Monotonic and Parallelizable Algorithm for Simultaneous Reconstruction of Activity/Attenuation using Emission data in PET

  • In PET(Positron Emission Tomography), it is necessary to use transmission scan data in order to estimate the attenuation map. Recently, there are several empirical studies in which one might be able to estimate attenuation map and activity distribution simultaneously with emissive sinogram alone without transmission scan. However, their algorithms are based on the model in which does not include the background counts term, and so is unrealistic. If the background counts component has been included in the model, their algorithm would introduce non-monotonic reconstruction algorithm which results in vain in practice. in this paper, we develop a monotonic and parallelizable algorithm for simultaneous reconstruction of both characteristics and present the validity through some simulations.

Effect of Gd-based MR contrast agents on CT attenuation of PET/CT for quantitative PET-MRI study

  • We evaluate the influence of MR contrast agent on positron emission tomography (PET) image using phantom, animal and human studies. Phantom consisted of 15 solutions with the mixture of various concentrations of Gd-based MR contrast agent and fixed activity of [$^{18}F$]FDG. Animal study was performed using rabbit and two kinds of MR contrast agents. After injecting contrast agent, CT or MRI scanning was performed at 1, 2, 5, 10, and 20 minutes. PET image was obtained using clinical PET/CT scan, and attenuation correction was performed using the all CT images. The values of HU, PET activity and MRI intensity were obtained from ROIs in each phantom and organ regions. In clinical study, patients (n=20) with breast cancer underwent sequential acquisitions of early [$^{18}F$]FDG PET/CT, MRI and delayed PET/CT. In phantom study, as the concentration increased, the CT attenuation and PET activity also increased. However, there was no relationship between the PET activity and the concentration in the clinical dose range of contrast agent. In animal study, change of PET activity was not significant at all time point of CT scan both MR contrast agents. There was no significant change of HU between early and delayed CT, except for kidney. Early and delayed SUV in tumor and liver showed significant increase and decrease, respectively (P<0.05). Under the condition of most clinical study (< 0.2 mM), MR contrast agent did not influence on PET image quantitation.

Characteristics of Tissue Dose of High Dose Rate Ir-192 Source Substitution for Co-60 Brachytherapy Source (코발트-60 선원 대체용 고선량률 Ir-192 선원의 조직선량특성)

  • To achieve the 2D dose distribution around the designed high dose rate Ir-192 source substitution for Co-60 brachytherapy source, we determined the exposure rate constant and tissue attenuation factors as a large depth as a 20 cm from source center. The exposure rate constant is used for apparent activity in designed source with self-absorption and encapsulation steel wall. The tissue dose delivered from the 4401 segments of 2.5 mm in a diameter and 2.5 mm height of disk-type source layer. In the experiments, the tissue attenuation factors include the tissue attenuation and multiple scattering in a medium surrounding the source. The fitted the polynomial regression with 4th order for the tissue attenuation factors are very closed to the experimental measurement data within ${\pm}$1% discrepancy. The Meisberger's constant showed the large uncertainty in large distance from source. The exposure rate constant 4.69 Rcm$^2$/mCi-hr was currently used for determination of apparent activity of source and air kerma strength was obtained 0.973 for tissue absorbed dose from the energy spectrum of Ir-192 source. In our experiments with designed high dose rate brachytherapy source, the apparent activity of Ir-192 source was delivered from the 54.6 % of actual physical source activity through the self-absorption and encapsulation wall attenuations. This paper provides the 2-dimensional dose tabulation from unit apparent activity in a water medium for dose planning includes the multiple scattering, source anisotropy effect and geometric factors.

Improved Activity Estimation using Combined Scatter and Attenuation Correction in SPECT (단일광자방출단층촬영 영상에서 산란 및 감쇠 보정에 위한 절대방사능 측정)

  • Purpose: The purpose of this study was to evaluate the accuracy of radioactivity quantitation in Tc-99m SPECT by using combined scatter and attenuation correction. Materials and Methods: A cylindrical phantom which simulates tumors (T) and normal tissue (B) was filled with varying activity ratios of Tc-99m. We acquired emission scans of the phantom using a three-headed SPECT system (Trionix, Inc.) with two energy windows (photopeak window: $126{\sim}154keV$ and scatter window: $101{\sim}123keV$). We performed the scatter correction with dual-energy window subtraction method (k=0.4) and Chang attenuation correction. Three sets of SPECT images were reconstructed using combined scatter and attenuation correction (SC+AC), attenuation correction (AC) and without any correction (NONE). We compared T/B ratio, image contrast [(T-B)/(T+B)] and absolute radioactivity with true values. Results: SC+AC images had the highest mean values of T/B ratios. Image contrast was 0.92 in SC+AC, which was close to the true value of 1, and higher than AC (0.77) or NONE (0.80). Errors of true activity by SPECT images ranged from 1 to 11% for SC+AC, $22{\sim}47%$ for AC, and $2{\sim}16%$ for NONE in a phantom which was located 2.4cm from the phantom surface. In a phantom located 10.0cm from the surface, SC+AC underestimated by 24%, NONE 40%. However, AC overestimated by 10%. Conclusion: We conclude that accurate SPECT activity quantitation of Tc-99m distribution can be achieved by dual window scatter correction combind with attenuation correction.

Bioremediation of Diesel-Contaminated Soils by Natural Attenuation, Biostimulation and Bioaugmentation Employing Rhodococcus sp. EH831 (Natural attenuation, biostimulation 및 Rhodococcus sp. EH831을 이용한 bioaugmentation에 의한 디젤 오염 토양의 정화)

  • Three bioremediation methods, natural attenuation (NA), biostimulation (BS) and bioaugmentation (BA) were applied to remediate diesel-contaminated soil, with their remediation efficiencies and soil microbial activities compared both with and without surfactant (Tween 80). BA treatment employing Rhodococcus sp. EH831 was the most effective for the remediation of diesel-contaminated soil at initial remediation stage. On the addition of surfactant, no significant effect on the remediation performance was observed. A negative correlation was found between the dehydrogenase activity (DHA) and residual concentration of total petroleum hydrocarbons (TPHs) at below 20,000 mg-$TPHs{\cdot}kg$-dry $soil^{-1}$, as follows: DHA (${\mu}g$-TPF(Triphenylformazan)${\cdot}g$-dry $soil^{-1}\;d^{-1}$) = -0.02 ${\times}$ TPHs concentration (mg-$TPHs{\cdot}kg$-dry $soil^{-1}$) + 425.76 (2500 ${\leq}$ TPHs concentration ${\leq}$ 20000, p < 0.01).

  • Diffusive transport of volatile organic compounds(VOCs) and their degradation by bacteria in unsaturated soils are couple by poorly understood mass transfer kinetics at the gas/water interface. Determination of the fate of VOCs in unsaturated soil is necessary to evaluate the feasibility of natural attenuation as a VOC remediation strategy. The objective of this study was to develop a mechanistically based mathematical model that would consider the interdependence of VOC transport, microbial activity, and sorptive interaction in a moist, unsaturated soil. Because the focus of the model was on description of natural attenuation, the advective VOC transport that is induced in engineered remediation processes such as vapor extraction was not considered. The utility of the model was assessed through its ability to describe experimental observations form diffusion experiments using toluene as a representative VOC in well-defined soil columns that contained a toluene degrading bacterium, Pseudomonas Putida, as the sole active microbial species. The coefficient for gas-liquid mass-transfer, K$\sub$LA/, was found to be a key parameter controlling the ability of bacteria to degrade VOCs. This finding indicates that soil size and geometry are likely to be important parameters in assessing the possible success of natural attenuation of VOCs in contaminated unsaturated soils.

Effects of Ginsenoside Total Saponins on Experimental Irritable Bowel Syndrome in Rats

  • In the previous study, we reported that the in viかo inhibitory effect of ginsenosides, active ingredient of Panax ginseng, on $5-HT_{3A}$ receptor channel activity is coupled to in vivo anti-vomiting and anti-nausea effect. In the present study, we further investigated that the inhibitory effect of ginsenosides, active ingredient of Panax ginseng, on 5-HT3A receptor channel activity is also coupled to attenuation of irritable bowel syndrome (IBS), which is induced by colorectal distention (CRD) and $0.6\%$ acetic acid treatment. The CRD-induced visceral pains induced by CRD and acetic acid treatment are measured by frequency of contractions of the external oblique muscle in conscious rats. Treatment of GTS significantly inhibited CRD-induced visceral pain with dose-dependent manner. The $EC_{50}$ was $5.5{\pm}4.7$ mg/kg ($95\%$ confidence intervals: 1.2-15.7) and the antinociceptive effect of GTS on visceral pain was persistent for 4 h. We also compared the effects of protopanaxadiol (PD) ginsenosides and protopanaxatriol (PT) ginsenosides with saline on acetic acid-and CRD-induced visceral pain, and found that protopanaxatriol (PT) ginsenosides was much more potent than PD ginsenosides in attenuating CRD-induced visceral pain. These results indicate that U ginsenosides of Panax ginseng are components far attenuation of experimentally CRD-induced visceral pains.

Determination of Attenuation Collection Methods According to the Type of Radioactive Waste Drums (방사성폐기물드럼 종류별 감쇠보정방법의 결정)

  • The measured radioactivity of gamma-emitting radionuclides in each radioactive waste drum using the non-destructive waste assay method is underestimated than real radioactivity in radioactive waste drum because the gamma-rays are attenuated within the medium. Therefore, the measured radioactivity should be corrected for the attenuation of gamma-rays. For the correction of the attenuation of gamma-rays, the attenuation correction method should be applied differently by considering the distribution and density of medium in radioactive wastes drum generated from nuclear power plants. In this study, the model drums were fabricated for simulating five types of radioactive waste drums generated from nuclear power plant and the optimum methods of the attenuation correction were experimentally determined to analyze the activity of radionuclides in the waste drum accurately using the segmented gamma scanning system. With the determination of the attenuation correction methods from the experimental results the transmission method and the average density method for the miscellaneous waste drum, the transmission method and the differential peak absorption method for the shielded miscellaneous waste drum were used to measure the density of medium in waste drums. Also, the average density method and the differential peak absorption method for the spent resin drum, the paraffin solidified drum, and the spent filter drum were used.

Protective Effect of Whagan-Jeon (huaganjian) on Acetaminophen-induced Hepatotoxicity (화간전이 아세트아미노펜에 의한 간독성에 미치는 영향)

  • Objective : This study was performed to investigate the activity of Whagan-Jeon (huaganjian) in protection against acetaminophen (AAP)-induced hepatotoxicity and the possible mechanisms in vivo. Methods : The following were performed : Serum ALT, depletion of hepatic glutathione (GSH) levels, the microsomal p. nitrophenol hydroxylation activity, microsomal aniline hydroxylation activity, genomic DNA fragmentation and its reversal, hepatic glutathione-S-transferase (GST) activity, and hepatic NAD(P)H:quinone oxidoreductase (QR) activity Results : Whagan-Jeon (huaganjian) protected against AAP-inducedhepatotoxicity by the increase of GSH levels, inhibition of P450 2E1-specific metabolic activities, attenuation of hepatic DNA damage, and induction of GST and QR activities in vivo. Conclusions : In conclusion, Whagan-Jeon (huaganjian) was effective in protection against AAP-induced hepatoxicity.

Physiologic and epigenetic effects of nutrients on disease pathways

  • BACKGROUND/OBJECTIVES: Epigenetic regulation by nutrients can influence the development of specific diseases. This study sought to examine the effect of individual nutrients and nutrient families in the context of preventing chronic metabolic diseases via epigenetic regulation. The inhibition of lipid accumulation and inflammation by nutrients including proteins, lipids, vitamins, and minerals were observed, and histone acetylation by histone acetyltransferase (HAT) was measured. Correlative analyses were also performed. MATERIALS/METHODS: Nutrients were selected according to information from the Korean Ministry of Food and Drug Safety. Selected nutrient functionalities, including the attenuation of fatty acid-induced lipid accumulation and lipopolysaccharide-mediated acute inflammation were evaluated in mouse macrophage Raw264.7 and mouse hepatocyte AML-12 cells. Effects of the selected nutrients on in vitro HAT inhibition were also evaluated. RESULTS: Nitric oxide (NO) production correlated with HAT activity, which was regulated by the amino acids group, suggesting that amino acids potentially contribute to the attenuation of NO production via the inhibition of HAT activity. Unsaturated fatty acids tended to attenuate inflammation by inhibiting NO production, which may be attributable to the inhibition of in vitro HAT activity. In contrast to water-soluble vitamins, the lipid-soluble vitamins significantly decreased NO production. Water- and lipid-soluble vitamins both exhibited significant inhibitory activities against HAT. In addition, calcium and manganese significantly inhibited lipid accumulation, NO production, and HAT activity. CONCLUSIONS: Several candidate nutrients and their family members may have roles in the prevention of diseases, including hepatic steatosis and inflammation-related diseases (i.e., nonalcoholic steatohepatitis) via epigenetic regulation. Further studies are warranted to determine which specific amino acids, unsaturated fatty acids and lipid-soluble vitamins or specific minerals influence the development of steatosis and inflammatory-related diseases.
