  • The activities of s-amylase, ${\alpha}-amylase$ and protease of three combination products containing digestive enzymes, antacids and herbal drugs on the Korean market were estimated. The effects of antacids and herbal drugs on the activities of digestive enzymes were investigated. Starch-saccarifying activity of s-amylase, starch-dextrinizing activity of ${\alpha}-amylase$ and protein-peptic activity of protease were estimated by Somogy, Mc'Credy, and Casein-Folin method, respectivley. The optimal pH of s-amylase, ${\alpha}-amylase$ and protease were pH 5.0, 4.8 and 7.0, rcspectively. The digestive activities at optimal pH continued about eight hours. The digestive activities of individual enzymes were reduced to 40-90% by antacids and were affected somewhat positively or negatively by herbal drugs. Enzyme activities of the combination products were also affected by pH and reaction time.

판크레아틴의 규격 표준화 연구 (Standardization of Pancreatin)

  • Pancreatin is a enzyme mixture breaking down carbohydrates, proteins and lipids. Most pancreatin used in Korea is imported from foreign countries. However, guideline of each country for pancreatin produced from each country is different. Therefore, guideline for pancreatin imported from several countries, such as Europe, Japan and America, it is standardized to control its quality. Assay of enzyme activity for pancreatin in KP is similar to tat in JP, but it is significantly different from those in FP ad in USP. We measured pancreatin digestive activities of 17 commercial products. Activity assay of digestive enzymes, starch- and lipid-digestive enzymes, for pancreatin by KP method (including JP) was difficult compared to those by FIP ad USP methods. Particularly, activity assays of starch- and lipid-digestive enzymes by KP method were mistakable, ad varied in diluted samples than those by FIP. However, activity assay of protein-digestive enzyme by KP method was similar to that by FIP. Starch-digestive enzyme activities of 17 commercial pancreatins by KP method were lower 0.079-fold compared to those by FIP method. Their protein-digestive enzyme activities by KP method were higher 75.7-fold than those by FIP method. Their lipid-digestive enzyme activities by KP method were lower 0.234-fold compared to those by FIP method.

Effects of Enzyme Supplementation on Growth, Intestinal Content Viscosity, and Digestive Enzyme Activities in Growing Pigs Fed Rough Rice-based Diet

  • The purpose of the present study was to investigate the effects of exogenous non-starch polysaccharides (NSP) enzymes on performance, intestinal content viscosity and digestive enzyme activities of growing pigs fed a rough rice-based diet. A total of 60 crossbred barrows with an initial body weight of 35.16 kg (SD = 0.82) were blocked by body weight and randomly assigned to two treatments with three replications. Each group was fed the diet based on rice with or without exogenous NSP enzymes (2 g/kg of diet). During the 70 days of the feeding trial, all pigs were given free access to feed and water. At the end of the feeding trial, six pigs from each treatment were randomly selected and slaughtered to collect intestinal digesta, intestinal mucosa, and pancreas. The addition of NSP enzymes improved average daily gain (p<0.05) and feed:gain (p<0.05), and decreased viscosity of digesta in the jejunum (p<0.001) and ileum (p<0.01) of pigs. The supplementation of NSP enzymes increased activities of protease (p<0.01), trypsin (p<0.01) and ${\alpha}$-amylase (p<0.05) in duodenal contents. However, digestive enzymes in the pancreas, jejunal and ileal mucosa were unaffected by the supplemental NSP enzymes (p>0.10). The results indicate that the addition of NSP enzymes to rough rice-based diets improved performance of pigs, reduced viscosity and increased digestive activity in the small intestine.

Effects of Flower Thinning Formulation on Activities of Digestive Enzymes and Acetylcholine Esterase in Honey bee Apis mellifera

  • The effects of a newly developed flower thinning formulation (FTF) on the vitality of the honey bee Apis mellifera were examined by measuring the activities of various digestive enzymes in adult worker bees. First, direct spraying of the FTF solution did not cause any behavioral changes or lethal effects for the honey bees based on 24 h observation. Second, oral ingestion of a sugar solution containing the FTF did not produce any significant change in the activities of amylase, proteinases, lipase, or acetylcholine esterase (AChE) in the worker bees 6 h or 24 h after treatment. Meanwhile, a commercial formulation containing sulfur compounds showed slightly reduced activities for several digestive enzymes and AChE, although no behavioral disturbance. Thus, the results of the present study suggest that the FTF is not toxic for honey bees, in terms of contact and ingestion. Therefore, this newly developed FTF can be used for flower thinning without any detrimental effects on pollinating insects.

Changes in Antioxidant Enzyme Activities in the Gill And Digestive Glands of the Manila Clam Ruditapes philippinarum exposed to Cu

  • We investigated the effect of Cu exposure on the activities of protective antioxidant enzymes in the gills and digestive glands of the manila clam Ruditapes phillippinarum exposed to subchronic concentrations (0, 20, 40, and $80{\mu}gL^{-1}$) of waterborne Cu. No mortality occurred during the experimental period, and no significant condition index differences were observed in any exposure group compared with the control. No significant differences were observed in the digestive glands and gills of the clams observed during 15 days of exposure, but after 30 days, the SOD activity in the gill showed a significant difference between the $80{\mu}gL^{-1}$ Cu-exposed group and the control. GPx activities in the digestive glands and gills were significantly lower after 30 days of Cu exposure. Gill GR activity in the high-exposure group ($80{\mu}gL^{-1}$) was significantly elevated compared with that in the control group. GST activities in the digestive glands of all groups did not change over 30 days. However, GST activity in the gill at $80{\mu}gL^{-1}$ Cu was significantly higher after 15 and 30 days of exposure. GSH activities in the gill showed patterns similar to those of GST activities during exposure periods. In the digestive glands, GSH activity was higher only at $80{\mu}gL^{-1}$ after 30 days exposure. In digestive glands and gills, the MDA levels of clams exposed to $80{\mu}gL^{-1}$ Cu were significantly higher after 30 days of exposure.

이매패류 3종 소화맹낭의 소화효소 구성 및 활성도 (Digestive Enzymatic Compositions and Activities of the Digestive Diverticula in Three Species of Bivalves)

  • 본 연구는 3종의 이매패류를 대상으로 소화맹낭의 소화효소 구성 및 활성도에 대해 조사하였다. 본 연구에 사용된 이매패류는 지중해담치, 개조개 및 꼬막이며, 이들은 한국 남해안에서 2010년 11월에 채집하였다. 지중해담치, 개조개, 꼬막의 소화맹낭을 구성하는 소화효소는 amylase와 cellulase가 약 95%로 대부분을 차지하였다. 소화맹낭에서 amylase와 cellulase 활성도는 지중해담치는 2.6과 0.8 U/mg, 개조개는 2.4와 8.8 U/mg, 꼬막은 7.3과 11.8 U/mg였다. 그리고 소화맹낭의 소화효소 가운데 protease의 활성도는 지중해담치, 개조개, 꼬막에서 각각 0.00019, 0.00028, 0.00022 U/mg로 가장 낮았다.

Effects of Different Levels of Supplementary Alpha-amylase on Digestive Enzyme Activities and Pancreatic Amylase mRNA Expression of Young Broilers

  • Four hundred and forty 1-day-old Arbor Acre broilers were fed commercial starter diets with 0, 250, 750 and 2,250 mg/kg of an alpha-amylase preparation from 1 to 21 days of age to investigate the effects of an exogenous enzyme on growth performance, activities of digestive enzymes in the pancreas and anterior intestinal contents and pancreatic amylase mRNA expression. Body weight gain (BWG) and average daily gain (ADG) increased linearly (p<0.01) with increasing levels of supplementary amylase but feed conversion ratio (FCR) was not affected. There was a negative quadratic change of protease and amylase in the small intestinal contents with the increase of supplementary amylase level. The activity of intestinal trypsin was also increased (p<0.05). Lipase was unaffected by amylase supplementation (p>0.05). The pancreatic protease, trypsin, and lipase were not affected by exogenous amylase levels. Consistent with the tendency for a linear depression of amylase activity, pancreatic ${\alpha}$-amylase mRNA was down-regulated by dietary amylase supplementation. The present study suggested that oral administration of exogenous amylase affected activities of intestinal enzymes and the production of pancreatic digestive enzymes in a dose-dependent manner.

Rotifer Brachionus rotundiformis의 개체군 성장과 소화효소 활성의 관계 (Relationship between Population Growth and Digestive Enzyme Activity of Rotifer Brachionus rotundiformis)

  • 본 연구의 목적은 rotifer Brachionus rotundiformis의 영양강화와 자어의 소화기능을 높이기 위해 소화효소 활성이 높은 rotifer의 선택방법을 제시하는 것이다. 소화효소 활성을 측정하는 개체군은 성장과정에 있는 개체군들 중 임의로 표본 수집하여 소화효소활성을 측정하여 결정하였다. 선택된 개체군에서 개체밀도와 성장률의 관계가 RD=5865 SGR-350.08(P<0.001)로 나타났다. 포란률은 성장률과 개체밀도와의 관계가 각각 F=-36.147 SGR+61.652(P<0.05)와 F=-0.0085 RD+66.38(P<0.001)로 나타났다. 성장률과 소화효소 활성의 rotifer 개체당 활성은 Amyl=-1.6482 SGR+3.2498(P<0.05), TAP=-0.8115 SGR+1.1361(P<0.001) 및 TGL+0.0055 SGR+0.0079(P=0.239)로 나타나 TG-lipase 활성에서는 성장률과 관계없는 것으로 나타났다. 또한 포란률과 rotifer 개체당 소화효소 활성의 관계는 Amyl=0.0296 F+1.0981(P<0.001). TAP=0.0252 F+0.0975(P<0.001) 및 TGL=-6E-06 F+0.0113(P=0.915)으로 나타나 TG-lipase 활성이 포란률과 관계없는 것으로 나타났다. 반면 TG-lipase의 단백질 비활성과 포란률, 성장률 및 개체밀도와의 관계는 각각 TGL=-0.0024 F0.2332(P=0.132), TGL=0.1267 SGR+0.005(P<0.01) 및 TGL=0.0002 F-0.0594(P<0.001)로 나타나 TG-lipase 단백질 비활성은 포란률과의 관계를 제외하고 각각 상관된 변화를 보였다. 따라서 rotifer의 영양강화효과와 자어에게 외부기인 소화효소 전달을 위한 높은 소화효소 활성을 보이는 rotifer 개체군이 개체밀도가 높은 개체군보다는 포란률이 높은 경우에 나타났기 때문에 포란률이 높은 개체군을 선택하여 영양강화하고 자어에 공급하는 것이 보다 자어에 유익할 것으로 판단된다.

상기생(Loranthus parasiticus) 추출물의 탄수화물 소화 효소 및 식후 고혈당 저해 효과 (Inhibitory Effects of Loranthus Parasiticus Extract on Carbohydrate Digestive Enzymes and Postprandial Hyperglycemia)

  • 상기생(Loranthus parasiticus)은 뽕나무에 기생하는 겨우살이로 전세계에 널리 분포되어 있으며, 수 세기 동안 전통 의학의 성분으로 사용되어 왔고, 최근까지 항암, 항산화, 항비만, 항염증 효과 등이 연구되었으나, 탄수화물 효소나 식후 혈당 수치에 미치는 영향에 관한 연구는 부족한 실정이다. 본 연구는 Loranthus parasiticus 추출물 (LPE)이 탄수화물 분해 효소(α-glucosidase, α-amylase) 활성 억제와 streptozotocin (STZ)으로 유도된 당뇨병 마우스에서 식후 고혈당 완화 효과를 조사하였다. 그 결과 LPE는 농도에 비례하여 α-glucosidase와 α-amylase 활성을 억제 하였고, 각각의 IC50 값으로 0.121±0.007 및 0.157±0.004 mg/ml을 나타내어 양성 대조군인 acabose 보다 유의하게 강한 억제 효과가 있음을 보여주었다(p<0.05). 또한 STZ으로 유도된 당뇨병 마우스에서는 대조군의 높은 혈당과 달리 LPE 첨가군에서는 혈당이 유의하게 감소하였다(p<0.05). 따라서, 이들 결과는 LPE가 α-glucosidase와 α-amylase 억제 효과로 식후 고혈당을 감소시킬 수 있는 천연 항고혈당 식품으로 사용 가능성을 시사한다.

Development of Gastric and Pancreatic Enzyme Activities and Their Relationship with Some Gut Regulatory Peptides in Grazing Sheep

  • Forty-four Gansu Alpine Fine-wool lambs were used to study changes in the activities of three gastric and five pancreatic enzymes under grazing conditions between 0 and 56 days of age. The lambs were slaughtered on days 0, 3, 7, 14, 21, 28, 42 and 56, the abomasal contents, mucosa and pancreas were immediately removed and placed into liquid nitrogen and enzyme activities were determined. Gastric enzyme (chymosin, pepsin and pregastrc esterase) activities were relatively high at birth, especially chymosin, but decreased quickly between day 0 and 21. The activity of pepsin changed insignificantly with increasing age. There was no significant change in the pancreatic enzyme activities (trypsin, chymotrypsin, ${\alpha}$-amylase, lipase and lactase). The activity of trypsin was relatively higher than that of the other pancreatic enzymes, and lactase activity was low. These ontogenic patterns might be under the control of many gut regulatory peptides, the plasma concentrations of which changed simultaneously. Some gastric and pancreatic enzymes were correlated with plasma concentrations of these gut regulatory peptides.
