine and 1,3-diaminopropane under anaerobic conditions resulted in a decrease in spermidine contents. Correlated with the decrease in spermidine contents, there was a drastic increase in spermidine dehydrogenase. The molecular weight of the purified enzyme estimated by Sephacryl S-300 gel column and SDS gel electrophoresis were 130,000 dalton and 65,000 dalton, respectively, indicating that the enzyme is a dimer. The optimal pH for activity was 9.3. The $K_m$ value for spermidine was 0.61 mM. Neither metal ions nor polyamine and derivatives affected enzymatic activity, but the enzyme was inhibited by DTNB, NEM and PCMB, suggesting that a cysteine residue of the enzyme is associated with or involved in enzyme activity. To our knowledge, this is the first report describing properties of the enzyme from plants. Considered together, the data in this paper indicate that both spermidine and 1,3-diaminopropane, novel activators, enhance the spermidine dehydrogenase activity and control the intracellular spermidine contents.

A Study on e-Trade Activity Relationship Commitment of Trade Potal Site 6C's (무역 포털사이트 6C's의 관계결속이 전자무역 활용에 미치는 연구)

  • This paper describes a theoretical model for investigating the relationship commitment of trade portal site 6C's. This research is the first to empirical test the causal relationships in Trade portal site. The relationship commitment for e-Trade portal site are tested here using data from 195 samples. Research methodology used in this study was mainly survey and questionnaire. Structural equation model is used in the hypotheses test. the results support all hypotheses involving hypothesis 1 and 6. The major finding of this paper would be summarized as followed: 1) Perceived relationship commitment of e-Trade site 6C's is positively related to contents, community, communication, customization in e-trade activity level and activity range of trade practical 2) Perceived commerce of electronic trade site is negatively related to the relationship commitment of trade companies. 3) Perceived e-trade site in electronic trade connection is positively related to the re-activity intention. In examining the relationships commitment of the factors, e-trade potal 6C's show in significantly by Contents, Community, Communication, Customization, Re-activity intention, but that show in not significantly by Commerce factor.

A Study on the β-glucan, Ginsenoside Content, 2,2-diphenyl-1-picrylhydrazyl (DPPH) Free Radical Scavenging Activity and Single Dose Toxicity Assessment of Modified Kyungohkgo (경옥고가미방의 베타글루칸, 진세노사이드 함량, 2,2-diphenyl-1-picrylhydrazyl (DPPH) Free Radical 소거 활성 및 단회 투여 독성 연구)

  • Objectives This study was conducted to investigate the beta-glucan, ginsenoside content, antioxidant activity and safety of modified Kyungohkgo added to Sparassis crispa and Hericium erinaceum. Methods The marker compounds contents, antioxidant activity and safety of modified Kyungohkgo were tested. The contents of beta-glucan and ginsenoside Rb1, Rg1, and Rg3 marker compounds were measured, the antioxidant activity was measured using 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, and a safety test was conducted via single dose toxicity assessment. Results Analyzing the contents of marker compounds showed 351.75 mg/g of beta-glucan, 0.0327 mg/g of ginsenoside Rb1 and 0.0802 mg/g of ginsenosai Rg3. In the DPPH free radical scavenging activity, the inhibition concentration 50% of modified Kyungohkgo was 0.2880%. The scavenging activity of modified Kyungohkgo was 5.49% activity at 0.05% concentration, 89.66% activity at 0.5% concentration, 94.68% activity at 1% concentration, and 96.06% activity at 5% concentration. In the single dose toxicity test of modified Kyungohkgo, a dose of 2,000 mg/kg B.W. was set at its highest capacity and observed after oral administration to female and male rats. No toxicological findings were recognized. It was observed that the resulting lethal dose can be set to 2,000 mg/kg B.W. or higher for both females and males. Conclusions The results of the experiment on modified Kyungohkgo showed that the marker compounds contents were beta-glucan and ginsenoside Rb1 and Rg3, that antioxidant activity was observed through the DPPH free radical scavenging activity, and safety was confirmed through the single dose toxicity assessment.

  • In line with policy support and demand, mobile videos about science and technology have growth potential. For activation of domestic mobile video contents about science and technology, this article aims to analysis current global leading contents and draw implications for benchmarking. Therefore, this paper attempted to analyzed types and storytelling strategy of Science and technology channels on YouTube, which has the most influence among the mobile video media. The results are as follows. Fist, the YouTube contents about science technology are categorized into three types, activity-, explain-, review-centered type. Second, activity type channels are classified into 4 sub-types. Third, The core storytelling strategy of activity type channels is a combination of familiarity and unfamiliarity to deal with everyday material through specialized scientific and technological methods. Based on the results of this research, we expect to plan and produce global killer contents.
